人参皂苷Rg3增加卵巢癌细胞对顺铂敏感性及机制研究

目的:探究人参皂苷Rg3对卵巢癌细胞SKOV3的增殖及顺铂增敏功能的效果及其作用机制。方法:CCK-8法测人参皂苷Rg3和顺铂对卵巢癌细胞的细胞活度;倒置显微镜观察细胞形态;划痕实验探究细胞迁移;克隆形成实验探讨人参皂苷Rg3对SKOV3细胞克... 目的:探究人参皂苷Rg3对卵巢癌细胞SKOV3的增殖及顺铂增敏功能的效果及其作用机制。方法:CCK-8法测人参皂苷Rg3和顺铂对卵巢癌细胞的细胞活度;倒置显微镜观察细胞形态;划痕实验探究细胞迁移;克隆形成实验探讨人参皂苷Rg3对SKOV3细胞克隆产生能力的作用;Westernblot实验探讨人参皂苷Rg3对SKOV3细胞PI3K/Akt/mTOR信号通路表达水平的效力。结果:人参皂苷Rg3对SKOV3细胞呈时间和剂量依赖性抑制,人参皂苷Rg3可增加SKOV3细胞对顺铂的敏感性。在镜下查看细胞,结果发现和单一使用DDP相比,加药后细胞变圆,凋亡细胞增多。克隆形成数据得出,顺铂的浓度抬升,SKOV3细胞的克隆形成数量显著削减,加入人参皂苷Rg3能进一步抑制;划痕实验实验结果可以显示,人参皂苷Rg3能够进一步抑制DDP作用下的卵巢癌细胞的迁移效果。Western blot探究数据看出,人参皂苷Rg3可使PI3K/Akt/mTOR信号通路蛋白表达量降低。结论:人参皂苷Rg3可控制SKOV3细胞的增殖和进展,可-1-www.ivypub.org/bf以促进细胞对顺铂的敏感性,对PI3K/Akt/mTOR信号通路表达具有抑制效果,并通过抑制该信号通路表达增加对顺铂的敏感性。 展开更多

BACKGROUND Acute urinary retention(AUR)is rare during pregnancy.CASE SUMMARY We report on three pregnant women with AUR between the 10^(th) and 18^(th) wk of gestation.Case 1 was first diagnosed as urinary tract infection and developed a urinary tract infection due to urinary retention caused by urethral obstruction.Case 2 had a history of previous abdominal surgery for pelvic tuberculosis,leading to severe adhesions and a persistent retroverted uterus.In case 3,healthcare providers focused on the patient’s gastrointestinal symptoms and did not investigate her inability to void.Case 1 required manual disimpaction of the uterus and the knee-chest position.The other cases required immediate catheterization.The condition resolved in cases 1 and 2;these patients had normal pregnancies.Case 3 had severe complications at the time of consultation,leading to an abortion.CONCLUSION Retroverted uterus is the most common cause of AUR.Prompt recognition and diagnosis are required.Clinicians should be aware of the risk factors,etiology,and clinical presentation of AUR in the first and second trimester of pregnancy. 展开更多
