Polo-like kinase 1(PLK1)plays a crucial role in cell mitosis and has been associated with necroptosis.However,the role of PLK1 and necroptosis in lung adenocarcinoma(LA)remains unclear.In this study,we analyzed The Cancer Genome Atlas(TCGA)and Genotype-Tissue Expression databases to evaluate the prognostic value and mechanistic role of PLK1 in LA.PLK1 was found to be highly expressed in LA and was positively associated with advanced disease staging and poor survival outcomes.Functional enrichment analysis showed that PLK1 was involved in cell mitosis,neurotransmitter transmission,and drug metabolism.Further analysis using single-sample gene set enrichment analysis and ESTIMATE algorithm revealed a correlation between PLK1 expression and immune infiltration in LA.Silencing of PLK1 using miRNA transfection in LA cells reduced cell proliferation and increased apoptosis,as well as upregulating the expression of necroptosis-related proteins,such as RIPK1,RIPK3,and MLKL.Additionally,nude mouse transplantation tumor experiments demonstrated that silencing PLK1 reduced the growth capacity of LA cells.These findings suggest that PLK1 plays a critical role in LA progression by regulating necroptosis and immune infiltration,and may serve as a potential therapeutic target for immunotherapy.Furthermore,PLK1 expression can be used as a prognostic biomarker for LA patients. 展开更多

Heavy-duty machine tools are composed of many subsystems with different functions,and their reliability is governed by the reliabilities of these subsystems.It is important to rank the weaknesses of subsystems and identify the weakest subsystem to optimize products and improve their reliabilities.However,traditional ranking methods based on failure mode effect and critical analysis(FMECA)does not consider the complex maintenance of products.Herein,a weakness ranking method for the subsystems of heavy-duty machine tools is proposed based on generalized FMECA information.In this method,eight reliability indexes,including maintainability and maintenance cost,are considered in the generalized FMECA information.Subsequently,the cognition best worst method is used to calculate the weight of each screened index,and the weaknesses of the subsystems are ranked using a technique for order preference by similarity to an ideal solution.Finally,based on the failure data collected from certain domestic heavy-duty horizontal lathes,the weakness ranking result of the subsystems is obtained to verify the effectiveness of the proposed method.An improved weakness ranking method that can comprehensively analyze and identify weak subsystems is proposed herein for designing and improving the reliability of complex electromechanical products. 展开更多

The energy-intensive Haber-Bosch process currently dominants the production of ammonia (NH3), an indispensable chemical forhumans. For the sustainable development of society, highly efficient and green strategies to convert nitrogen (N2) to NH3 are urgentlyrequired. Electrocatalytic N2 reduction reaction (eNRR) is universally regarded as a promising strategy owing to the mild operatingconditions and renewable energy supply. The key for eNRR is the high-performance catalysts, which activate the inert N-N triple bondand thus decrease the energy barrier. Herein, the recent theoretical and experimental progress on eNRR catalysts at room temperatureand ambient pressure is summarized, aiming to provide a reference for future design of high-performance eNRR catalysts. 展开更多

作为能源密集型Haber-Bosch工艺合成氨的一种新兴替代品,电化学硝酸盐还原反应(NO_(3)RR)在可持续合成氨和废水处理方面受到了关注.然而,由于缺乏有效的电催化剂,NO_(3)RR目前仍然面临氨产率低和选择性差的问题.本文报道了通过脱合金法... 作为能源密集型Haber-Bosch工艺合成氨的一种新兴替代品,电化学硝酸盐还原反应(NO_(3)RR)在可持续合成氨和废水处理方面受到了关注.然而,由于缺乏有效的电催化剂,NO_(3)RR目前仍然面临氨产率低和选择性差的问题.本文报道了通过脱合金法制备的纳米双模式孔Ru掺杂Cu催化剂作为NO_(3)RR的电催化剂,在-0.2 V versus RHE的电位下表现出29.63±0.74 mg h^(-1)mg_(cat.)^(-1)的超高氨产率与97.3%±2.5%的法拉第效率,优于大多数报道的催化剂.密度泛函理论计算表明,在Cu中掺杂Ru可以优化中间体的吸附能,降低NO_(3)RR速控步骤的能垒.此外,Ru原子可以促进H2O的吸附/解离,为含N中间体氢化为NH_(3)提供活性氢.这项工作为NO_(3)RR等过程进行高性能催化剂的合理设计提供了新的途径. 展开更多
