目的探讨TAK-242对β-淀粉样蛋白25-35(Aβ_(25-35))诱导的大鼠海马神经元损伤的作用及其相关机制。方法用Aβ_(25-35)注射大鼠双侧海马复制阿尔兹海默病的动物模型,TAK-242腹腔注射进行治疗,Nissl染色观察大鼠海马CA3区神经元的形态和... 目的探讨TAK-242对β-淀粉样蛋白25-35(Aβ_(25-35))诱导的大鼠海马神经元损伤的作用及其相关机制。方法用Aβ_(25-35)注射大鼠双侧海马复制阿尔兹海默病的动物模型,TAK-242腹腔注射进行治疗,Nissl染色观察大鼠海马CA3区神经元的形态和数量,Western blot检测大鼠海马Toll样受体4(TLR4)和髓样分化因子88(My D88)蛋白的表达,酶联免疫吸附法检测大鼠海马白细胞介素1β(IL-1β)和肿瘤坏死因子α(TNF-α)表达水平。结果大鼠海马注射Aβ_(25-35)后引起大鼠海马CA3区神经元破坏和数量减少,而TAK-242可以抵抗Aβ_(25-35)所造成的神经毒性并保护海马神经元,同时TAK-242可降低Aβ_(25-35)所引起的海马组织内TLR4、My D88、IL-1β和TNF-α表达升高。结论 TAK-242可以通过抑制TLR4/My D88信号通路,降低炎症因子IL-1β和TNF-α水平,从而保护大鼠海马神经元抵抗Aβ_(25-35)诱导的神经毒性。 展开更多

Objective:To explore the effects of different MⅡstage oocytes zona pellucida birefringence on pregnancy outcome.Methods:A total of 46 couples with infertile which induced by single cause received in-vitro fertilization treatment were analyzed retrospectively,and randomly divided into the high zona birefringence(HZB)/HZB group.HZB/low zona birefringeuce(LZB) group and LZB/LZB group according to different oocytes zona pellucida birefringence.Intracytoplasmic sperm injection outcome was analyzed and compared.Results:The proportion of HZB oocytes, implantation rate and the pregnancy rate were decreased in three groups(HZB/HZB group】HZB/ LZB group】LZB/LZB group)(P【0.05).But there was no significantly different between the number of oocytes and fertilization rate of these groups(P】0.05).Logistic regression analysis showed that factors allecl MⅡstage oocytes zona pellucida birefringence were age.basal FSH level and the LH level on the day of HCG injection.Age and KSH levels were negatively correlated with the single oocyte zona pellucida birefringence:While the LH level on the day of hCC injection was positively correlated with the single oocylc zona pellucida birefringence.Conclusions:The primary influence factors on MⅡstage oocytes zona pellucida are age.basal FSH level and the LH level on the day of hCG injection.The birefringence value of zona pellucida can affect the pregnancy outcome. 展开更多

Objective To investigate the feasibility of vitrification of blastocysts following blastomere biopsy. Methods Among patients undergoing pre-implantation genetic diagnosis (PGD), artificial shrinkage of the blastocoelic cavity and subsequent vitrification of applicable surplus blastocysts after day-3 blastomere biopsy were performed. According to patient requirements, thawed blastocysts were transferred into patients due to pregnancy failure after fresh embryo transfer, ectopic pregnancy, ovarian hyperstimulation. Results Twenty-four PGD cycles were carried out. According to genetic diagnosis and the development of blastocysts, transfer was cancelled in 7 cycles due to absence of applicable embryos or ovarian hyperstimulation. In the remaining 17 cycles, 26 blastocysts were thawed and transferred, which resulted in 13 implanted (50.0%). Clinical pregnancies were observed in 11 patients (64.71%). Following transfer, 30 applicable blastocysts in 10 cycles were cryopreserved. Six patients received transfer of thawed blastocysts. All 8 thawed embryos survived and were transferred, and singleton pregnancies occurred in 5 patients. Two women delivered healthy infants and 3 pregnancies are ongoing. Conclusion Vitrification with artificial shrinkage is effective for preserving blastocysts following blastomere biopsy. 展开更多
