Objective: To construct green fluorescent protein(GFP) retroviral vector(pLgXSN), and to investigate the expression of GFP in primary rat myoblast. Methods: GFP cDNA was subcloned into the plasmid pLgXSN, and the recombinant vector was transfected into packaging cell PT67. G418 was used to select positive colony. Myoblasts were infected by a high-titer viral supernatant. The recombinant retroviral plasmid vector was identified by restriction endonuclease analysis and DNA sequence analysis. Confocal microscopy and flow cytometry were used to detect the expression of GFP. Results: The GFP cDNA sequence was identical to that of GenBank. Recombinant retroviral plasmid vector pLgGFPSN was constructed successfully. The titer of the packaged recombinant retrovirus was 1×10^6 cfu/ml. Bright green fluorescence of the transfected cells was observed under confocal microscope 48 h after transfection. The transfection rate was 33%. The effective expression of GFP in myoblast infected by recombinant retrovirus lasted for 6 weeks. Conclusion: GFP gene could be effectively and stably expressed in myoblast, which suggests that GFP could act as a marker for studies on myoblast. 展开更多

Wearable electronics powered by flexible energy storage devices have captured global attention.Under low-temperature conditions,unfortunately,solidification of flexible hydrogel electrolyte and decreased pseudo-capacity of these devices largely hamper their practical use.In this study,photothermally-active Prussian blue(PB)was introduced onto poly(3,4-ethylenedioxythiophene)/polyacrylamide(PEDOT/PAM)networks to address the challenges of electrolyte solidification and degraded pseudo-capacitance for flexible all-in-one device at low temperatures.The as-constructed PB/PEDOT/PAM hydrogel device delivers stable electrochemical performance and remarkable mechanical property with 1652%elongation.Importantly,this hydrogel device well retains its flexibility in cold environment with a freezing point below−30℃.The incorporation of PB extends the voltage range to 1.5 V as a single device,thus significantly enhancing the electrochemical performance as an all-inone integrated device.Benefitting from the outstanding photo-to-thermal conversion ability of embedded PB nanocubes,the temperature of the assembled all-in-one PB/PEDOT/PAM device increased from^(−2)0 to 17.7℃ after solar-light irradiation for only 5 min.Moreover,the degraded pseudo-capacitance was subsequently boosted to 287.1%of its original capacitance at−20℃.This study establishes a connection between flexible all-solid-state hydrogel devices and photothermally enhanced pseudocapacitors in freezing environments,thereby expanding the potential applications of multi-functional pseudo-capacitors. 展开更多

This study focuses on the microstructure and mechanical properties of the joints of Q235 mild steel,which was formed by the friction stir welding(FSW).The results indicated that,after the FSW,the heat-affected zone(HAZ)of the retreating side(HAZRS)and the HAZ of the advancing side(HAZAS)recovered under the influence of the heating cycle.The transformation of the phases in the thermo-mechanically affected zone(TMAZ)of the retreating side(TMAZRS),the stir zone(SZ)and the TMAZ of the advancing side(TMAZAS)generated the pearlite and acicular ferrite.The continuous dynamic recrystallization occurred in all the three zones,whereas the grains were refined.The SZ mainly consisted of D1,D2 and F shear textures,while the TMAZAS was made up of only the F shear texture.The fine-grained structure,pearlite and the acicular ferrite improved the hardness and tensile strength of the joint.Its ultimate tensile strength was 479 MPa,which was 1.3% higher than that of the base metal.However,the uniform elongation was 16%,which showed a decrease of 33%.The fracture was a ductile fracture with the appearance of dimples.Besides,the joints of the FSW showed an excellent bending performance. 展开更多

In order to obtain the superalloy with excellent properties, graphene reinforced K418 nickel base superalloy(GNPs/K418 composite) was prepared by selective laser melting technique in this study. Through systematically comparing and analyzing the microstructure and mechanical property of K418 superalloy and GNPs/K418 composite, it is found that the percentage of small-diameter grain(≤ 15 μm) increased from 84% to 90%, and the max strength of grain orientation(<001>) reduce from 5.76 to 4.17 due to the addition of GNPs. And GNPs can also improve the height and the full width at the half peak of the strong diffraction peak of GNPs/K418 composite. Besides, GNPs/K418 composite is a kind of sandwiched structure, which is consist of GNPs, carbides, and K418 matrix. Therefore, the hardness of the GNPs/K418 composite is 4.1% and 6.9% higher than that of the K418 matrix in the transverse and vertical direction, respectively. The room temperature tensile strength of the GNPs/K418 composite is 9% higher than that of the K418 matrix. And the 600℃ and 900℃ tensile strengths of the GNPs/K418 composite are 7.6% and 10.4% higher than that of the K418 matrix, respectively. It is inferred that the effect of graphene on K418 matrix strengthening is mainly fine grain strengthening and Orowan strengthening. However, the elongation rate of the composite material is reduced, which is attributed to crack sprouting at the interface between the matrix and GNPs under high stress. 展开更多
