Background:Glucocorticoid receptor(GR)mediated corticosterone-induced fatty liver syndrome(FLS)in the chicken by transactivation of Fat mass and obesity associated gene(FTO),leading to demethylation of N6-methyladenosine(m^(6)A)and post-transcriptional activation of lipogenic genes.Nutrition is considered the main cause of FLS in the modern poultry industry.Therefore,this study was aimed to investigate whether GR and m^(6)A modification are involved in high-energy and low protein(HELP)diet-induced FLS in laying hens,and if true,what specific m^(6)A sites of lipogenic genes are modified and how GR mediates m^(6)A-dependent lipogenic gene activation in HELP diet-induced FLS in the chicken.Results:Laying hens fed HELP diet exhibit excess(P<0.05)lipid accumulation and lipogenic genes activation in the liver,which is associated with significantly increased(P<0.05)GR expression that coincided with global m^(6)A demethylation.Concurrently,the m^(6)A demethylase FTO is upregulated(P<0.05),whereas the m^(6)A reader YTHDF2 is downregulated(P<0.05)in the liver of FLS chickens.Further analysis identifies site-specific demethylation(P<0.05)of m^(6)A in the mRNA of lipogenic genes,including FASN,SREBP1 and SCD.Moreover,GR binding to the promoter of FTO gene is highly enriched(P<0.05),while GR binding to the promoter of YTHDF2 gene is diminished(P<0.05).Conclusions:These results implicate a possible role of GR-mediated transcriptional regulation of m^(6)A metabolic genes on m^(6)A-depenent post-transcriptional activation of lipogenic genes and shed new light in the molecular mechanism of FLS etiology in the chicken. 展开更多

Assessment of complete lymph node dissection in patients with melanoma:A systemic review and meta-analysis

Background: Complete lymph node dissection(CLND) for patients with melanoma remains controversial. This meta-analysis aimed to compare the prognoses and complications between the CLND and control groups(patients who receive adjuvant treatment or observation only) in patients with sentinel lymph node(SLN)-positive melanoma.Methods: The Pub Med, Embase, Cochrane, and Web of Science databases were searched for cohort studies and randomized clinical trials(RCTs) conducted between 1964 and 2022, and the quality of the studies was assessed using the Cochrane risk-of-bias tool and Newcastle-Ottawa Scale. Hazard ratios(HR) or risk ratios(RR) with 95%confidence intervals(CIs) were calculated for each outcome. Heterogeneity and sensitivity tests were also conducted, and publication bias tests were performed when the pooled number of studies was >10.Results: Fifteen studies, including 11 cohort studies and 4 RCTs, were enrolled and assessed for quality. Analysis of overall survival showed no significant difference between the CLND and control groups(HR=1.02, 95% CI:0.69–1.51, P=0.922). Similarly, recurrence-free survival(HR=0.84, 95% CI: 0.6–1.16, P=0.287), disease-free survival(HR=1.06, 95% CI: 0.65–1.72, P=0.82), and disease-specific survival(HR=0.84, 95% CI: 0.59–1.21,P=0.355) showed no difference between the two groups. CLND did not reduce the risk of recurrence(RR=0.98,95% CI: 0.8–1.2, P=0.851).Conclusion: Remarkably, patients who underwent CLND were more likely to have complications such as flap necrosis and lymphedema than the controls. CLND does not improve patient prognosis and may increase the incidence of complications. 展开更多

Anthocyanins are important natural plant pigments and play diverse roles in plant growth and adaptation.Anthocyanins function as screens to protect photosynthetic tissues from photoinhibition.However,the regulatory mechanisms underlying the biosynthesis and spatial accumulation pattern of anthocyanins remain some unresolved issues.Here,we demonstrate that the GARP-type transcription factor GOLDEN2-LIKE 1(GLK1)functions as a positive factor in anthocyanin accumulation.GLK1 enhances the transcriptional activation activities of MYB75,MYB90,and MYB113 via direct proteinprotein interactions to increase the expression of anthocyanin-specific biosynthetic genes.Anthocyanins accumulate in an acropetal manner in Arabidopsis.We also found that the expression pattern of GLK1 overall mimicked the accumulation pattern of anthocyanin from the base of the main stem to the shoot apex.Based on these findings,we established a working model for the role of GLK1 in anthocyanin accumulation and propose that GLK1mediates the spatial distribution pattern of anthocyanins by affecting the transcriptional activation activities of MYB75,MYB90,and MYB113. 展开更多

The expanding development of the world economy has triggered various approaches to address the impending environmental problems,among which docximentary serves as one of the most effective and attractive ways to cultivate eco-harmony awareness.This article aims to probe how attitudinal resources are being distributed and what Sino-British ideologies have conveyed in the Chinese documentary,Wild China,and the British documentary,Climate Change:The Facts,under the framework of appraisal theory with the help of UAM CorpusTool 3.3.The results from this study indicate that Wild China tends to employ more dis/inclination resources to show that China is willful to collaborate with other countries for environmental protection.On the other hand,the United Kingdom(UK)tends to adopt dis/satisfaction resources to explicitly or implicitly convey their disagreement with the promotion of the economy under the cost of the environment.The appraisal resources employed by both documentaries convey the ideology of opposing humans5 over-exploitation of natural resources.The findings of this study have both theoretical and practical implications in enriching the application of appraisal theory on the analysis of ecological discourse and further encourage the world to take action in order to prevent the over-exploitation of natural resources. 展开更多

This article discusses about the cultivation of foreign language talents,through which it can be appreciated that the emphasis is to be laid on improving their comprehensive qualities.Only in this way can foreign language talents adapt to the needs of the market,integrate into the ranks of technical personnel in the field of engineering,and be warmly welcomed by employers or employing units.Nevertheless,in order to achieve this aim,three factors should be overcome to avoid the hindrance faced by technical colleges and universities from cultivating foreign language talents with comprehensive qualities.This article probes into the three negative factors:first,students are not fluent in their mother tongue;second,foreign language majors lack basic knowledge of science and technology so much so that they are not able to do well in such translation;third,they lack the ability to perform simultaneous interpretation Simultaneous interpreting.In regard to those factors,this article then provides several suggestions as countermeasures to those three negative factors,so as to cultivate talents that can satisfy the needs of the market. 展开更多

Effect of Mesoscale Land Use Change on Characteristics of Convective Boundary Layer:Semi-Idealized Large Eddy Simulations over Northwest China 被引量:4

Although large-scale topography and land use have been properly considered in weather and climate models, the effect of mesoscale and microscale heterogeneous land use on convective boundary layer(CBL) has not been fully understood yet. In this study, the influence of semi-idealized strip-like patches of oases and deserts, which resemble irrigated land use in Northwest China, on the CBL characteristics, is investigated based on the Weather Research and Forecasting(WRF)-large eddy simulation(LES) driven by observed land surface data. The influences of soil water content in oases on aloft CBL flow structure, stability, turbulent kinetic energy(TKE), and vertical fluxes are carefully examined through a group of sensitivity experiments. The results show that secondary circulation(SC)/turbulent organized structures(TOS) is the strongest/weakest when soil water content in oases is close to saturation(e.g.,when the oases are irrigated). With the decrease of soil water content in oases(i.e., after irrigation), SC(TOS) becomes weak(strong) in the lower and middle CBL, the flux induced by SC and TOS becomes small(large), which has a dramatic impact on point measurement of eddy covariance(EC) fluxes. The flux induced by SC and TOS has little influence on EC sensible heat flux, but great influence on EC latent heat flux. Under this circumstance, the area averaged heat flux cannot be represented by point measurement of flux by the EC method, especially just after irrigation in oases. Comparison of imbalance ratio(i.e., contribution of SC and TOS to the total flux) reveals that increased soil moisture in oases leads to a larger imbalance ratio as well as enhanced surface heterogeneity. Moreover,we found that the soil layer configuration at different depths has a negligible impact on the CBL flux properties. 展开更多

配置管理是支撑云服务提供商管理大规模容器集群的重要基础设施.这一规模的集群通常包含百万量级的容器实例,如何根据不同业务场景的需求,及时可靠地将配置更新推送至对其感兴趣的容器实例,是亟待解决的关键问题.然而,现有方法仍存在不... 配置管理是支撑云服务提供商管理大规模容器集群的重要基础设施.这一规模的集群通常包含百万量级的容器实例,如何根据不同业务场景的需求,及时可靠地将配置更新推送至对其感兴趣的容器实例,是亟待解决的关键问题.然而,现有方法仍存在不足之处.保障顺序一致性的共识算法限制了集群的扩展能力,难以适用于大规模集群.反熵算法存在长尾现象,时延难以保障,不适用于推送关键的配置更新.为了应对上述挑战,本文提出了一种面向大规模集群的柔性配置更新推送方法.这一方法基于发布/订阅机制,引入基于完全N叉树拓扑的可定义的多层次推送,并使用订阅者的部分计算资源协助推送,以提升推送性能;引入容错机制以应对节点失效和网络分区,保障网络分区时多分区读写可用.方法的拓扑参数和策略可根据业务场景对性能、可靠性等维度的不同需求而调整.实验结果表明,与现有的方法相比,我们的方法可以有效降低更新推送的时延,并可有效应对节点失效和网络分区场景. 展开更多

本研究收集了2014年1月1日—2018年12月31日甘肃省妇幼保健院和2015年1月1日—2018年12月31日天水市第一医院儿童腹泻病门诊数据, 以标准化降水指数(SPI)和气象干旱综合指数(MCI)作为干旱指标, 采用quasi-Poisson分布广义相加模型分析... 本研究收集了2014年1月1日—2018年12月31日甘肃省妇幼保健院和2015年1月1日—2018年12月31日天水市第一医院儿童腹泻病门诊数据, 以标准化降水指数(SPI)和气象干旱综合指数(MCI)作为干旱指标, 采用quasi-Poisson分布广义相加模型分析干旱与儿童腹泻门诊量的关联情况。研究期间, 兰州市和天水市干旱天数依据SPI-1分别为298和379 d, 依据MCI分别为303和398 d;甘肃省妇幼保健院和天水市第一医院0~6岁儿童腹泻门诊量分别57 147和18 703例。基于月降水量的SPI(SPI-1)和MCI与儿童腹泻门诊量呈负相关。与非干旱时期相比, 依据SPI-1划分, 中旱与儿童腹泻病门诊量关联性最强, 兰州市和天水市儿童腹泻病门诊量分别增加了13.4%(95%CI:7.9%~19.3%)和20.0%(95%CI:12.7%~27.8%);依据MCI划分, 特旱导致天水儿童腹泻门诊量增加60.5%(95%CI:3.4%~149.0%)。 展开更多

Herein we provide a novel high-efficiency nanocomposite for bacterial capture based on mixed metal oxides(MMOs)with deleterious chromium properties.With both the layer structure of layered double hydroxides(LDHs)and the magnetic properties of Fe,MMOs enrich the location of ionic forms on the surface,providing a good carrier for adsorption of the heavy metal Cr(VI).The capacity for adsorption of Cr(VI)by MMOs can be as high as 98.80 mg/g.The prepared Cr(VI)-MMOs achieved extremely expeditious location of gram-negative antibiotic-resistant E.coliNDM-1 by identifying lipid bilayers.Cr-MMOs with a Cr loading of 19.70 mg/g had the best bactericidal effect,and the concentration of E.coliNDM-1 was decreased from~10^(8) to~10^(3) CFU/mL after 30 min of reaction.The binding of nitrogen and phosphorus hydrophilic groups to chromate generated realistic models for density functional theory(DFT)calculations.The specific selectivity of MMOs toward bacterial cells was improved by taking Cr(VI)as a transferable medium,thereby enhancing the antibacterial activity of Cr-MMOs.Under the combined action of chemical and physical reactions,Cr(VI)-MMOs achieved high capacity for inactivation of bacteria.Moreover,the metallic elements ratio in Cr-MMOs remained stable in their initial valence states after inactivation.This guaranteed high removal efficiency for both heavy metals and bacteria,allowing recycling of the adsorbent in practical applications. 展开更多
