心肌肥大(cardiac hypertrophy)是由外周组织对血流动力学需求增加而发生的一种代偿性反应。在心肌肥大过程中,不同时期的不同类型的基因表达受到生理和病理信号的多级转录调控。组蛋白乙酰化作为最广泛的翻译后修饰方式,受相互拮抗的... 心肌肥大(cardiac hypertrophy)是由外周组织对血流动力学需求增加而发生的一种代偿性反应。在心肌肥大过程中,不同时期的不同类型的基因表达受到生理和病理信号的多级转录调控。组蛋白乙酰化作为最广泛的翻译后修饰方式,受相互拮抗的组蛋白乙酰化酶(histone acetyltransferases,HAT)和组蛋白去乙酰化酶(histone deacetylases,HDACs)的精细控制。近年来研究表明,HDACs作为一类抑制转录过程并含有高度保守的脱乙酰酶结构域家族酶,通过多种作用途径调控心肌肥大过程中的基因表达。本文主要综述了组蛋白去乙酰化酶调节心肌肥大过程的相关研究进展,通过阐明不同种类HDACs在心肌肥大中的作用和分子机制,为不同类型心肌肥大和心衰的发病治疗提供新的思路,为新药设计提供分子靶点。 展开更多

钾离子通道在心肌细胞动作电位复极过程中起着重要作用。钾离子通道蛋白种类繁多,已知钾离子通道蛋白KCNQ和HERG/eag参与心脏动作电位的形成,调节心脏收缩节律。钾离子通道蛋白Shaker是果蝇(Drosophila)体内发现的第一个电压门控钾离子... 钾离子通道在心肌细胞动作电位复极过程中起着重要作用。钾离子通道蛋白种类繁多,已知钾离子通道蛋白KCNQ和HERG/eag参与心脏动作电位的形成,调节心脏收缩节律。钾离子通道蛋白Shaker是果蝇(Drosophila)体内发现的第一个电压门控钾离子通道,维持神经元和肌肉细胞的电兴奋性,但是目前其在成人心脏功能中的作用仍不清楚。本研究以果蝇为模型,高频电刺激模拟心脏应激状态,观察钾离子通道蛋白shaker基因突变体的心衰发生率。同时,利用心脏特异性启动子hand4.2Gal4特异性敲低钾离子通道蛋白Shaker的表达;果蝇成体心脏生理学功能分析系统分析了1、3、5周龄特异性敲低钾离子通道蛋白Shaker的心脏表型。结果表明,shaker基因突变将严重影响果蝇心脏抗应激能力,表现在高频电刺激后的心力衰竭发生率显著性升高;心脏特异性敲低shaker基因导致5周龄果蝇心律失常发生率显著性增加;心脏特异性敲低HDAC3将显著降低果蝇寿命。综上所述,本研究推测钾离子通道蛋白Shaker在衰老过程中维护果蝇正常的心脏功能。 展开更多

以西藏年楚河流域的典型危险性冰湖——黄湖及其下游区域为研究对象,采用二维溃坝洪水演进数值模型,进行了模型的率定和不同工况下的洪水演进模拟。设置了瞬时全部溃决和逐渐溃决2种工况,模拟结果表明:洪水发生后,预计最大淹没面积约为8... 以西藏年楚河流域的典型危险性冰湖——黄湖及其下游区域为研究对象,采用二维溃坝洪水演进数值模型,进行了模型的率定和不同工况下的洪水演进模拟。设置了瞬时全部溃决和逐渐溃决2种工况,模拟结果表明:洪水发生后,预计最大淹没面积约为89.25 km2,溃决洪水到达江孜的时间约为33.6 h,江孜处的洪峰流量约为1777 m3/s,大于江孜地区暴雨洪水万年一遇洪峰流量1000 m3/s。根据预测结果做好预警工作,能一定程度上减小灾害损失。本研究的研究结果对于完善江孜地区的潜在危险冰湖数据库和科学制定洪水预警策略具有重要的现实意义。 展开更多

A metal-insulator-metal(MIM)-based arc-shaped resonator coupled with a rectangular stub(MARS) structure is proposed. This structure can generate two tunable Fano resonances originating from two different mechanisms. The structure has the advantage of being sensitive to the refractive index, and this feature makes it favorable for application in various microsensors. The relationship between the structural parameters and Fano resonance is researched using the finite element method(FEM) based on the software COMSOL Multiphysics 5.4. The simulation reveals that the sensitivity reaches1900 nm/refractive index unit(RIU), and the figure of merit(FOM) is 23.75. 展开更多

Magnetoreception is essential for magnetic orientation in animal migration. The molecular basis for magnetoreception has recently been elucidated in fruitfly as complexes between the magnetic receptor magnetoreceptor(Mag R) and its ligand cryptochrome(Cry). Mag R and Cry are present in the animal kingdom. However, it is unknown whether they perform a conserved role in diverse animals. Here we report the identification and expression of zebrafish Mag R and Cry homologs towards understanding their roles in lower vertebrates. A single magr gene and 7 cry genes are present in the zebrafish genome. Zebrafish has four cry1 genes(cry1aa, cry1 ab, cry1 ba and cry1bb) homologous to human CRY1 and a single ortholog of human CRY2 as well as 2 cry-like genes(cry4 and cry5). By RT-PCR, magr exhibited a high level of ubiquitous RNA expression in embryos and adult organs, whereas cry genes displayed differential embryonic and adult expression. Importantly, magr depletion did not produce apparent abnormalities in organogenesis. Taken together, magr and cry2 exist as a single copy gene, whereas cry1 exists as multiple gene duplicates in zebrafish. Our result suggests that magr may play a dispensable role in organogenesis and predicts a possibility to generate magr mutants for analyzing its role in zebrafish. 展开更多

HNULP1,a new member of the basic helixloop-helix transcription factors,contains a DUF654 domain in its C-terminus and is highly conserved from Drosophilae,yeast,zebrafish to mouse.The function of this motif,however,is currently unknown.In this research,we fused five deletion fragments of the DUF654 domain to the GAL4 DNA-binding domain and then co-transfected with plasmids L8G5-Luc and VP-16.The analysis of the GAL4 luciferase reporter gene indicated that fragments from 228 to 407 amino acids in the DUF654 domain had a strong transcription repression activity.Therefore,this study lays a solid foundation for research on the mechanism of hnulp1 transcriptional regulation and the function of the DUF654 domain. 展开更多
