Microbial production of monoterpenoid indole alkaloids(MIAs)provides a sustainable and eco-friendly means to obtain compounds with high pharmaceutical values.However,efficient biosynthesis of MIAs in heterologous microorganisms is hindered due to low supply of key precursors such as geraniol and its derivative 8-hydroxygeraniol catalyzed by geraniol 8-hydroxylase(G8H).In this study,we developed a facile evolution platform to screen strains with improved yield of geraniol by using the SCRaMbLE system embedded in the Sc2.0 synthetic yeast and confirmed the causal role of relevant genomic targets.Through genome mining,we identified several G8H enzymes that perform much better than the commonly used CrG8H for 8-hydroxygeraniol production in vivo.We further showed that the N-terminus of these G8H enzymes plays an important role in cellular activity by swapping experiments.Finally,the combination of the engineered chassis,optimized biosynthesis pathway,and utilization of G8H led to the final strain with more than 30-fold improvement in producing 8-hydroxygeraniol compared with the starting strain.Overall,this study will provide insights into the construction and optimization of yeast cells for efficient biosynthesis of 8-hydroxygeraniol and its derivatives. 展开更多

Green building development in China relies mainly on compulsory measures that lack incentive policies to motivate the enthusiasm of real estate developers.A floor area ratio bonus is encouraged by the Chinese State Council.In order to ensure the feasibility of a reward quota,residential buildings that met the requirement for energy efficiency during the official assessment in Ningbo in 2014 were selected as research objects.The amount of energy and water savings in terms of the Assessment Standard for Green Building is converted into carbon emissions.Carbon emissions of different star-rated green buildings are then measured in accordance with the actual water and power consumption of residential dwellings in 2014.A regression equation is set up concerning the floor area ratio and index for residential land per capita.A carbon emission-based method is proposed for measuring the reward quota associated with floor area ratio and recommendations are given for development using green building. 展开更多

医学专业新生智能手机成瘾与心理健康关系研究

目的了解医学专业新生手机成瘾和心理健康现状,并对手机成瘾与心理健康关系进行研究,为医学专业新生智能手机成瘾的预防及干预提供依据。方法采用分层随机抽样的方法选取滨州医学院医学专业大一新生279人为研究对象,选用修订的智能手机... 目的了解医学专业新生手机成瘾和心理健康现状,并对手机成瘾与心理健康关系进行研究,为医学专业新生智能手机成瘾的预防及干预提供依据。方法采用分层随机抽样的方法选取滨州医学院医学专业大一新生279人为研究对象,选用修订的智能手机成瘾量表(smartphone addiction scale,SAS-C)、心理健康症状自评量表(mental health symptom self-rating checklist scale,SCL-90)进行测查。采用SPSS 19.0软件进行t检验。结果29.71%的医学专业新生存在智能手机成瘾问题,总体处于中等偏上水平。医学专业新生手机成瘾组的SCL-90总分与躯体化、人际关系敏感、抑郁、焦虑、敌对、精神病性等因子的得分均高于非手机成瘾组。医学专业新生智能手机成瘾总分与SCL-90总分存在相关性,除日常生活干扰因子外,其他因子与SCL-90各因子均呈正相关性。人际关系敏感和强迫症状对手机成瘾有显著的预测作用(β=0.24、0.26,P<0.01),是影响医学专业新生手机成瘾的重要变量。结论医学专业部分新生中存在智能手机成瘾问题,智能手机成瘾会产生一系列心理问题,特别是强迫症状、人际关系敏感、焦虑和精神病性等。 展开更多
