p53 is a key transcription factor to regulate gene transcription.However,the molecular mechanism of chromatin-associated p53 on gene transcription remains elusive.Here,using unbiased protein affinity purification,we f... p53 is a key transcription factor to regulate gene transcription.However,the molecular mechanism of chromatin-associated p53 on gene transcription remains elusive.Here,using unbiased protein affinity purification,we found that the RNF20/40 complex associated with p53 on the chromatin.Further analyses indicated that p53 mediated the recruitment of the RNF20/40 complex to p53 target gene loci including p21 and PUMA loci and regulated the transcription of p21 and PUMA via the RNF20/40 complex-dependent histone H2B ubiquitination(ubH2B).Lacking the RNF20/40 complex suppressed not only ubH2B but also the generation of the mature mRNA of p21 and PUMA.Moreover,ubH2B was recognized by the ubiquitin-binding motif of pre-mRNA processing splicing factor 8(PRPF8),a subunit in the spliceosome,and PRPF8 was required for the maturation of the mRNA of p21 and PUMA.Our study unveils a novel p53-dependent pathway that regulates mRNA splicing for tumor suppression. 展开更多

An eight-node solid-beam element based on absolute nodal coordinate formulation(ANCF)which uses cubic interpolation at the longitudinal direction and linear at the transverse direction is proposed.The element can accurately discretize the geometry represented by the Bézier volume with the same basis function order.The continuity property of the proposed beam element is verified.Advantages of the proposed element can be found in the application of leaf spring modeling.The parabolically varying thickness of the leaf can be accurately described.Components in leaf spring such as the spring eye and the rubber bushing can be assembled efficiently since the proposed element can be connected at any direction.The ANCF reference node is used to represent the rigid components and the revolute joints in the vehicle suspension system.Static I-shaped cross-section cantilever beam and flexible pendulum model are used to test the accuracy and dynamic performance of the proposed solid-beam element.The leaf spring model shows the flexibility of the element in modeling the complex mechanical system and the balance between the accuracy and the efficiency. 展开更多

At present,current filters can basically solve the filtering problem in target tracking,but there are still many problems such as too many filtering variants,too many filtering forms,loosely coupled with the target motion model,and so on.To solve the above problems,we carry out crossapplication research of artificial intelligence theory and methods in the field of tracking filters.We firstly analyze the computation graphs of typical a-βand Kalman.Through analysis,it is concluded that a-βand Kalman have the same computation structures analogous to a typical recurrent neural network and can be considered as a kind of recurrent neural network with constrained weights.Then,given this and considering that a recurrent neural network has the recognition capability for target motion patterns,a new filter is developed in a unified neural network architecture and specifically constructed using feedforward neural network,recurrent neural network,and attention mechanism.And the unified tracking filter proposed in this paper can generate three aspects of unity:a unified target motion model,an adaptive filter method,and an overall track filtering framework.Finally,Simulation results show that the proposed filter is effective and useful,of which the overall performance is superior to those of compared filters. 展开更多
