Analysis of the Effect of GINS4 Regarding the Proliferation and Invasion of Breast Cancer Cells

Objective: To analyze the role and influence of the GINS4 gene in breast cancer progression and to explore its expression in triple-negative and non-triple-negative breast cancer cell lines. Methods: Single-gene analysis of GINS4 was performed by breast cancer RNA transcriptome data from The Cancer Genome Atlas (TCGA). Real-time quantitative polymerase chain reaction (PCR) was used to detect the expression of GINS4 in triple-negative and non-triple-negative breast cancer cell lines. The knockdown effects of GINS4 in MDA-MB-231 and MCF-7 cell lines on the proliferation and invasion of breast cancer cells were examined by cell counting kit 8 (CCK8) and Transwell assays. Results: Bioinformatics analysis showed that the expression of GINS4 in breast cancer was significantly higher than that in normal breast tissues (P > 0.05). At the same time, cell experiments confirmed that GINS4 was highly expressed in human breast cancer cell lines with normal breast cells as reference and in MDA-MB-231 and MCF-7 cell lines as reference, where the ability of proliferation and invasion of MDA-MB-231 and MCF-7 cells decreased after GINS4 knockdown. Conclusion: GINS4 is a gene associated with breast cancer malignancy, which can act as a novel tumor marker and has the potential as a new therapeutic target for breast cancer. 展开更多

Gaudich.) and the tolerance response in ramie. [Methods] A pot experiment was conducted to study the effects of Sb stress on root growth and Sb accumulation and transport of the root system of cultivated ramie Zhongzhu No.1, as well as on the physiological characteristics of ramie leaves. [Results] The plant height and root dry weight and volume of Zhongzhu No.1 showed an effect of "promoting at low concentrations and inhibiting at high concentrations" with the increase of Sb concentration, and decreased significantly at the concentration of 4 000 mg/kg, but no obvious toxic growth symptoms were observed. The content of Sb in roots(289.7-508.6 mg/kg) and the root-shoot transfer factor(0.09-0.57) of Zhongzhu No.1 increased with the increase of soil Sb concentration, but the change of Sb bioconcentration factor in roots was opposite, indicating that high concentrations of Sb in soil could promote the absorption of Sb in roots and the transport of Sb to the aboveground part, but the Sb enrichment capacity of roots was relatively reduced with the increase of soil Sb. Sb stress had a certain impact on the physiological characteristics of ramie leaves. With the increase of Sb treatment concentration, MDA, POD and SOD showed a change trend of "first increasing and then decreasing", while CAT gradually increased, indicating that Sb stress caused changes in the physiological characteristics of ramie leaves, thereby affecting plant growth and development. [Conclusions] This study provides a theoretical basis for ecological restoration of ramie in mining areas. 展开更多

Large intelligent surface(LIS)is considered as a new solution to enhance the performance of wireless networks[1].LIS comprises low-cost passive elements which can be well controlled.In this paper,a LIS is invoked in the vehicular networks.We analyze the system performance under Weibull fading.We derive a novel exact analytical expression for outage probability in closed form.Based on the analytical result,we discuss three special scenarios including high SNR case,low SNR case,as well as weak interference case.The corresponding approximations for three cases are provided,respectively.In order to gain more insights,we obtain the diversity order of outage probability and it is proved that the outage probability at high SNR depends on the interference,threshold and fading parameters which leads to 0 diversity order.Furthermore,we investigate the ergodic achievable rate of LIS-assisted vehicular networks and present the closed-form tight bounds.Similar to the outage performance,three special cases are studied and the asymptotic expressions are provided in simple forms.A rate ceiling is shown for high SNRs due to the existence of interference which results 0 high SNR slope.Finally,we give the energy efficiency of LIS-assisted vehicular network.Numerical results are presented to verify the accuracy of our analysis.It is evident that the performance of LIS-assisted vehicular networks with optimal phase shift scheme exceeds that of traditional vehicular networks and random phase Received:Aug.6,2020 Revised:Nov.17,2020 Editor:Caijun Zhong shift scheme significantly. 展开更多

Emotional Mechanisms in Supervisor-Student Relationship:Evidence from Machine Learning and Investigation

How to cultivate innovative talents has become an important educational issue nowadays.In China’s long-term mentorship education environment,supervisor-student relationship often affects students’creativity.From the perspective of students’psychology,we explore the influence mechanism of supervisor-student relationship on creativity by machine learning and questionnaire survey.In Study 1,based on video interviews with 16 postgraduate students,we use the machine learning method to analyze the emotional states exhibited by the postgraduate students in the videos when associating them with the supervisor-student interaction scenario,finding that students have negative emotions in bad supervisor-student relationship.Subsequently,we further explore the impact of supervisor-student relationship on postgraduate students’development in supervisor-student interaction scenarios at the affective level.In Study 2,a questionnaire survey is conducted to explore the relationship between relevant variables,finding that a good supervisor-student relationship can significantly reduce power stereotype threat,decrease emotional labor surface behaviors,and promote creativity expression.The above results theoretically reveal the internal psychological processes by which supervisor-student relationship affects creativity,and have important implications for reducing emotional labor and enhancing creativity expression of postgraduate students. 展开更多

Bacterial cellulose(BC)is an exopolysaccharide with unique properties that has been applied in various fields.However,the dense and intertwined nature of BC fibers limits its use in certain applications,including 3D printing scaffolds for bone regeneration.In this work,a controllable BC-based bio-ink for 3D printing was successfully prepared by modifying the neat BC through maleic acid(MA)treatment,aiming to promote bone tissue regeneration.To achieve homogeneous BC dispersions while preserving its crystalline and chemical properties,BC was modified by MA solution(60%,w/V)with solid-liquid ratio from 1꞉5 to 1꞉50(w/V)to obtain MA-BC dispersions.The analysis results from microstructure,chemical group,crystallinity,and wettability indicated that the BC/MA solution with ratio of 1꞉30 demonstrated the best pre-treatment performance to obtain MA-BC.Subsequently,by combining MA-BC with gelatin,we successfully formulated MA-BC-GEL gels with favorable rheological properties and compression modulus,which can be used as promising bio-inks for 3D bioprinting applications.In vitro tests demonstrated 1꞉30 MA-BC possessed excellent biocompatibility,a significant ability to express the alkaline phosphatase gene and osteogenic-related genes,and facilitated the formation of mineralized nodules.The utilization of this novel bio-ink in scaffold preparation for bone regeneration highlights the promising application of modified BC in bone tissue engineering field. 展开更多
