Damage to Wistar rat livers after hypo-fractionated radiation and oxaliplatin

Objective The purpose of this study was to clarify whether hypo-fractionated radiation therapy combined with oxaliplatin can aggravate liver damage, in order to determine its safety for clinical application. Methods Eighty Wistar rats were randomly divided into four groups: the control group, the chemotherapy treatment group, the radiation treatment group, and the concurrent chemoradiotherapy group. The rats' liver tissues were then collected for histological evaluation at the first, second, fourth, sixth, and eight week after irradiation. The tissues were histologically evaluated using hematoxylin and eosin staining, and immunohistochemistry to analyze the expression of Bcl-2 and Bax. Results Histological examination revealed swollen hepatocellular cells in the experimental groups, with visible liver degeneration and necrosis. Alanine aminotransferase and aspartate aminotransferase levels were significantly different between the groups(F = 85.869 and 214.663; P < 0.001). The intra-group expressions of Bcl-2 and Bax were also significantly different between each time point(F = 6.047 and 43.344; P < 0.05). Bax expression was significantly different between each group(F = 8.122; P < 0.05), although no inter-group differences were observed for Bcl-2 expression(F = 0.808; P > 0.05). Conclusion Chemoradiotherapy may aggravate liver injury, possible via overexpression of Bcl-2 and reduced expression of Bax. Therefore, this treatment should be used carefully in the clinic. 展开更多

Objective: The aim of our study was to examine whether irradiation combined with pemetrexed can exacerbate pulmonary injury. Methods: Two groups of male Wister Rats were subjected to bilateral apex of lungs irradiation(a single dose of 12 Gy), with or without pemetrexed(20 mg/kg) by intraperitoneal injection at the same time; a third group of weightand age- matched animals were treated with pemetrexed alone, as the same dose scheme, time and root of injection. The fourth group served as control. The whole lung mounts were dissected to histological evaluation, while serum cytokine transforming growth factor-β1(TGF-β1) analysis were compared at 1, 7, 21, 35, 49 days post-irradiation after irradiation. Results: Histological examination showed a thickening of alveolar septal, accumulation of inflammatory cells. The irradiation treatment group and the radiation-chemo treatment group showed a statistically significant higher level of TGF-β1(P < 0.05) than other two groups, but there were no differences between these two irradiation groups. Conclusion: These results demonstrated that pemetrexed can not aggravate pulmonary injury and it could be safely used in concurrent or sequential radio-chemotherapy in lung adenocarcinoma. 展开更多

Objective: The aim of our study was to investigate the impact of abdominal hypo-fraction irradiation on liver damage in rats so as to provide a reference for its clinical application. Methods: A total of 100 Wistar rats were equally randomized to five groups as control, 4 Gy, 6 Gy, 8 Gy and 12 Gy group, and the corresponding fractionated doses were offered. Liver functions were examined at the 2nd, 4th, 6th, 8th and 10th week after irradiation. Morphological changes were observed by HE staining. Expressions of Bcl-2 and Bax were examined by immunohistochemical technique. Results: In all irradiation groups, hepatocellular swell, degeneration, necrosis and even hepatic fibrosis could be seen. The differences of the liver coefficient, Glutamyl pyruvic transaminase (GPT), Glutamyl oxaloacetic transaminase (GOT) were significant among the groups and different time points (F = 11.833-781.972, F = 20.857-264.692, P < 0.001). Expressions of Bcl-2 and Bax were significantly different between each group (F = 211.607, 116.577; P < 0. 001), and between each time point (F = 54.083, 68.749; P < 0. 001). Conclusion: Compare with conventional fraction, abdominal hypo-fraction irradiation may cause radiation damage to rat liver, being dose-and-time dependent. Up-regulation of activating apoptosis protein Bax and down-regulation of inhibiting apoptosis protein Bcl-2 may involve in the process. 展开更多
