The present report included three patients (one male and two females; age range, 25-51 years) with hepatic PEComas. The collected data included the clinical manifestations, diagnosis, management, treatment, and prognosis. Since it is difficult to diagnose hepatic PEComas by imaging, the patients were diagnosed by tumor tissue examination such as immunohistochemistry, which was positive for HMB-45, Melan-A, and SMA on all slides. The tumor was composed of diverse tissues including smooth muscle, adipose tissue, and thick-walled blood vessels. During the follow-up period, one of the tumors was malignant (double-positive for CD34 and Ki-67) and recurred 3 months after surgery. In addition, malignant hepatic PECo- mas were reviewed in the literature, indicating that the majority of hepatic PEComas are benign, but few hepatic PEComas exhibit malignant behaviors in older female patients (〉50 years of age) with abdominal discomfort and pain, larger tumor size (〉10 cm), or positive staining for CD34 and Ki-67. In conclusion, there is no effective method to diagnose PEComas. Currently, the diagnosis of PEComas depends on immunohistochemical staining. Tumor resection and close follow-up are the principal methods for the management of PEComas.展开更多

Despite early diagnosis and best therapeutic regimens available, the prognosis of malignant or metastatic breast cancer patients is still not optimistic. Hedgehog signaling, a classical pathway indispensable to embryonic development, participates in the growth of a variety of tumors. In the present study,the effect of Sonic Hedgehog(Shh) on breast cancer cells was investigated. We identified that Shh signal stimulated the migration of MCF-7 breast cancer cells. Smo and Gli1 were involved in Shh-stimulated migration of MCF-7 cells. Activating Smo and Gli1 induced cell migration, which was blocked by their specific antagonists.The effect of Shh signaling on MCF-7 cells was independent of Wnt5 a, Dvl2 and Rab35, but directly dependent on Rac1. In conclusion, our study suggested that Shh promotes breast cancer cell migration via Rac1 independently of the non-canonical Wnt signaling pathway, which may represent a rational molecular target for combination medication in breast cancer.展开更多
Objective:To analyze the clinical characteristics and laboratory examinations of patients with new coronavirus(COVID-19)and compare the effect and prognosis of patients with diabetes mellitus(DM)on COVID-19.Methods:56...Objective:To analyze the clinical characteristics and laboratory examinations of patients with new coronavirus(COVID-19)and compare the effect and prognosis of patients with diabetes mellitus(DM)on COVID-19.Methods:56 cases of COVID-19 with DM and 85 age-matched admitted patients without DM who were admitted to the People's Hospital of Wuhan University from January 31,2020 to March 15,2020 were analyzed through a retrospective cohort study.Patients data were collected through electronic cases,and used SPSS 26.0 and Graphpad Prism 8.0 statistical software to compare changes in various indicators of patients in DM and non-DM groups and to draw the survival curves.Results:Compared with the non-DM group,the DM group has a higher percentage of mechanical ventilation,higher mortality and severe patients,with a higher proportion of initial symptoms of nausea and hypertension(P<0.05).There were no significant differences in the gender,initial symptoms(except nausea),the combined underlying diseases(except hypertension)between the two groups(P<0.05);The white blood cell,centroblast count,CRP,PCT,PT,D-dimer,total bilirubin,Urea,LDH,CK,MB,hs-TnI,CK-MB,NT-proBNP were higher in the DM group(P<0.05),and lymphocytes,single Nuclear cell,CD3,CD4,and CD8 counts were low(P<0.05).There was no statistically significant difference in platelet,IL-6,APTT,ALT,AST,Cr and CD19 counts between the two groups.Conclusion:Compared with COVID-19 patients with non-DM,COVID-19 patients with DM have a higher proportion of mortality and severe cases.Heart function,liver and kidney function,immune function damage and coagulation dysfunction are more obvious.Therefore,the monitoring of such patients should be strengthened,and active treatment should be performed to improve the prognosis.展开更多

Amino acid intake plays a crucial role in the Warburg effect of cancer.Gut microbes can regulate intestinal amino acid metabolism.However,it is still unknown whether probiotic therapy can protect the host from intesti...Amino acid intake plays a crucial role in the Warburg effect of cancer.Gut microbes can regulate intestinal amino acid metabolism.However,it is still unknown whether probiotic therapy can protect the host from intestinal tumor invasion by reducing amino acid intake.With in vitro methods,three acid-tolerant strains from fermented pickles were screened out.Using azoxymethane/dextran sulfate sodium-induced colon cancer models,we evaluated the therapeutic effects of Lactobacillus brevis CLB3,Lactobacillus plantarum XLP,and Lactobacillus johnsonii CM on model mice.Their functional mechanisms were further explained through anatomy section,quantitative reverse transcription polymerase chain reaction(qRT-PCR),Western blot,and immunohistochemical staining analyses as well as database mining and gut culturomics.The Lactobacillus brevis(L.brevis)CLB3 treatment significantly improved the clinical signs and symptoms of colon cancer,alleviated colon damage,and inhibited colon carcinogenesis in mice.In addition,this treatment significantly increased gut cultivable Lactobacillus abundance,inhibited the expression and translation levels of the tumor metabolism-related solute carrier(SLC)amino acid transporter including SLC7A5 and SLC7A11,lowered circulating interleukin-6(IL-6)and interleukin-17A(IL-17A)levels,and improved the accumulation of tumor-infiltrating lymphocytes and cancer proliferation factors.These findings suggest that L.brevis CLB3 can reduce amino acid transport,inhibit mammalian target of rapamycin(mTOR)signaling and enhance intestinal anti-tumor immune responses,which provides a potential targeting amino acid transporter strategy for preventing colorectal cancer.展开更多
