Thus the development of highly sele...Bisulfite(HSO_3) is an important sulfur dioxide(SO_2) derivative, which plays a major role in many physiological processes and is also closely associated with a variety of diseases. Thus the development of highly selective and sensitive fluorescent probes is essential to detect HSO_3 in living cells. In this work,we report the synthesis and analysis of a ratiometric fluorescent probe for selective detection of HSO_3 based on the 1,4-nucleophilic addition reaction with the carbazole as an electron donor(D) and aldehyde group as an electron acceptor(A). The addition of HSO_3 and other ions to our probe can be observed by UV vis and fluorescence spectrometry. Our investigation proved that the probe is highly selective and sensitive for HSO_3 and ratiometric changes. Moreover, the probe has good cell permeability and was successfully applied to the detection of exogenous HSO_3 in Hela cells and C. elegans.展开更多
We constructed a reaction-based near-infrared fluorescent probe(Niap) to specifically identify alkaline phosphatase(ALP) with fast red fluorescence enhancement.Based on the positive concentrationdependent manner betwe...We constructed a reaction-based near-infrared fluorescent probe(Niap) to specifically identify alkaline phosphatase(ALP) with fast red fluorescence enhancement.Based on the positive concentrationdependent manner between the fluorescent intensity of the Niap and ALP,probe Niap was used to study the ALP enrichment and variation in golden apple snails(Pomacea canaliculata) exposed to the molluscicide candidate PPU06.After treatment with different concentrations of PPU06 over various times,three organs of the surviving snails,liver,stomach and plantaris,were frozen and sectioned for fluorescent imaging experiments.With increased PPU06 concentration,red fluorescence substantially increased in the liver and reached a maximum within 24 h when the PPU06 co ncentration was 0.75 mg/L.No obvious changes in the stomach or foot plantaris were found.It showed PPU06 caused liver injury and stimulated the increase of ALP in the liver of P.canaliculata.This study demonstrates a rapid ALP fluorescent identification method that can be used to study the effects of PPU06 on P.canaliculata.It also provides optical evidence that may aid in the discovery of new chemistry for snail control.展开更多

Dear Editor,Small-cell lung cancer(SCLC),one of the most-deadly malignant tumors,accounts for∼15%of all lung cancers.1,2 Although highly responsive to the standard chemoradiotherapy,the recurrence rate of SCLC is nea...Dear Editor,Small-cell lung cancer(SCLC),one of the most-deadly malignant tumors,accounts for∼15%of all lung cancers.1,2 Although highly responsive to the standard chemoradiotherapy,the recurrence rate of SCLC is nearly unity,with median survival time ranging from 2–6 months.Almost all SCLC patients are inoperable,leading to the difficulty in obtaining adequate biopsy tissue for research.3 Consequently,there is an urgent need to find noninvasive biomarkers to predict the risk and provide early diagnosis of SCLC.We aimed to identify potential epigenomic markers for SCLC development in blood DNA and reveal differentially methylated genes through a multi-omics approach,from genome-wide DNA methylation profiling,followed by pathway enrichment analyses and target gene validation utilizing Gene Expression Omnibus(GEO)and Oncomine microarray databases,to TCGA PanCancer cohorts.It is known that virtually all patients with SCLC are victims of tobacco smoking,4 but it is unknown whether stopping smoking before SCLC diagnosis would influence the epigenetic biomarkers under investigation.We here report a study that identified aberrantly methylated genes between current and former smokers among SCLC patients,revealing a set of candidate biomarkers in peripheral blood DNA for better stratifying patients with high risk.展开更多
