目的:将来源于龟裂链霉菌(Streptomyces rimosus)M527的otr A基因在天蓝色链霉菌M145中异源表达,通过考察宿主菌对土霉素以及氨基糖苷类抗生素的抗性、放线紫红素的合成、菌株形态等方面的变化鉴定otr A基因的功能。创新点:S.rimosus中... 目的:将来源于龟裂链霉菌(Streptomyces rimosus)M527的otr A基因在天蓝色链霉菌M145中异源表达,通过考察宿主菌对土霉素以及氨基糖苷类抗生素的抗性、放线紫红素的合成、菌株形态等方面的变化鉴定otr A基因的功能。创新点:S.rimosus中的otrA基因与转录延伸因子EF-G具有很高的同源性,被认为是土霉素的抗性基因之一,但其具体的生物学功能目前尚未有研究报道。本文首次实现otrA基因在天蓝色链霉菌M145中的异源表达,不仅提高了宿主对土霉素以及氨基糖苷类抗生素的抗性,还促进了菌株产孢和放线紫红素的合成,从而初步证实了OTRA生物学功能。方法:克隆来源于S.rimosus M527的otrA基因,将其置于链霉菌整合型载体pIB139强启动子PermE*的下游,构建重组质粒pIB139-otrA;通过接合转移将其转入天蓝色链霉菌M145获得重组菌M145-OA,实现otrA基因在天蓝色链霉菌M145中的异源表达;通过扫描电镜观察菌株的形态变化;通过含有不同浓度的不同抗生素的抗性平板筛选测试菌株的抗性水平变化;通过5-L发酵罐发酵实验考察次级代谢产物放线紫红素的合成能力变化;通过荧光定量PCR考察放线紫红素合成途径中的调控基因actII-orf4的转录水平变化。结论:来源于S.rimosus M527的otrA基因在天蓝色链霉菌M145中实现异源表达。一方面对宿主天蓝色链霉菌形态分化、放线紫红素产量的影响表明otrA可能作为一种类似延伸因子的发挥重要作用;另一方面宿主对土霉素及氨基糖苷类抗生素抗性的提高可能归因于OTRA对核糖体的保护作用。 展开更多

A series of novel diaryl ureas containing 4-[(2-amino-6-trifluromethyl)pyrimidine-4-yl]piperazine-l-yl group were synthesized and evaluated for their cytotoxic activities in a panel of human cancer cell lines. Compared with the reference drug Sorafenib,some compounds showed more potent and a broader spectrum of anti-cancer activities.Among them,compound 2p demonstrated significant inhibitory activities against MDA-MB-231,HT-29 and MCF-7 cell lines with IC_(50) values of 0.016,0.63,0.001μmol/L, respectively. 展开更多

The planting of the vegetation on the floodplain helps the ecological restoration,which is the main form of the river’s ecological corridor.Therefore,the current research of the river dynamics focuses on the water movement under a compound channel with the vegetated floodplains.Two simulated vegetation species are selected in this paper for the flume simulation experiments of the floodplain vegetation,and the compound channel is divided into three subregions in the transverse direction.The Navier-Stokes equation and the eddy viscosity theory are applied to obtain the transverse distribution of the depth-averaged velocity and the results agree well with the experimental data.This paper proposes a new method based on the analytical solution of the flow velocity distribution to calculate the average flow velocity in each section.Calculation results can effectively simulate the average flow velocity of the measured sections.The description of the pollutant transport processes in a moving stream requires a refined determination of the dispersion coefficients in the compound channel.The process of the pollutant concentrations in each zone and the reasons for their occurrence are elucidated on the basis of the experimental results.Simultaneously,the measured values of the longitudinal dispersion coefficients are obtained by the“routing procedure,”and a two-zone model of the pollutant dispersion is constructed on the basis of the hydrodynamic study.The prediction method for the longitudinal dispersion coefficients is also presented.Applying the predicted and measured section average flow velocities to the two-zone model to predict the longitudinal dispersion coefficient,and the average relative errors are only 4.17%,7.15%,respectively.This result indicates that the two-zone model can effectively predict the longitudinal dispersion coefficients.The calculation methods for the longitudinal dispersion coefficients from—various studies are compared.The results reveal that the predicted values of these calculation methods are all larger than the measured values,indicating that the vegetation has a considerable influence on the dispersion process.This study comprehensively shows the dispersion features of the pollutants and provides a theoretical basis for the planning and the design of the vegetated ecological corridors. 展开更多

Based on our previous work,a series of hydrazone moiety-bearing aminopyrimidines were synthesized.The compounds were evaluated for inhibitory activities against EGFRT790M/L858 R and antiproliferative activities against H1975 and A549 NSCLC cell lines harboring different forms of EGFR.Compounds 7f and7 k exhibited potent and selective activity in inhibition of gefitinib-resistant H1975 cancer cells(IC50;0.45,0.2μmol/L) while were much less active on A549 cancer cells(IC50;52.83,〉100μmol/L).Both compounds could be served as promising lead compounds for further investigation. 展开更多
