·AIM: To study the role of immature dendritic cells (imDCs) on immune tolerance in rat penetrating keratoplasty (PKP) in high -risk eyes and to investigate the mechanism of immune hyporesponsiveness induced by do...·AIM: To study the role of immature dendritic cells (imDCs) on immune tolerance in rat penetrating keratoplasty (PKP) in high -risk eyes and to investigate the mechanism of immune hyporesponsiveness induced by donor-derived imDCs. ·METHODS: Seventy-five SD rats (recipient) and 39 Wistar rats (donor) were randomly divided into 3 groups: control, imDC and mature dendritic cell (mDC) group respectively. Using a model of orthotopic corneal transplantation in which allografts were placed in neovascularized high -risk eyes of recipient rat. Corneal neovascularization was induced by alkaline burn in the central cornea of recipient rat. Recipients in imDC group or mDC group were injected donor bone marrow-derived imDCs or mDCs of 1 ×10 6 respectively 1 week before corneal transplantation tail vein. Control rat received the same volume of PBS. In each group, 16 recipients were kept for determination of survival time and other 9 recipients were executed on day 3, 7 and 14 after transplantation. Cornea was harvested for hematoxylin eosin staining and acute rejection evaluation, Western blot was used to detect the expression level of Foxp3. ·RESULTS: The mean survival time of imDC group was significantly longer than that of control and mDC groups (all 【0.05). The expression level of Foxp3 on CD4 + CD25 + T cells of imDC group (2.24 ±0.18) was significantly higher than that in the control (1.68 ±0.09) and mDC groups (1.46±0.13) (all 【0.05).·CONCLUSION: Donor -derived imDC is an effective treatment in inducing immune hyporesponsiveness in rat PKP. The mechanism of immune tolerance induced by imDC might be inhibit T lymphocytes responsiveness by regulatory T cells. ·展开更多

In this study, surveill...As a novel experimental animal model, tree shrews have received increasing attention in recent years. Despite this, little is known in regards to the time phases of their embryonic development. In this study, surveillance systems were used to record the behavior and timing of copulations; embryos at different post-copulation stages were collected and cultured in vitro; and the developmental characteristics of both early-stage and in vitro cultured embryos were determined. A total of 163 females were collected following effective copulation, and 150 were used in either unilateral or bilateral oviduct embryo collections, with 307 embryos from 111 females obtained (conception rate=74%). Among them, 237 embryos were collected from 78 females, bilaterally, i.e., the average embryo number per female was 3.04; 172 fertilized eggs collected from 55 females, bilaterally, were cultured for 24-108 h in vitro for developmental observations; finally, 65 embryos from 23 bilateral cases and 70 embryos from 33 unilateral cases were used in embryo transplantation.展开更多
AIM:To explore the protective effects of amino-guanidine(AG) on retinal apoptosis in mice with oxygeninduced retinopathy(OIR).·METHODS:A total of 80 C57BL/6J mice,aged 7 days,were randomly divided into four group...AIM:To explore the protective effects of amino-guanidine(AG) on retinal apoptosis in mice with oxygeninduced retinopathy(OIR).·METHODS:A total of 80 C57BL/6J mice,aged 7 days,were randomly divided into four groups:normal,high oxygen,high oxygen saline and high oxygen treated with AG.In the normal group,mice were housed in normoxic conditions from postnatal day P7 to P17.Mice in the other 3 groups were placed under hyperoxic conditions(75 ±2% O2) in an oxygen-regulated chamber for 5 days and subsequently placed in normoxic conditions for 5days.Mice in the AG group were treated once daily,from P12 to P17,with AG hemisulfate(100mg/kg body weight,intraperitoneally) dissolved in physiological saline.An equivalent amount of 0.9% physiological saline was administered,as above,to mice in the high oxygen saline group.Ten mice were randomly selected from each group on P14 and on P17,euthanized and the retinas examined.Apoptotic cells in the retina were detected using the terminal-deoxynucleoitidyl transferase mediated nick end labeling(TUNEL) method.The expression of nitric oxide synthase(iNOS) in the retina was detected by immunohistochemistry and changes in rod cells were observed using electron microscopy.·RESULTS:TUNEL-positive cells and iNOS immunoreactive neurons were present in the inner nuclear and ganglion cell retinal layers of mice in the high oxygen group.The number of TUNEL-positive cells was significantly greater in the high oxygen group compared with the normal group(t =-20.81,P14d【0.05;t =-15.05,P17d【0.05).However,the number of TUNEL-positive cells in the AG treatment group was significantly lower(t =-13.21,P14d【0.05;t =-6.61,P17d【0.05) compared with thehigh oxygen group.The expression of iNOS was significantly higher in the high oxygen group compared with the normal group(t =-21.95,P14d【0.05;t =-17.30,P17d【0.05).However,the expression of iNOS in the AG treatment group was significantly lower(t =-12.17,P14d【0.05;t =-10.30,P17d【0.05) compared with the high oxygen group.The outer segments of the rods were disorganized and short in the high oxygen group.Rod morphology appeared to be slightly improved in the AG group.·CONCLUSION:AG may protect retinal neurons in OIR by inhibiting apoptosis.The mechanism may be related to iNOS.展开更多

All eyes were evaluated with transmission electron microscopy(TEM) at 1, 2, 3,and 4wk follow-up, then all corneas were tested by hematoxylin and eosin staining histology.· RESULTS: The corneal grafts in Group A were transparent, while those in Group B showed corneal stromal edema and loosely arranged collagen fibers. One week after treatment, TEM revealed the intercellular desmosomes in the epithelial layers and intact non-dissolving nuclei in Group A. At week 4, the center of the corneas in Group A was transparent with regularly arranged collagen fibers and fibroblasts in the stroma. In Group B, squamous cells were observed on the corneal surface and some epithelial cells were detached.· CONCLUSION: Femtosecond laser-assisted deep lamellar keratoplasty can suppress inflammatory responses, prevent toxic substance-induced injury to the corneal endothelium and inner tissues with quicker recovery and better visual outcomes.展开更多

The fractional anisotropy (FA) derived from sequence C (b=1000s/mm^2, spatial resolution=0.55 mm×0.55 mm×2.5 mm, direction number=33, NSA=3) was too fuzzy to be used in tracking white matter fibers. By comparison, the high resolution and the FA with high contrast of gray matter and white matter derived from sequence D (b=800 s/mm^2, spatial resolution=1.0 mm×1.0 mm ×1.0 mm, numbers of direction=33, NSA=3) qualified in its application in tracking both thick and thin fibers, making it an optimal DTI setting for rhesus macaques.展开更多

Prefrontal dysfunction is closely associated with the symptoms of schizophrenia. In addition to the features typical of schizophrenia, patients also present with aspects of cognitive disorders. Based on these relationships, a monkey model mimicking the cognitive symptoms of schizophrenia has been made using treatment with the non-specific competitive N-methyI-D-aspartate receptor antagonist, phencyclidine. The symptoms are ameliorated by atypical antipsychotic drugs such as clozapine. The beneficial effects of clozapine on behavioral .impairment might be a specific indicator of schizophrenia-related cognitive impairment.展开更多
Apolygus lucorum is the dominant pathogenic insect attacking Bacillus thuringiensis(Bt)cotton in China.Additionally,20-hydroxyecdysone(20E)has important functions in many biological processes,including insect reproduc...Apolygus lucorum is the dominant pathogenic insect attacking Bacillus thuringiensis(Bt)cotton in China.Additionally,20-hydroxyecdysone(20E)has important functions in many biological processes,including insect reproduction.Phospholipase C(PLC),which is an essential enzyme for phosphoinositide metabolism,is involved in 20E signal transduction,but its function in 20E-mediated reproduction in A.lucorum remains unclear.In this study,20E increased A/PLCγ transcription as well as the abundance and activity of the encoded protein during molting and metamorphosis.The 20E treatment also induced the considerable accumulation of two second messengers,inositol triphosphate and diacylglycerol.The expression levels of genes encoding vitellogenin(AlVg)and soluble trehalase(AlTre-1)were similar to those of AlPLCy,and were upregulated in response to 20C.The silencing of AlPLCγ resulted in downregulated expression o f AITre-γ smdAlVg.However,the silencing of AlTre-1 and AlVg did not affect AlPLCγ expression.Moreover,the silencing of AlVg did not alter AlTre-1 expression.Furthermore,an examination of the insect specimens indicated that AlPLCy is required for female adult reproduction,and that downregulated expression of this gene is associated with decreases in fecundity,adult longevity,and egg hatching rate as well as delayed oocyte maturation.We propose that 20E regulates AlTre-1 expression via AlPLCy and affects Vg expression as well as ovary development to facilitate the reproductive activities of A.lucorum females.展开更多

文摘Three new pyridine type alkaloids,(-)-vinmajpyridines A–C(1–3),along with two known alkaloids,have been isolated from the aerial parts of Vinca major cultivated in Pakistan.Their structures have been elucidated by means of NMR and HRESIMS spectroscopic data.The new alkaloids were evaluated for their cytotoxicity against glioma initiating cell lines(GITC-3#and GITC-18#),glioblastoma cell lines(U-87MG and T98G),and lung cancer cell line A-549,but none of them was active at 20 lg/mL concentration.

文摘The prefrontal cortex is implicated in cognitive functioning and schizophrenia. Prefrontal dysfunction is closely associated with the symptoms of schizophrenia. In addition to the features typical of schizophrenia, patients also present with aspects of cognitive disorders. Based on these relationships, a monkey model mimicking the cognitive symptoms of schizophrenia has been made using treatment with the non-specific competitive N-methyI-D-aspartate receptor antagonist, phencyclidine. The symptoms are ameliorated by atypical antipsychotic drugs such as clozapine. The beneficial effects of clozapine on behavioral .impairment might be a specific indicator of schizophrenia-related cognitive impairment.

文摘Apolygus lucorum is the dominant pathogenic insect attacking Bacillus thuringiensis(Bt)cotton in China.Additionally,20-hydroxyecdysone(20E)has important functions in many biological processes,including insect reproduction.Phospholipase C(PLC),which is an essential enzyme for phosphoinositide metabolism,is involved in 20E signal transduction,but its function in 20E-mediated reproduction in A.lucorum remains unclear.In this study,20E increased A/PLCγ transcription as well as the abundance and activity of the encoded protein during molting and metamorphosis.The 20E treatment also induced the considerable accumulation of two second messengers,inositol triphosphate and diacylglycerol.The expression levels of genes encoding vitellogenin(AlVg)and soluble trehalase(AlTre-1)were similar to those of AlPLCy,and were upregulated in response to 20C.The silencing of AlPLCγ resulted in downregulated expression o f AITre-γ smdAlVg.However,the silencing of AlTre-1 and AlVg did not affect AlPLCγ expression.Moreover,the silencing of AlVg did not alter AlTre-1 expression.Furthermore,an examination of the insect specimens indicated that AlPLCy is required for female adult reproduction,and that downregulated expression of this gene is associated with decreases in fecundity,adult longevity,and egg hatching rate as well as delayed oocyte maturation.We propose that 20E regulates AlTre-1 expression via AlPLCy and affects Vg expression as well as ovary development to facilitate the reproductive activities of A.lucorum females.
