Glioblastoma(GBM)is a highly vascularized malignant brain tumor with poor clinical outcomes.Vasculogenic mimicry(VM)formed by aggressive GBM cells is an alternative approach for tumor blood supply and contributes to t...Glioblastoma(GBM)is a highly vascularized malignant brain tumor with poor clinical outcomes.Vasculogenic mimicry(VM)formed by aggressive GBM cells is an alternative approach for tumor blood supply and contributes to the failure of anti-angiogenic therapy.To date,there is still a lack of effective drugs that target VM formation in GBM.In the present study,we evaluated the effects of the plant cyclopeptide moroidin on VM formed by GBM cells and investigated its underlying molecular mechanisms.Moroidin significantly suppressed cell migration,tube formation,and the expression levels ofα-smooth muscle actin and matrix metalloproteinase-9 in human GBM cell lines at sublethal concentrations.The RNA sequencing data suggested the involvement of the epithelialmesenchymal transition(EMT)pathway in the mechanism of moroidin.Exposure to moroidin led to a concentration-dependent decrease in the expression levels of the EMT markers N-cadherin and vimentin in GBM cells.Moreover,moroidin significantly reduced the level of phosphorylated extracellular signal-regulated protein kinase(p-ERK)and inhibited the activation of β-catenin.Finally,we demonstrated that the plant cyclopeptide moroidin inhibited VM formation by GBM cells through inhibiting the ERK/β-catenin-mediated EMT.Therefore,our study indicates a potential application of moroidin as an anti-VM agent in the treatment of GBM.展开更多
Growing evidence suggests that the presence of cancer stem cells(CSCs)is a major challenge in current tumor treatments,especially the transition from non-CSCs to differentiation of CSCs for evading conventional therap...Growing evidence suggests that the presence of cancer stem cells(CSCs)is a major challenge in current tumor treatments,especially the transition from non-CSCs to differentiation of CSCs for evading conventional therapies and driving metastasis.Here we propose a therapeutic strategy of synergistic differentiation therapy and phototherapy to induce differentiation of CSCs into mature tumor cells by differentiation inducers and synergistic elimination of them and normal cancer cells through phototherapy.In this work,we synthesized a biomimetic nanoplatform loaded with IR-780 and all-trans retinoic acid(ATRA)via biomineralization.This method can integrate aluminum ions into small-sized protein carriers to form nanoclusters,which undergo responsive degradation under acidic conditions and facilitate deep tumor penetration.With the help of CSC differentiation induced by ATRA,IR-780 inhibited the self-renewal of CSCs and cancer progression by generating hyperthermia and reactive oxygen species in a synergistic manner.Furthermore,ATRA can boost immunogenic cell death induced by phototherapy,thereby strongly causing a systemic anti-tumor immune response and efficiently eliminating CSCs and tumor cells.Taken together,this dual strategy represents a new paradigm of targeted eradication of CSCs and tumors by inducing CSC differentiation,improving photothermal therapy/photodynamic therapy and enhancing antitumor immunity.展开更多
To support the explosive growth of Information and Communications Technology(ICT),Mobile Edge Comput-ing(MEC)provides users with low latency and high bandwidth service by offloading computational tasks to the network...To support the explosive growth of Information and Communications Technology(ICT),Mobile Edge Comput-ing(MEC)provides users with low latency and high bandwidth service by offloading computational tasks to the network’s edge.However,resource-constrained mobile devices still suffer from a capacity mismatch when faced with latency-sensitive and compute-intensive emerging applications.To address the difficulty of running computationally intensive applications on resource-constrained clients,a model of the computation offloading problem in a network consisting of multiple mobile users and edge cloud servers is studied in this paper.Then a user benefit function EoU(Experience of Users)is proposed jointly considering energy consumption and time delay.The EoU maximization problem is decomposed into two steps,i.e.,resource allocation and offloading decision.The offloading decision is usually given by heuristic algorithms which are often faced with the challenge of slow convergence and poor stability.Thus,a combined offloading algorithm,i.e.,a Gini coefficient-based adaptive genetic algorithm(GCAGA),is proposed to alleviate the dilemma.The proposed algorithm optimizes the offloading decision by maximizing EoU and accelerates the convergence with the Gini coefficient.The simulation compares the proposed algorithm with the genetic algorithm(GA)and adaptive genetic algorithm(AGA).Experiment results show that the Gini coefficient and the adaptive heuristic operators can accelerate the convergence speed,and the proposed algorithm performs better in terms of convergence while obtaining higher EoU.The simulation code of the proposed algorithm is available:https://github.com/Grox888/Mobile_Edge_Computing/tree/GCAGA.展开更多

仙灵骨葆口服制剂是我国治疗骨病的中药大品种,但近年来国家食品药品监督管理总局通报其易引起肝损伤。传统毒理学研究很难评价其肝损伤作用,我们前期发现仙灵骨葆肝损伤具有特异质属性,拆方研究发现补骨脂和淫羊藿是其导致特异质肝损...仙灵骨葆口服制剂是我国治疗骨病的中药大品种,但近年来国家食品药品监督管理总局通报其易引起肝损伤。传统毒理学研究很难评价其肝损伤作用,我们前期发现仙灵骨葆肝损伤具有特异质属性,拆方研究发现补骨脂和淫羊藿是其导致特异质肝损伤的主要药味,且配伍后其特异质肝损伤加重,呈现七情配伍中“相反”的特点。进一步研究发现,TNF-α介导的免疫应激是其重要诱因,免疫促进成分和肝损伤易感成分存在是其另一重要诱因。然而,具体机制尚不清楚。本研究通过建立动物模型,考察了免疫促进淫羊藿次苷Ⅱ联合肝损伤易感成分补骨脂甲素对小鼠肝损伤的影响;借助非靶向代谢组学技术评价了两个成分联合对肝损伤代谢标志物的影响;利用16S r RNA测序技术探讨了肠道菌群的物种组成和相对丰度的变化。结果表明在TNF-α诱导的免疫应激小鼠模型上发现,单独给药补骨脂甲素能够引起明显肝损伤,而淫羊藿次苷Ⅱ组却无明显变化,但是淫羊藿次苷Ⅱ能够协同补骨脂甲素导致特异质肝损伤;同时代谢组学结果揭示补骨脂甲素联合淫羊藿次苷Ⅱ能够引起小鼠肝脏中甲基氨基甲酰PAF、吲哚-3-醋酸甲酯等代谢物的水平升高,而甘氨酸-酪氨酸(Gly-Tyr)、L-亮氨酰-L-甘氨酸(Leu-Gly)和L-色氨酸-L-丝氨酸(Trp-Ser)等代谢物的水平则下降。这些与肝损伤相关的差异表达代谢物主要富集在鞘脂代谢、鞘脂信号通路和坏死等代谢途径。值得注意的是,补骨脂甲素联合淫羊藿次苷Ⅱ可以诱导肝损伤小鼠肠道中的乳酸杆菌和脱硫弧菌科丰度显著增加。相关性分析结果表明,类杆菌科和脱硫弧菌科与甲基氨基甲酰基PAF和吲哚-3-醋酸甲酯呈正相关,而与Gly-Tyr、Leu-Gly和Trp-Ser呈负相关。本研究初步阐明了补骨脂配伍淫羊藿引起特异质型药物性肝损伤的物质基础和机制,为中成药临床合理使用提供了科学依据。展开更多
Flower type is an important and extremely complicated trait of chrysanthemum.The corolla tube merged degree(CTMD)and the relative number of ray florets(RNRF)are the two key factors affecting chrysanthemum flower type....Flower type is an important and extremely complicated trait of chrysanthemum.The corolla tube merged degree(CTMD)and the relative number of ray florets(RNRF)are the two key factors affecting chrysanthemum flower type.However,few reports have clarified the inheritance of these two complex traits,which limits directed breeding for flower-type improvement.In this study,305 F1 hybrids were obtained from two parents with obvious differences in CTMD and RNRF performance.Using specific-locus amplified fragment sequencing(SLAF-seq)technology,we constructed a high-density genetic linkage map with an average map distance of 0.76 cM.Three major QTLs controlling CTMD and four major QTLs underlying RNRF were repeatedly detected in the 2 years.Moreover,the synteny between the genetic map and other Compositae species was investigated,and weak collinearity was observed.In QTL regions with a high degree of genomic collinearity,eight annotated genes were probed in the Helianthus annuus L.and Lactuca sativa L.var.ramosa Hort.genomes.Furthermore,20 and 11 unigenes were identified via BLAST searches between the SNP markers of the QTL regions and the C.vestitum and C.lavandulifolium transcriptomes,respectively.These results lay a foundation for molecular marker-assisted breeding and candidate gene exploration in chrysanthemum without a reference assembly.展开更多
Efficient and stable bifunctional electrocatalysts for water splitting is essential for producing hydrogen and alleviating huge energy consumption.Meanwhile,charge transfer engineering is an efficient approach to modu...Efficient and stable bifunctional electrocatalysts for water splitting is essential for producing hydrogen and alleviating huge energy consumption.Meanwhile,charge transfer engineering is an efficient approach to modulate the localized electronic properties of catalysts and tune the electrocatalytic performance.Herein,we tactfully fabricate PtFeNi alloys/NiFe layered double hydroxides(LDHs)heterostructure by an easily electrochemical way with a small amount of Pt.The experimental and theoretical results unravel that the charge transfer on the alloy clusters modulated by the defective substrates(NiFe LDHs),which synergistically optimizes the adsorption energy of the reaction intermediates.The electrocatalyst exhibits an ultra‐low overpotential of 81 and 243 mV at the current density of 100 mA cm^(–2) for hydrogen evolution and oxygen evolution,respectively.Furthermore,the overall water splitting indicates that PtFeNi alloys/NiFe LDHs presents an ultra‐low overpotential of 265 and 406 mV to reach the current density of 10 and 300 mA cm^(–2),respectively.It proves that the PtFeNi alloys/NiFe LDHs catalyst is an excellent dual‐function electrocatalyst for water splitting and promising for industrialization.This work provides a new electrochemical approach to construct the alloy heterostructure.The prepared heterostructures act as an ideal platform to investigate the charge re‐distribution behavior and to improve the electrocatalytic activity.展开更多
Surface segregation is ubiquitous in multi-component materials and is of great important for catalysis but little is known on the surface structure under graphene encapsulation.Here,we show that the graphene encapsula...Surface segregation is ubiquitous in multi-component materials and is of great important for catalysis but little is known on the surface structure under graphene encapsulation.Here,we show that the graphene encapsulated CoCu performs well for the electrocatalytic oxidation of 5-hydroxymethylfurfural(HMF)to2,5-furandicarboxylic acid(FDCA)with the onset potential before 1.23 VRHEand a nearly 100%selectivity of FDCA under 1.4 VRHE.From the experimental results,the unprecedented catalytic performance was attributed to local structural distortion and sub-nanometer lattice composition of the CoCu surface.We accurately show the dispersed Cu doped Co_(3)O_(4) nano-islands with a lot of edge sites on the bimetallic Co-Cu surface.While,the gradient components effectively facilitate the establishment of built-in electric field and accelerate the charge transfer.Theoretical and experimental results reveal that the surface Co and neighbouring Cu atoms in sub-nanometer lattice synergistically promote the catalysis of HMF.This work offers new insights into surface segregation in tuning the element spatial distribution for catalysis.展开更多
Grain number and seed-setting rate are components of crop yield.Cytokinin influences grain yield.However,emerging studies suggest that high cytokinin signals often lead to reduced branching or seed-setting rate,leadin...Grain number and seed-setting rate are components of crop yield.Cytokinin influences grain yield.However,emerging studies suggest that high cytokinin signals often lead to reduced branching or seed-setting rate,leading to reduced grain yield,although the mechanisms remain unclear.In this study,we identified and characterized the rice(Oryza sativa L.)gene LONELY GUY-LIKE 8(LOGL8),based on analysis of the LOGL8-pm(promoter mutant of LOGL8)mutant,which harbors a T-DNA insertion in the promoter of this gene.The mutation in LOGL8-pm causes ectopic hyperexpression of LOGL8 in inflorescence organs,resulting in plants with smaller panicles and defective ovules lacking archesporial cells and integuments.Knockout of LOGL8 caused pollen abortion,leading to a reduced seed-setting rate.LOGL8 encodes a putative cytokinin-activating enzyme.Our results showed that LOGL8 directly catalyzes the biosynthesis of bioactive cytokinins.Therefore,we propose that the ectopic expression of LOGL8 disrupts cytokinin spatiotemporal distribution and causes inhibition of LONELY GUY(LOG),which affects panicle branching and female organ development.These findings reveal the important role of LOGL8 in male development,and highlight the delicate balance of local cytokinin levels during panicle branching and female organ development.展开更多

However, due to the added complexity in the two-crystal assembly in which the spectra might be diffracted by two crystals, some additional impurity lines were identified. For example,tungsten(W) lines in different ionization states were diffracted by the He-like and H-like crystal.Additional molybdenum(Mo) lines in the wavelength range of He-like and H-like argon spectra lines were also summarized. The existence of these additional lines caused the fitted temperature to be different from the true values. This paper presents the identified lines through a comparison with available database, which should be included in the fitting procedure.展开更多
The equilibrium reconstruction is important to study the tokamak plasma physical processes.To analyze the contribution of fast ions to the equilibrium,the kinetic equilibria at two time-slices in a typical H-mode disc...The equilibrium reconstruction is important to study the tokamak plasma physical processes.To analyze the contribution of fast ions to the equilibrium,the kinetic equilibria at two time-slices in a typical H-mode discharge with different auxiliary heatings are reconstructed by using magnetic diagnostics,kinetic diagnostics and TRANSP code.It is found that the fast-ion pressure might be up to one-third of the plasma pressure and the contribution is mainly in the core plasma due to the neutral beam injection power is primarily deposited in the core region.The fast-ion current contributes mainly in the core region while contributes little to the pedestal current.A steep pressure gradient in the pedestal is observed which gives rise to a strong edge current.It is proved that the fast ion effects cannot be ignored and should be considered in the future study of EAST.展开更多

Methods: Immun...Objective: The aim of the study was to investigate the human epidermal growth factor receptor 2(HER2) gene amplification and protein expression and interpretation points in the stomach mixed carcinomas. Methods: Immunohistochemistry(IHC) and fluorescence in situ hybridization(FISH) technique were used to detect HER2 gene amplification and expression of HER2 protein in 442 cases of gastric mixed carcinoma. Results: The expression rate of HER2 protein was 41.2%(182/442): the HER2 protein expression IHC 3+ extensive type in 18 cases, partial type in 21 cases, focal type in 8 cases, accounting for 10.6%(47/442); the HER2 protein expression IHC 2+ extensive type in 23 cases, partial type in 28 cases, focal type in 11 cases, accounting for 14.0%(62/442); the HER2 protein expression IHC 1+ extensive type in 27 cases, partial type in 31 cases, focal type in 15 cases, accounting for 16.5%(73/442). HER2 gene amplification rate of 442 cases was 16.1%(71/442). In 182 cases of HER2 protein positive expression, the HER2 gene cluster amplification rate was 14.8%(27/182), large granular amplification rate 11.0%(20/182), punctate amplification rate 6.0%(11/182) and high polysomy 7.1%(13/182). In 71 cases of HER2 gene amplification, there was 42 cases of HER2 protein expression IHC 3+, 22 cases of HER2 protein expression IHC 2+, and 7 cases of IHC 1+. Conclusion: HER2 detection of gastric mixed carcinoma has great heterogeneity, HER2 protein positive expression is divided into extensive type, partial type and focal type, and HER2 gene positive amplification is divided into cluster amplification, large granular amplification, punctate amplification and high polysomy. These typing of HER2 protein expression and HER2 gene amplification provide reference index to quantify for targeted therapeutic effect of anticancer drugs.展开更多

The expression rates of p16, p53, Bcl-2, and c-erb B-2 in cervical squamous cell carcinoma tissues were 60.0%(12/20), 60.0%(12/20), 75.0%(15/20), and 65.0%(13/20), respectively. The expression rates of p16, p53, Bcl-2, and c-erb B-2 in normal cervical tissues were 0.0%(0/20), 0.0%(0/20), 35.0%(7/20), 0.0%(0/20), respectively. In comparison to normal tissues, the differential expressions of p16, p53, and Bcl-2 in squamous cell carcinoma and CIN were statistically significant(P <0.001), whereas the difference between the squamous cell carcinoma and CIN was not significant(P >0.05). In comparison to normal tissues, the differential expressions of c-erb B-2 and p53 in squamous cell carcinoma and CIN were statistically significant(P >0.05). The differential expressions of Bcl-2, c-erb B-2, and p53 in CIN 3 were statistically significant in comparison to CIN 1 and CIN 2(P <0.05). Conclusion Overexpression of Bcl-2 occurs early in the development of cervical cancer, whereas p16 and c-erb B-2 overexpression are markers for cell malignancy. The expression of p53 is correlated with the development of cervical cancer.展开更多
The current landscape of chiral covalent organic frameworks(COFs)predominantly centered on constructing asymmetric molecular-scale chirality,often introducing an inherent contradiction to the COF symmetry and limiting...The current landscape of chiral covalent organic frameworks(COFs)predominantly centered on constructing asymmetric molecular-scale chirality,often introducing an inherent contradiction to the COF symmetry and limiting diversity.Herein,we overcome these challenges by achieving chiral transfer between one-dimensional(1D)imine linear polymers(LPs)and two-dimensional(2D)networkβ-ketoenamine COFs composed of achiral monomers.We successfully synthesize several 1D imine LPs with mesoscopic helical chirality,comprising achiral C2-symmetric terephthalaldehyde and diamine linkers in a chiral supramolecular transcription system.Leveraging the irreversible tautomerism mechanism within the linker replacement approach,terephthalaldehyde(TPA)units in these helical 1D LPs are substituted with C3-symmetric 1,3,5-triformylphloroglucinol(TP),yielding the corresponding 2D networkβ-ketoenamine COFs.Crystallinity and helicity of the resultantβ-ketoenamine COFs intimately hinge on reaction conditions,including the aldehyde stoichiometry of Tp and TPA,as well as the quantity and concentration of the catalyst employed.Under optimized conditions,the nucleation and growth were precisely governed,achieving a harmonious equilibrium of crystallinity and helicity within the generated 2D networkβ-ketoenamine COFs,even with covalent bond rupture,recombination,and topological transition(from[C2+C2]to[C3+C2]).Impressively,the ground state chirality inherent to helical 1D LPs seamlessly transfers to helical 2D networkβ-ketoenamine COFs.This study not only offers new perspectives on the development of chiral functional COFs,but also provides fresh insights into the precise control of COFs'microscopic morphology.展开更多

Objective:A typical case of Xianling Gubao(XLGB)Tablets-induced liver injury was systematically studied in the clinic and the laboratory.Methods:A patient with herb-induced liver injury(HILI)and a history of taking XL...Objective:A typical case of Xianling Gubao(XLGB)Tablets-induced liver injury was systematically studied in the clinic and the laboratory.Methods:A patient with herb-induced liver injury(HILI)and a history of taking XLGB Tablets before disease onset was engaged as the study subject,and the case was diagnosed according to the updated Roussel Uclaf Causality Assessment Method(RUCAM)and the integrated evidence chain(iEC)method recommended by the Guidelines for Diagnosis and Treatment of Herb-induced Liver Injury(HILI Guidelines).Results:Clinical history,biochemical indexes and imaging tests were used to exclude the influence of fundamental diseases and confusing liver diseases such as viral,alcoholic and autoimmune liver diseases on the diagnosis.Based on an investigation of the patient’s medication history,she was suspected to have HILI caused by XLGB Tablets,as the patient was only taking an oral preparation of XLGB Tablets,and the influence of other drugs on the diagnosis was excluded.This patient with alanine aminotransferase(ALT)3 upper limit of normal(ULN)and a calculated R of 6 was diagnosed with possible acute drug-induced hepatocellular injury.The relationship was considered“highly probable”(score of 9)using the updated RUCAM of 2016.Moreover,the fingerprint similarity between the preparation taken by the patient and a commercially available preparation was 0.99,suggesting that the patient was consuming XLGB Tablets rather than another drug.LC-MS technology and the Agilent Fake TCM-Drugs database were used to investigate the drug,and no chemical additions were found.Examination of the drug for pesticide residues,heavy metals,aflatoxins and other exogenous substances indicated compliance with the content limits of the Chinese Pharmacopoeia.Conclusion:In summary,the final diagnosis of XLGB-induced liver injury reached the clinical diagnosis of HILI and was acute severe hepatocellular injury type by the updated RUCAM and iEC.Therefore,this study provides scientific evidence regarding the causality evaluation of compound preparations of traditional Chinese medicines-induced liver injury.展开更多

文摘Glioblastoma(GBM)is a highly vascularized malignant brain tumor with poor clinical outcomes.Vasculogenic mimicry(VM)formed by aggressive GBM cells is an alternative approach for tumor blood supply and contributes to the failure of anti-angiogenic therapy.To date,there is still a lack of effective drugs that target VM formation in GBM.In the present study,we evaluated the effects of the plant cyclopeptide moroidin on VM formed by GBM cells and investigated its underlying molecular mechanisms.Moroidin significantly suppressed cell migration,tube formation,and the expression levels ofα-smooth muscle actin and matrix metalloproteinase-9 in human GBM cell lines at sublethal concentrations.The RNA sequencing data suggested the involvement of the epithelialmesenchymal transition(EMT)pathway in the mechanism of moroidin.Exposure to moroidin led to a concentration-dependent decrease in the expression levels of the EMT markers N-cadherin and vimentin in GBM cells.Moreover,moroidin significantly reduced the level of phosphorylated extracellular signal-regulated protein kinase(p-ERK)and inhibited the activation of β-catenin.Finally,we demonstrated that the plant cyclopeptide moroidin inhibited VM formation by GBM cells through inhibiting the ERK/β-catenin-mediated EMT.Therefore,our study indicates a potential application of moroidin as an anti-VM agent in the treatment of GBM.

文摘Growing evidence suggests that the presence of cancer stem cells(CSCs)is a major challenge in current tumor treatments,especially the transition from non-CSCs to differentiation of CSCs for evading conventional therapies and driving metastasis.Here we propose a therapeutic strategy of synergistic differentiation therapy and phototherapy to induce differentiation of CSCs into mature tumor cells by differentiation inducers and synergistic elimination of them and normal cancer cells through phototherapy.In this work,we synthesized a biomimetic nanoplatform loaded with IR-780 and all-trans retinoic acid(ATRA)via biomineralization.This method can integrate aluminum ions into small-sized protein carriers to form nanoclusters,which undergo responsive degradation under acidic conditions and facilitate deep tumor penetration.With the help of CSC differentiation induced by ATRA,IR-780 inhibited the self-renewal of CSCs and cancer progression by generating hyperthermia and reactive oxygen species in a synergistic manner.Furthermore,ATRA can boost immunogenic cell death induced by phototherapy,thereby strongly causing a systemic anti-tumor immune response and efficiently eliminating CSCs and tumor cells.Taken together,this dual strategy represents a new paradigm of targeted eradication of CSCs and tumors by inducing CSC differentiation,improving photothermal therapy/photodynamic therapy and enhancing antitumor immunity.
文摘To support the explosive growth of Information and Communications Technology(ICT),Mobile Edge Comput-ing(MEC)provides users with low latency and high bandwidth service by offloading computational tasks to the network’s edge.However,resource-constrained mobile devices still suffer from a capacity mismatch when faced with latency-sensitive and compute-intensive emerging applications.To address the difficulty of running computationally intensive applications on resource-constrained clients,a model of the computation offloading problem in a network consisting of multiple mobile users and edge cloud servers is studied in this paper.Then a user benefit function EoU(Experience of Users)is proposed jointly considering energy consumption and time delay.The EoU maximization problem is decomposed into two steps,i.e.,resource allocation and offloading decision.The offloading decision is usually given by heuristic algorithms which are often faced with the challenge of slow convergence and poor stability.Thus,a combined offloading algorithm,i.e.,a Gini coefficient-based adaptive genetic algorithm(GCAGA),is proposed to alleviate the dilemma.The proposed algorithm optimizes the offloading decision by maximizing EoU and accelerates the convergence with the Gini coefficient.The simulation compares the proposed algorithm with the genetic algorithm(GA)and adaptive genetic algorithm(AGA).Experiment results show that the Gini coefficient and the adaptive heuristic operators can accelerate the convergence speed,and the proposed algorithm performs better in terms of convergence while obtaining higher EoU.The simulation code of the proposed algorithm is available:https://github.com/Grox888/Mobile_Edge_Computing/tree/GCAGA.

文摘Ultrafast charge exchange recombination spectroscopy(UF-CXRS)has been developed on the EAST tokamak(Yingying Li et al 2019 Fusion Eng.Des.146522)to measure fast evolutions of ion temperature and toroidal velocity.Here,we report the preliminary diagnostic measurements after relative sensitivity calibration.The measurement results show a much higher temporal resolution compared with conventional CXRS,benefiting from the usage of a prismcoupled,high-dispersion volume-phase holographic transmission grating and a high quantum efficiency,high-gain detector array.Utilizing the UF-CXRS diagnostic,the fast evolutions of the ion temperature and rotation velocity during a set of high-frequency small-amplitude edgelocalized modes(ELMs)are obtained on the EAST tokamak,which are then compared with the case of large-amplitude ELMs.

文摘仙灵骨葆口服制剂是我国治疗骨病的中药大品种,但近年来国家食品药品监督管理总局通报其易引起肝损伤。传统毒理学研究很难评价其肝损伤作用,我们前期发现仙灵骨葆肝损伤具有特异质属性,拆方研究发现补骨脂和淫羊藿是其导致特异质肝损伤的主要药味,且配伍后其特异质肝损伤加重,呈现七情配伍中“相反”的特点。进一步研究发现,TNF-α介导的免疫应激是其重要诱因,免疫促进成分和肝损伤易感成分存在是其另一重要诱因。然而,具体机制尚不清楚。本研究通过建立动物模型,考察了免疫促进淫羊藿次苷Ⅱ联合肝损伤易感成分补骨脂甲素对小鼠肝损伤的影响;借助非靶向代谢组学技术评价了两个成分联合对肝损伤代谢标志物的影响;利用16S r RNA测序技术探讨了肠道菌群的物种组成和相对丰度的变化。结果表明在TNF-α诱导的免疫应激小鼠模型上发现,单独给药补骨脂甲素能够引起明显肝损伤,而淫羊藿次苷Ⅱ组却无明显变化,但是淫羊藿次苷Ⅱ能够协同补骨脂甲素导致特异质肝损伤;同时代谢组学结果揭示补骨脂甲素联合淫羊藿次苷Ⅱ能够引起小鼠肝脏中甲基氨基甲酰PAF、吲哚-3-醋酸甲酯等代谢物的水平升高,而甘氨酸-酪氨酸(Gly-Tyr)、L-亮氨酰-L-甘氨酸(Leu-Gly)和L-色氨酸-L-丝氨酸(Trp-Ser)等代谢物的水平则下降。这些与肝损伤相关的差异表达代谢物主要富集在鞘脂代谢、鞘脂信号通路和坏死等代谢途径。值得注意的是,补骨脂甲素联合淫羊藿次苷Ⅱ可以诱导肝损伤小鼠肠道中的乳酸杆菌和脱硫弧菌科丰度显著增加。相关性分析结果表明,类杆菌科和脱硫弧菌科与甲基氨基甲酰基PAF和吲哚-3-醋酸甲酯呈正相关,而与Gly-Tyr、Leu-Gly和Trp-Ser呈负相关。本研究初步阐明了补骨脂配伍淫羊藿引起特异质型药物性肝损伤的物质基础和机制,为中成药临床合理使用提供了科学依据。

文摘Flower type is an important and extremely complicated trait of chrysanthemum.The corolla tube merged degree(CTMD)and the relative number of ray florets(RNRF)are the two key factors affecting chrysanthemum flower type.However,few reports have clarified the inheritance of these two complex traits,which limits directed breeding for flower-type improvement.In this study,305 F1 hybrids were obtained from two parents with obvious differences in CTMD and RNRF performance.Using specific-locus amplified fragment sequencing(SLAF-seq)technology,we constructed a high-density genetic linkage map with an average map distance of 0.76 cM.Three major QTLs controlling CTMD and four major QTLs underlying RNRF were repeatedly detected in the 2 years.Moreover,the synteny between the genetic map and other Compositae species was investigated,and weak collinearity was observed.In QTL regions with a high degree of genomic collinearity,eight annotated genes were probed in the Helianthus annuus L.and Lactuca sativa L.var.ramosa Hort.genomes.Furthermore,20 and 11 unigenes were identified via BLAST searches between the SNP markers of the QTL regions and the C.vestitum and C.lavandulifolium transcriptomes,respectively.These results lay a foundation for molecular marker-assisted breeding and candidate gene exploration in chrysanthemum without a reference assembly.
文摘Efficient and stable bifunctional electrocatalysts for water splitting is essential for producing hydrogen and alleviating huge energy consumption.Meanwhile,charge transfer engineering is an efficient approach to modulate the localized electronic properties of catalysts and tune the electrocatalytic performance.Herein,we tactfully fabricate PtFeNi alloys/NiFe layered double hydroxides(LDHs)heterostructure by an easily electrochemical way with a small amount of Pt.The experimental and theoretical results unravel that the charge transfer on the alloy clusters modulated by the defective substrates(NiFe LDHs),which synergistically optimizes the adsorption energy of the reaction intermediates.The electrocatalyst exhibits an ultra‐low overpotential of 81 and 243 mV at the current density of 100 mA cm^(–2) for hydrogen evolution and oxygen evolution,respectively.Furthermore,the overall water splitting indicates that PtFeNi alloys/NiFe LDHs presents an ultra‐low overpotential of 265 and 406 mV to reach the current density of 10 and 300 mA cm^(–2),respectively.It proves that the PtFeNi alloys/NiFe LDHs catalyst is an excellent dual‐function electrocatalyst for water splitting and promising for industrialization.This work provides a new electrochemical approach to construct the alloy heterostructure.The prepared heterostructures act as an ideal platform to investigate the charge re‐distribution behavior and to improve the electrocatalytic activity.

文摘Surface segregation is ubiquitous in multi-component materials and is of great important for catalysis but little is known on the surface structure under graphene encapsulation.Here,we show that the graphene encapsulated CoCu performs well for the electrocatalytic oxidation of 5-hydroxymethylfurfural(HMF)to2,5-furandicarboxylic acid(FDCA)with the onset potential before 1.23 VRHEand a nearly 100%selectivity of FDCA under 1.4 VRHE.From the experimental results,the unprecedented catalytic performance was attributed to local structural distortion and sub-nanometer lattice composition of the CoCu surface.We accurately show the dispersed Cu doped Co_(3)O_(4) nano-islands with a lot of edge sites on the bimetallic Co-Cu surface.While,the gradient components effectively facilitate the establishment of built-in electric field and accelerate the charge transfer.Theoretical and experimental results reveal that the surface Co and neighbouring Cu atoms in sub-nanometer lattice synergistically promote the catalysis of HMF.This work offers new insights into surface segregation in tuning the element spatial distribution for catalysis.

文摘Grain number and seed-setting rate are components of crop yield.Cytokinin influences grain yield.However,emerging studies suggest that high cytokinin signals often lead to reduced branching or seed-setting rate,leading to reduced grain yield,although the mechanisms remain unclear.In this study,we identified and characterized the rice(Oryza sativa L.)gene LONELY GUY-LIKE 8(LOGL8),based on analysis of the LOGL8-pm(promoter mutant of LOGL8)mutant,which harbors a T-DNA insertion in the promoter of this gene.The mutation in LOGL8-pm causes ectopic hyperexpression of LOGL8 in inflorescence organs,resulting in plants with smaller panicles and defective ovules lacking archesporial cells and integuments.Knockout of LOGL8 caused pollen abortion,leading to a reduced seed-setting rate.LOGL8 encodes a putative cytokinin-activating enzyme.Our results showed that LOGL8 directly catalyzes the biosynthesis of bioactive cytokinins.Therefore,we propose that the ectopic expression of LOGL8 disrupts cytokinin spatiotemporal distribution and causes inhibition of LONELY GUY(LOG),which affects panicle branching and female organ development.These findings reveal the important role of LOGL8 in male development,and highlight the delicate balance of local cytokinin levels during panicle branching and female organ development.

文摘We have numerically and experimentally observed the soliton pulsation with obvious breathing behavior in the anomalous fiber laser mode-locked by a nonlinear polarization rotation technique.The numerical study of the soliton pulsation with breathing behavior was analyzed through the split-step Fourier method at first,and it was found that the phase difference caused by the polarization controller would affect the breathing characteristics.Then,taking advantage of the dispersive Fourier transform technique,we confirmed the breathing characteristic of soliton pulsation in the same fiber laser as the simulation model experimentally.These results complement the research on the breathing characteristic of soliton pulsation.

文摘To simultaneously measure the He-like and H-like argon spectra, a two-crystal assembly has been deployed to replace the previous single crystal on the tangential x-ray crystal spectrometer.By selecting appropriate crystals with similar Bragg angles, plasma temperature in the range of 0.5 keV≤Te≤10 keV and rotation can be diagnosed based on the He-like and H-like argon spectra. However, due to the added complexity in the two-crystal assembly in which the spectra might be diffracted by two crystals, some additional impurity lines were identified. For example,tungsten(W) lines in different ionization states were diffracted by the He-like and H-like crystal.Additional molybdenum(Mo) lines in the wavelength range of He-like and H-like argon spectra lines were also summarized. The existence of these additional lines caused the fitted temperature to be different from the true values. This paper presents the identified lines through a comparison with available database, which should be included in the fitting procedure.

文摘The equilibrium reconstruction is important to study the tokamak plasma physical processes.To analyze the contribution of fast ions to the equilibrium,the kinetic equilibria at two time-slices in a typical H-mode discharge with different auxiliary heatings are reconstructed by using magnetic diagnostics,kinetic diagnostics and TRANSP code.It is found that the fast-ion pressure might be up to one-third of the plasma pressure and the contribution is mainly in the core plasma due to the neutral beam injection power is primarily deposited in the core region.The fast-ion current contributes mainly in the core region while contributes little to the pedestal current.A steep pressure gradient in the pedestal is observed which gives rise to a strong edge current.It is proved that the fast ion effects cannot be ignored and should be considered in the future study of EAST.

文摘The excitation condition of reversed shear Alfven eigenmodes(RSAEs)has been investigated during sawtoothlike oscillation in the EAST tokamak.The sawtooth-like phenomena can be reproduced in the configuration of reversed magnetic shear,and the threshold gradient of electron temperature is formed accordingly,together with the increasing of the confinement of thermal particles.The distribution function of energetic ions density is altered dramatically when the neutral beam is switched from NBI1 L(tangent)to NBI1 R(perpendicular),which can be captured by the measurement of radial neutron camera.The RSAEs are excited easily in the vicinity of q_(min)(1.99 m≤R≤2.06 m)for the injection of neutral beam with perpendicular direction,which should be excited by the steep gradient of energetic ions density.Furthermore,the excitation of RSAEs and the formation of threshold gradient of electron temperature can take place concurrently,which means that the neutral beam with perpendicular injection is beneficial for the establishment of internal transport barrier.

文摘Coagulation Factor Xa(FXa)is the crucial enzyme at the convergent point of the intrinsic and extrinsic coagulation pathways.The inhibition of FXa is an effective approach against thrombotic diseases.In the present study,a specific strategy is reported to discover 10 novel FXa inhibitors based on ligand-based(pharmacophore)virtual screening and molecular docking analysis from a dataset of specs(containing 220000 molecules).The binding modes analysis provide insights into the contribution of particular structural moieties of the compounds towards their activity against FXa,and 10 novel structural compounds were discovered as potent candidate molecules.This work could be helpful in further design and development of FXa inhibitors.

文摘Objective: The aim of the study was to investigate the human epidermal growth factor receptor 2(HER2) gene amplification and protein expression and interpretation points in the stomach mixed carcinomas. Methods: Immunohistochemistry(IHC) and fluorescence in situ hybridization(FISH) technique were used to detect HER2 gene amplification and expression of HER2 protein in 442 cases of gastric mixed carcinoma. Results: The expression rate of HER2 protein was 41.2%(182/442): the HER2 protein expression IHC 3+ extensive type in 18 cases, partial type in 21 cases, focal type in 8 cases, accounting for 10.6%(47/442); the HER2 protein expression IHC 2+ extensive type in 23 cases, partial type in 28 cases, focal type in 11 cases, accounting for 14.0%(62/442); the HER2 protein expression IHC 1+ extensive type in 27 cases, partial type in 31 cases, focal type in 15 cases, accounting for 16.5%(73/442). HER2 gene amplification rate of 442 cases was 16.1%(71/442). In 182 cases of HER2 protein positive expression, the HER2 gene cluster amplification rate was 14.8%(27/182), large granular amplification rate 11.0%(20/182), punctate amplification rate 6.0%(11/182) and high polysomy 7.1%(13/182). In 71 cases of HER2 gene amplification, there was 42 cases of HER2 protein expression IHC 3+, 22 cases of HER2 protein expression IHC 2+, and 7 cases of IHC 1+. Conclusion: HER2 detection of gastric mixed carcinoma has great heterogeneity, HER2 protein positive expression is divided into extensive type, partial type and focal type, and HER2 gene positive amplification is divided into cluster amplification, large granular amplification, punctate amplification and high polysomy. These typing of HER2 protein expression and HER2 gene amplification provide reference index to quantify for targeted therapeutic effect of anticancer drugs.
文摘Objective To explore the clinical pathological features and immunophenotypes of cervical intraepithelial neoplasia(CIN).Methods The protein expression of p16, p53, Bcl-2, and c-erb B-2 in 59 cases of CIN, 20 cases of cervical squamous cell carcinoma, and 20 cases of normal cervical tissues were tested using immunohistochemistry staining.Results The expression rates of p16, p53, Bcl-2, and c-erb B-2 in CIN tissues were 76.3%(45/59), 28.85(17/59), 61.0%(36/59), and 40.0%(23/59), respectively. The expression rates of p16, p53, Bcl-2, and c-erb B-2 in cervical squamous cell carcinoma tissues were 60.0%(12/20), 60.0%(12/20), 75.0%(15/20), and 65.0%(13/20), respectively. The expression rates of p16, p53, Bcl-2, and c-erb B-2 in normal cervical tissues were 0.0%(0/20), 0.0%(0/20), 35.0%(7/20), 0.0%(0/20), respectively. In comparison to normal tissues, the differential expressions of p16, p53, and Bcl-2 in squamous cell carcinoma and CIN were statistically significant(P <0.001), whereas the difference between the squamous cell carcinoma and CIN was not significant(P >0.05). In comparison to normal tissues, the differential expressions of c-erb B-2 and p53 in squamous cell carcinoma and CIN were statistically significant(P >0.05). The differential expressions of Bcl-2, c-erb B-2, and p53 in CIN 3 were statistically significant in comparison to CIN 1 and CIN 2(P <0.05). Conclusion Overexpression of Bcl-2 occurs early in the development of cervical cancer, whereas p16 and c-erb B-2 overexpression are markers for cell malignancy. The expression of p53 is correlated with the development of cervical cancer.

文摘The current landscape of chiral covalent organic frameworks(COFs)predominantly centered on constructing asymmetric molecular-scale chirality,often introducing an inherent contradiction to the COF symmetry and limiting diversity.Herein,we overcome these challenges by achieving chiral transfer between one-dimensional(1D)imine linear polymers(LPs)and two-dimensional(2D)networkβ-ketoenamine COFs composed of achiral monomers.We successfully synthesize several 1D imine LPs with mesoscopic helical chirality,comprising achiral C2-symmetric terephthalaldehyde and diamine linkers in a chiral supramolecular transcription system.Leveraging the irreversible tautomerism mechanism within the linker replacement approach,terephthalaldehyde(TPA)units in these helical 1D LPs are substituted with C3-symmetric 1,3,5-triformylphloroglucinol(TP),yielding the corresponding 2D networkβ-ketoenamine COFs.Crystallinity and helicity of the resultantβ-ketoenamine COFs intimately hinge on reaction conditions,including the aldehyde stoichiometry of Tp and TPA,as well as the quantity and concentration of the catalyst employed.Under optimized conditions,the nucleation and growth were precisely governed,achieving a harmonious equilibrium of crystallinity and helicity within the generated 2D networkβ-ketoenamine COFs,even with covalent bond rupture,recombination,and topological transition(from[C2+C2]to[C3+C2]).Impressively,the ground state chirality inherent to helical 1D LPs seamlessly transfers to helical 2D networkβ-ketoenamine COFs.This study not only offers new perspectives on the development of chiral functional COFs,but also provides fresh insights into the precise control of COFs'microscopic morphology.

文摘To improve the estimation accuracy,a novel time delay estimation(TDE)method based on the closed-form offset compensation is proposed.Firstly,we use the generalized cross-correlation with phase transform(GCC-PHAT)method to obtain the initial TDE.Secondly,a signal model using normalized cross spectrum is established,and the noise subspace is extracted by eigenvalue decomposition(EVD)of covariance matrix.Using the orthogonal relation between the steering vector and the noise subspace,the first-order Taylor expansion is carried out on the steering vector reconstructed by the initial TDE.Finally,the offsets are compensated via simple least squares(LS).Compared to other state-of-the-art methods,the proposed method significantly reduces the computational complexity and achieves better estimation performance.Experiments on both simulation and real-world data verify the efficiency of the proposed approach.

文摘With its more sustainable production mode and higher healthy food supplying,cultivated meat is gradually regarded as the promising alternative meat food in future by human beings.Most of the countries in the planet had participated in the cut-throat competition for sustainable protein searching,especially for the regions with low food self-sufficiency rate.In this paper,by reviewing the commercial market participation,product development stage,and technical research progress related to cultivated meat in detail,we demonstrated the industrialization progress of cultivated meat and explored the challenges of cultivated meat in the near future.1)Seeded cell and serum free medium were the basic raw materials,which were directly related to the food safety regulation,production cost control and productivity improvement;2)Bioreactor and regulation were the cornerstone of industrialization,which were closely related to the capacity expansion,product marketing;3)Technological improvements in taste,texture,safety,scale,cost were the foundation of realizing product iterative upgrade and market acceptance.

文摘Objective:A typical case of Xianling Gubao(XLGB)Tablets-induced liver injury was systematically studied in the clinic and the laboratory.Methods:A patient with herb-induced liver injury(HILI)and a history of taking XLGB Tablets before disease onset was engaged as the study subject,and the case was diagnosed according to the updated Roussel Uclaf Causality Assessment Method(RUCAM)and the integrated evidence chain(iEC)method recommended by the Guidelines for Diagnosis and Treatment of Herb-induced Liver Injury(HILI Guidelines).Results:Clinical history,biochemical indexes and imaging tests were used to exclude the influence of fundamental diseases and confusing liver diseases such as viral,alcoholic and autoimmune liver diseases on the diagnosis.Based on an investigation of the patient’s medication history,she was suspected to have HILI caused by XLGB Tablets,as the patient was only taking an oral preparation of XLGB Tablets,and the influence of other drugs on the diagnosis was excluded.This patient with alanine aminotransferase(ALT)3 upper limit of normal(ULN)and a calculated R of 6 was diagnosed with possible acute drug-induced hepatocellular injury.The relationship was considered“highly probable”(score of 9)using the updated RUCAM of 2016.Moreover,the fingerprint similarity between the preparation taken by the patient and a commercially available preparation was 0.99,suggesting that the patient was consuming XLGB Tablets rather than another drug.LC-MS technology and the Agilent Fake TCM-Drugs database were used to investigate the drug,and no chemical additions were found.Examination of the drug for pesticide residues,heavy metals,aflatoxins and other exogenous substances indicated compliance with the content limits of the Chinese Pharmacopoeia.Conclusion:In summary,the final diagnosis of XLGB-induced liver injury reached the clinical diagnosis of HILI and was acute severe hepatocellular injury type by the updated RUCAM and iEC.Therefore,this study provides scientific evidence regarding the causality evaluation of compound preparations of traditional Chinese medicines-induced liver injury.
