该研究通过综合分析对甘蔗(ROC 22)茎尖离体培养褐变不同条件因素的影响以及褐变细胞区室结构的变化,探讨了甘蔗茎尖离体培养褐变的机理机制。结果表明:不同芽位茎尖诱导成活率具有明显差异,随着芽位的增加,诱导成活率不断降低;不同季... 该研究通过综合分析对甘蔗(ROC 22)茎尖离体培养褐变不同条件因素的影响以及褐变细胞区室结构的变化,探讨了甘蔗茎尖离体培养褐变的机理机制。结果表明:不同芽位茎尖诱导成活率具有明显差异,随着芽位的增加,诱导成活率不断降低;不同季节取芽对外植体茎尖总酚类物质含量无明显影响;但不同芽位及不同催芽天数,外植体芽的总多酚含量明显不同,随着催芽天数的增加,不同芽位的多酚含量呈现由低升高的趋势;蔗芽在培养4周时多酚含量较低,适宜进行采芽接种培养;从褐变甘蔗茎尖的解剖结构变化分析,褐变甘蔗茎尖细胞离体培养初期细胞核结构出现变形,线粒体有肿胀拉长,部分液泡膜开始分解;中后期质壁分离更为严重,胞质中出现大量溶酶体,线粒体等细胞器全分解,细胞膜、液泡膜、核膜、线粒体膜的双层膜结构出现破损和缺口;而正常发育的茎尖细胞,能基本保持细胞核的形态结构,只有少量的溶酶体出现。因此,可以推测细胞核和线粒体结构变形以及膜系统的大量破损是甘蔗茎尖培养褐变死亡的原因。 展开更多

为优化咖啡生豆中绿原酸的提取工艺条件,采用超声波法、BBD中心组合实验和响应面法进行提取。从甲醇、乙醇、聚乙二醇200(PEG200)、聚乙二醇400(PEG400)和聚乙二醇600(PEG600)中优选出最佳提取剂为PEG400,最佳浓度为40%;通过单因素和BB... 为优化咖啡生豆中绿原酸的提取工艺条件,采用超声波法、BBD中心组合实验和响应面法进行提取。从甲醇、乙醇、聚乙二醇200(PEG200)、聚乙二醇400(PEG400)和聚乙二醇600(PEG600)中优选出最佳提取剂为PEG400,最佳浓度为40%;通过单因素和BBD实验考察了3个因素的影响程度,经实验确定最佳工艺条件为:固液比1∶28(g/mL)、提取时间50min、提取温度60℃,绿原酸得率达到13.68mg/g,与响应曲面拟合所得方程的预测值13.6986mg/g符合良好,此模型合理可靠。这3个影响因素的主次顺序为固液比>提取温度>提取时间。 展开更多

In order to study the molecular mechanism of injury in rat organs induced by methylmercury, and the relationship between neurotransmitter and oxidative damage in the toxicity process of rat injury by methylmercury was studied. The control group was physiological saline of 0.9%, the concentration of exposure groups were 5 mg/(kg5d) and 10 mg/(kg5d) respectively. The content of AChE, ACh, NOS, NO, MDA, SOD, GSH-Px and GSH in different organs of rats were determined with conventional methods. The results showed that after exposure to methylmercury for 7 d, the mercury content in brain of exposure groups increased clearly and had significant difference compared with the control group(P<0.01). In rat's brain, serum, liver and kidney, the content of ACh and AChE were all decreased; the content of NOS and NO were all increased; the content of MDA was increased compared with the control group, the exposure groups had significant difference (P<0.01); the content of SOD, GSH and GSH-Px was decreased compared with the control group, the exposure groups had significant difference(P<0.01). It could be concluded that methylmercury did effect the change of neurotransmitter and free radical. They participated in the toxicity process of injury by methylmercury. The damage of neurotransmitter maybe cause the chaos of free radical and the chaos of free radical may also do more damage to neurotransmitter vice versa. 展开更多

The neurotransmitter level changes of ducks exposed 8-month in a mercury-polluted site(Wanshan, China) and a reference site(Shanghai, China) were examined. Chemical analyses showed both higher mercury and selenium concentrations in the organ of Wanshan ducks. An increased content of acetylcholine(ACh) in brain and blood and a decreased activity of acetylcholinesterase(AChE) in blood were observed. Moreover, there was an increasing trend for nitric oxide synthase(NOS) activity and nitric oxide(NO) production in duck brain, but a reduction of NOS activity in duck serum. The possible explanations were due to the interactive effect of selenium accumulation and the sublethal exposure level of mercury in Wanshan area. The present study showed that AChE and NOS were sensitive to mercury contamination of real circumstance, suggesting that these two indexes have the potential to be biomarkers in assessment of health effects by mercury contamination. 展开更多

目的:研究心理干预对社区高血压患者血压及焦虑抑郁情绪的影响,探索在社区中对高血压患者进行心理干预的实用方法和模式。方法:在北京西城区选择由二级以上医院确诊的原发性高血压患者,分级为轻中度高血压患者,年龄40~75岁,干预组80人,... 目的:研究心理干预对社区高血压患者血压及焦虑抑郁情绪的影响,探索在社区中对高血压患者进行心理干预的实用方法和模式。方法:在北京西城区选择由二级以上医院确诊的原发性高血压患者,分级为轻中度高血压患者,年龄40~75岁,干预组80人,对照组80人,两组患者均自由服用抗高血压药物。两组患者都以抑郁自评量表、焦虑自评量表进行心理测量;研究起始测量血压,并进行慢性病行为危险因素和健康知识问卷调查;干预组每月进行两次心理干预讲座或活动,连续干预8个月,对照组不进行任何干预。8个月后分别对两组患者进行抑郁和焦虑评分,测量血压。结果:干预组干预前后相比,焦虑分数差异具有统计学意义(43.57±11.57 vs 39.73±8.07,t=2.31,P=0.02),抑郁分数差异亦具有统计学意义(0.61±0.16 vs0.50±0.13,t=4.14,P=0.00),收缩压差异具有统计学意义(138.71±13.77 vs 132.49±14.38(mm Hg),t=4.34,P=0.00),舒张压的差异亦具有统计学意义(77.68±9.25 vs 74.71±9.54(mm Hg),t=2.64,P=0.01)。而对照组研究前后比较,焦虑分数、抑郁分数、收缩压、舒张压差异都没有统计学意义。结论:心理干预可以缓解高血压患者的焦虑抑郁情绪,并对高血压患者降低血压有显著的效果,可以作为社区高血压患者的辅助治疗手段。 展开更多

Enzymatic synthesis of cefaclor from 7-aminodesacetoxymethyl-3-chlorocephalosporainc acid(6-ACCA) and phenylglycine derivatives using penicillin G acylase was studied .Many factors that affect the conversion of 7-ACCA to cefaclor were examined.The immobilized enzyme from Bacillis megaterium gave a better catalytic properties and the higher conversion was obtained using phenylglycine methyl ester(PGME) as acyl donor.And the external mass transfer limitation could be eliminated when the stirring rate was more than 150r/min.Low temperature was beneficial for the synthesis and the results showed that the synthetase activity was hardly influenced by temperature while the amidase activity was affected greatly by temperature.The optimum reaction conditions were determined at pH 6.5 and 10℃,respectively.The best 7-ACCA conversion of 56% was achieved when the intial concentration of 7-ACCA and PGME was at 50 mM and 150mM,respectively. 展开更多

To investigate the significance of the SARS-associated coronavirus (SARS-CoV) antibody,detected by ELISA and indirect immunofluorescence assays (IFA) for the SARS-CoV Vero E6 cell lysates,in non-SARS subjects, 114 serum samples from healthy controls and 104 serum specimens from autoimmune disease patients were collected.The results of ELISA showed that among 114 sera from healthy controls,4 (3.5%) were positive of SARS-CoV-IgG antibody and 114 (100%) were all negative of SARS-CoV-IgM antibody;the specificity of SARS-CoV-IgG antibody for SARS patients was 96.5%,but the specificity of both SARS-CoV-IgG and -IgM antibodies for SARS patients was 100%.In 58 cases with SLE,positive rates of SARS-CoV-IgG and -IgM antibodies were 32.8% (19/58) and 8.6% (5/58),respectively,in which 11 cases (19%) were positive of both SARS-CoV-IgG and -IgM antibodies;in 10 cases with SS,positive rate of both SARS-CoV-IgG and -IgM antibodies was 10% (1/10);in 16 cases with MCTD,positive rate of SARS-CoV-IgG was 37.5% (6/16),positive rate of both SARS-CoV-IgG and -IgM antibodies was 6.3% (1/16);in 20 cases with RA,one case was positive (5%) of SARS-CoV-IgG.However,of all samples with positive SARS-CoV-IgG and -IgM antibodies for autoimmune diseases and healthy controls,SARS-CoV RNA and antibodies were all negative by RT-PCR and IFA.All sera for negative or positive ELISA results were also negative or positive results using ELISA with Vero E6 cells lysates.These studies showed that SARS-CoV Vero E6 cell lysates for the ELISA to detect SARS-CoV antibodies could lead to the false-positive reactions or cross-reactions of SARS-CoV antibodies in non-SARS diseases and healthy controls,and the false-positive reactions or cross-reactions were related to Vero E6 cell lysates and autoantibodies in non-SARS population.Cellular & Molecular Immunology.2004;1(4): 304-307. 展开更多

Based on generalized characteristic polynomials andprojective transformations, algebraic conditions are derived for detecting the various positionalrelationships between two planar conies, namely, outer separation, exterior contact, intersection,interior contact and inclusion. Then the results are applied to detecting the positionalrelationships between a cylinder (or a cone) and a quadric. The criteria is very effective andeasier to use than other known methods. 展开更多
