Background Global warming leading to heat stress(HS)is becoming a major challenge for broiler production.This study aimed to explore the protective effects of seaweed(Enteromorpha prolifera)polysaccharides(EPS)on the ...Background Global warming leading to heat stress(HS)is becoming a major challenge for broiler production.This study aimed to explore the protective effects of seaweed(Enteromorpha prolifera)polysaccharides(EPS)on the intestinal barrier function,microbial ecology,and performance of broilers under HS.A total of 144 yellow-feathered broilers(male,56 days old)with 682.59±7.38 g were randomly assigned to 3 groups:1)TN(thermal neutral zone,23.6±1.8℃),2)HS(heat stress,33.2±1.5℃ for 10 h/d),and 3)HSE(HS+0.1%EPS).Each group contained 6 replicates with 8 broilers per replicate.The study was conducted for 4 weeks;feed intake and body weights were measured at the end of weeks 2 and 4.At the end of the feeding trial,small intestine samples were collected for histomorphology,antioxidant,secretory immunoglobulin A(s Ig A)content,apoptosis,gene and protein expression analysis;cecal contents were also collected for microbiota analysis based on 16S r DNA sequencing.Results Dietary EPS promoted the average daily gain(ADG)of broilers during 3–4 weeks of HS(P<0.05).At the end of HS on broilers,the activity of total superoxide dismutase(T-SOD),glutathione S-transferase(GST),and the content of s Ig A in jejunum were improved by EPS supplementation(P<0.05).Besides,dietary EPS reduced the epithelial cell apoptosis of jejunum and ileum in heat-stressed broilers(P<0.05).Addition of EPS in HS group broilers'diet upregulated the relative m RNA expression of Occludin,ZO-1,γ-GCLc and IL-10 of the jejunum(P<0.05),whereas downregulated the relative m RNA expression of NF-κB p65,TNF-αand IL-1βof the jejunum(P<0.05).Dietary EPS increased the protein expression of Occludin and ZO-1,whereas it reduced the protein expression of NF-κB p65 and MLCK(P<0.01)and tended to decrease the protein expression of TNF-α(P=0.094)in heat-stressed broilers.Furthermore,the proportions of Bacteroides and Oscillospira among the three groups were positively associated with jejunal apoptosis and pro-inflammatory cytokine expression(P<0.05)and negatively correlated with jejunal Occludin level(P<0.05).However,the proportions of Lactobacillus,Barnesiella,Subdoligranulum,Megasphaera,Collinsella,and Blautia among the three groups were positively related to ADG(P<0.05).Conclusions EPS can be used as a feed additive in yellow-feathered broilers.It effectively improves growth performance and alleviates HS-induced intestinal injury by relieving inflammatory damage and improving the tight junction proteins expression.These beneficial effects may be related to inhibiting NF-κB/MLCK signaling pathway activation and regulation of cecal microbiota.展开更多
Due to the limitations of a priori knowledge and convolution operation,the existing image restoration techniques cannot be directly applied to the cultural relics mural restoration,in order to more accurately restore ...Due to the limitations of a priori knowledge and convolution operation,the existing image restoration techniques cannot be directly applied to the cultural relics mural restoration,in order to more accurately restore the original appearance of the cultural relics mural images,an image restoration based on the denoising diffusion probability model(Denoising Diffusion Probability Model(DDPM))and the Transformer method.The process involves two steps:in the first step,the damaged mural image is firstly utilized as the condition to generate the noise image,using the time,condition and noise image patch as the inputs to the noise prediction network,capturing the global dependencies in the input sequence through the multi-attentionmechanismof the input sequence and feedforward neural network processing,and designing a long skip connection between the shallow and deep layers in the Transformer blocks between the shallow and deep layers using long skip connections to fuse the feature information of global and local outputs to maintain the overall consistency of the restoration results;In the second step,taking the noisy image as a condition to direct the diffusion model to back sample to generate the restored image.Experiment results show that the PSNR and SSIM of the proposedmethod are improved by 2%to 9%and 1%to 3.3%,respectively,which are compared to the comparison methods.This study proposed synthesizes the advantages of the diffusionmodel and deep learningmodel to make themural restoration results more accurate.展开更多

Objective: To investigate the impacts of mild hypothermia on lipopolysaccharide (LPS)-mediated TLR4/NF-κB signaling pathway in microglia. Method: The cultured microglia cells in vitro were divided into the NS group and the LPS group at 33?C and 37?C, respectively;quantitative RT-PCR was performed to detect the expressions of TLR4 and NF-κB mRNA in the microglia, Western blot was used to detect the expressions of TLR4 and NF-κB protein in the microglia, and ELISA was performed to detect the levels of tumor necrosis factor α (TNF-α) and interleukin-10 (IL-10) in the culture medium. Terpenes are the largest and most diverse class of plant specialized metabolites.Sesterterpenes(C25),which are derived from the plastid methylerythritol phosphate pathway,were recently characterized in plants.In Arabi...Terpenes are the largest and most diverse class of plant specialized metabolites.Sesterterpenes(C25),which are derived from the plastid methylerythritol phosphate pathway,were recently characterized in plants.In Arabidopsis thaliana,four genes encoding geranylfarnesyl diphosphate synthase(GFPPS)(AtGFPPS1 to 4)are responsible for the production of GFPP,which is the common precursor for sesterterpene biosynthesis.However,the interplay between sesterterpenes and other known terpenes remain elusive.Here,we first provide genetic evidence to demonstrate that GFPPSs are responsible for sesterterpene production in Arabidopsis.Blockage of the sesterterpene pathway at the GFPPS step increased the production of geranylgeranyl diphosphate(GGPP)-derived terpenes.Interestingly,co-expression of sester TPSs in GFPPSOE(overexpression)plants rescued the phenotypic changes of GFPPS-OE plants by restoring the endogenous GGPP.We further demonstrated that,in addition to precursor(DMAPP/IPP)competition by GFPPS and GGPP synthase(GGPPS)in plastids,GFPPS directly decreased the activity of GGPPS through protein-protein interaction,ultimately leading to GGPP deficiency in planta.Our study provides a new regulatory mechanism of the plastidial terpenoid network in plant cells.展开更多

The phenol-TCT derivatives were readily prepared using grinding method in short time without further purification. This catalytic system allowed the facile C-O cleavage of phenol-TCT derivatives under mild reaction conditions with high efficiency and good functional group tolerance. Gram-scale reaction was also achieved. Particularly, sequential functionalization of phenol-TCT derivatives followed by C-O bond reduction could also be realized,affording the high value-added products in moderate to good yields.展开更多
Aims Despite the importance of stem cO_(2)efflux(Es)in ecosystem carbon cycling and energy balance,little is known about temporal variation in the temperature coefficient(Q10)and sapwood nitrogen concen-trations([N])a...Aims Despite the importance of stem cO_(2)efflux(Es)in ecosystem carbon cycling and energy balance,little is known about temporal variation in the temperature coefficient(Q10)and sapwood nitrogen concen-trations([N])and their intrinsic links with Es.The objectives of this study were:(i)to examine the response of Es to temperature in a subalpine region and(ii)to explore the influence of stem diameter and[N]on Es.Also,we will test the hypothesis that(i)Es in trees has thermal acclimation and(ii)Es will be well correlated with diameter at breast height(DBH)and[N].Methods Here,a horizontally oriented soil chamber technique was applied to measure Es of Abies fabri in two subalpine forest stands in Southwestern china from May to December 2014.We also exam-ined the variability in Es,Q10 and[N]in trees and monitored the relationship between temperature,[N],DBH and Es.Important Findings During the measurement period,Es showed an apparent seasonal trend,following the change in air temperature,increasing from May and peaking in July,then continuously decreasing until December.The mean Es for the growing and dormant seasons were 1.45 and 0.25μmol·m^(−2)·s^(−1),respectively,and Es in the mature forest was sig-nificantly higher than in the immature forest.The area-based Es was positively correlated with DBH and sapwood width(SW),while volume-based Es showed negative relationship with DBH and SW.Across the five diameter classes,69.8-89.0%of the variation in Es could be explained by air temperature.The temperature sensitiv-ity(Q10)of Es ranged from 2.98 to 5.61 during the measurement period,with a higher Q10 appearing in the growing season than in the dormant season.There was a significant linear relationship(P<0.01)between[N]and Es(expressed based on two different units).Additionally,exponential models of Es against[N]and air temperature were applied to estimate Es.展开更多
