Background:Wear particles-induced osteolysis is a major long-term complication after total joint arthroplasty.Up to now,there is no effective treatment for wear particles-induced osteolysis except for the revision sur...Background:Wear particles-induced osteolysis is a major long-term complication after total joint arthroplasty.Up to now,there is no effective treatment for wear particles-induced osteolysis except for the revision surgery,which is a heavy psychological and economic burden to patients.A metabolite of gut microbiota,short chain fatty acids(SCFAs),has been reported to be beneficial for many chronic inflammatory diseases.This study aimed to investigate the therapeutic effect of SCFAs on osteolysis.Methods:A model of inflammatory osteolysis was established by applying CoCrMo alloy particles to mouse calvarium.After two weeks of intervention,the anti-inflammatory effects of SCFAs on wear particle-induced osteolysis were evaluated by micro-CT analysis and immunohistochemistry staining.In vitro study,lipopolysaccharide(LPS)primed bone marrow-derived macrophages(BMDMs)and Tohoku hospital pediatrics-1(THP-1)macrophages were stimulated with CoCrMo particles to activate inflammasome in the presence of acetate(C2),propionate(C3),and butyrate(C4).Western blotting,enzyme-linked immunosorbent assay,and immunofluorescence were used to detect the activation of NLRP3 inflammasome.The effects of SCFAs on osteoclasts were evaluate by qRT-PCR,Western blotting,immunofluorescence,and tartrate-resistant acid phosphatase(TRAP)staining.Additionally,histone deacetylase(HDAC)inhibitors,agonists of GPR41,GPR43,and GPR109A were applied to confirm the underlying mechanism of SCFAs on the inflammasome activation of macrophages and osteoclastogenesis.Results:C3 and C4 but not C2 could alleviate wear particles-induced osteolysis with fewer bone erosion pits(P<0.001),higher level of bone volume to tissue volume(BV/TV,P<0.001),bone mineral density(BMD,P<0.001),and a lower total porosity(P<0.001).C3 and C4 prevented CoCrMo alloy particles-induced ASC speck formation and nucleationinduced oligomerization,suppressing the cleavage of caspase-1(P<0.05)and IL-1β(P<0.05)stimulated by CoCrMo alloy particles.C3 and C4 also inhibited the generation of gasdermin D-N-terminal fragment(GSDMD-NT)to regulate pyroptosis.Besides,C3 and C4 have a negative impact on osteoclast differentiation(P<0.05)and its function(P<0.05),affecting the podosome arrangement and morphologically normal podosome belts formation.Conclusions:Our work showed that C3 and C4 are qualified candidates for the treatment of wear particle-induced osteolysis.展开更多

[Methods] Colon cancer cells treated with different concentrations of EriB were detected by CCK-8 assay;cell scratch assay and crystal violet staining were used to detect the invasion and migration of colon cancer cell;cell apoptosis was detected by Annexin V/PI double staining, and cell cycle was detected by PI staining;Western Blotting was used to detect epithelial-mesenchymal transition and apoptosis-related proteins in colon cancer cells treated with EriB. [Results] After EriB treatment, the proliferation, migration and apoptosis of colon cancer cells were significantly inhibited, and the ratio of P-ERK1/2 to ERK was significantly decreased. [Conclusions] EriB can effectively inhibit the proliferation of colon cancer cells and promote the apoptosis of colon cancer cells through ERK1/2 pathway.展开更多

The results show...The passivation process of X80 pipeline steel in bicarbonate solutions was investigated using potentiodynamic, dynamic electro-chemical impedance spectroscopy (DEIS), and Mott-Schottky measurements. The results show that the shape of polarization curves changes with concentration. The critical 'passive' concentration is 0.009 mol/L for X80 pipeline steel in bicarbonate solutions. No anodic current peak exists in solutions when the concentration is lower than 0.009 mol/L, whereas there are one and two anodic current peaks when the concentration ranges from 0.009 to 0.05 mol/L and is higher than 0.1 mol/L, respectively. DEIS measurements show that there exist active dissolution range, transition range, pre-passive range, passive layer formation range, passive range, and trans-passive range for X80 pipeline steel in the 0.1 mol/L solutions. The results of DEIS measurements are in complete agreement with the potentiodynamic diagram. An equivalent circuit containing three sub-layers is used to explain the Nyquist plots in the passive range. Analyses are well made for explaining the corresponding fitted capacitance and impedance. The Mott-Schottky plots show that the passive film of X80 pipeline steel is an n-type semiconductor, and capacitance measurements are in good accordance with the results of DEIS experiment.展开更多
The present study aimed to clarify the role of SENEX in malignant cell proliferation in diffuse large B-cell lymphoma(DLBCL).22 DLBCL patients(6 newly diagnosed cases,7 cases at complete remission,and 9 relapsed cases...The present study aimed to clarify the role of SENEX in malignant cell proliferation in diffuse large B-cell lymphoma(DLBCL).22 DLBCL patients(6 newly diagnosed cases,7 cases at complete remission,and 9 relapsed cases)were included in the study.Our results indicated that both SENEX gene and protein were significantly increased in peripheral blood mononuclear cells(PBMCs)and tumor cells of relapsed DLBCL patients,accompanied by overexpression of p21,p16,and phosphorylated retinoblastoma(Rb).Silencing the SENEX gene in a DLBCL cell line caused a significant decrease in cell proliferation and inhibited cell cycle progression in the G1 phase.Phosphorylated Rb and E2F1 were also decreased,and activation of the Rb/E2F1 pathway was obviously suppressed.To conclude,the SENEX gene promotes proliferation in PBMC and tumor cells of DLBCL patients by activating the Rb/E2F1 pathway,in a manner suggesting that increased SENEX expression affects the relapse of DLBCL and may serve as an important target for DLBCL therapy.展开更多
As an economically important marine fish,the large yellow croaker Larimichthys crocea suffered from marine white spot disease caused by the ectoparasite Cryptocaryon irritans in recent years.This disease not only coul...As an economically important marine fish,the large yellow croaker Larimichthys crocea suffered from marine white spot disease caused by the ectoparasite Cryptocaryon irritans in recent years.This disease not only could result in physiological damage,but also lead to secondary bacterial invasion.Reports indicated some AMPs(antimicrobial peptides)were of antiparasitic activity to C.irritans.Hepcidin-like(Lc-HepL)was one of the significant differential expression genes excavated from the transcriptome following a challenge with C.irritans.In this study,we characterized this AMP’s bioactivity based on the levels of mRNA and protein.After challenged by C.irritans,qRT-PCR showed Lc-HepL was significantly upregulated in six tissues,including gill,muscle,liver,head kidney and spleen during theront infection,trophont falling off,and secondary bacterial invasion stages,which implicated a role Lc-HepL played in the immune defense against C.irritans and secondary bacterial infection.Recombinant Lc-HepL(rLc-HepL)was induced and purified successfully.rLc-HepL exhibited antibacterial activity to certain bacteria in a dose-and time-dependent manners.Anti-C.irritans activity was explored for the first time and found it could cause the theronts membrane rupture and contents leakage.These results provided the first evidence that Lc-HepL had strong antiparasitic activity against marine fish ectoparasites C.irritans theronts.Together,data indicated that Lc-HepL might be an important component in the innate immune system against C.irritans and has the potential to be employed in future drug development.展开更多

A draft genomic sequence is expected to be publicly available in the near future, which means that an explosion of gene expression studies awaits. Thus, the work of selecting appropriate reference genes for q PCR normalization in different tissues or under various experimental conditions is extremely valuable. In this study, ten candidate genes were analyzed in B. luminifera subjected to different abiotic stresses and at various flowering stages.The expression stability of these genes was evaluated using three distinct algorithms implemented using ge Norm,Norm Finder and Best Keeper. The best-ranked reference genes varied across different sample sets, though RPL39,MDH and EF1 a were determined as the most stable by the three programs among all tested samples. RPL39 and EF1 a should be appropriate for normalization in N-starved roots,while the combination of RPL39 and MDH should be appropriate for N-starved stems and EF1 a and MDH should be appropriate in N-starved leaves. In PEG-treated(osmotic) roots, MDH was the most suitable, whereas EF1 a was suitable for PEG-treated stems and leaves. TUA was also stably expressed levels in PEG-treated plants. The combination of RPL39 and TUB should be appropriate for heat-stressed leaves and flowering stage. For reference gene validation, the expression levels of SOD and NFYA-3were investigated. This work will be beneficial to future studies on gene expression under different abiotic stress conditions and flowering status in B. luminifera.展开更多
Myelodysplastic syndromes(MDS)are highly heterogeneous myeloid neoplasms,and a large number of patients are difficult to diagnose and classify by blood and bone marrow examination.As a surface marker of granulocyte,st...Myelodysplastic syndromes(MDS)are highly heterogeneous myeloid neoplasms,and a large number of patients are difficult to diagnose and classify by blood and bone marrow examination.As a surface marker of granulocyte,studies have shown CD10 can be used to define the degree of granulocyte maturation in MDS patients.However,whether it can be used for differential diagnosis of MDS and other hematological diseases remains inconclusive.To explore the value of CD10 for differential diagnosis of MDS,60 newly diagnosed MDS,20 aplastic anemia(AA)patients,and 35 iron-deficient anemia(IDA)patients were selected for this study.Bone marrow(BM)specimens were processed for surface marker analysis and labeled with pre-conjugated monoclonal antibodies.Stained cells were detected by flow cytometry.Our results indicated that CD10-positive granulocytes were significantly decreased in BM of MDS patients than AA and IDA patients,and the level of CD10-positive mature granulocytes was not associated with the clinical stages of malignancy.Receiver operating characteristic(ROC)areas under the curve(AUC)of CD10-positive granulocytes was 0.86 and 0.85,respectively,in MDS patients than the IDA group and AA group with good specificity and sensitivity.Further,CD10-positive granulocytes were increased after effective treatment.In conclusion,we found the decrease in CD10-positive granulocytes has a differential diagnostic value of MDS.展开更多

This study aimed to use a computerized docking approach to examine the interactions between the human AR and phytooestrogens (genistein, daidzein, and flavone) and xeno-oestrogens (bisphenol A, 4-nonylphenol, dichlorodiphenyl trichloroethane [DDT], diethylstilbestrol [DES]). The predicted three-dimensional structure of AR and androgens was established using X-ray diffraction. The binding of four xeno-oestrogens and three phyto-oestrogens to AR was analysed. The steroids estradiol and dihydrotestosterone (DHT) were used as positive controls and thyroxine as negative control. All the ligands shared the same binding site except for thyroxine. The endogenous hormones DHT and 17β-oestradiol showed the strongest binding with the lowest affinity energy (〈 -10 kcal mol-1). All three phyto- oestrogens and two xeno-oestrogens (bisphenol A and DES) showed strong binding to AR. The affinities offlavone, genistein, and daidzein were between -8.8 and -8.5 kcal mol 1, while that of bisphenol A was -8.1 kcal mol-l and DES -8.3 kcal mol-1. Another two xeno-oestrogens, 4-nonylphenol and DDT, although they fit within the binding domain of AR, showed weak affinity (-6.4 and -6.7 kcal mol 1, respectively). The phyto-oestrogens genistein, daidzein and flavone, and the xeno-oestrogens bisphenol A and DES can be regarded as androgenic effectors. The xenooestrogens DDT and 4-nonylphenol bind only weakly to AR.展开更多
Objective:To investigate the effect of intestinal flora from Henoch-Schönlein purpura(HSP)on visceral sensitivity,gastrointestinal hormones and cytokines in pseudo-sterile rats.Methods:The pseudo-sterile rat mode...Objective:To investigate the effect of intestinal flora from Henoch-Schönlein purpura(HSP)on visceral sensitivity,gastrointestinal hormones and cytokines in pseudo-sterile rats.Methods:The pseudo-sterile rat model was established.The rats were was given fecal microbiota solutions of children with abdominal HSP and healthy children,respectively.The visceral sensitivity was determined by abdominal withdrawal reflex(AWR)which was induced by rectal balloon distention in all the rats.And serum gastrin(Gas),motilin(MTL),cholecystokinin(CCK),substance P(SP),tumor necrosis factor(TNF)-αand interleukin(IL)-6 levels in rats were measured with ELISA method.Results:The volume of rectum water injection under the score 3 of AWR in the rats administrated with fecal microbiota solution from HSP children(HSP group)was significantly decreased compared with that in the rats administrated with fecal microbiota solution from healthy children(HC group),and there was significant difference between these two groups(P<0.05).The serum Gas,MTL,CCK and SP levels were higher in HSP group than those in HC group.And serum MTL,CCK and SP levels in HSP group were significantly different from those in the HC group.The serum TNF-αandIL-6 levels were higher in HSP group than those in HC group,there was significant difference between these two groups(P<0.05).Conclusion:Intestinal flora from HSP can induce the production of visceral sensitivity,inhibit gastrointestinal hormone secretion and prompt cytokine production.展开更多

Microwave absorption materials are prone to degradation in extremely humid and salty environments,and it is still challenging to develop a dense and firm interface to protect microwave absorbers.Herein,a robust FeSiAl...Microwave absorption materials are prone to degradation in extremely humid and salty environments,and it is still challenging to develop a dense and firm interface to protect microwave absorbers.Herein,a robust FeSiAl@PUA@SiO2(PUA:acrylic polyurethane)gradient hybrid was prepared through plasma-enhanced chemical vapor deposition(PECVD)to achieve efficient microwave absorption and anti-corrosion properties.The organic/inorganic dual coat of PUA/SiO2 not only facilitated the interface polarization but also effectively reduced the dielectric constant and optimized impedance matching.Owing to the unique hybrid structure,the(PECVD-FeSiAl@PUA)@SiO_(2) exhibited highly efficient microwave absorbing performance in frequency bands covering almost the entire Ku-bands(12-18 GHz)with a minimum reflection loss(RLmin)of−47 dB with a matching thickness of 2.3 mm.The organic/inorganic dual protection effectively shields against the corrosive medium,as the corrosion potential and the polarization resistance increased from−0.167 to−0.047 V and 8,064 to 16,273Ω·cm^(2),respectively.While the corrosion current decreased from 3.04×10^(−6) to 2.16×10^(−6) A/cm^(2).Hence,the plasma-enhanced densification of PUA created a strong bridge to integrate FeSiAl and organic/inorganic components acquiring dual-function of efficient microwave absorption and anticorrosion,which opened a promising platform for potential practical absorbers.展开更多
A novel carbon quantum dots decorated C-doped a-Bi_(2)O_(3)photocatalyst(CBO/CQDs)was synthesized by solvothermal method.The synergistic effect of adsorption and photocatalysis highly improved contaminants removal eff...A novel carbon quantum dots decorated C-doped a-Bi_(2)O_(3)photocatalyst(CBO/CQDs)was synthesized by solvothermal method.The synergistic effect of adsorption and photocatalysis highly improved contaminants removal efficiencies.The ceftriaxone sodium degradation rate constant(k)of CBO/CQDs was 11.4 and 3.2 times that of pure a-Bi2O3 and C-doped a-Bi_(2)O_(3),respectively.The interstitial carbon doping generated localized states above the valence band,which enhanced the utilization of visible light and facilitated the separation of photogenerated electrons and holes;the loading of CQDs improved the charge carrier separation and extended the visible light response;the reduced particle size of CBO/CQDs accelerated the migration of photogenerated carriers.The·O2-and ht were identified as the dominant reactive species in ceftriaxone sodium degradation,and the key role of·O2-was further investigated by NBT transformation experiments.The Fukui index was applied to ascertain the molecular bonds of ceftriaxone sodium susceptible to radical attack,and intermediates analysis was conducted to explore the possible degradation pathways.The toxicity evaluation revealed that some degradation intermediates possessed high toxicity,thus the contaminants require sufficient mineralization to ensure safe discharge.The present study makes new insights into synchronous carbon dopping and CQDs decoration on modification of a-Bi2O3,which provides references for future studies.展开更多
The large yellow croaker(Larimichthys crocea)is one of the most economically valuable marine fsh in China and is a notable species in ecological studies owing to a serious collapse of wild germplasm in the past few de...The large yellow croaker(Larimichthys crocea)is one of the most economically valuable marine fsh in China and is a notable species in ecological studies owing to a serious collapse of wild germplasm in the past few decades.The stock division and species distribution,which have important implications for ecological protection,germplasm recovery,and fshery resource management,have been debated since the 1960s.However,it is still uncertain even how many stocks exist in this species.To address this,we evaluated the fne-scale genetic structure of large yellow croaker populations distributed along the eastern and southern Chinese coastline based on 7.64 million SNP markers.Compared with the widely accepted stock boundaries proposed in the 1960s,our results revealed that a climate-driven habitat change probably occurred between the Naozhou(Nanhai)Stock and the Ming-Yuedong(Mindong)Stock.The boundary between these two stocks might have shifted northwards from the Pearl River Estuary to the northern area of the Taiwan Strait,accompanied by highly asymmetric introgression.In addition,we found divergent landscapes of natural selection between the stocks inhabiting northern and southern areas.The northern population exhibited highly agminated signatures of strong natural selection in genes related to developmental processes,whereas moderate and interspersed selective signatures were detected in many immune-related genes in the southern populations.These fndings establish the stock status and genome-wide evolutionary landscapes of large yellow croaker,providing a basis for conservation,fsheries management and further evolutionary biology studies.展开更多

Nuclear receptor subfamily 2 (NR2F2, also known as COUP-TFU) is involved in the development of many types of cancers, but its role in gastric cancer remains elusive. In this experiment, oncomine and Kaplan-meier database revealed that NR2F2 was up-regulated in gastric cancer and that the high NR2F2 expression contributed to poor survival. MicroRNA-2Tb was targeted and down-regulated by NR2F2 in human gastric cancer tissues and cells. The ectopic expression of miR-27b inhibited gastric cancer cell proliferation and tumor growth in vitro and in vivo. Assays suggested that the overexpression of miR-27b could promote MGC-803 cells' migration and invasion and retard their metastasis to the liver. In addition, down-regulation of miR-27b enhanced GES-1 cells' proliferation and metastasis in vitro. These findings reveal that miR-27b is a tumor suppressor in gastric cancer and a biomarker for improving patients' survival.展开更多
With the rapid development of electronic information technology,the waste electromagnetic radiation generated by the wireless and various other electronic devices has caused serious harm to human health.The developmen...With the rapid development of electronic information technology,the waste electromagnetic radiation generated by the wireless and various other electronic devices has caused serious harm to human health.The development of high-efficiency flexible electromagnetic interference(EMI)shielding materials has become one of the most effective ways to mitigate the increasingly serious problem of electromagnetic pollution.Flexible carbon fiber composites possess most of the physical requirements needed for EMI shielding applications,including high electrical conductivity,large specific surface area,light weight,flexibility,porous structure,and adjustable physical and chemical properties,and various morphology of carbon fibers-based composites applied EMI.In this review,the primary EMI shielding mechanism of carbonfiber-based materials is presented.Since carbon fiber suffers from a low absorption efficiency when used for EMI shielding by itself,it is usually combined with additional EMI filling materials.Therefore,the methods of adding EMI filler materials to the carbon fiber framework and the influence of different fillers on the EMI shielding performance are also discussed.In addition,the novel structural designs of flexible carbon fiber-based EMI shielding materials,including fiber orientation,core-shell structure,and multilayer heterostructure,are summarized in terms of their classification,which includes composites with metal,polymer,nano-carbon,and MXenes.The review concludes with a brief discussion of the challenges,research directions,and future prospects of carbon-fiber-based EMI shielding materials.展开更多

[Results] After EriB treatment, the proliferation, migration and apoptosis of colon cancer cells were significantly inhibited, and the ratio of P-ERK1/2 to ERK was significantly decreased. [Conclusions] EriB can effectively inhibit the proliferation of colon cancer cells and promote the apoptosis of colon cancer cells through ERK1/2 pathway.

文摘The passivation process of X80 pipeline steel in bicarbonate solutions was investigated using potentiodynamic, dynamic electro-chemical impedance spectroscopy (DEIS), and Mott-Schottky measurements. The results show that the shape of polarization curves changes with concentration. The critical 'passive' concentration is 0.009 mol/L for X80 pipeline steel in bicarbonate solutions. No anodic current peak exists in solutions when the concentration is lower than 0.009 mol/L, whereas there are one and two anodic current peaks when the concentration ranges from 0.009 to 0.05 mol/L and is higher than 0.1 mol/L, respectively. DEIS measurements show that there exist active dissolution range, transition range, pre-passive range, passive layer formation range, passive range, and trans-passive range for X80 pipeline steel in the 0.1 mol/L solutions. The results of DEIS measurements are in complete agreement with the potentiodynamic diagram. An equivalent circuit containing three sub-layers is used to explain the Nyquist plots in the passive range. Analyses are well made for explaining the corresponding fitted capacitance and impedance. The Mott-Schottky plots show that the passive film of X80 pipeline steel is an n-type semiconductor, and capacitance measurements are in good accordance with the results of DEIS experiment.

文摘The present study aimed to clarify the role of SENEX in malignant cell proliferation in diffuse large B-cell lymphoma(DLBCL).22 DLBCL patients(6 newly diagnosed cases,7 cases at complete remission,and 9 relapsed cases)were included in the study.Our results indicated that both SENEX gene and protein were significantly increased in peripheral blood mononuclear cells(PBMCs)and tumor cells of relapsed DLBCL patients,accompanied by overexpression of p21,p16,and phosphorylated retinoblastoma(Rb).Silencing the SENEX gene in a DLBCL cell line caused a significant decrease in cell proliferation and inhibited cell cycle progression in the G1 phase.Phosphorylated Rb and E2F1 were also decreased,and activation of the Rb/E2F1 pathway was obviously suppressed.To conclude,the SENEX gene promotes proliferation in PBMC and tumor cells of DLBCL patients by activating the Rb/E2F1 pathway,in a manner suggesting that increased SENEX expression affects the relapse of DLBCL and may serve as an important target for DLBCL therapy.

文摘As an economically important marine fish,the large yellow croaker Larimichthys crocea suffered from marine white spot disease caused by the ectoparasite Cryptocaryon irritans in recent years.This disease not only could result in physiological damage,but also lead to secondary bacterial invasion.Reports indicated some AMPs(antimicrobial peptides)were of antiparasitic activity to C.irritans.Hepcidin-like(Lc-HepL)was one of the significant differential expression genes excavated from the transcriptome following a challenge with C.irritans.In this study,we characterized this AMP’s bioactivity based on the levels of mRNA and protein.After challenged by C.irritans,qRT-PCR showed Lc-HepL was significantly upregulated in six tissues,including gill,muscle,liver,head kidney and spleen during theront infection,trophont falling off,and secondary bacterial invasion stages,which implicated a role Lc-HepL played in the immune defense against C.irritans and secondary bacterial infection.Recombinant Lc-HepL(rLc-HepL)was induced and purified successfully.rLc-HepL exhibited antibacterial activity to certain bacteria in a dose-and time-dependent manners.Anti-C.irritans activity was explored for the first time and found it could cause the theronts membrane rupture and contents leakage.These results provided the first evidence that Lc-HepL had strong antiparasitic activity against marine fish ectoparasites C.irritans theronts.Together,data indicated that Lc-HepL might be an important component in the innate immune system against C.irritans and has the potential to be employed in future drug development.

文摘Betula luminifera is a commercial tree species that is emerging as a new model system for tree genomics research. A draft genomic sequence is expected to be publicly available in the near future, which means that an explosion of gene expression studies awaits. Thus, the work of selecting appropriate reference genes for q PCR normalization in different tissues or under various experimental conditions is extremely valuable. In this study, ten candidate genes were analyzed in B. luminifera subjected to different abiotic stresses and at various flowering stages.The expression stability of these genes was evaluated using three distinct algorithms implemented using ge Norm,Norm Finder and Best Keeper. The best-ranked reference genes varied across different sample sets, though RPL39,MDH and EF1 a were determined as the most stable by the three programs among all tested samples. RPL39 and EF1 a should be appropriate for normalization in N-starved roots,while the combination of RPL39 and MDH should be appropriate for N-starved stems and EF1 a and MDH should be appropriate in N-starved leaves. In PEG-treated(osmotic) roots, MDH was the most suitable, whereas EF1 a was suitable for PEG-treated stems and leaves. TUA was also stably expressed levels in PEG-treated plants. The combination of RPL39 and TUB should be appropriate for heat-stressed leaves and flowering stage. For reference gene validation, the expression levels of SOD and NFYA-3were investigated. This work will be beneficial to future studies on gene expression under different abiotic stress conditions and flowering status in B. luminifera.

文摘Myelodysplastic syndromes(MDS)are highly heterogeneous myeloid neoplasms,and a large number of patients are difficult to diagnose and classify by blood and bone marrow examination.As a surface marker of granulocyte,studies have shown CD10 can be used to define the degree of granulocyte maturation in MDS patients.However,whether it can be used for differential diagnosis of MDS and other hematological diseases remains inconclusive.To explore the value of CD10 for differential diagnosis of MDS,60 newly diagnosed MDS,20 aplastic anemia(AA)patients,and 35 iron-deficient anemia(IDA)patients were selected for this study.Bone marrow(BM)specimens were processed for surface marker analysis and labeled with pre-conjugated monoclonal antibodies.Stained cells were detected by flow cytometry.Our results indicated that CD10-positive granulocytes were significantly decreased in BM of MDS patients than AA and IDA patients,and the level of CD10-positive mature granulocytes was not associated with the clinical stages of malignancy.Receiver operating characteristic(ROC)areas under the curve(AUC)of CD10-positive granulocytes was 0.86 and 0.85,respectively,in MDS patients than the IDA group and AA group with good specificity and sensitivity.Further,CD10-positive granulocytes were increased after effective treatment.In conclusion,we found the decrease in CD10-positive granulocytes has a differential diagnostic value of MDS.

文摘The androgen receptor (AR) plays a critical role in prostate cancer development and progression. This study aimed to use a computerized docking approach to examine the interactions between the human AR and phytooestrogens (genistein, daidzein, and flavone) and xeno-oestrogens (bisphenol A, 4-nonylphenol, dichlorodiphenyl trichloroethane [DDT], diethylstilbestrol [DES]). The predicted three-dimensional structure of AR and androgens was established using X-ray diffraction. The binding of four xeno-oestrogens and three phyto-oestrogens to AR was analysed. The steroids estradiol and dihydrotestosterone (DHT) were used as positive controls and thyroxine as negative control. All the ligands shared the same binding site except for thyroxine. The endogenous hormones DHT and 17β-oestradiol showed the strongest binding with the lowest affinity energy (〈 -10 kcal mol-1). All three phyto- oestrogens and two xeno-oestrogens (bisphenol A and DES) showed strong binding to AR. The affinities offlavone, genistein, and daidzein were between -8.8 and -8.5 kcal mol 1, while that of bisphenol A was -8.1 kcal mol-l and DES -8.3 kcal mol-1. Another two xeno-oestrogens, 4-nonylphenol and DDT, although they fit within the binding domain of AR, showed weak affinity (-6.4 and -6.7 kcal mol 1, respectively). The phyto-oestrogens genistein, daidzein and flavone, and the xeno-oestrogens bisphenol A and DES can be regarded as androgenic effectors. The xenooestrogens DDT and 4-nonylphenol bind only weakly to AR.

文摘Objective:To investigate the effect of intestinal flora from Henoch-Schönlein purpura(HSP)on visceral sensitivity,gastrointestinal hormones and cytokines in pseudo-sterile rats.Methods:The pseudo-sterile rat model was established.The rats were was given fecal microbiota solutions of children with abdominal HSP and healthy children,respectively.The visceral sensitivity was determined by abdominal withdrawal reflex(AWR)which was induced by rectal balloon distention in all the rats.And serum gastrin(Gas),motilin(MTL),cholecystokinin(CCK),substance P(SP),tumor necrosis factor(TNF)-αand interleukin(IL)-6 levels in rats were measured with ELISA method.Results:The volume of rectum water injection under the score 3 of AWR in the rats administrated with fecal microbiota solution from HSP children(HSP group)was significantly decreased compared with that in the rats administrated with fecal microbiota solution from healthy children(HC group),and there was significant difference between these two groups(P<0.05).The serum Gas,MTL,CCK and SP levels were higher in HSP group than those in HC group.And serum MTL,CCK and SP levels in HSP group were significantly different from those in the HC group.The serum TNF-αandIL-6 levels were higher in HSP group than those in HC group,there was significant difference between these two groups(P<0.05).Conclusion:Intestinal flora from HSP can induce the production of visceral sensitivity,inhibit gastrointestinal hormone secretion and prompt cytokine production.

文摘Microwave absorption materials are prone to degradation in extremely humid and salty environments,and it is still challenging to develop a dense and firm interface to protect microwave absorbers.Herein,a robust FeSiAl@PUA@SiO2(PUA:acrylic polyurethane)gradient hybrid was prepared through plasma-enhanced chemical vapor deposition(PECVD)to achieve efficient microwave absorption and anti-corrosion properties.The organic/inorganic dual coat of PUA/SiO2 not only facilitated the interface polarization but also effectively reduced the dielectric constant and optimized impedance matching.Owing to the unique hybrid structure,the(PECVD-FeSiAl@PUA)@SiO_(2) exhibited highly efficient microwave absorbing performance in frequency bands covering almost the entire Ku-bands(12-18 GHz)with a minimum reflection loss(RLmin)of−47 dB with a matching thickness of 2.3 mm.The organic/inorganic dual protection effectively shields against the corrosive medium,as the corrosion potential and the polarization resistance increased from−0.167 to−0.047 V and 8,064 to 16,273Ω·cm^(2),respectively.While the corrosion current decreased from 3.04×10^(−6) to 2.16×10^(−6) A/cm^(2).Hence,the plasma-enhanced densification of PUA created a strong bridge to integrate FeSiAl and organic/inorganic components acquiring dual-function of efficient microwave absorption and anticorrosion,which opened a promising platform for potential practical absorbers.

文摘A novel carbon quantum dots decorated C-doped a-Bi_(2)O_(3)photocatalyst(CBO/CQDs)was synthesized by solvothermal method.The synergistic effect of adsorption and photocatalysis highly improved contaminants removal efficiencies.The ceftriaxone sodium degradation rate constant(k)of CBO/CQDs was 11.4 and 3.2 times that of pure a-Bi2O3 and C-doped a-Bi_(2)O_(3),respectively.The interstitial carbon doping generated localized states above the valence band,which enhanced the utilization of visible light and facilitated the separation of photogenerated electrons and holes;the loading of CQDs improved the charge carrier separation and extended the visible light response;the reduced particle size of CBO/CQDs accelerated the migration of photogenerated carriers.The·O2-and ht were identified as the dominant reactive species in ceftriaxone sodium degradation,and the key role of·O2-was further investigated by NBT transformation experiments.The Fukui index was applied to ascertain the molecular bonds of ceftriaxone sodium susceptible to radical attack,and intermediates analysis was conducted to explore the possible degradation pathways.The toxicity evaluation revealed that some degradation intermediates possessed high toxicity,thus the contaminants require sufficient mineralization to ensure safe discharge.The present study makes new insights into synchronous carbon dopping and CQDs decoration on modification of a-Bi2O3,which provides references for future studies.

文摘The large yellow croaker(Larimichthys crocea)is one of the most economically valuable marine fsh in China and is a notable species in ecological studies owing to a serious collapse of wild germplasm in the past few decades.The stock division and species distribution,which have important implications for ecological protection,germplasm recovery,and fshery resource management,have been debated since the 1960s.However,it is still uncertain even how many stocks exist in this species.To address this,we evaluated the fne-scale genetic structure of large yellow croaker populations distributed along the eastern and southern Chinese coastline based on 7.64 million SNP markers.Compared with the widely accepted stock boundaries proposed in the 1960s,our results revealed that a climate-driven habitat change probably occurred between the Naozhou(Nanhai)Stock and the Ming-Yuedong(Mindong)Stock.The boundary between these two stocks might have shifted northwards from the Pearl River Estuary to the northern area of the Taiwan Strait,accompanied by highly asymmetric introgression.In addition,we found divergent landscapes of natural selection between the stocks inhabiting northern and southern areas.The northern population exhibited highly agminated signatures of strong natural selection in genes related to developmental processes,whereas moderate and interspersed selective signatures were detected in many immune-related genes in the southern populations.These fndings establish the stock status and genome-wide evolutionary landscapes of large yellow croaker,providing a basis for conservation,fsheries management and further evolutionary biology studies.
文摘Increasing attention is focused on the down-regulation of miRNAs in cancer process. Nuclear receptor subfamily 2 (NR2F2, also known as COUP-TFU) is involved in the development of many types of cancers, but its role in gastric cancer remains elusive. In this experiment, oncomine and Kaplan-meier database revealed that NR2F2 was up-regulated in gastric cancer and that the high NR2F2 expression contributed to poor survival. MicroRNA-2Tb was targeted and down-regulated by NR2F2 in human gastric cancer tissues and cells. The ectopic expression of miR-27b inhibited gastric cancer cell proliferation and tumor growth in vitro and in vivo. Assays suggested that the overexpression of miR-27b could promote MGC-803 cells' migration and invasion and retard their metastasis to the liver. In addition, down-regulation of miR-27b enhanced GES-1 cells' proliferation and metastasis in vitro. These findings reveal that miR-27b is a tumor suppressor in gastric cancer and a biomarker for improving patients' survival.

文摘With the rapid development of electronic information technology,the waste electromagnetic radiation generated by the wireless and various other electronic devices has caused serious harm to human health.The development of high-efficiency flexible electromagnetic interference(EMI)shielding materials has become one of the most effective ways to mitigate the increasingly serious problem of electromagnetic pollution.Flexible carbon fiber composites possess most of the physical requirements needed for EMI shielding applications,including high electrical conductivity,large specific surface area,light weight,flexibility,porous structure,and adjustable physical and chemical properties,and various morphology of carbon fibers-based composites applied EMI.In this review,the primary EMI shielding mechanism of carbonfiber-based materials is presented.Since carbon fiber suffers from a low absorption efficiency when used for EMI shielding by itself,it is usually combined with additional EMI filling materials.Therefore,the methods of adding EMI filler materials to the carbon fiber framework and the influence of different fillers on the EMI shielding performance are also discussed.In addition,the novel structural designs of flexible carbon fiber-based EMI shielding materials,including fiber orientation,core-shell structure,and multilayer heterostructure,are summarized in terms of their classification,which includes composites with metal,polymer,nano-carbon,and MXenes.The review concludes with a brief discussion of the challenges,research directions,and future prospects of carbon-fiber-based EMI shielding materials.
