BACKGROUND As an active ingredient derived from Dioscorea zingiberensis C.H.Wright,deltonin has been reported to show anti-cancer effects in a variety of malignancies.AIM To investigate the role and mechanism of actio...BACKGROUND As an active ingredient derived from Dioscorea zingiberensis C.H.Wright,deltonin has been reported to show anti-cancer effects in a variety of malignancies.AIM To investigate the role and mechanism of action of deltonin in promoting gastric carcinoma(GC)cell apoptosis and chemosensitivity to cisplatin.METHODS The GC cell lines AGS,HGC-27,and MKN-45 were treated with deltonin and then subjected to flow cytometry and 3-(4,5-dimethylthiazol-2-yl)-3,5-diphenyltet-razolium bromide assays for cell apoptosis and viability determination.Western blot analysis was conducted to examine alterations in the expression of apoptosis-related proteins(Bax,Bid,Bad,and Fas),DNA repair-associated proteins(Rad51 and MDM2),and phosphatidylinositol 3-kinase/protein kinase B/mammalian target of the rapamycin(PI3K/AKT/mTOR)and p38-mitogen-activated protein kinase(MAPK)axis proteins.Additionally,the influence of deltonin on GC cell chemosensitivity to cisplatin was evaluated both in vitro and in vivo.RESULTS Deltonin treatment weakened viability,enhanced apoptosis,and dampened DNA repair in GC cell lines in a dose-dependent pattern.Furthermore,deltonin mitigated PI3K,AKT,mTOR,and p38-MAPK phosphorylation.HS-173,an inhibitor of PI3K,attenuated GC cell viability and abolished deltonin inhibition of GC cell viability and PI3K/AKT/mTOR and p38-MAPK pathway activation.Deltonin also promoted the chemosensitivity of GC cells to cisplatin via repressing GC cell proliferation and growth and accelerating apoptosis.CONCLUSION Deltonin can boost the chemosensitivity of GC cells to cisplatin via inactivating p38-MAPK and PI3K/AKT/mTOR signaling.展开更多
Four intelligent optimization algorithms are compared by searching for control pulses to achieve the preparation of target quantum states for closed and open quantum systems, which include differential evolution(DE), ...Four intelligent optimization algorithms are compared by searching for control pulses to achieve the preparation of target quantum states for closed and open quantum systems, which include differential evolution(DE), particle swarm optimization(PSO), quantum-behaved particle swarm optimization(QPSO), and quantum evolutionary algorithm(QEA).We compare their control performance and point out their differences. By sampling and learning for uncertain quantum systems, the robustness of control pulses found by these four algorithms is also demonstrated and compared. The resulting research shows that the QPSO nearly outperforms the other three algorithms for all the performance criteria considered.This conclusion provides an important reference for solving complex quantum control problems by optimization algorithms and makes the QPSO be a powerful optimization tool.展开更多

Seventy male rats were rando...AIM To investigate the effect and mechanism of moxibustion in rats with ulcerative colitis.METHODS A rat colitis model was established by administering 4% dextran sulphate sodium solution. Seventy male rats were randomly divided into seven groups: Healthy controls(HC), ulcerative colitis model group(UC), UC with 7 d of moxibustion(UC-7), UC with 14 d of moxibustion(UC-14), UC with mesalazine gavage(UC-W), HC with 7 d of moxibustion(HC-7), HC with 14 d of moxibustion(HC-14). Moxibustion was applied to the bilateral Tianshu(ST25). Gut microbiome profiling was conducted by 16 S r RNA amplicon sequencing, and PCR and ELISA determined the expression of inflammatory cytokines in colon mucosa and serum, respectively. RESULTS Moxibustion treatment restored the colonic mucosa and decreased submucosal inflammatory cell infiltration in colitis rats. Rats treated with moxibustion and mesalazine had significantly lower levels of the dominant phyla Proteobacteria and the genera Saccharibacteria, Sphingomonas and Barnesiella than colitis rats, and they could restore the microbiome to levels similar to those observed in healthy rats. UC rats had reduced alpha diversity, which could be alleviated by moxibustion therapy, and UC-7 had a higher alpha diversity than UC-14. This finding suggests that short-term(7 d) but no longer term(14 d) moxibustion treatment may significantly affect the gut microbiome. The potential bacterial functions affected by moxibustion may be ascorbate and aldarate metabolism, and amino acid metabolism. Compared with HC group, the levels of the cytokines interleukin-12(IL-12)(P < 0.05) and IL-6, IL-17, IL-23, interferon-γ, lipopolysaccharide, Ig A, tumour necrosis factor-α and its receptors 1(TNFR1) and TNFR2(P < 0.01) were all increased, whereas anti-inflammatory cytokine IL-2 and IL-10(P < 0.01) and transforming growth factor-β(P < 0.05) were decreased in UC rats. These changes were reversed by moxibustion.CONCLUSION Our findings suggest that moxibustion exerts its therapeutic effect by repairing mucosal tissue damage and modulating the gut microbiome and intestinal mucosal immunity.展开更多
Casing corrosion during CO2 injection or storage results in significant economic loss and increased production risks.Therefore,in this paper,a corroded casing risk assessment model based on analytic hierarchy process ...Casing corrosion during CO2 injection or storage results in significant economic loss and increased production risks.Therefore,in this paper,a corroded casing risk assessment model based on analytic hierarchy process and fuzzy comprehensive evaluation is established to identify potential risks in time.First,the corrosion rate and residual strength characteristics are analyzed through corrosion tests and numerical simulations,respectively,to determine the risk factors that may lead to an accident.Then,an index system for corroded casing risk evaluation is established based on six important factors:temperature,CO2 partial pressure,flow velocity,corrosion radius,corrosion depth and wellhead pressure.Subsequently,the index weights are calculated via the analytic hierarchy process.Finally,the risk level of corroded casing is obtained via the fuzzy comprehensive evaluation.The corroded casing risk assessment model has been verified by a case well,which shows that the model is valuable and feasible.It provides an effective decision-making method for the risk evaluation of corroded casing in CO2 injection well,which is conductive to improve the wellbore operation efficiency.展开更多
BACKGROUND Ulcerative colitis(UC)is a chronic,nonspecific intestinal inflammatory disease.Acupuncture and moxibustion is proved effective in treating UC,but the mechanism has not been clarified.Proteomic technology ha...BACKGROUND Ulcerative colitis(UC)is a chronic,nonspecific intestinal inflammatory disease.Acupuncture and moxibustion is proved effective in treating UC,but the mechanism has not been clarified.Proteomic technology has revealed a variety of biological markers related to immunity and inflammation in UC,which provide new insights and directions for the study of mechanism of acupuncture and moxibustion treatment of UC.AIM To investigate the mechanism of electroacupuncture(EA)and herb-partitioned moxibustion(HM)on UC rats by using proteomics technology.METHODS Male Sprague-Dawley rats were randomly divided into the normal(N)group,the dextran sulfate sodium(DSS)-induced UC model(M)group,the HM group,and the EA group.UC rat model was prepared with 3%DSS,and HM and EA interventions at the bilateral Tianshu and Qihai acupoints were performed in HM or EA group.Haematoxylin and eosin staining was used for morphological evaluation of colon tissues.Isotope-labeled relative and absolute quantification(iTRAQ)and liquid chromatography-tandem mass spectrometry were performed for proteome analysis of the colon tissues,followed by bioinformatics analysis and protein-protein interaction networks establishment of differentially expressed proteins(DEPs)between groups.Then western blot was used for verification of selected DEPs.RESULTS The macroscopic colon injury scores and histopathology scores in the HM and EA groups were significantly decreased compared to the rats in the M group(P<0.01).Compared with the N group,a total of 202 DEPs were identified in the M group,including 111 up-regulated proteins and 91 down-regulated proteins,of which 25 and 15 proteins were reversed after HM and EA interventions,respectively.The DEPs were involved in various biological processes such as biological regulation,immune system progression and in multiple pathways including natural killer cell mediated cytotoxicity,intestinal immune network for immunoglobulin A(IgA)production,and FcγR-mediated phagocytosis.The Kyoto Encyclopedia of Genes and Genomes pathways of DEPs between HM and M groups,EA and M groups both included immuneassociated and oxidative phosphorylation.Network analysis revealed that multiple pathways for the DEPs of each group were involved in protein-protein interactions,and the expression of oxidative phosphorylation pathway-related proteins,including ATP synthase subunit g(ATP5L),ATP synthase beta subunit precursor(Atp5f),cytochrome c oxidase subunit 4 isoform 1(Cox4i1)were down-regulated after HM and EA interventions.Subsequent verification of selected DEPs(Synaptic vesicle glycoprotein 2A;nuclear cap binding protein subunit 1;carbamoyl phosphate synthetase 1;Cox4i1;ATP synthase subunit b,Atp5f1;doublecortin like kinase 3)by western blot confirmed the reliability of the iTRAQ data,HM and EA interventions can significantly downregulate the expression of oxidative phosphorylation-associated proteins(Cox4i1,Atp5f1)(P<0.01).CONCLUSION EA and HM could regulate the expression of ATP5L,Atp5f1,Cox4i1 that associated with oxidative phosphorylation,then might regulate immune-related pathways of intestinal immune network for IgA production,FcγR-mediated phagocytosis,thereby alleviating colonic inflammation of DSSinduced UC rats.展开更多
AIM To investigate and compare the analytical and clinical performance of Tian Long automatic hypersensitive hepatitis B virus(HBV) DNA quantification system and Roche CAP/CTM system.METHODS Two hundred blood samples ...AIM To investigate and compare the analytical and clinical performance of Tian Long automatic hypersensitive hepatitis B virus(HBV) DNA quantification system and Roche CAP/CTM system.METHODS Two hundred blood samples for HBV DNA testing, HBV-DNA negative samples and high-titer HBV-DNA mixture samples were collected and prepared. National standard materials for serum HBV and a worldwide HBV DNA panel were employed for performance verification. The analytical performance, such as limit of detection, limit of quantification, accuracy, precision, reproducibility, linearity, genotype coverage and cross-contamination, was determined using the Tian Long automatic hypersensitive HBV DNA quantification system(TL system). Correlation and Bland-Altman plot analyses were carried out to compare the clinical performance of the TL system assay and the CAP/CTM system. RESULTS The detection limit of the TL system was 10 IU/m L, and its limit of quantification was 30 IU/m L. The differences between the expected and tested concentrations of the national standards were less than ± 0.4 Log10 IU/m L, which showed high accuracy of the system. Results of the precision, reproducibility and linearity tests showed that the multiple test coefficient of variation(CV) of the same sample was less than 5% for 102-106 IU/m L; and for 30-108 IU/m L, the linear correlation coefficient r2 = 0.99. The TL system detected HBV DNA(A-H) genotypes and there was no cross-contamination during the "checkerboard" test. When compared with the CAP/CTM assay, the two assays showed 100% consistency in both negative and positive sample results(15 negative samples and 185 positive samples). No statistical differences between the two assays in the HBV DNA quantification values were observed(P > 0.05). Correlation analysis indicated a significant correlation between the two assays, r2 = 0.9774. The Bland-Altman plot analysis showed that 98.9% of the positive data were within the 95% acceptable range, and the maximum difference was-0.49.CONCLUSION The TL system has good analytical performance, and exhibits good agreement with the CAP/CTM system in clinical performance.展开更多
Objective:To investigate the efficacy and safety of Kanglaite injection(KLT)combined with chemotherapy in the treatment of pancreatic cancer.Methods:PubMed,EMBASE,the Cochrane Library,China National Knowledge Infrastr...Objective:To investigate the efficacy and safety of Kanglaite injection(KLT)combined with chemotherapy in the treatment of pancreatic cancer.Methods:PubMed,EMBASE,the Cochrane Library,China National Knowledge Infrastructure(CNKI),Wanfang Database,VIP Data,and Chinese Biomedical Database(CBM)were searched to get the studies about KLT plus chemotherapy for pancreatic cancer(from established to May 2019).Data extraction and bias risk assessment were carried out by two authors independently according to the retrieval method.RevMan(version 5.3)were employed for data analysis.Results:A total of 151 literatures were retrieved and 11 literatures were finally included.A total of 614 patients were included,including 308 in the treatment group and 306 in the control group.The results of meta-analysis showed that compared with chemotherapy alone,KLT combined with chemotherapy could improve the effective rate[Porr=0.0009,OR=1.96,95%CI(1.32,2.92)]and disease control rate[Pdcr<0.00001,OR=2.53,95%CI(1.76,3.62)],improve KPS score[P<0.00001,OR=3.59,95%CI(2.00,6.44)]and body mass indexes[P=0.0003,OR=3.45,95%CI(1.78,6.69)],prolong progression free survival(PFS)and overall survival(OS),reduce the rate of myelosuppression[P=0.03,OR=0.54,95%CI(0.30,0.95)],but could not reduce the occurrence of neurotoxicity[P=0.49,OR=0.80,95%CI(0.42,1.51)]and digestive tract reaction[P=0.51,OR=0.83,95%CI(0.48,1.44)].Conclusion:KLT combined with chemotherapy can improve the curative effect of pancreatic cancer,improve the quality of life of patients,prolong the survival of patients,and reduce the incidence of bone marrow suppression.However,due to the limitation of the quality and quantity of included literatures,this conclusion needs to be verified by high-quality study.展开更多
Background:We investigated possible biomarkers for endometriosis (EM) using the ClinProt technique and proteomics methods.Methods:We enrolled 50 patients with EM,34 with benign ovarian neoplasms and 40 healthy vol...Background:We investigated possible biomarkers for endometriosis (EM) using the ClinProt technique and proteomics methods.Methods:We enrolled 50 patients with EM,34 with benign ovarian neoplasms and 40 healthy volunteers in this study.Serum proteomic spectra were generated by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MS) combined with weak cationic exchange (WCX) magnetic beads.Possible biomarkers were analyzed by a random and repeat pattern model-validation method that we designed,and ClinProtools software,results were refined using online liquid chromatography-tandem MS.Results:We found a cluster of 5 peptides (4210,5264,2660,5635,and 5904 Da),using 3 peptides (4210,5904,2660 Da) to discriminate EM patients from healthy volunteers,with 96.67% sensitivity and 100% specificity.We selected 4210 and 5904 m/z,which differed most between patients with EM and controls,and identified them as fragments of ATP1B4,and the fibrinogen alpha (FGA) isoform 1/2 of the FGA chain precursor,respectively.Conclusions:ClinProt can identify EM biomarkers,which-most notably-distinguish even early-stage or minimal disease.We found 5 stable peaks at 4210,5264,2660,5635,and 5904 Da as potential EM biomarkers,the strongest of which were associated with ATP1B4 (4210 Da) and FGA (5904 Da); this indicates that ATP1B4 and FGA are associated with EM pathogenesis.展开更多

In this work, fully con...Donor-acceptor (D-A) type fully conjugated block copolymer systems have been rarely reported due to the challenges in synthetic approaches to prepare well-defined low-polydispersity products. In this work, fully conjugated block copolymers are synthesized in a one-pot reaction through Stille coupling polycondensation, by utilizing the end-functional polymer copolymerization method. End-functional P3HT are copolymerized with AA (2,7-dihromo-9-(heptadecan-9-yl)-9H- carbazole) and BB (4,7-bis(5-(trimethylstannyl)thiophen-2-yl)benzo[c][1,2,5]thiadiazole, TBT) type monomers, respectively. The orthogonal solubility between the very soluble P3HT donor and the insoluble PCDTBT acceptor block improves the purity of block copolymers as well as distinct nano-scale phase-separation compared with other reports on miscibility of donor and acceptor polymer block. Further purification via preparative GPC is carried out to remove the excess of unreacted P3HT and free PCDTBT as well as to achieve low polydispersity of block copolymers. The chemical structure of the P3HT- b-PCDTBT block copolymers are verified via IH-NMR, and further confirmed by FTIR spectra. The block copolymer shows broad absorption and moderate optical band gap of 1.8 eV. Furthermore, the fully conjugated block copolymer films exhibit significant fine structures, much smoother film morphology compared to P3HT/PCDTBT polymer blends. By adding a small amount of block copolymer P3HT-b-PCDTBT as a compatibilizer into the bulk-heterojunction of P3HT:PC61BM blends, polymer solar ceils with an 8% increase of short circuit current (Jse) and 10% increase of power conversion efficiency (PCE) are achieved owing to the improvement of the active-layer film morphology. To the best of our knowledge, this is the first report on donor-acceptor type fully conjugated block copolymer as an effective ternary additive in polymer: fullerene bulk heterojunction solar cells.展开更多

By now, dynamic reliability research mainly concentrates on the reliability assessment; the methods mainly include dynamic fault tree, extension of event sequence diagram and Monte Carlo simulation, and et al. The paper aims to research the dynamic reliability optimization. On the basis of analysis of the four quality influence factors in the construction engineering, a method based on gray correlation degree is employed to calculate the weights of factors affecting construction process quality. Then the weights are added into the reliability improvement feasible index (RIFI). Furthermore, a novel nonlinear programming mathematic optimization model is established. In the Insight software environment, the Adaptive Simulated Annealing (ASA) algorithm is used to get a more accurate construction subsystem optimal reliability under different RIFI conditions. In addition, the relationship between construction quality and construction system reliability is analyzed, the proposed methods and detailed processing can offer a useful reference for improving the construction system quality level.展开更多
A series of random terpolymers P2-P5 were designed and synthesized by randomly embedding 5 mol%, 10 mol%, 15 mol% and 25 mol% feed ratios of low cost 2,2-bithiophene as the third monomer to the famous donor-acceptor ...A series of random terpolymers P2-P5 were designed and synthesized by randomly embedding 5 mol%, 10 mol%, 15 mol% and 25 mol% feed ratios of low cost 2,2-bithiophene as the third monomer to the famous donor-acceptor (D-A) type copolymer PTBT-Th (P1). All polymers showed similar molecular weight with number-average molecular weight (Mn) and weight-average molecular weight (Mw) in the range of (59-74) and (93-114) kg·mol-1, respectively, to ensure a fair comparison on the structure-property relationships. Compared with the control copolymer PTBT-Th, the random terpolymers exhibited enhanced absorption intensity in a wide range from 400 nm to 650 nm in both solution and film as well as in polymer/PC71BM blends. From grazing incident wide-angle X-ray diffraction (GIWAXS), compared with the regularly alternated copolymer PTB7-Th, the random terpolymers demonstrated mild structural disorder with reduced (100) lamellar stacking and slightly weakened (010) π-π stacking for the polymers as well as slightly reduced PC71BM aggregation in polymer/PC71BM blends. However, the measured hole mobility for terpolymers ((1.20-3.73) × 10 -4 cm2·V-1·s-1) was evaluated to be comparable or even higher than 1.35 × 10 -4 cm2·V-1 ·s-1 of the alternative copolymer. Enhanced average power conversion efficiency (PCE) from 7.35% to 8.11% and 7.79% to 8.37% was observed in both conventional and inverted device architectures from copolymer P1 to terpolymers P4, while further increasing the 2,2-bithiophene feed ratio decreased the PCE.展开更多
A series of novel hyperbranched polymers (HBPs) consisting ofa 2,7-subsituted 9-(heptadecan-9-yl)-9H-carbazole unit (A2+A2') and a tetra-substituted green thermally activated delayed fluorescence (TADF) dye ...A series of novel hyperbranched polymers (HBPs) consisting ofa 2,7-subsituted 9-(heptadecan-9-yl)-9H-carbazole unit (A2+A2') and a tetra-substituted green thermally activated delayed fluorescence (TADF) dye of 2,3,5,6-tetra(9H- carbazol-9-yl)-4-pyridinecarbonitrile (4CzCNPy, B4) have been synthesized via Suzuki cross-coupling reaction following an "Az+A2'+B4" method. The polymers are named according to the polymerization ratio of 4CzCNPy monomer (5 mol%, 10 mol% and 15 mol% for HBPs of P2-P4 respectively, and 0 mol% for the control linear polymer P1). Their thermal, optoelectronic and electrochemical properties have been characterized by a combination of techniques. All the polymers exhibit high thermal stability with the decomposition temperatures (Ta) above 400 ℃ and glass transition temperatures (Tg) up to 98℃. Unfortunately, the incorporation of TADF moiety into these HBP materials induced non-TADF characteristics. However, when the HBPs functionalized as the host for our previously developed 4CzCNPy TADF dopant in solution processed devices, maximum external quantum efficiency of 5.7% and current efficiency of 17.9 cd/A have been achieved in P3-based device, which is significantly higher than those of 1.5% and 4.2 cd/A for the linear polymer P1.展开更多

文摘AIM To investigate the effect and mechanism of moxibustion in rats with ulcerative colitis.METHODS A rat colitis model was established by administering 4% dextran sulphate sodium solution. Seventy male rats were randomly divided into seven groups: Healthy controls(HC), ulcerative colitis model group(UC), UC with 7 d of moxibustion(UC-7), UC with 14 d of moxibustion(UC-14), UC with mesalazine gavage(UC-W), HC with 7 d of moxibustion(HC-7), HC with 14 d of moxibustion(HC-14). Moxibustion was applied to the bilateral Tianshu(ST25). Gut microbiome profiling was conducted by 16 S r RNA amplicon sequencing, and PCR and ELISA determined the expression of inflammatory cytokines in colon mucosa and serum, respectively. RESULTS Moxibustion treatment restored the colonic mucosa and decreased submucosal inflammatory cell infiltration in colitis rats. Rats treated with moxibustion and mesalazine had significantly lower levels of the dominant phyla Proteobacteria and the genera Saccharibacteria, Sphingomonas and Barnesiella than colitis rats, and they could restore the microbiome to levels similar to those observed in healthy rats. UC rats had reduced alpha diversity, which could be alleviated by moxibustion therapy, and UC-7 had a higher alpha diversity than UC-14. This finding suggests that short-term(7 d) but no longer term(14 d) moxibustion treatment may significantly affect the gut microbiome. The potential bacterial functions affected by moxibustion may be ascorbate and aldarate metabolism, and amino acid metabolism. Compared with HC group, the levels of the cytokines interleukin-12(IL-12)(P < 0.05) and IL-6, IL-17, IL-23, interferon-γ, lipopolysaccharide, Ig A, tumour necrosis factor-α and its receptors 1(TNFR1) and TNFR2(P < 0.01) were all increased, whereas anti-inflammatory cytokine IL-2 and IL-10(P < 0.01) and transforming growth factor-β(P < 0.05) were decreased in UC rats. These changes were reversed by moxibustion.CONCLUSION Our findings suggest that moxibustion exerts its therapeutic effect by repairing mucosal tissue damage and modulating the gut microbiome and intestinal mucosal immunity.

文摘Casing corrosion during CO2 injection or storage results in significant economic loss and increased production risks.Therefore,in this paper,a corroded casing risk assessment model based on analytic hierarchy process and fuzzy comprehensive evaluation is established to identify potential risks in time.First,the corrosion rate and residual strength characteristics are analyzed through corrosion tests and numerical simulations,respectively,to determine the risk factors that may lead to an accident.Then,an index system for corroded casing risk evaluation is established based on six important factors:temperature,CO2 partial pressure,flow velocity,corrosion radius,corrosion depth and wellhead pressure.Subsequently,the index weights are calculated via the analytic hierarchy process.Finally,the risk level of corroded casing is obtained via the fuzzy comprehensive evaluation.The corroded casing risk assessment model has been verified by a case well,which shows that the model is valuable and feasible.It provides an effective decision-making method for the risk evaluation of corroded casing in CO2 injection well,which is conductive to improve the wellbore operation efficiency.

文摘BACKGROUND Ulcerative colitis(UC)is a chronic,nonspecific intestinal inflammatory disease.Acupuncture and moxibustion is proved effective in treating UC,but the mechanism has not been clarified.Proteomic technology has revealed a variety of biological markers related to immunity and inflammation in UC,which provide new insights and directions for the study of mechanism of acupuncture and moxibustion treatment of UC.AIM To investigate the mechanism of electroacupuncture(EA)and herb-partitioned moxibustion(HM)on UC rats by using proteomics technology.METHODS Male Sprague-Dawley rats were randomly divided into the normal(N)group,the dextran sulfate sodium(DSS)-induced UC model(M)group,the HM group,and the EA group.UC rat model was prepared with 3%DSS,and HM and EA interventions at the bilateral Tianshu and Qihai acupoints were performed in HM or EA group.Haematoxylin and eosin staining was used for morphological evaluation of colon tissues.Isotope-labeled relative and absolute quantification(iTRAQ)and liquid chromatography-tandem mass spectrometry were performed for proteome analysis of the colon tissues,followed by bioinformatics analysis and protein-protein interaction networks establishment of differentially expressed proteins(DEPs)between groups.Then western blot was used for verification of selected DEPs.RESULTS The macroscopic colon injury scores and histopathology scores in the HM and EA groups were significantly decreased compared to the rats in the M group(P<0.01).Compared with the N group,a total of 202 DEPs were identified in the M group,including 111 up-regulated proteins and 91 down-regulated proteins,of which 25 and 15 proteins were reversed after HM and EA interventions,respectively.The DEPs were involved in various biological processes such as biological regulation,immune system progression and in multiple pathways including natural killer cell mediated cytotoxicity,intestinal immune network for immunoglobulin A(IgA)production,and FcγR-mediated phagocytosis.The Kyoto Encyclopedia of Genes and Genomes pathways of DEPs between HM and M groups,EA and M groups both included immuneassociated and oxidative phosphorylation.Network analysis revealed that multiple pathways for the DEPs of each group were involved in protein-protein interactions,and the expression of oxidative phosphorylation pathway-related proteins,including ATP synthase subunit g(ATP5L),ATP synthase beta subunit precursor(Atp5f),cytochrome c oxidase subunit 4 isoform 1(Cox4i1)were down-regulated after HM and EA interventions.Subsequent verification of selected DEPs(Synaptic vesicle glycoprotein 2A;nuclear cap binding protein subunit 1;carbamoyl phosphate synthetase 1;Cox4i1;ATP synthase subunit b,Atp5f1;doublecortin like kinase 3)by western blot confirmed the reliability of the iTRAQ data,HM and EA interventions can significantly downregulate the expression of oxidative phosphorylation-associated proteins(Cox4i1,Atp5f1)(P<0.01).CONCLUSION EA and HM could regulate the expression of ATP5L,Atp5f1,Cox4i1 that associated with oxidative phosphorylation,then might regulate immune-related pathways of intestinal immune network for IgA production,FcγR-mediated phagocytosis,thereby alleviating colonic inflammation of DSSinduced UC rats.
文摘AIM To investigate and compare the analytical and clinical performance of Tian Long automatic hypersensitive hepatitis B virus(HBV) DNA quantification system and Roche CAP/CTM system.METHODS Two hundred blood samples for HBV DNA testing, HBV-DNA negative samples and high-titer HBV-DNA mixture samples were collected and prepared. National standard materials for serum HBV and a worldwide HBV DNA panel were employed for performance verification. The analytical performance, such as limit of detection, limit of quantification, accuracy, precision, reproducibility, linearity, genotype coverage and cross-contamination, was determined using the Tian Long automatic hypersensitive HBV DNA quantification system(TL system). Correlation and Bland-Altman plot analyses were carried out to compare the clinical performance of the TL system assay and the CAP/CTM system. RESULTS The detection limit of the TL system was 10 IU/m L, and its limit of quantification was 30 IU/m L. The differences between the expected and tested concentrations of the national standards were less than ± 0.4 Log10 IU/m L, which showed high accuracy of the system. Results of the precision, reproducibility and linearity tests showed that the multiple test coefficient of variation(CV) of the same sample was less than 5% for 102-106 IU/m L; and for 30-108 IU/m L, the linear correlation coefficient r2 = 0.99. The TL system detected HBV DNA(A-H) genotypes and there was no cross-contamination during the "checkerboard" test. When compared with the CAP/CTM assay, the two assays showed 100% consistency in both negative and positive sample results(15 negative samples and 185 positive samples). No statistical differences between the two assays in the HBV DNA quantification values were observed(P > 0.05). Correlation analysis indicated a significant correlation between the two assays, r2 = 0.9774. The Bland-Altman plot analysis showed that 98.9% of the positive data were within the 95% acceptable range, and the maximum difference was-0.49.CONCLUSION The TL system has good analytical performance, and exhibits good agreement with the CAP/CTM system in clinical performance.

文摘Objective:To investigate the efficacy and safety of Kanglaite injection(KLT)combined with chemotherapy in the treatment of pancreatic cancer.Methods:PubMed,EMBASE,the Cochrane Library,China National Knowledge Infrastructure(CNKI),Wanfang Database,VIP Data,and Chinese Biomedical Database(CBM)were searched to get the studies about KLT plus chemotherapy for pancreatic cancer(from established to May 2019).Data extraction and bias risk assessment were carried out by two authors independently according to the retrieval method.RevMan(version 5.3)were employed for data analysis.Results:A total of 151 literatures were retrieved and 11 literatures were finally included.A total of 614 patients were included,including 308 in the treatment group and 306 in the control group.The results of meta-analysis showed that compared with chemotherapy alone,KLT combined with chemotherapy could improve the effective rate[Porr=0.0009,OR=1.96,95%CI(1.32,2.92)]and disease control rate[Pdcr<0.00001,OR=2.53,95%CI(1.76,3.62)],improve KPS score[P<0.00001,OR=3.59,95%CI(2.00,6.44)]and body mass indexes[P=0.0003,OR=3.45,95%CI(1.78,6.69)],prolong progression free survival(PFS)and overall survival(OS),reduce the rate of myelosuppression[P=0.03,OR=0.54,95%CI(0.30,0.95)],but could not reduce the occurrence of neurotoxicity[P=0.49,OR=0.80,95%CI(0.42,1.51)]and digestive tract reaction[P=0.51,OR=0.83,95%CI(0.48,1.44)].Conclusion:KLT combined with chemotherapy can improve the curative effect of pancreatic cancer,improve the quality of life of patients,prolong the survival of patients,and reduce the incidence of bone marrow suppression.However,due to the limitation of the quality and quantity of included literatures,this conclusion needs to be verified by high-quality study.

文摘Background:We investigated possible biomarkers for endometriosis (EM) using the ClinProt technique and proteomics methods.Methods:We enrolled 50 patients with EM,34 with benign ovarian neoplasms and 40 healthy volunteers in this study.Serum proteomic spectra were generated by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MS) combined with weak cationic exchange (WCX) magnetic beads.Possible biomarkers were analyzed by a random and repeat pattern model-validation method that we designed,and ClinProtools software,results were refined using online liquid chromatography-tandem MS.Results:We found a cluster of 5 peptides (4210,5264,2660,5635,and 5904 Da),using 3 peptides (4210,5904,2660 Da) to discriminate EM patients from healthy volunteers,with 96.67% sensitivity and 100% specificity.We selected 4210 and 5904 m/z,which differed most between patients with EM and controls,and identified them as fragments of ATP1B4,and the fibrinogen alpha (FGA) isoform 1/2 of the FGA chain precursor,respectively.Conclusions:ClinProt can identify EM biomarkers,which-most notably-distinguish even early-stage or minimal disease.We found 5 stable peaks at 4210,5264,2660,5635,and 5904 Da as potential EM biomarkers,the strongest of which were associated with ATP1B4 (4210 Da) and FGA (5904 Da); this indicates that ATP1B4 and FGA are associated with EM pathogenesis.

文摘Donor-acceptor (D-A) type fully conjugated block copolymer systems have been rarely reported due to the challenges in synthetic approaches to prepare well-defined low-polydispersity products. In this work, fully conjugated block copolymers are synthesized in a one-pot reaction through Stille coupling polycondensation, by utilizing the end-functional polymer copolymerization method. End-functional P3HT are copolymerized with AA (2,7-dihromo-9-(heptadecan-9-yl)-9H- carbazole) and BB (4,7-bis(5-(trimethylstannyl)thiophen-2-yl)benzo[c][1,2,5]thiadiazole, TBT) type monomers, respectively. The orthogonal solubility between the very soluble P3HT donor and the insoluble PCDTBT acceptor block improves the purity of block copolymers as well as distinct nano-scale phase-separation compared with other reports on miscibility of donor and acceptor polymer block. Further purification via preparative GPC is carried out to remove the excess of unreacted P3HT and free PCDTBT as well as to achieve low polydispersity of block copolymers. The chemical structure of the P3HT- b-PCDTBT block copolymers are verified via IH-NMR, and further confirmed by FTIR spectra. The block copolymer shows broad absorption and moderate optical band gap of 1.8 eV. Furthermore, the fully conjugated block copolymer films exhibit significant fine structures, much smoother film morphology compared to P3HT/PCDTBT polymer blends. By adding a small amount of block copolymer P3HT-b-PCDTBT as a compatibilizer into the bulk-heterojunction of P3HT:PC61BM blends, polymer solar ceils with an 8% increase of short circuit current (Jse) and 10% increase of power conversion efficiency (PCE) are achieved owing to the improvement of the active-layer film morphology. To the best of our knowledge, this is the first report on donor-acceptor type fully conjugated block copolymer as an effective ternary additive in polymer: fullerene bulk heterojunction solar cells.

文摘The clinical efficacy of acupuncture-moxibustion(AM)mainly depends on acupoints,but the relationship between the acupoints and AM remains unclear.Improving clinical efficacy and clarifying the mechanisms of AM is critical.We found that the specificity and sensitivity of acupoints,the skill of operation,and reasonable amount of stimulation can significantly improve the efficacy of AM.In addition,some studies have shown that Neural-Endocrine-Immune network and metabolites are involved in this process,clarifying the therapeutic mechanism of acupoints.Therefore,how to effectively use acupoints to improve the clinical efficacy of AM is one of the key issues in need of an urgent solution.In summary,this article reviewed the Chinese and English databases on the clinical efficacy and underlying mechanisms of acupoints to clarify the relationship between acupoints and AM efficacy and ultimately improve clinical efficacy through effective and rational use of acupoints.
文摘Dynamic reliability is a very important issue in reliability research. The dynamic reliability analysis for the project is still in search of domestic and international research in the exploration stage. By now, dynamic reliability research mainly concentrates on the reliability assessment; the methods mainly include dynamic fault tree, extension of event sequence diagram and Monte Carlo simulation, and et al. The paper aims to research the dynamic reliability optimization. On the basis of analysis of the four quality influence factors in the construction engineering, a method based on gray correlation degree is employed to calculate the weights of factors affecting construction process quality. Then the weights are added into the reliability improvement feasible index (RIFI). Furthermore, a novel nonlinear programming mathematic optimization model is established. In the Insight software environment, the Adaptive Simulated Annealing (ASA) algorithm is used to get a more accurate construction subsystem optimal reliability under different RIFI conditions. In addition, the relationship between construction quality and construction system reliability is analyzed, the proposed methods and detailed processing can offer a useful reference for improving the construction system quality level.

文摘A series of random terpolymers P2-P5 were designed and synthesized by randomly embedding 5 mol%, 10 mol%, 15 mol% and 25 mol% feed ratios of low cost 2,2-bithiophene as the third monomer to the famous donor-acceptor (D-A) type copolymer PTBT-Th (P1). All polymers showed similar molecular weight with number-average molecular weight (Mn) and weight-average molecular weight (Mw) in the range of (59-74) and (93-114) kg·mol-1, respectively, to ensure a fair comparison on the structure-property relationships. Compared with the control copolymer PTBT-Th, the random terpolymers exhibited enhanced absorption intensity in a wide range from 400 nm to 650 nm in both solution and film as well as in polymer/PC71BM blends. From grazing incident wide-angle X-ray diffraction (GIWAXS), compared with the regularly alternated copolymer PTB7-Th, the random terpolymers demonstrated mild structural disorder with reduced (100) lamellar stacking and slightly weakened (010) π-π stacking for the polymers as well as slightly reduced PC71BM aggregation in polymer/PC71BM blends. However, the measured hole mobility for terpolymers ((1.20-3.73) × 10 -4 cm2·V-1·s-1) was evaluated to be comparable or even higher than 1.35 × 10 -4 cm2·V-1 ·s-1 of the alternative copolymer. Enhanced average power conversion efficiency (PCE) from 7.35% to 8.11% and 7.79% to 8.37% was observed in both conventional and inverted device architectures from copolymer P1 to terpolymers P4, while further increasing the 2,2-bithiophene feed ratio decreased the PCE.

文摘A series of novel hyperbranched polymers (HBPs) consisting ofa 2,7-subsituted 9-(heptadecan-9-yl)-9H-carbazole unit (A2+A2') and a tetra-substituted green thermally activated delayed fluorescence (TADF) dye of 2,3,5,6-tetra(9H- carbazol-9-yl)-4-pyridinecarbonitrile (4CzCNPy, B4) have been synthesized via Suzuki cross-coupling reaction following an "Az+A2'+B4" method. The polymers are named according to the polymerization ratio of 4CzCNPy monomer (5 mol%, 10 mol% and 15 mol% for HBPs of P2-P4 respectively, and 0 mol% for the control linear polymer P1). Their thermal, optoelectronic and electrochemical properties have been characterized by a combination of techniques. All the polymers exhibit high thermal stability with the decomposition temperatures (Ta) above 400 ℃ and glass transition temperatures (Tg) up to 98℃. Unfortunately, the incorporation of TADF moiety into these HBP materials induced non-TADF characteristics. However, when the HBPs functionalized as the host for our previously developed 4CzCNPy TADF dopant in solution processed devices, maximum external quantum efficiency of 5.7% and current efficiency of 17.9 cd/A have been achieved in P3-based device, which is significantly higher than those of 1.5% and 4.2 cd/A for the linear polymer P1.
