Liver regeneration following injury aids the restoration of liver mass and the recovery of liver function.In the present study we investigated the contribution of megakaryocytic leukemia 1(MKL1),a transcriptional modu...Liver regeneration following injury aids the restoration of liver mass and the recovery of liver function.In the present study we investigated the contribution of megakaryocytic leukemia 1(MKL1),a transcriptional modulator,to liver regeneration.We report that both MKL1 expression and its nuclear translocation correlated with hepatocyte proliferation in cell and animal models of liver regeneration and in liver failure patients.Mice with MKL1 deletion exhibited defective regenerative response in the liver.Transcriptomic analysis revealed that MKL1 interacted with E2F1 to program pro-regenerative transcription.MAPKAPK2 mediated phosphorylation primed MKL1 for its interaction with E2F1.Of interest,phospholipase d2 promoted MKL1 nuclear accumulation and liver regeneration by catalyzing production of phosphatidic acid(PA).PA administration stimulated hepatocyte proliferation and enhanced survival in a MKL1-dependent manner in a pre-clinical model of liver failure.Finally,PA levels was detected to be positively correlated with expression of pro-regenerative genes and inversely correlated with liver injury in liver failure patients.In conclusion,our data reveal a novel mechanism whereby MKL1 contributes to liver regeneration.Screening for small-molecule compounds boosting MKL1 activity may be considered as a reasonable approach to treat acute liver failure.展开更多
Protolignin extraction can facilitate structure elucidation and valorization of lignin in biorefinery,but is rather challenging due to the complex chemical bonds present.Here,we developed the in situ generated NH3-rel...Protolignin extraction can facilitate structure elucidation and valorization of lignin in biorefinery,but is rather challenging due to the complex chemical bonds present.Here,we developed the in situ generated NH3-reline(IGNR)system to realize one-pot protolignin extraction from lignocellulose.In the IGNR system,reline consisting of choline chloride and urea acted as both a solvent and a nucleophile generator,and the nucleophilic addition-elimination mechanism was verified by model compound studies.The in situ generated NH3 could precisely cleave the lignin-carbohydrate esters in lignocellulose with a near-quantitative retention of carbohydrates.The extracted IGNR-Protolignin exhibited native lignin substructure with high molecular weight and highβ-O-4′content(41.5 per 100 aromatic units).In addition,the up-scaled kilogram reaction demonstrated the feasibility of the IGNR system for potential industrial application in a green and sustainable pathway.This work represents a breakthrough toward protolignin extraction in practice with the future goal of achieving total biorefinery.展开更多
Currently,renewable energy generation has received more and more attention.This article focuses on wind energy generation,one of the renewable energy sources.Aiming at the intermittent and unpredictable wind power pro...Currently,renewable energy generation has received more and more attention.This article focuses on wind energy generation,one of the renewable energy sources.Aiming at the intermittent and unpredictable wind power problems,according to the day ahead bidding mechanism in the power market,this paper introduces the energy storage system to maximize wind power merchants profit based on the newsvendor model.First,this paper focuses on the wind farms combined with storage system to put forward the optimal bidding decision of selling or buying electricity to the market one day in advance and the optimal bidding amount.Then,we analyze the relationship between the optimal bid amount and the market penalty price coefficient,government subsidy,storage system charge and discharge efficiency,and other parameters.Based on the analysis of various parameters,power generation companies can make reasonable bidding decisions and amount to maximize profits.展开更多
