Recent clinical evidence has shown the benefits o...Background: Concurrent chemoradiotherapy(CCRT) significantly increases the survival rate of esophageal squa?mous cell carcinoma(ESCC) patients with malignant fistulae. Recent clinical evidence has shown the benefits of enteral nutrition for malnourished cancer patients. In this study, we aimed to validate that, with the support of enteral nutrition, ESCC patients who develop malignant fistulae might be able to complete CCRT and achieve long?term survival.Methods: We reviewed the medical records of 652 patients with ESCC who received definitive CCRT at Sun Yat?sen University Cancer Center between January 2010 and December 2012. Treatment outcome and toxicity were ret?rospectively evaluated in 40 ESCC patients with malignant fistulae. All the 40 patients were treated with CCRT and evaluated by clinical nutritionists using nutrition risk screening(NRS) before, during, and after treatment. Twenty?two patients received a nasogastric tube, and 18 underwent percutaneous endoscopic gastrostomy feeding. The median energy intake was 2166 kcal/day. Treatment response was evaluated at 3 months after the completion of CCRT.Results: With a median follow?up of 18 months(range, 3–39 months), patients' 1?year overall survival(OS) rate was 62.5%, and the estimated OS time was 25.5 months. Univariate analysis showed that the NRS score(P n NRS score(P se to treatment(P < 0.001) were sig= 0.003), increase i= 0.024), fistula closure(P = 0.011), and responnifi?cantly associated with OS. Multivariate analysis showed that tumor response(P = 0.044) and increase in NRS score(P = 0.044) were independent predictors of OS. Grade 3 vomiting was observed in 8 patients(20.0%), grade 3 neutro?penia was observed in 11 patients(27.5%), and grade 3 cough was observed in 13 patients(32.5%); 2 patients(5.0%) died of massive bleeding during treatment.Conclusions: CCRT combined with enteral nutrition support is effective for ESCC patients with malignant fistulae. Patients have an increased potential to be cured, especially those who experience complete response and have an increase in NRS score. Careful observation and nutrition support are required for patients with advanced T?category ESCC who undergo CCRT.展开更多

The results show that most of the microwave power is absorbed around the resonance surface near the antenna when the plasma density is below the cutoff density of 7.4 × 10^(16) m^(-3) for 2.45 GHz microwaves, and the simulation results also show that the plasma density is strongly influenced by the position and shape of the resonance surface, where a larger resonance surface would improve the plasma density.展开更多

Infrared and ultraviolet-visible spectra were obtained and compared with experiments. The ultraviolet-visible spectrum was also analyzed by the molecular orbital population. The Lamiaceae family is renowned for its terpenoid-based medicinal components,but Leonurus,which has traditional medicinal uses,stands out for its alkaloid-rich composition.Leonurine,the principal active compound foun...The Lamiaceae family is renowned for its terpenoid-based medicinal components,but Leonurus,which has traditional medicinal uses,stands out for its alkaloid-rich composition.Leonurine,the principal active compound found in Leonurus,has demonstrated promising effects in reducing blood lipids and treating strokes.However,the biosynthetic pathway of leonurine remains largely unexplored.Here,we present the chromosome-level genome sequence assemblies of Leonurus japonicus,known for its high leonurine production,and Leonurus sibiricus,characterized by very limited leonurine production.By integrating genomics,RNA sequencing,metabolomics,and enzyme activity assay data,we constructed the leonurine biosynthesis pathway and identified the arginine decarboxylase(ADC),uridine diphosphate glucosyltransferase(UGT),and serine carboxypeptidase-like(SCPL)acyltransferase enzymes that catalyze key reactions in this pathway.Further analyses revealed that the UGT–SCPL gene cluster evolved by gene duplication in the ancestor of Leonurus and neofunctionalization of SCPL in L.japonicus,which contributed to the accumulation of leonurine specifically in L.japonicus.Collectively,our comprehensive study illuminates leonurine biosynthesis and its evolution in Leonurus.展开更多
This study examined the inhibitory effect of triptolid(TP)on tumor necrosis factor-α(TNF-α)secreted from peripheral blood monocular cells(PBMCs)and the association of the inhibitory effect with TNF-α-308 gene polym...This study examined the inhibitory effect of triptolid(TP)on tumor necrosis factor-α(TNF-α)secreted from peripheral blood monocular cells(PBMCs)and the association of the inhibitory effect with TNF-α-308 gene polymorphisms in rheumatoid arthritis(RA)patients.Gene polymorphism at A-G site 308 in the promoter region of TNF-αgene was detected in 42 RA patients by using allele specific polymerase chain reaction(AS-PCR)assay.PBMCs were harvested from these patients and treatedfirst with lipopolysaccharides(LPS)and then with different doses of TP(1,5.4 and 15 ng/mL).The TNF-αlevel in the supernatants was measured by enzyme-linked immuno-sorbent assay(ELISA).The results showed that TNF-αlevel in the supernatants of TP(1 ng/mL)-treated PBMCs was decreased by 3.80%and 4.91%,respectively,in the patients with AA and AG genotypes,when compared with those treated with LPS alone(P>0.05).Moreover,the TNF-αlevel in the patients with GG genotype was reduced by 20.74%(P<0.05).When PBMCs were treated with TP at 5.4 ng/mL,TNF-αlevels in the patients with AA,AG,and GG genotypes were decreased by 20.42%,34.73%,and 41.69%,respectively(P<0.05).The TNF-αlevel was slightly higher in the PBMCs treated with 15 ng/mL of TP than those in the two TP groups in the patients carrying AA,AG,and GG genotypes(P>0.05).It was concluded that gene polymorphism at TNF-α-308 sites may relate to the secretion of TNF-αin RA patients.TP has different inhibitory effects on the secretion of TNF-αin the patients harboring different genotypes,which may be one of the reasons for individual variation in response to TP.展开更多

By analyzing training set molecules, our MDS method combined with a PROXSCAL algorithm can predict inhibitory activity of most compounds correctly. Thus, a new sample's activity can be estimated and judged conveniently, and whether it should be synthesized can be known. The MDS method is applicable to optimize the structure for a compound and to provide suggestions for drug design.展开更多
