The corresponding adsorption capacity of immobilized ssDNA immunoadsorbent for IgGRF was enhanced from 46% to 86%, This result indicates the tryptophan residue is essential for the interaction between ssDNA and IgGRF .and ionic-bonding plays an important role in adsorption.展开更多

At different temperatures, pyridoxal 5--phosphate (PLP) was added in a 4:1 molar-ratio to the Hb tetramer followed by reduction with NaBH4 under N2 for 16 hrs subsequenl crosslinking by glutaraldehyde in presence of lysine for 14--16 hrs. Yielded soluble pyridoxalated polyhemoglobin (Poly Hb--P). 5%--20% gradient SDS--PAGE showed the change of the Hb molecular weight (MW) after crosslinking. Gel chromatography showed that the poly Hb--P had a continuous distribution of molecular weight with the range from about 65,000 to 600,000 Dalton. The poly Hb--P solution provided a normal oxygen carrying with 23.1 of P50, at pH 7.24 or 26.1 of P50 at pH 7.24展开更多

The degree of substitution of cholesteryl moiety in dextran main line is 3-5 cholesterols the 100 glucose units. We have prepared water solution of CHD using probe type sonifier and N-Phenyl-α-naphthylamine (PNA) as a fluorescent probe to study CHD self-aggregate process .For each solution of two samples, we found that the maximum emission of PNA in CHD solution was shifted to the lower wavelength and the intensity drastically increased above a critical CHD concentration. This change corresponds to the formation of micelle-like clusters self-aggregated by the cholesterol moiety once the CHD concentration exceeds 0.01 mg/ml.展开更多

T...A tumor-targeting gene vector G250mAb-PEI-PEG has been prepared by modification of polyethylenimine (PEI) with polyethyleneglycol (PEG) and G250, a monoclonal antibody against the G250 antigen on tumor cell surface. The transfection efficiency was as high as 70% in G250 positive HeLa cells, whereas the transfection efficiency was relatively low (30%) in normal NIH3T3 cells. A plasmid encoding the short hairpin RNA (shRNA) specific for nucleostemin gene (NS) was efficiently transfected into the HeLa cells with this nonviral gene vector. RNA interference down-regulated the expression of NS gene in HeLa cells, inhibited cells proliferation and induced apoptosis. However, the growth and activity of the NIH3T3 cells were not affected under the same treatment. These results indicate that the reported nonviral gene vector, G250mAb-PEI-PEG, can target and efficiently deliver genes into HeLa cells, and has the potential for the cervical cancer treatment.展开更多
