Objective:To explore the value of flow cytometry(FCM)in detecting the level of exfoliated cells in pleural effusion in the differential diagnosis of non-small cell lung cancer and benign lung diseases.Methods:Clinical data of patients with non-small cell lung cancer who were hospitalized in Hebei hospital from June 2019 to March 2022 were collected.A total of 98 patients were included,and 63 patients with alveolar lung disease were screened during the same period,and the two groups of patients were analyzed.Results:Compared with alveolar lung disease group,FCM detection and analysis showed that the level of exfoliated cells in the pleural effusion of non-small cell lung cancer(NSCLC)patients was 99(3-969)/100,000,and patients with alveolar lung disease was 4(0~19)/100,000.Additionally,compared with the alveolar lung disease group,the level of exfoliated cells in the pleural effusion of patients with non-small cell lung cancer(NSCLC)was significantly increased(P<0.001).The diagnostic efficacy of FCM for detecting pleural fluid exfoliated cells in non-small cell lung cancer was assessed using ROC curves and using 95%CI(-11.1,-13.2)with a sensitivity of 0.75 and specificity of 0.94,and the diagnostic efficacy of FCM for detecting pleural fluid exfoliated cells in alveolar lung disease was assessed using 95%CI(-11.1,-13.2)with a sensitivity of 0.71 and specificity of 0.87.Conclusion:Flow cytometry has a wider range of clinical applications,simple operation,low cost,and high sensitivity,which makes it of great significance in disease diagnosis. 展开更多

Objective:To investigate the effect of combined detection of serum carcinoembryonic antigen(CEA),cytokeratin 19 fragment(CYFRA21-1),cancer antigen 125(CA125),and neuron-specific enolase(NSE)in patients with lung cancer by fluorescence flow cytometry.Methods:From August 2019 to July 2022,200 patients with lung cancer diagnosed by pathology in our hospital were retrospectively analyzed.2 mL venous blood was collected in a fasting state and centrifuged to separate the serum(containing human chorionic gonadotropin antibody[anti-hCG antibody],hepatitis B surface antibody[anti-HBs antibody],and CEA).Results:The sensitivities of CEA and CYFRA21-1 detected via enzyme-linked immunosorbent assay(ELISA)were 100%,and the detection limits were 0.5 ng/mL and 0.1 ng/mL,respectively;the sensitivities of CA125 and NSE detected via flow cytometry were 100%,and the detection limits were 10 U/mL and 2 ng/mL,respectively.Compared with ELISA,the sensitivities of CA125 and NSE detected via flow cytometry were higher.When the concentration of CEA was 10-40 ng/mL,the sensitivities of the three markers CYFRA21-1,CA125,and NSE showed no significant changes(P>0.05);when the concentration of CEA was 40-80 ng/mL,the sensitivity of CEA significantly decreased(P<0.01),but the sensitivities of the three markers CYFRA21-1,CA125,and NSE showed no significant changes(P>0.05);when the concentration of CEA was 80-200 ng/mL,the sensitivities of all four markers showed no significant changes(P>0.05).Conclusion:Compared with the double-antibody sandwich ELISA,fluorescence flow cytometry has certain advantages,including high sensitivity,good precision,short detection time,low sample usage,and low medical cost;thus,it is worthy of clinical promotion. 展开更多

However, due to the geographical location, cultural and historical reasons, the waterfront space of Macao has a development gap in the use of urban waterfront space. The internal reasons for the differences in the waterfront space construction in which regions of the island are more perfect are the issues to be studied in this article. It is hoped that through the study of the current situation of Macao’s local waterfront space infrastructure construction, the main points that affect advanced cases will be sorted out. From the location, industry, and infrastructure, find out its enlightenment to other waterfront spaces in Macao that are relatively backward in construction. 展开更多

Core decompression(CD)with the elimination of osteonecrotic bone is the most common strategy for treating early-stage nontraumatic osteonecrosis of the femoral head(ONFH).Adjuvant treatments are widely used in combination with CD as suitable methods of therapy.Existing augmentations have to be fabricated in advance.Here,we report a novel injectable glycerin-modified polycaprolactone(GPCL)that can adapt to the shape of the CD cavity.GPCL shows great flowability at 52.6℃.After solidification,its compressive modulus was 120 kPa at body temperature(37℃).This excellent characteristic enables the polymer to provide mechanical support in vivo.In addition,GPCL acts as a carrier of the therapeutic agent zoledronic acid(ZA),demonstrating sustained release into the CD region.ZA-loaded GPCL was injected into ONFH lesions to treat early-stage nontraumatic cases.Compared to that in the CD group,CD+ZA-loaded GPCL injection preserved bone density and increased the collagen level in the femoral head.At the interface between the GPCL and CD tunnel wall,osteogenesis was significantly promoted.In addition,morphological evaluations revealed that the femoral heads in the CD+ZA-GPCL group exhibited improved pressure resistance.These results suggest a strategy effective to preserve the bone density of the femoral head,thus decreasing the possibility of femoral head collapse.This novel injectable polymer has,therefore,considerable potential in clinical applications. 展开更多

Phylogenetic diversity and bioactivity of culturable deepsea-derived fungi from Okinawa Trough

Deep-sea sediments are now recognized as a home for rich and largely microbial community.Recently,it has been believed in an increasing number of studies that bacteria could be abundant in deepsea sediments of many types;however,fungi in deep-sea sediments remain relatively unknown.The phylogenetic diversity and bioactivity of culturable deep-sea-derived fungi from Okinawa Trough sediments were investigated in traditional method combined with fungal identification of molecular biology in this study.A total of 76 isolates belonged to 15 fungal taxa were recovered in a harsh condition of low nutrient and low temperature,indicating that the fungal communities in deep-sea sediments from Okinawa Trough were relatively abundant and diversified.Aspergillus,Cladosporium,and Penicillium were the dominant fungal genera,while Mycosphaerella,Purpureocillium,and Schizophyllum were relatively rare in the deep-sea sediments from Okinawa Trough.Among the six genera recovered,Mycosphaerella was firstly recovered from deep-sea sediments in this study.Moreover,about 75%of the extracts from the 15 fungal representative isolates displayed distinct bioactivity against at least one indicator bacterium or marine macrofouler,emphasizing the potentials of these deep-sea-derived fungi from Okinawa Trough as producers of bioactive metabolites.Notably,isolates Cladosporium oxysporum SCSIO z001 and Penicillium citrinum SCSIO z049 displayed a wide spectrum of bioactivities,isolates Cladosporium cladosporioides SCSIO z015,Cladosporium sphaerospermum SCSIO z030,and Penicillium verruculosum SCSIO z007 exhibited a strong anti-bacterial-growth activity,and isolate Penicillium chrysogenum SCSIO z062 displayed a strong anti-larval-settlement activity.These results suggest that these isolates deserved further study as potential sources of novel bioactive metabolites. 展开更多

In order to evaluate the health status of pigs in time,monitor accurately the disease dynamics of live pigs,and reduce the morbidity and mortality of pigs in the existing large-scale farming model,pig detection and tracking technology based on machine vision are used to monitor the behavior of pigs.However,it is challenging to efficiently detect and track pigs with noise caused by occlusion and interaction between targets.In view of the actual breeding conditions of pigs and the limitations of existing behavior monitoring technology of an individual pig,this study proposed a method that used color feature,target centroid and the minimum circumscribed rectangle length-width ratio as the features to build a multi-target tracking algorithm,which based on joint probability data association and particle filter.Experimental results show the proposed algorithm can quickly and accurately track pigs in the video,and it is able to cope with partial occlusions and recover the tracks after temporary loss. 展开更多

RNA-remodeling proteins,including RNA helicases and chaperones,play vital roles in the remodeling of structured RNAs.During viral replication,viruses require RNA-remodeling proteins to facilitate proper folding and/or re-folding the viral RNA elements.Coxsackieviruses B3(CVB3)and Coxsackieviruses B5(CVB5),belonging to the genus Enterovirus in the family Picornaviridae,have been reported to cause various infectious diseases such as hand-foot-and-mouth disease,aseptic meningitis,and viral myocarditis.However,little is known about whether CVB3 and CVB5 encode any RNA remodeling proteins.In this study,we showed that 2C proteins of CVB3 and CVB5 contained the conserved SF3 helicase A,B,and C motifs,and functioned not only as RNA helicase that unwound RNA helix bidirectionally in an NTP-dependent manner,but also as RNA chaperone that remodeled structured RNAs and facilitated RNA strand annealing independently of NTP.In addition,we determined that the NTPase activity and RNA helicase activity of 2C proteins of CVB3 and CVB5 were dependent on the presence of divalent metallic ions.Our findings demonstrate that 2C proteins of CVBs possess RNA-remodeling activity and underline the functional importance of 2C protein in the life cycle of CVBs. 展开更多

In order to solve the problem that target tracking frames are lost during the visual tracking of pigs,this research proposed an algorithm for multi target pigs tracking loss correction based on Faster R-CNN.The video of live pigs was processed by Faster R-CNN to get the object bounding box.Then,the SURF and background difference method were combined to predict whether the target pig will be occluded in the next frame.According to the occlusion condition,the maximum value of the horizontal and vertical coordinate offset of the bounding box in the adjacent two frames of the frame image in continuous N(N is the value of the video frame rate)were calculated.When bounding boxes in a video frame are merged into one bounding box,this maximum value was used to correct the current tracking frame offset in order to achieve the purpose of solving the tracking target loss problem.The experiment results showed that the success rate range of RP Faster-RCNN in the data set was 80%-97% while in term of Faster-RCNN was 40%-85%.And the average center point error of RP Faster-RCNN was 1.46 lower than Faster-RCNN which was about 2.60.The new algorithm was characterized by good robustness and adaptability,which could solve the problem of missing tracking target and accurately track multiple targets when the targets occlude each other. 展开更多
