Exosomes:the next-generation therapeutic platform for ischemic stroke

Current therapeutic strategies for ischemic stroke fall short of the desired objective of neurological functional recovery.Therefore,there is an urgent need to develop new methods for the treatment of this condition.Exosomes are natural cell-derived vesicles that mediate signal transduction between cells under physiological and pathological conditions.They have low immunogenicity,good stability,high delivery efficiency,and the ability to cross the blood–brain barrier.These physiological properties of exosomes have the potential to lead to new breakthroughs in the treatment of ischemic stroke.The rapid development of nanotechnology has advanced the application of engineered exosomes,which can effectively improve targeting ability,enhance therapeutic efficacy,and minimize the dosages needed.Advances in technology have also driven clinical translational research on exosomes.In this review,we describe the therapeutic effects of exosomes and their positive roles in current treatment strategies for ischemic stroke,including their antiinflammation,anti-apoptosis,autophagy-regulation,angiogenesis,neurogenesis,and glial scar formation reduction effects.However,it is worth noting that,despite their significant therapeutic potential,there remains a dearth of standardized characterization methods and efficient isolation techniques capable of producing highly purified exosomes.Future optimization strategies should prioritize the exploration of suitable isolation techniques and the establishment of unified workflows to effectively harness exosomes for diagnostic or therapeutic applications in ischemic stroke.Ultimately,our review aims to summarize our understanding of exosome-based treatment prospects in ischemic stroke and foster innovative ideas for the development of exosome-based therapies. 展开更多

Removal of cloud cover on the satellite remote sensing image can effectively improve the availability of remote sensing images. For thin cloud cover, support vector value contourlet transform is used to achieve multi-scale decomposition of the area of thin cloud cover on remote sensing images. Through enhancing coefficients of high frequency and suppressing coefficients of low frequency, the thin cloud is removed. For thick cloud cover, if the areas of thick cloud cover on multi-source or multi-temporal remote sensing images do not overlap, the multi-output support vector regression learning method is used to remove this kind of thick clouds. If the thick cloud cover areas overlap, by using the multi-output learning of the surrounding areas to predict the surface features of the overlapped thick cloud cover areas, this kind of thick cloud is removed. Experimental results show that the proposed cloud removal method can effectively solve the problems of the cloud overlapping and radiation difference among multi-source images. Potassium ion batteries(PIBs)are emerging as potential next-generation energy storage systems on account of their low cost and high theoretical energy density.Nevertheless,they also face challenges of low specific capacity and suboptimal cycling stability.Herein,we synthesize a cocoon-like P3-type K_(0.5)Mn_(0.7)Ni_(0.3)O_(2)(KMNO)cathode material by a self-template method.The KMNO cocoons possess a hierarchical layered architecture composed of nanoparticle stacking,which can accelerate the transport kinetics of potassium ions,mitigate the stress caused by K^(+)intercalation and deintercalation,and improve structural stability.In addition,Ni can not only alleviate the Jahn-Teller distortion and suppress the phase transition to stabilize the structure,but also act as an electrochemically active element,providing the capacity of two electrons from Ni2+to Ni4+.Combining the advantages of structure and nickel substitution,the P3-type KMNO cocoons are used for electrochemical performance testing of PIB cathodes,delivering an excellent rate capability of 57.1 m A h g^(-1)at 500 m A g^(-1)and a remarkable cycling stability of 77.0%over 300 cycles at 100 m A g^(-1).Impressively,the KMNO cocoons//pitch-derived soft carbon assembled full battery exhibits superior electrochemical performance with a reversible capacity of 79.7 m A h g^(-1)at 50 m A g^(-1).Moreover,ex-situ XRD also further reveals a solid solution phase reaction with a volume change of only 1.46%.This work furnishes a suitable approach to fabricating highperformance layered oxide cathodes for PIBs with outstanding cycling stability and rate capability. 展开更多

To investigate the effect of different dietary energy and protein levels on meat performance and meat quality of Jinghai yellow chickens, 480 43-day old Jinghai yellow chickens with similar weight were randomly divided into four experimental groups: experimental group 1 (protein 15%, metabolic energy 9.95 MJ/kg), experimental group 2 (protein 16%, metabolic energy 10.95 MJ/kg), experimental group 3 (protein 17%, metabolic energy 12.65 MJ/kg) and experimental group 4 ( protein 18%, metabolic energy 13.95 MJ/kg), respectively. All chickens were slaughtered at 112-day old. The breast and leg muscles of Jinghai yellow chickens were collected, to determine the slaughter performance, conventional meat quality and muscle chemical indicators. The results indicated that dressing-out percentage and eviscerated yield percentage in four experimental groups were above 87.27% and 67.00%, respectively; other slaughter performance indicators exhibited no significant differences among various groups (P 〉 0.05 ) ; breast muscle color of hens in experimental group 4 varied significantly from that in other three groups ( P 〈 0.05 ) ; leg muscle color of hens in experimental group 2 varied extremely significantly from that in other three groups ( P 〈 0.01 ) ; water-holding capacity of breast muscles of hens in experimental group 3 was significantly higher than that in experimental group 4 (P 〈 0.05 ) ; thiamine content of breast muscles of cocks in experimental group 3 was significandy higher than that in experimental group 2 ( P 〈 0.05 ) ; however, other properties exhibited no significant differenees among various groups (P 〉 0.05 ). 展开更多

It can increase the stability of the drug and enhance drug bioavailability. The selective targeting of nanoparticles can be achieved through enhanced permeability and retention effect and a conjugated specific ligand or through the effects of physiological conditions, such as pH and temperature. Nanoparticles can be prepared by using a wide range of materials and can be used to encapsulate chemotherapeutic agents to reduce toxicity, which can be used for imaging, therapy, and diagnosis. In this research, recent progress on nanoparticles as a targeted drug delivery system will be reviewed, including positive-targeting, negative-targeting, and physicochemical-targeting used as anticancer drug carriers. 展开更多

High crystallinity of TiOwas prepared by a modified alcohothermal method, in which titanium isopropoxide was used as the titania precursor, absolute ethanol as the reaction medium, and NHHCOas the raw materials for release of water, ammonia and carbon dioxides via in-situ decomposition. The X-ray powder diffraction(XRD) and transmission electron microscope(TEM) measurements showed that water and ammonia from the in-situ decomposition of NHHCOplayed an important role in conducting the size, shape, crystallinity and microstructure of TiO. The photoluminescence spectroscopy and photocurrent measurements indicated that enhanced crystallinity could hinder the recombination and promote the separation of electron-hole pairs in TiO, which contribute to the improvement of photocatalytic activity.Methyl orange photodegradation under UV light confirmed that high crystallinity of TiOdid present a high photocatalytic activity due to the effective separation of photoinduced charges. 展开更多

[Objectives]The paper was to investigate the distribution characteristics of soil total nitrogen in low-efficiency forest land in the northern windy desert area of Jingbian County.[Methods]The distribution of soil total nitrogen in the 0-40 cm soil layer of 5 towns in the northern windy desert area of Jingbian County was studied through field sampling and laboratory detection.[Results]The average soil total nitrogen contents of Hongdunjie Town,Haizetan Town,Huanghaojie Town,Ningtiaoliang Town,Dongkeng Town and windy desert area in the 0-20 cm soil layer were 0.259,0.224,0.242,0.248,0.431 and 0.275 g/kg,respectively.The soil total nitrogen content in Dongkeng Town was higher than those in other towns,while there was little difference among other regions.The average total nitrogen contents in the 20-40 cm soil layer were 0.239,0.285,0.113,0.262,0.349 and 0.241 g/kg,respectively.The soil total nitrogen content in Huanghaojie Town was slightly lower than those in other towns,while that in Dongkeng town was higher.The variation coefficient of soil total nitrogen content in the survey area was greater than 30%,and there was great difference in spatial distribution.With the increase of soil depth,there was little change in soil total nitrogen content,namely the soil total nitrogen contents in 5 towns and windy desert area were not statistically different in the 0-40 cm soil layer.According to the nutrient grading standard of the second national soil survey,the soil total nitrogen content in the survey area was in the deficiency grade.[Conclusions]The research will provide a scientific guidance for the healthy and sustainable development of vegetation in arid regions. 展开更多

Biocrusts(BSCs)are widely distributed in frozen ground regions on the Qinghai-Tibet Plateau,and they are considered an important component of cold ecosystems.However,the specific impacts of BSCs on frozen soil remains relatively unclear.The aim of our study was to clarify the influence of BSCs(light BSCs and dark BSCs in two different succession stages)on the physical properties and ecological stoichiometry characteristics of frozen soil.Our results showed that BSCs increased the silt particle content in 20–40 cm soil layer,leading to a decrease in soil bulk density.And the field water capacity increased about 10%–40% compared to bare land.Additionally,BSCs significantly increased the contents of soil organic carbon(SOC,22.6–30.8 g kg^(-1))and total nitrogen(TN,2.1–2.8 g kg^(-1))in the upper 40 cm soil layer,both of them were approximately 1.3–2.0 and 1.3–4.0 times higher than those observed in bare land.However,BSCs did not have significant influence on soil total phosphorus(TP).BSCs had a significant impact on the stoichiometric ratios within 40 cm.The C/N ratios of the two types of BSCs ranged from 8.8 to 13.5,the C/P ratios ranged from 6.6 to 13.8,and the N/P ratios ranged from 0.6 to 1.2,which were all higher than those of the bare land.There were no significant differences among the C/N,C/P,and N/P ratios between two types of BSCs.However,the increment of C/P and N/P ratios of dark BSCs were significantly higher than those of light BSCs within 0–30 cm,which indicated that a reduction in the availability of phosphorus during the later stages of BSCs succession.These findings provided a theoretical basis for further research on the ecological functions of BSCs in frozen ground regions. 展开更多

In this review, we examine the current state of research on ginseng, including the main active ingredient ginsenoside, its pharmacological effects on the cardiovascular system, and mechanisms of action. We focus on what is known of the effects of ginseng against atherosclerosis, arrhythmia, myocardial ischemia, and its inhibition of ventricular remodeling, providing a basis for expanding the clinical applications of ginseng. 展开更多

Helicobacter pylori(H.pylori),creating a global infection rate over 50%,presents great challenges in clinical therapies due to its complex pathological microenvironment in vivo.To improve the eradication efficacy,herein we fabricated a pharmaceutical vesicle RHL/Cl-Ch-cal where cholesterol-PEG,calcitriol and first-line antibiotic clarithromycin were co-loaded in the rhamnolipid-composed outer lipid layer.RHL/Cl-Ch-cal could quickly penetrate through gastric mucus layer to reach H.pylori infection sites,and then effectively destroyed the architecture of H.pylori biofilms,killed dispersed H.pylori and inhibited the re-adhesion of residual bacteria(called biofilms eradication tetralogy).Moreover,RHL/Cl-Ch-cal activated the host immune response to H.pylori by replenishing cholesterol to repair lipid raft on the cell membrane of host epithelial cells.Finally,RHL/Cl-Ch-cal killed the intracellular H.pylori through recovering the lysosomal acidification and assisting degradation.In experiments,RHL/Cl-Ch-cal demonstrated prominent anti-H.pylori efficacy in the classical H.pylori-infected mice model.Therefore,the study provides a“comprehensive attack”strategy for anti-H.pylori therapies including biofilms eradication tetralogy,immune activation and intracellular bacteria killing. 展开更多

The diversity of body plans of mammals accelerates the innovation of lifestyles and the extensive adaptation to different habitats,including terrestrial,aerial and aquatic habitats.However,the genetic basis of those phenotypic modifications,which have occurred during mammalian evolution,remains poorly explored.In the present study,we synthetically surveyed the evolutionary pattern of Hox clusters that played a powerful role in the morphogenesis along the head-tail axis of animal embryos and the main regulatory factors(Mll,Bmi1 and E2f6)that control the expression of Hox genes.A deflected density of repetitive elements and lineage-specific radical mutations of Mll have been determined in marine mammals with morphological changes,suggesting that evolutionary changes may alter Hox gene expression in these lineages,leading to the morphological modification of these lineages.Although no positive selection was detected at certain ancestor nodes of lineages,the increasedωvalues of Hox genes implied the relaxation of functional constraints of these genes during the mammalian evolutionary process.More importantly,49 positively-selected sites were identified in mammalian lineages with phenotypic modifications,indicating adaptive evolution acting on Hox genes and regulatory factors.In addition,3 parallel amino acid substitutions in some Hox genes were examined in marine mammals,which might be responsible for their streamlined body. 展开更多

Well-dispersed BaSO4 nanoparticles were synthesized in the presence of sodium polyacrylate (PAAS) by a simple precipitation method, with BaCl2 and (NH4)2SO4 as reactants. The different roles performed by PAAS in the synthesis of BaSO4 nanoparticles were investigated using X-ray diffractometry, Fourier transform infrared spectroscopy, and transmission electron microscopy. The results indicate that the assynthesized BaSO4 nanoparticles were spheres with an average diameter of 30 nm and that their surfaces were affected by the PAAS. Under a typical procedure employed, PAAS reacted with BaCl2 to yield an intermediate, serving as a control releasing agent and separating the nucleation and crystal growth processes of the BaSO4 nuclei. During formation of the BaSO4 nanospheres, the intermediate slowly dissolved and released barium and polyacrylate ions, inhibiting the growth and aggregation of newly formed BaSO4 seeds and resulting in particles of narrow diameter distribution and improved dispersibility. Moreover, these polyacrylate ions further modified the surfaces of the BaSO4 nanoparticles. 展开更多

This study evaluated the effects of commercial starter cultures on the microbial counts,protein degradation,biogenic amines and volatile compounds in fermented fish-chili paste.Four inoculated groups with different starter cultures and one group without inoculation were investigated.The results revealed that the application of mixed starter cultures promoted the growth of lactic acid bacteria and yeast,while effectively inhibited the Enterbacteriaceae counts compared with the spontaneously fermented group.The inoculation with mixed starter cultures resulted in higher levels of umami-tasting free amino acids(FAAs)in all inoculated groups.Highest content of sweet-tasting FAAs was found in group HS(Pichia anomala+Staphylococcus xylosus+Staphylococcus carnosus+Pediococcus pentosaceus+Pediococcus acidilactici),and much lower bitter-tasting components were presented in group HW(Pichia anomala+Staphylococcus xylosus+Staphylococcus carnosus),HS and HV(Pichia anomala+Lactobacillus plantarum).In addition,total levels of biogenic amines detected in all inoculated samples were significantly lower compared with control,especially the content of tyramine(P<0.05).For the volatile compounds,more esters as well as less undesirable odor substances such as aldehydes,were exhibited in four inoculated groups. 展开更多

Metal sulfides have been widely investigated as promising electrode materials for potassium-ion batteries(PIBs)due to their high theoretical capacities.However,the practical application of metal sulfides in PIBs is still hindered by their intrinsic shortcomings of low conductivity and severe volume changes during the potassiation/depotassiation process.Herein,a simple template-based two-step annealing strategy is proposed to impregnate CoS_(2) nanoparticles in the well-structured carbon nanocubes(denoted CoS_(2)/CNCs)as an advanced anode material for PIBs.The ex-situ XRD measurements reveal the K+storage mechanism in CoS_(2)/CNCs.Benefiting from the unique structures,including abundant active interfacial sites,high electronic conductivity,and significantly alleviated volume variation,CoS_(2)/CNCs present a high specific capacity(537.3 mAh g^(−1) at 0.1 A g^(−1)),good cycling stability(322.4 mAh g^(−1) at 0.5 A g^(−1) after 300 cycles),and excellent rate capability(153.1 mAh g^(−1) at 5 A g^(−1)).Moreover,the obtained nanocomposite shows superior potassium storage properties in K-ion full cells when it is coupled with a KVPO4F cathode. 展开更多

Tongue diagnosis is a diagnostic method to understand the physiological functions and pathological changes of the human body by observing human tongue quality,tongue coating,and sublingual collateral veins.Tongue elephant research is an important part of objective Chinese medicine.Based on the research of tongue image analysis method,the design of a tongue image collection analysis system based on the smartphone platform was demonstrated.This research uses a smartphone camera to collect tongue images,upload them to the server and complete tongue image analysis on the server.Finally,the tongue image analysis results are generated,and the results are transmitted from the server to the smartphone mobile terminal.We used HTTP protocol for communication between mobile client and server.We asked professional TCM doctors to evaluate the results of tongue analysis in 100 cases,and the final anastomosis rate was above 80%.This study fills in the gaps in the mobile platform of objectification of tongue diagnosis,which is conducive to the use of mobile phones to collect tongue images anytime,anywhere,and to conveniently share the results of tongue diagnosis with physicians.With an Internet connection,users can use their smartphones to remotely diagnose their tongues,who can store and generate their own personal diagnostic reports. 展开更多
