Background:Traditional Chinese medicine is promising for managing challenging and complex disorders,including cancer,and in particular,saffron is applied in treating various cancer types.However,its potential therapeu...Background:Traditional Chinese medicine is promising for managing challenging and complex disorders,including cancer,and in particular,saffron is applied in treating various cancer types.However,its potential therapeutic efficacy and active components in managing squamous cell carcinoma of the head and neck(HNSCC)remain unclear yet.Methods:Using network pharmacology approaches,active ingredients of saffron,their target genes,and HNSCC-related genes were identified.Enrichment analyses were conducted for determining molecular functions and pathways enriched by genes that overlapped between the saffron target gene set and the HNSCC gene set.Among the four known active ingredients of saffron,crocetin was found to have the strongest inhibitory impact on HNSCC,based on the findings of cell viability and migration assays.Therefore,the potential target genes of crocetin in HNSCC cells were examined using molecular docking experiments and were confirmed by qPCR.Result s:Four active ingredients of saffron and 184 of their target genes were identified.Further,a total of 34 overlapping saffron-/HNSCC-associated targets related to the four active ingredients were screened,and crocetin was chosen for further investigation because it had the strongest inhibitory effect on HNSCC cells.Molecular docking experiments indicated that ESR1 and CCND1 were the target genes of crocetin.These results were confirmed through qPCR analysis,in which crocetin was found to lower the expression of the ESR1 and CCND1 genes in AMC-HN-8 and FaDu cells.Conclusion:According to our results,crocetin is a primary active anti-cancer component of saffron that may have potential in the development of novel HNSCC-treating medications.However,more thorough molecular research is necessary for confirming these results and elucidating the anti-cancer mechanism underlying saffron.展开更多

AIM:To evaluate the application value of multi-slice spiral computed tomography(MSCT)for imaging determination of metastatic lymph nodes of gastric cancer and to explore reasonable diagnostic criteria.METHODS:Sixty pa...AIM:To evaluate the application value of multi-slice spiral computed tomography(MSCT)for imaging determination of metastatic lymph nodes of gastric cancer and to explore reasonable diagnostic criteria.METHODS:Sixty patients with gastric cancer underwent 64 MSCT scans before operation.Gastric cancer samples and perigastric lymph nodes were obtained after operation,formalin fixation and haematoxylineosin staining.The metastatic conditions of gastric cancer and perigastric lymph nodes were determined under a light microscope.A total of 605 lymph nodes were grouped and assessed according to distribution,size,shape and degree of lymph node enhancement.Then,the findings were compared with the postoperative pathological results.RESULTS:Among 605 lymph nodes,358 were confirmed as metastatic,accounting for 59.2%.A total of535 lymph nodes were detected in original axis images combined with multiplanar reconstruction images of MSCT.The metastatic lymph nodes had specific signs in computed tomography.This study showed that the long diameter of lymph nodes≥8 mm indicated metastasis;the sensitivity and specificity were 79.6%and78.8%,respectively.The difference of the mean value of lymph node enhancement density≥80 Hu indicated metastasis;the sensitivity and specificity were81.6%and 75.6%,respectively.The ratio of short diameter to long diameter of lymph nodes≥0.7 indicated metastasis;the sensitivity and specificity were85.6%and 71.8%,respectively.CONCLUSION:MSCT is a non-invasive and reliable method for preoperative examination of gastric cancer.Sensitivity and specificity for prediction of lymph node metastasis are high.展开更多

Objective To investigate the expressions of chemokine receptors and interleukin (IL) receptors on the peripheral blood mononuclear cells (PBMCs) from systemic lupus erythematosus (SLE) patients and their correla...Objective To investigate the expressions of chemokine receptors and interleukin (IL) receptors on the peripheral blood mononuclear cells (PBMCs) from systemic lupus erythematosus (SLE) patients and their correlations with clinical features as well as SLE disease activity index (SLEDAI). Methods The mRNA expressions of chemokine receptors and IL receptors on PBMCs of 93 SLE patients and 30 healthy controls were detected by reverse transcription-polymerase chain reaction, including CCR2, CCR3, CCR4, CCR5, CCR6, CCR8, CXCR3, CXCR5, CX3CR1, XCR1, IL-4R, and IL-10R. The clinical features of SLE patients were recorded. The correlations of chemokine receptors and IL receptors mRNA expressions with clinical features as well as SLEDAI were assayed using linear regression analysis. Results The level of CCR5 mRNA in SLE patients (including active and inactive SLE) was signifi- cantly higher than that in healthy controls (P〈0.05), and there was no significant difference between active and inactive patients in this respect (P〉0.05). CX3CR1 mRNA expression significantly increased from healthy control to inactive SLE to active SLE in sequence. The others (except for CCR8, CXCR3, and IL-1 OR) in active SLE patients weresignificantly higher than those in both inactive SLE patients and healthy controls (all P〈0.05). There were positive correlations between SLEDAI and CCR2 (r=0.424, t=4.313, P〈0.001), CCR3 (r=0.518, t=5.410, P〈0.001), CCR4 (r=0.376, t=3.851, P〈0.001), CCR6 (r=0.457, t=4.513,P〈0.001), CXCR5 (r=0.455, t=4.629, P〈0.001), CX3CR1 (r=0.44-5, t=4.523, P〈0.001), as well as XCRI (r=0.540, t=5.445, P〈0.001). And CCR5 mRNA expression level was positively correlated with IL-4R mRNA (r=0.313, t=2.353, P〈0.05). The patients with myositis and cutaneous vasculitis simultaneously showed lower levels of CCR5 and CX3CRI, and CCR5 expression was negatively correlated with the scores of SLEDAI in SLE cases accompanied by photosensitivity (r=0.426, t=- 2.155, P〈0.05). Conclusion Increased expressions of CCR5 and CX3CRI on PBMCs may be indicators in clinical survey for SLE.展开更多
Animals that live in seasonal environments adjust their reproduction cycle to optimize seasonal forage quality.Giant pandas(Ailuropoda melanoleuca)are seasonal altitudinal migrants that feed on bamboo shoots and leave...Animals that live in seasonal environments adjust their reproduction cycle to optimize seasonal forage quality.Giant pandas(Ailuropoda melanoleuca)are seasonal altitudinal migrants that feed on bamboo shoots and leaves with different nutritional quality.However,the importance of bamboo shoots to giant pandas,especially small and isolated populations,is not fully appreciated.Here,we explored whether mating time of giant pandas is shaped by bamboo shoot phenology.We also assessed the intensity of ongoing bamboo shoot harvesting by local communities in 42 giant panda reserves based on questionnaire surveys.Varying intensity and protection levels of bamboo shoot harvesting were found.From these data,we developed a priority ranking scheme of protection areas for this key food resource.Our study showed that pandas time their mating behavior to coincide with bamboo shoot phenology due to the high nutritional demands associated with mating and pregnancy.We also found that bamboo shoots were not well protected in many places.Liangshan,Daxiangling,and Xiaoxiangling,containing the most isolated panda populations,were identified as the areas with the most urgent need of protection.Furthermore,equal attention should be paid to the Qiong L-B population,as this region holds considerable potential to serve as a corridor between the Minshan and Qionglai populations.To address the challenges posed by bamboo shoot harvesting,we recommend establishing more practical bamboo shoot management policies,increasing public awareness of bamboo shoot protection,and providing alternative sources of income for local communities.展开更多

Fifty-six male SD rats were randomly divided into control group, stress group and post-stress group. The stress group was divided into 1, 2 and 4 h stress subgroups. The post-stress group was divided into 24, 48 and 72 h subgroups. The pH value of gastric juice, ulcer index (UI) of gastric mucosa and H^+, K^+- ATPase activity of gastric parietal cells were measured. Ultrastructural change of parietal cells was observed under transmission electron microscope (TEM).RESULTS: The pH value of gastric juice decreased time-dependently in stress group and increased in post-stress group. The H^+, K^+-ATPase activity of gastric parietal cells and the UI of gastric mucosa increased time-dependently in stress group and decreased in poststress group. Compared to control group, the pH value decreased remarkably (P = 0.0001), the UI and H^+, K^+- ATPase activity increased significantly (P = 0.0001, P = 0.0174) in 4 h stress subgroup. UI was positively related with stress time (r = 0.9876, P 〈 0.01) but negatively with pH value (r = -0.8724, P 〈 0.05). The parietal cells became active in stress group, especially in 4 h stress subgroup, in which plenty of intracellular canalicular and mitochondria were observed under TEM. In post-stress group, the parietal cells recovered to resting state.CONCOUSION: The acid secretion of parietal cells is consistent with their ultrastructural changes during the development and healing of stress ulcer induced by WRS and the degree of gastric mucosal lesions, suggesting gastric acid play an important role in the development of stress ulcer and is closely related with the recovery of gastric mucosal lesions induced by WRS.展开更多
Human induced pluripotent stem cells(hiPSCs)are invaluable resources for producing high-quality differentiated cells in unlimited quantities for both basic research and clinical use.They are particularly useful for st...Human induced pluripotent stem cells(hiPSCs)are invaluable resources for producing high-quality differentiated cells in unlimited quantities for both basic research and clinical use.They are particularly useful for studying human disease mechanisms in vitro by making it possible to circumvent the ethical issues of human embryonic stem cell research.However,significant limitations exist when using conventional flat culturing methods especially concerning cell expansion,differentiation efficiency,stability maintenance and multicellular 3D structure establishment,differentiation prediction.Embryoid bodies(EBs),the multicellular aggregates spontaneously generated from iPSCs in the suspension system,might help to address these issues.Due to the unique microenvironment and cell communication in EB structure that a 2D culture system cannot achieve,EBs have been widely applied in hiPSC-derived differentiation and show significant advantages especially in scaling up culturing,differentiation efficiency enhancement,ex vivo simulation,and organoid establishment.EBs can potentially also be used in early prediction of iPSC differentiation capability.To improve the stability and feasibility of EB-mediated differentiation and generate high quality EBs,critical factors including iPSC pluripotency maintenance,generation of uniform morphology using micro-pattern 3D culture systems,proper cellular density inoculation,and EB size control are discussed on the basis of both published data and our own laboratory experiences.Collectively,the production of a large quantity of homogeneous EBs with high quality is important for the stability and feasibility of many PSCs related studies.展开更多

Methods The concentration values c...Objective To measure the acute toxicity of halogenated benzenes to bacteria in natural waters and to study quantitative relationships between the structure and activity of chemicals. Methods The concentration values causing 50% inhibition of bacteria growth (24h-IC50) were determined according to the bacterial growth inhibition test method. The energy of the lowest unoccupied molecular orbital and the net charge of carbon atom of 20 halogenated benzenes were calculated by the quantum chemical MOPAC program. Results The logl/IC50 values ranged from 4.79 for 2,4-dinitrochlorobenzene to 3.65 for chlorobenzene. A quantitative structure-activity relationship model was derived from the toxicity and structural parameters: logl/IC50 =-0.531(ELUMO)+1.693(Qc)+0.163(logP)+3.375. This equation was found to fit well (r^2=0.860, s=0.106), and the average percentage error was only 1.98%. Conclusion Halogenated benzenes and alkyl halogenated benzenes are non-polar narcotics, and have hydrophobicity-dependent toxicity. The halogenated phenols and anilines exhibit a higher toxic potency than their hydrophobicity, whereas 2,4-dinitrochlorobenzene is electrophile with the halogen acting as the leaving group.展开更多

Human pluripotent stem cells(hPSCs)have the distinct advantage of being able to differentiate into cells of all three germ layers.Target cells or tissues derived from hPSCs have many uses such as drug screening,diseas...Human pluripotent stem cells(hPSCs)have the distinct advantage of being able to differentiate into cells of all three germ layers.Target cells or tissues derived from hPSCs have many uses such as drug screening,disease modeling,and transplantation therapy.There are currently a wide variety of differentiation methods available.However,most of the existing differentiation methods are unreliable,with uneven differentiation efficiency and poor reproducibility.At the same time,it is difficult to choose the optimal method when faced with so many differentiation schemes,and it is time-consuming and costly to explore a new differentiation approach.Thus,it is critical to design a robust and efficient method of differentiation.In this review article,we summarize a comprehensive approach in which hPSCs are differentiated into target cells or organoids including brain,liver,blood,melanocytes,and mesenchymal cells.This was accomplished by employing an embryoid body-based three-dimensional(3D)suspension culture system with multiple cells co-cultured.The method has high stable differentiation efficiency compared to the conventional 2D culture and can meet the requirements of clinical application.Additionally,ex vivo co-culture models might be able to constitute organoids that are highly similar or mimic human organs for potential organ transplantation in the future.展开更多

Objective:This cross-sectional survey aimed to explore Chinese college students’perceptions regarding acne and its topical medications,and to evaluate whether these perceptions affect their motivation to seek and adh...Objective:This cross-sectional survey aimed to explore Chinese college students’perceptions regarding acne and its topical medications,and to evaluate whether these perceptions affect their motivation to seek and adhere to treatments.Methods:A cross section survey was performed using a 14-item questionnaire was administered via WeChat.among Chinese college students at Jiangsu University aged 18-24 years.Results:One thousand and four hundred and fifteen responses were received.The prevalence of self-diagnosed acne was 34.1%.The factors associated with a greater likelihood of seeking treatment were being female,having acne for a longer duration,and having severe acne.Nearly two-thirds of the respondents had discontinued topical medication use within 1 month,with only 4.8%continuing for over 6 months.The main reason for discontinuation was slow onset of action.Patients who considered acne a long-term condition were more likely to seek treatment and use medication for longer periods than those who considered it a short-term condition.Lack of knowledge about medication features,proper usage,and prevention of adverse effects may lead to early discontinuation.Furthermore,93.8%of the respondents hoped to obtain the above information from doctors.Conclusion:Detailed explanations of acne and medication guidance are necessary to help patients understand the necessity of continuous treatment and the expected efficacy.展开更多

文摘AIM:To evaluate the application value of multi-slice spiral computed tomography(MSCT)for imaging determination of metastatic lymph nodes of gastric cancer and to explore reasonable diagnostic criteria.METHODS:Sixty patients with gastric cancer underwent 64 MSCT scans before operation.Gastric cancer samples and perigastric lymph nodes were obtained after operation,formalin fixation and haematoxylineosin staining.The metastatic conditions of gastric cancer and perigastric lymph nodes were determined under a light microscope.A total of 605 lymph nodes were grouped and assessed according to distribution,size,shape and degree of lymph node enhancement.Then,the findings were compared with the postoperative pathological results.RESULTS:Among 605 lymph nodes,358 were confirmed as metastatic,accounting for 59.2%.A total of535 lymph nodes were detected in original axis images combined with multiplanar reconstruction images of MSCT.The metastatic lymph nodes had specific signs in computed tomography.This study showed that the long diameter of lymph nodes≥8 mm indicated metastasis;the sensitivity and specificity were 79.6%and78.8%,respectively.The difference of the mean value of lymph node enhancement density≥80 Hu indicated metastasis;the sensitivity and specificity were81.6%and 75.6%,respectively.The ratio of short diameter to long diameter of lymph nodes≥0.7 indicated metastasis;the sensitivity and specificity were85.6%and 71.8%,respectively.CONCLUSION:MSCT is a non-invasive and reliable method for preoperative examination of gastric cancer.Sensitivity and specificity for prediction of lymph node metastasis are high.

文摘With the continuous development of digital medicine,minimally invasive precision and safety have become the primary development trends in hepatobiliary surgery.Due to the specificity and complexity of hepatobiliary surgery,traditional preoperative imaging techniques such as computed tomography and magnetic resonance imaging cannot meet the need for identification of fine anatomical regions.Imaging-based three-dimensional(3D)reconstruction,virtual simulation of surgery and 3D printing optimize the surgical plan through preoperative assessment,improving the controllability and safety of intraoperative operations,and in difficult-to-reach areas of the posterior and superior liver,assistive robots reproduce the surgeon’s natural movements with stable cameras,reducing natural vibrations.Electromagnetic navigation in abdominal surgery solves the problem of conventional surgery still relying on direct visual observation or preoperative image assessment.We summarize and compare these recent trends in digital medical solutions for the future development and refinement of digital medicine in hepatobiliary surgery.

文摘Objective To investigate the expressions of chemokine receptors and interleukin (IL) receptors on the peripheral blood mononuclear cells (PBMCs) from systemic lupus erythematosus (SLE) patients and their correlations with clinical features as well as SLE disease activity index (SLEDAI). Methods The mRNA expressions of chemokine receptors and IL receptors on PBMCs of 93 SLE patients and 30 healthy controls were detected by reverse transcription-polymerase chain reaction, including CCR2, CCR3, CCR4, CCR5, CCR6, CCR8, CXCR3, CXCR5, CX3CR1, XCR1, IL-4R, and IL-10R. The clinical features of SLE patients were recorded. The correlations of chemokine receptors and IL receptors mRNA expressions with clinical features as well as SLEDAI were assayed using linear regression analysis. Results The level of CCR5 mRNA in SLE patients (including active and inactive SLE) was signifi- cantly higher than that in healthy controls (P〈0.05), and there was no significant difference between active and inactive patients in this respect (P〉0.05). CX3CR1 mRNA expression significantly increased from healthy control to inactive SLE to active SLE in sequence. The others (except for CCR8, CXCR3, and IL-1 OR) in active SLE patients weresignificantly higher than those in both inactive SLE patients and healthy controls (all P〈0.05). There were positive correlations between SLEDAI and CCR2 (r=0.424, t=4.313, P〈0.001), CCR3 (r=0.518, t=5.410, P〈0.001), CCR4 (r=0.376, t=3.851, P〈0.001), CCR6 (r=0.457, t=4.513,P〈0.001), CXCR5 (r=0.455, t=4.629, P〈0.001), CX3CR1 (r=0.44-5, t=4.523, P〈0.001), as well as XCRI (r=0.540, t=5.445, P〈0.001). And CCR5 mRNA expression level was positively correlated with IL-4R mRNA (r=0.313, t=2.353, P〈0.05). The patients with myositis and cutaneous vasculitis simultaneously showed lower levels of CCR5 and CX3CRI, and CCR5 expression was negatively correlated with the scores of SLEDAI in SLE cases accompanied by photosensitivity (r=0.426, t=- 2.155, P〈0.05). Conclusion Increased expressions of CCR5 and CX3CRI on PBMCs may be indicators in clinical survey for SLE.

文摘Animals that live in seasonal environments adjust their reproduction cycle to optimize seasonal forage quality.Giant pandas(Ailuropoda melanoleuca)are seasonal altitudinal migrants that feed on bamboo shoots and leaves with different nutritional quality.However,the importance of bamboo shoots to giant pandas,especially small and isolated populations,is not fully appreciated.Here,we explored whether mating time of giant pandas is shaped by bamboo shoot phenology.We also assessed the intensity of ongoing bamboo shoot harvesting by local communities in 42 giant panda reserves based on questionnaire surveys.Varying intensity and protection levels of bamboo shoot harvesting were found.From these data,we developed a priority ranking scheme of protection areas for this key food resource.Our study showed that pandas time their mating behavior to coincide with bamboo shoot phenology due to the high nutritional demands associated with mating and pregnancy.We also found that bamboo shoots were not well protected in many places.Liangshan,Daxiangling,and Xiaoxiangling,containing the most isolated panda populations,were identified as the areas with the most urgent need of protection.Furthermore,equal attention should be paid to the Qiong L-B population,as this region holds considerable potential to serve as a corridor between the Minshan and Qionglai populations.To address the challenges posed by bamboo shoot harvesting,we recommend establishing more practical bamboo shoot management policies,increasing public awareness of bamboo shoot protection,and providing alternative sources of income for local communities.

文摘AIM: To investigate the dynamic functional and ultrastructural changes of gastric parietal cells induced by water immersion-restraint stress (WRS) in rats.METHODS: WRS model of Sprague-Dawley (SD) rats was established. Fifty-six male SD rats were randomly divided into control group, stress group and post-stress group. The stress group was divided into 1, 2 and 4 h stress subgroups. The post-stress group was divided into 24, 48 and 72 h subgroups. The pH value of gastric juice, ulcer index (UI) of gastric mucosa and H^+, K^+- ATPase activity of gastric parietal cells were measured. Ultrastructural change of parietal cells was observed under transmission electron microscope (TEM).RESULTS: The pH value of gastric juice decreased time-dependently in stress group and increased in post-stress group. The H^+, K^+-ATPase activity of gastric parietal cells and the UI of gastric mucosa increased time-dependently in stress group and decreased in poststress group. Compared to control group, the pH value decreased remarkably (P = 0.0001), the UI and H^+, K^+- ATPase activity increased significantly (P = 0.0001, P = 0.0174) in 4 h stress subgroup. UI was positively related with stress time (r = 0.9876, P 〈 0.01) but negatively with pH value (r = -0.8724, P 〈 0.05). The parietal cells became active in stress group, especially in 4 h stress subgroup, in which plenty of intracellular canalicular and mitochondria were observed under TEM. In post-stress group, the parietal cells recovered to resting state.CONCOUSION: The acid secretion of parietal cells is consistent with their ultrastructural changes during the development and healing of stress ulcer induced by WRS and the degree of gastric mucosal lesions, suggesting gastric acid play an important role in the development of stress ulcer and is closely related with the recovery of gastric mucosal lesions induced by WRS.

文摘Human induced pluripotent stem cells(hiPSCs)are invaluable resources for producing high-quality differentiated cells in unlimited quantities for both basic research and clinical use.They are particularly useful for studying human disease mechanisms in vitro by making it possible to circumvent the ethical issues of human embryonic stem cell research.However,significant limitations exist when using conventional flat culturing methods especially concerning cell expansion,differentiation efficiency,stability maintenance and multicellular 3D structure establishment,differentiation prediction.Embryoid bodies(EBs),the multicellular aggregates spontaneously generated from iPSCs in the suspension system,might help to address these issues.Due to the unique microenvironment and cell communication in EB structure that a 2D culture system cannot achieve,EBs have been widely applied in hiPSC-derived differentiation and show significant advantages especially in scaling up culturing,differentiation efficiency enhancement,ex vivo simulation,and organoid establishment.EBs can potentially also be used in early prediction of iPSC differentiation capability.To improve the stability and feasibility of EB-mediated differentiation and generate high quality EBs,critical factors including iPSC pluripotency maintenance,generation of uniform morphology using micro-pattern 3D culture systems,proper cellular density inoculation,and EB size control are discussed on the basis of both published data and our own laboratory experiences.Collectively,the production of a large quantity of homogeneous EBs with high quality is important for the stability and feasibility of many PSCs related studies.

文摘Objective To measure the acute toxicity of halogenated benzenes to bacteria in natural waters and to study quantitative relationships between the structure and activity of chemicals. Methods The concentration values causing 50% inhibition of bacteria growth (24h-IC50) were determined according to the bacterial growth inhibition test method. The energy of the lowest unoccupied molecular orbital and the net charge of carbon atom of 20 halogenated benzenes were calculated by the quantum chemical MOPAC program. Results The logl/IC50 values ranged from 4.79 for 2,4-dinitrochlorobenzene to 3.65 for chlorobenzene. A quantitative structure-activity relationship model was derived from the toxicity and structural parameters: logl/IC50 =-0.531(ELUMO)+1.693(Qc)+0.163(logP)+3.375. This equation was found to fit well (r^2=0.860, s=0.106), and the average percentage error was only 1.98%. Conclusion Halogenated benzenes and alkyl halogenated benzenes are non-polar narcotics, and have hydrophobicity-dependent toxicity. The halogenated phenols and anilines exhibit a higher toxic potency than their hydrophobicity, whereas 2,4-dinitrochlorobenzene is electrophile with the halogen acting as the leaving group.

文摘Given the difficulties of degrading benzotriazole(BTA),this study used a one-pot hydrothermal method to prepareα-Fe_(2)O_(3)/Cu_(2)O(FC)composites for photoelectrocatalytic(PEC)degradation of BTA.The characterization of FC structure showed that Cu_(2)O in cubic crystals was loaded with circular sheets of Fe_(2)O_(3).Owing to this structure,FC showed efficient PEC degradation of BTA when exposed to ultraviolet light.The experimental results demonstrated that FC efficiently degraded BTA.When the PEC degradation continued for 60 min,100%degradation of BTA was achieved because FC enhanced the photoelectron-hole separation and the separation and transfer of articulated carriers.High per-formance liquid chromatography-mass spectrometry showed that intermediates formed during the PEC degradation of BTA.Finally,various pathways for degradation of BTA were postulated.This FC-based PEC system provides a harmless and effective method for degradation of BTA.
文摘Human pluripotent stem cells(hPSCs)have the distinct advantage of being able to differentiate into cells of all three germ layers.Target cells or tissues derived from hPSCs have many uses such as drug screening,disease modeling,and transplantation therapy.There are currently a wide variety of differentiation methods available.However,most of the existing differentiation methods are unreliable,with uneven differentiation efficiency and poor reproducibility.At the same time,it is difficult to choose the optimal method when faced with so many differentiation schemes,and it is time-consuming and costly to explore a new differentiation approach.Thus,it is critical to design a robust and efficient method of differentiation.In this review article,we summarize a comprehensive approach in which hPSCs are differentiated into target cells or organoids including brain,liver,blood,melanocytes,and mesenchymal cells.This was accomplished by employing an embryoid body-based three-dimensional(3D)suspension culture system with multiple cells co-cultured.The method has high stable differentiation efficiency compared to the conventional 2D culture and can meet the requirements of clinical application.Additionally,ex vivo co-culture models might be able to constitute organoids that are highly similar or mimic human organs for potential organ transplantation in the future.

文摘To the Editor:A 43-year-old woman presented with mulberry-like plaques on the left side of her back.At the same site,she had acquired herpes zoster approximately 3 months ago and been healed after treatments.The condition started as small plaques and pimples in irregular shapes,followed by the extension of the new lesions with acupuncture-like pain and tenderness upon palpation.

文摘To the Editor:A 42-year-old Chinese man presented with head trauma caused by a brief loss of consciousness and fell after a bee sting on his nasal root while cycling outdoors.After being rushed to the emergency room,he developed sweats,dizziness,headaches,bosom frowsty,shortness of breath,and urinary incontinence.Blood pressure was 74/47 mmHg,blood oxygen saturation was 81%,and pulse rate was 149 beats/min.Physical examination revealed mental confusion,bilateral pupil diameter at 3 mm,slightly pupil insensitivity to light reflection,and rigid abdomen.25 mg promethazine and 10 mg dexamethasone were administered immediately.

文摘Objective:This cross-sectional survey aimed to explore Chinese college students’perceptions regarding acne and its topical medications,and to evaluate whether these perceptions affect their motivation to seek and adhere to treatments.Methods:A cross section survey was performed using a 14-item questionnaire was administered via WeChat.among Chinese college students at Jiangsu University aged 18-24 years.Results:One thousand and four hundred and fifteen responses were received.The prevalence of self-diagnosed acne was 34.1%.The factors associated with a greater likelihood of seeking treatment were being female,having acne for a longer duration,and having severe acne.Nearly two-thirds of the respondents had discontinued topical medication use within 1 month,with only 4.8%continuing for over 6 months.The main reason for discontinuation was slow onset of action.Patients who considered acne a long-term condition were more likely to seek treatment and use medication for longer periods than those who considered it a short-term condition.Lack of knowledge about medication features,proper usage,and prevention of adverse effects may lead to early discontinuation.Furthermore,93.8%of the respondents hoped to obtain the above information from doctors.Conclusion:Detailed explanations of acne and medication guidance are necessary to help patients understand the necessity of continuous treatment and the expected efficacy.
