The fruit peel color is a crucial trait of cucumber.To better understand the molecular mechanisms underlying cucumber peel coloration,we compared the UPLC-ESI-MS/MS-based flavonoid metabolomic and RNA sequencing-based transcriptomic profiling of the brown peeled cucumber line‘PW’at six developmental stages.A total of 210 flavonoid metabolites were identified.Of which,117 flavonoid metabolites were differentially accumulated.In this study,weighted gene co-expression network analysis combined with Kyoto Encyclopedia of Gene and Genomes enrichment analysis revealed key genes coding for seven enzymes and eight transcription factors(TFs)associated with flavonoid biosynthesis.Among them,the R2R3MYB CsaV3_4G001130 is the best candidate gene that is responsible for controlling mature fruit colors in cucumber.Sanger sequencing revealed one nonsynonymous SNP in the exon of CsaV3_4G001130 among the selected 11 cucumber lines,which introduced a premature stop codon,generating a truncated protein in pale yellow or creamy peeled fruits.Yeast two-hybrid assays showed a direct interaction of CsaV3_4G001130 with the bHLH TF CsaV3_1G002260 and the WD40 protein CsaV3_5G001800.However,the interactions were influenced by the nonsynonymous SNP we identified.Our finding revealed that the integrated transcriptome and metabolome analysis further demonstrated that the abundance of some pigmented flavonoids(especially anthocyanins and chalcones)contributed to the coloration of‘PW’fruits.These findings pave the way for elucidation of flavonoid biosynthesis and improvement of cucumber peel color in the future. 展开更多

BACKGROUND As one of the most important members in clinical trials,the number of clinical research nurses(CRN)can't keep up with the growth of experimental projects,so it is urgent to build clinical research training and strengthen the background knowledge of nurses.AIM To construct CRN training program based on position competence,accelerate the construction of CRN talent pool,and provide scientific guidance significance for CRN training.METHODS Based on the position competence model,combined with literature research and qualitative interview results,the first draft was prepared of the CRN training program.Two rounds of correspondence with 16 experts were conducted using the Delphi method to determine the training program.RESULTS The effective recovery rate of the expert correspondence questionnaire was 100%and the authority coefficients of the 2 rounds of experts were 0.826 and 0.895.Finally,4 first-level indicators and determine 15 s-level indicators of training objectives.The training program included 4 first-level indicators,training requirements,content,methods,assessment and evaluation,15 s-level indicators,and 74 third-level indicators.CONCLUSION The CRN training program based on position competence is scientific and extendable,providing a basis for participation in CRN training. 展开更多

目的 分析血清降钙素原(PCT)、C-反应蛋白(CRP)、血清淀粉样蛋白A(SAA)和高迁移率族蛋白B1(HMGB1)在尿毒症并发血液透析导管相关性感染早期诊断中的应用价值。方法 选取2018年1月至2021年1月武汉科技大学附属天佑医院收治的62例尿毒症... 目的 分析血清降钙素原(PCT)、C-反应蛋白(CRP)、血清淀粉样蛋白A(SAA)和高迁移率族蛋白B1(HMGB1)在尿毒症并发血液透析导管相关性感染早期诊断中的应用价值。方法 选取2018年1月至2021年1月武汉科技大学附属天佑医院收治的62例尿毒症患者作为研究对象,所有患者均采用颈内静脉留置导管进行血液透析。根据其是否被感染分为感染组29例、非感染组33例。检测所有患者血清PCT、CRP、SAA和HMGB1水平,并采用受试者工作特征(ROC)曲线分析各指标对血液透析导管相关性感染的诊断价值。结果 与非感染组患者相比,感染组患者血清PCT、CRP、SAA和HMGB1水平均明显升高(P<0.05)。ROC曲线分析结果显示:PCT诊断尿毒症并发血液透析导管相关性感染的最佳临界值为7.32 ng/mL,曲线下面积(AUC)为0.872;CRP诊断的最佳临界值为12.00 mg/L,AUC为0.752;SAA诊断的最佳临界值为66.12 ng/mL,AUC为0.796;HMGB1诊断的最佳临界值为88.63 ng/mL,AUC为0.805。PCT和CRP两指标串联诊断的灵敏度和特异度分别为93.60%和90.80%,AUC为0.918;SAA和HMGB1两个指标串联诊断的灵敏度和特异度分别为91.20%和94.70%,AUC为0.913。结论 PCT和CRP串联、SAA和HMGB1串联对于尿毒症并发血液透析导管相关性感染的诊断具有较高的临床价值,可推广应用。 展开更多

为强化输配电网络中电量信息的应用时效性,提出一种基于数字化输配电网络的信息时效性判定方法。参照分布式配电网传输体系的执行需求,连接数字化应用电阻及输电信号调节器,完成数字化输配电网络环境的构建。在此基础上,统计传输电子量... 为强化输配电网络中电量信息的应用时效性,提出一种基于数字化输配电网络的信息时效性判定方法。参照分布式配电网传输体系的执行需求,连接数字化应用电阻及输电信号调节器,完成数字化输配电网络环境的构建。在此基础上,统计传输电子量条件,通过确定时效配电位置的方式,计算关联性物理判定系数,实现数字化输配电网络信息的时效性判定技术研究。对比实验结果表明,与GTHA判定手段相比,应用数字化判定方法后,电量信息的存储极限达到8.8×10^15 T,YLA传输敏感系数提升至75,输配电网络环境中的电子节点全部得到均匀分配处理,实现了强化电量信息应用时效性的初衷。 展开更多

The most frequent chromosomal rearrangement reported in acute promyelocytic leukemia (APL) is t(15;17)(q22;q21). The t(15;17) generates the PML/RARA fusion gene that blocks the transcription of genes involved in myeloid cell differentiation. A small number of simple and complex variants of the classical t(15;17) have been reported. We report two complex three-way translocation variants,t(3;17;15)(q27;q21;q22) and t(8;17;15)(q24.3;q12;q22) in which the PML/RARA fusion gene has been created on the derivative 15 chromosomes. Many of these variant translocations are suspected by conventional cytogenetics but need to be confirmed with additional molecular testing. We discuss the importance of supplementing conventional cytogenetic testing with FISH and RT-PCR to accurately diagnose APL variant patients. 展开更多

Nanocrystalline molybdenum carbide (Mo2C) was prepared via one simple route by the reaction of metallic magnesium powders with molybdenum trioxide and potassium acetate in an autoclave at the condition of 600℃ and 4 h. X-ray powder diffraction (XRD) pattern indicated that the product was hexagonal α-Mo2C, and the cell constant was a = 3.0091 ?, c = 4.7368 ?. Scanning electron microscopy (SEM) image showed that the sample consisted of particles with an average size of about 100 nm in diameter. The product was also studied by the thermogravimetric analysis (TGA). It had good thermal stability and oxidation resistance below 450℃ in air. 展开更多

In existing integrated circuit (IC) fabrication methods,the yield is typically limited by defects generated in the manufacturing process.In fact,the yield often shows a good correlation with the type and density of the defect.As a result,an accurate defect limited yield model is essential for accurate correlation analysis and yield prediction.Since real defects exhibit a great variety of shapes,to ensure the accuracy of yield prediction,it is necessary to select the most appropriate defect model and to extract the critical area based on the defect model.Considering the realistic outline of scratches introduced by the chemical mechanical polishing (CMP) process,we propose a novel scratch-concerned yield model.A linear model is introduced to model scratches.Based on the linear model,the related critical area extraction algorithm and defect density distribution are discussed.Owing to higher correspondence with the realistic outline of scratches,the linear defect model enables a more accurate yield prediction caused by scratches and results in a more accurate total product yield prediction as compared to the traditional circular model. 展开更多

Different defect morphologies in polyethylene crystal induced by surface physicochemical properties

The physicochemical properties of surfaces have a great effect on the micro-morphologies of the crystal structures which are in contact with them.Understanding the interaction mechanism between the internal driving forces of the crystal and external inducing forces of the surfaces is the prerequisite of controlling and obtaining the desirable morphologies.In this work,the dynamic density functional theory was applied to construct the free energy functional expression of polyethylene(PE)lattice,and the micro-dynamic evolution processes of PE lattice morphology near the surfaces with different properties were observed to reveal the interaction mechanism at atomic scale.The results showed that the physical and chemical properties of the external surfaces synergistically affect the morphologies in both the defect shapes and the distribution of the defect regions.In the absence of the contact surfaces,driven by the oriented interactions among different CH2 groups,PE lattices gradually grow and form a defect-free structure.Conversely,the presence of contact surfaces leads to lattice defects in the interfacial regions,and PE lattice shows different self-healing abilities around different surfaces. 展开更多

目的通过动物实验观察褪黑素(MT)对缺血再灌注后大鼠肾脏线粒体氧化应激及炎症状态的影响。方法60只SD大鼠随机分为假手术组(Sham组,n=10)、缺血再灌注组(IR组,n=25)及缺血再灌注+MT 3.0 mg/kg组(MT 3.0 mg组,n=25)。建立再灌注模型,于... 目的通过动物实验观察褪黑素(MT)对缺血再灌注后大鼠肾脏线粒体氧化应激及炎症状态的影响。方法60只SD大鼠随机分为假手术组(Sham组,n=10)、缺血再灌注组(IR组,n=25)及缺血再灌注+MT 3.0 mg/kg组(MT 3.0 mg组,n=25)。建立再灌注模型,于缺血再灌注6、24 h后留取静脉血检测血清尿素氮(BUN)、血清肌酐(Scr)水平;取一侧肾脏上极制作HE染色标本用于观察肾脏组织结构改变(并作Paller法肾小管损伤程度评分)和免疫组化法检测肿瘤坏死因子-α(TNF-α)及白介素6(IL-6)蛋白表达;取另一侧肾皮质制备匀浆、提取肾组织线粒体,用酶联免疫吸附法检测线粒体中TNF-α、高敏C反应蛋白(hs-CRP)、IL-6、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)水平。结果缺血再灌注6 h及24 h后,与Sham组各时点比较,IR组BUN、Scr水平,Paller评分,肾脏组织IL-6、TNF-α表达及肾脏线粒体中hs-CRP、TNF-α、IL-6、MDA水平均升高(P<0.01),GSH-Px、SOD水平均降低(P<0.01);与IR组各时点比较,加MT后的MT 3.0 mg组上述各指标均呈现反向变化,差异均有统计学意义(P<0.01)。结论MT能改善缺血再灌注时肾脏线粒体氧化应激及炎症状态,有效缓解缺血再灌注所致的大鼠肾脏损伤并改善肾功能。 展开更多

目的:分析不同年龄对角膜塑形镜配戴后视力矫正效果和安全性的影响。方法:回顾性临床研究。连续性纳入2021年6—10月于温州医科大学附属眼视光医院进行角膜塑形镜配戴治疗的近视患者176例(176眼)。根据患者年龄进行分组,≤13岁组78例(78... 目的:分析不同年龄对角膜塑形镜配戴后视力矫正效果和安全性的影响。方法:回顾性临床研究。连续性纳入2021年6—10月于温州医科大学附属眼视光医院进行角膜塑形镜配戴治疗的近视患者176例(176眼)。根据患者年龄进行分组,≤13岁组78例(78眼),>13~<18岁组53例(53眼)、≥18岁组45例(45眼)。收集各组基线和戴镜后1 d、1周、2周及1、3、6、9、12个月的等效球镜度(SE)、裸眼视力(UCVA)、角膜平坦曲率(FK)和角膜陡峭曲率(SK),并对戴镜后的安全性进行评价。取右眼数据用于统计分析,组间基线数据比较采用单因素方差分析或Kruska-WallsH检验方法,组间、组内多时间点的数据比较采用广义估计方程。结果:3组患者戴镜期间的SE、UCVA、FK、SK与其基线比较差异均有统计学意义(均P<0.05)。戴镜后1、3个月,3组间的客观验光SE改变百分比差异均有统计学意义(χ^(2)=9.17,P=0.010;χ^(2)=7.11,P=0.029),其中≥18岁组变化较≤13岁组缓慢(P=0.003、0.009)。戴镜后12个月,3组间的主觉验光SE改变百分比差异有统计学意义(χ^(2)=7.93,P=0.019),≥18岁组变化较≤13岁组更大(P=0.005)。戴镜1 d至6个月期间,3组间的UCVA差异均无统计学意义(均P>0.05);戴镜后9、12个月,3组间的UCVA差异有统计学意义(χ^(2)=6.40,P=0.041;χ^(2)=8.78,P=0.012),其中≥18岁组视力较≤13岁组更好、更稳定(P=0.017、0.003)。3组的FK改变量仅在3、9个月时差异有统计学意义(χ^(2)=6.25,P=0.044;χ^(2)=7.57,P=0.023),≥18岁组的FK改变量均小于≤13岁组(P=0.013、0.006),余时间差异均无统计学意义(均P>0.05)。戴镜1周至9个月期间,3组间的SK改变量差异均有统计学意义(χ^(2)=9.71~15.23,P<0.05),进一步分析发现戴镜后1周、3个月≥18岁组的SK改变量均小于≤13岁组(P=0.001、0.004),而戴镜后2周和1、6、9个月,≥18岁组的SK改变量小于>13~<18岁组和≤13岁组(均P<0.05)。戴镜期间3组患者眼压改变量、角膜厚度改变量及各种并发症组间比较差异均无统计学意义(均P>0.05)。结论:对于不同年龄近视患者,角膜塑形镜均能安全有效地压平角膜,降低屈光度和提升UCVA,但成年患者相较于儿童患者的角膜曲率改变更少,屈光度改变和UCVA更稳定。 展开更多

Stem cell based transplants effectively regenerate tissues;however, limitations such as immune rejection and teratoma formation prevent their application. Extracellular vesicles (EVs)-mediated acellular tissue regeneration is a promising alternative to stem cell based transplants. Although neural EGFL-like 1 (Nell1) is known to contribute to the osteogenic differentiation of bone marrow stem cells (BMSCs), it remains unknown whether EVs are involved in this process. Here, we present that EVs derived from Nell1-modified BMSCs (Nell1/EVs) have a stronger ability to promote BMSC osteogenesis owing to miR-25–5p downregulation. MiR-25–5p inhibits osteogenesis by targeting Smad2 and suppressing the SMAD and extracellular signal-related kinase 1 and 2 (ERK1/2) pathway activation. In addition, we demonstrate that the 3D-Nell1/EV-hydrogel system is beneficial for bone regeneration in vivo, probably stemming from a slow, continuous release and high concentration of EVs in the bone defect area. Thus, our results have shown the potential of Nell1/EVs as a novel acellular bone regeneration strategy. Mechanistically, the identification of miR-25-5p-SMAD2 signaling axis expands the knowledge of Nell1/EVs induced osteogenesis. 展开更多

Heavy alcohol drinking is a major public health problem,causing a large disease,social and economic burden in societies.Subjective response (SR) to alcohol is an intermediate characteristic of heavy drinking.A variety of candidate genes have been reported to be associated with SR to alcohol.In this study,we investigated nine single nucleotide polymorphisms (SNPs) related to SR to alcohol in healthy individuals from five Chinese ethnic groups,the Han,Hui,Tibetan,Mongolian and Uygur populations,and a total of 584 bloodstain samples were collected.The nine SNPs included four SNPs in alcohol-metabolizing genes (ADH1B,ADH1C,ALDH2 and CYP2E1*5B) and five SNPs in genes of neurobiological pathways (GABRA2,OPRM1,CHRNA3,HYKK and SLC6A4).A SNaPshot analysis method was developed to type these SNPs simultaneously,and all samples were typed successfully.Statistical analyses of the allele frequencies indicated that the frequencies of all SNPs,except for ADH1C,showed varying degrees of difference in the five studied ethnic groups.Tibetans showed the highest frequencies of risk alleles for heavy drinking at most loci.The genetic polymorphic differences found in this study revealed the variation in genetic susceptibility to heavy drinking in the studied populations. 展开更多
