Northeast Asian cut-off lows are crucial cyclonic systems that can bring temperature and precipitation extremes over large areas.Skillful subseasonal forecasting of Northeast Asian cut-off lows is of great importance.Using two dynamical forecasting systems,one from the Beijing Climate Center(BCC-CSM2-HR)and the other from the Met Office(GloSea5),this study assesses simulation ability and subseasonal prediction skill for early-summer Northeast Asian cut-off lows.Both models are shown to have good ability in representing the spatial structure of cut-off lows,but they underestimate the intensity.The skillful prediction time scales for cut-off low intensity are about 10.2 days for BCC-CSM2-HR and 11.4 days for GloSea5 in advance.Further examination shows that both models can essentially capture the initial Rossby wave train,rapid growth and decay processes responsible for the evolution of cut-off lows,but the models show weaker amplitudes for the three-stage processes.The underestimated simulated strength of both the Eurasian midlatitude and East Asian subtropical jets may lead to the weaker local eddy-mean flow interaction responsible for the cut-off low evolution. 展开更多

Social isolation, loneliness and subsequent risk of major adverse cardiovascular events among individuals with type 2 diabetes mellitus

Background Individuals with type 2 diabetes mellitus(T2DM)are more vulnerable to social disconnection compared with the general population;however,there are few relevant studies investigating this issue.Aims To investigate whether social isolation or loneliness may be associated with subsequent risk of developing major adverse cardiovascular events,whether these associations vary according to fatal and non-fatal outcomes and how behavioural,psychological and physiological factors mediate these associations.Methods This longitudinal analysis included data from 19360 individuals with T2DM at baseline(2006-2010)from the UK Biobank.Social isolation and loneliness were measured using self-report questionnaires.The study outcomes included the first events of myocardial infarction(MI)or stroke(n=2273)and all-cause(n=2820)or cardiovascular disease-related mortality through linked hospital data ordeath registries.Results Over a median follow-up of 12.4 years(interquartile range(IQR):11.6-13.3 years),participants who were more socially isolated(most social isolation vs least social isolation)experienced increased risks for all-cause(hazard ratio(HR):1.33,95%confidence interval(Cl):1.19 to 1.47)and cardiovascular disease(HR:1.36,95%Cl:1.17 to 1.59)mortality but not first Ml or stroke.Loneliness(yes vs no)was associated with a greater risk for a composite of incident MI or stroke(HR:1.37,95%Cl:1.19 to 1.57)but not mortality.Social isolation was associated with fatal Ml and stroke,whereas loneliness was associated with non-fatal Ml and stroke.The significant associations of social isolation and loneliness with outcomes were mainly mediated by behavioural factors(mediating proportion:17.8%-28.2%and 17.6%-17.8%,respectively).Conclusions Among individuals with T2DM,social isolation and loneliness are associated with a greater risk of developing major adverse cardiovascular events,with differences in both risks stratified according to fatal and non-fatal events and underlying mediating factors. 展开更多

目的探讨右美托咪定(Dex)对老年髋部手术患者术后认知功能及脑氧代谢影响的适宜剂量。方法选取择期行全髋关节置换术老年患者120例。年龄≥62岁,体重54~79 kg,ASA分级Ⅰ、Ⅱ级,随机分为4组:右美托咪定组(D1、D2、D3组)和对照组(R组),D1... 目的探讨右美托咪定(Dex)对老年髋部手术患者术后认知功能及脑氧代谢影响的适宜剂量。方法选取择期行全髋关节置换术老年患者120例。年龄≥62岁,体重54~79 kg,ASA分级Ⅰ、Ⅱ级,随机分为4组:右美托咪定组(D1、D2、D3组)和对照组(R组),D1、D2、D3组在给予1μg/kg负荷量的Dex后分别继续维持泵注0.2、0.5和0.8μg/(kg·h)至手术结束前30 min,R组给予等量生理盐水。于麻醉诱导前(T0)、术毕即刻(T1)、术后1 d(T2)、术后3 d(T3)、术后7 d(T4)测定血清S100β蛋白及神经元特异性烯醇化酶(NSE)水平,并于T0、T1和T2时计算脑氧摄取率(CERO2)。利用蒙特利尔认知功能评分量表(Mo CA)对患者术前1 d、术后1、3、7 d时实施评分,观察患者术后认知功能障碍(POCD)的发生率。结果与T0时比较,4组T1~T3时血清S100β与NSE浓度均升高(P<0.05);与R组比较,D1、D2、D3组T1~T4时血清S100β与NSE浓度均降低(P<0.05)。与T0时比较,4组T1、T2时CERO2降低(P<0.05),与R组比较,D1、D2、D3组T2、T3时CERO2降低(P<0.05)。D1、D2、D3组术后1、3和7 d Mo CA评分均高于R组(P<0.05);与D1组比较,D2、D3组术后1、3 d和D2组术后7 d MoCA评分均升高(P<0.05)。R组、D1、D2和D3组POCD发生率分别为40%、23%、10%和17%。结论持续静脉输注Dex可有效降低POCD的发生率,其机制可能与降低S100β、NSE浓度及脑代谢率相关,其中以0.5μg/(kg·h)维持剂量最佳。 展开更多

Machine perfusion for liver transplantation:A concise review of clinical trials 被引量:6

Background: With the increased use of extended-criteria donors, static cold storage has failed to provide optimal preservation of liver grafts, resulting in early allograft dysfunction and long-term complications Machine perfusion(MP) is a beneficial alternative preservation strategy for donor livers, particularly fo those considered to be of suboptimal quality, and could expand the limited donor pool. Data sources: A comprehensive search in Pub Med, EMBASE, Ovid databases and Clinical Trials.gov website was conducted using the medical subject heading terms "machine perfusion", "machine preservation""liver transplantation", combined with free text terms such as "hypothermic", "normothermic" and "sub normothermic". The deadline for the search was September 30, 2017. Results: MP can be classified as hypothermic, subnormothermic, and normothermic with the tempera ture maintained at 0–12 °C, 25–34 °C and 35–38 °C, respectively. Twelve clinical trials of MP have been reported in recent years. MP effectively decreased AST/ALT level and the incidence of early allograft dys function. However, the graft and patient survival rate after MP were similar to static cold storage. The detailed clinical characteristics such as liver function, graft survival, patient survival and early allograf dysfunction were reviewed.Conclusions: Clinical trial results showed that MP improves delayed graft function, primary non-function and biliary strictures. However, MP still requires validation in large clinical trials and the key parameters during MP still require optimization. 展开更多

To analyze the effects of width and thickness of each composition element of mixed steel U-rib-stiffened plates on the welding residual stress distribution, the distribution of the U-rib and the plate residual stress was calculated using a simplified calculation method. The method involved welding the mixed steel U-rib-stiffened plates for a structure with different sizes and different strength ratios of U-rib to plate. Based on a welding residual stress numerical simulation method validated by the blind hole method test, the distribution law of the mixed steel U-rib stiffened plate was studied. The results showed that the change of plate width has little impact on the welding residual stress and that the ratio of the thicknesses of the plate to U-rib stiffeners, the thickness of the plate, and the thickness of the U-rib has a great influence on the distribution of the welding residual stress. The thickness of plate and steel strength also greatly influenced the distribution width of the residual tensile stress. While analyzing the compression capacity of U-rib-stiffened plates, the simplified distribution of welding residual stress was used. 展开更多

Establishment of vaginal immune defenses at the mucosal interface layer through gene vaccines promise to prevent infectious diseases among females.Mucosal barriers composed of a flowing mucus hydrogel and tightly conjugated epithelial cells(ECs),which represent the main technical difficulties for vaccine development,reside in the harsh,acidic human vaginal environment.Different from frequently employed viral vectors,two types of nonviral nanocarriers were designed to concurrently overcome the barriers and induce immune responses.Differing design concepts include the charge-reversal property(DRLS)to mimic a virus that uses any cells as factories,as well as the addition of a hyaluronic acid coating(HA/RLS)to directly target dendritic cells(DCs).With a suitable size and electrostatic neutrality,these two nanoparticles penetrate a mucus hydrogel with similar diffusivity.The DRLS system expressed a higher level of the carried human papillomavirus type 16 L1 gene compared to HA/RLS in vivo.Therefore it induced more robust mucosal,cellular,and humoral immune responses.Moreover,the DLRS applied to intravaginal immunization induced high IgA levels compared with intramuscularly injected DNA(naked),indicating timely protection against pathogens at the mucus layer.These findings also offer important approaches for the design and fabrication of nonviral gene vaccines in other mucosal systems. 展开更多

The reversal of surface air temperature anomalies in China between early and late winter 2021/2022:Observations and predictions

During winter of 2021/2022,the temperature in China is characterized by a warm-to-cold transition,and the average temperature anomaly in February 2022 is−1.6℃,the coldest February in 2013-2022.We revealed the circulation regimes and physical mechanisms associated with this reversal event and demonstrated the advantage of a regional model downscaling over the use of the global model alone in predicting.In early winter,the warm anomalies are mainly related to an anomalous anticyclonic system downstream of a PNA-like(Pacific-North America)Rossby-wave train induced by La Niña.In late winter,due to the circulation response to the central Pacific warming and negative tropical Indian Ocean Dipole(TIOD),two‘−+−’Rossby-wave trains from high latitudes and the tropical Indian Ocean jointly lead to an anomalous cyclonic system in China.Meanwhile,an anticyclonic blocking system on the northern side of Baikal brings strong and cold air to China.These two systems together cause a significant drop in surface air temperature anomaly in China during the late winter.The Beijing Climate Center climate system model(BCC_CSM1.1 m)can essentially predict this temperature reversal in China about five months in advance.However,the reversal amplitude is weaker due to warm deviations over the tropical Pacific Ocean and equatorial Indian Ocean.Using dynamic downscaling,a regional Climate-Weather Research and Forecasting(CWRF)model correctly predicts the cold SAT anomalies in late winter 2021/2022.The regional model depicts more realistic circulation patterns in East Asia;the anomalous cyclonic system in Inner Mongolia accompanied by the northerly anomalies contribute to a lower-than-normal SAT over China.This study reveals the cooperative effect of wave trains from high latitudes and the tropics on the subseasonal temperature reversal and demonstrates a possible solution to improve the forecast skill by dynamic downscaling according to precise characterization of local surface information. 展开更多

30% Fe CN/ZIS(30% Fe doped g-C_(3)N_(4)composited ZnIn_(2)S_(4)) was synthesized by a simple water bath method, via in-situ growth of abundant well-dispersed ZnIn_(2)S_(4)nanosheets on the Fe doped g-C_(3)N_(4)surface... 30% Fe CN/ZIS(30% Fe doped g-C_(3)N_(4)composited ZnIn_(2)S_(4)) was synthesized by a simple water bath method, via in-situ growth of abundant well-dispersed ZnIn_(2)S_(4)nanosheets on the Fe doped g-C_(3)N_(4)surface. Experimental results showed the optimized 30% Fe CN/ZIS achieved the best photoreduction of Cr(VI)performance within a wide p H range, which was 9.5 times and 700 times higher than that of pure ZnIn_(2)S_(4)and 30% Fe CN(Fe doped g-C_(3)N_(4)). This is due to the intense synergy between the Fe-Nxbond and close interface contact produces a high-speed charge transfer channel, thus significantly improving the efficiency of optical carrier separation and migration. Meanwhile, UV-vis diffuse reflection spectra and photoluminescence spectroscopy showed that iron doping significantly narrowed the bandgap of gC_(3)N_(4), preventing electron-hole pair recombination. Further, the microstructures and charge separation properties were analyzed by scanning electron microscope, Photoluminescence Spectroscopy and timeresolved photoluminescence, which revealed the structure-activity relationship of composite structure and the synergistic mechanism of each functional component. This research should provide a viable technique for creating composites with high photocatalytic activity for the treatment of chromium-containing wastewater. 展开更多

Climate variability modes, usually known as primary climate phenomena, are well recognized as the most important predictability sources in subseasonal–interannual climate prediction. This paper begins by reviewing the research and development carried out, and the recent progress made, at the Beijing Climate Center(BCC) in predicting some primary climate variability modes. These include the El Ni?o–Southern Oscillation(ENSO), Madden–Julian Oscillation(MJO), and Arctic Oscillation(AO), on global scales, as well as the sea surface temperature(SST) modes in the Indian Ocean and North Atlantic, western Pacific subtropical high(WPSH), and the East Asian winter and summer monsoons(EAWM and EASM, respectively), on regional scales. Based on its latest climate and statistical models, the BCC has established a climate phenomenon prediction system(CPPS) and completed a hindcast experiment for the period 1991–2014. The performance of the CPPS in predicting such climate variability modes is systematically evaluated. The results show that skillful predictions have been made for ENSO, MJO, the Indian Ocean basin mode, the WPSH, and partly for the EASM, whereas less skillful predictions were made for the Indian Ocean Dipole(IOD) and North Atlantic SST Tripole, and no clear skill at all for the AO, subtropical IOD, and EAWM. Improvements in the prediction of these climate variability modes with low skill need to be achieved by improving the BCC's climate models, developing physically based statistical models as well as correction methods for model predictions.Some of the monitoring/prediction products of the BCC-CPPS are also introduced in this paper. 展开更多

African swine fever virus(ASFV),as a member of the large DNA viruses,may regulate autophagy and apoptosis by inhibiting programmed cell death.However,the function of ASFV proteins has not been fully elucidated,especially the role of autophagy in ASFV infection.One of three Pyrroline-5-carboxylate reductases(PYCR),is primarily involved in conversion of glutamate to proline.Previous studies have shown that depletion of PYCR2 was related to the induction of autophagy.In the present study,we found for the first time that ASFV E199 L protein induced a complete autophagy process in Vero and HEK-293 T cells.Through co-immunoprecipitation coupled with mass spectrometry(Co IP-MS)analysis,we firstly identified that E199 L interact with PYCR2 in vitro.Importantly,our work provides evidence that E199 L down-regulated the expression of PYCR2,resulting in autophagy activation.Overall,our results demonstrate that ASFV E199 L protein induces complete autophagy through interaction with PYCR2 and down-regulate the expression level of PYCR2,which provide a valuable reference for the role of autophagy during ASFV infection and contribute to the functional clues of PYCR2. 展开更多

Heart regeneration is a promising strategy to prevent cardiac injury and protect against heart failure.^(1,2,3)Distinguishing from adult mammals,neonatal mouse heart can regenerate completely after cardiac injury,such as apical resection(AR)or myocardial infarction(MI).Our previous study reveals that acute inflammation is essential for initiation of neonatal mouse heart regeneration,4 providing an insight to understand the role of immune system in cardiac repair.Macrophage is a prominent inflammatory cell in injured heart during acute immune response;5 here we revealed that genetically deletion of CD11b-positive macrophages in CD11bDTR mice with diphtheria toxin(DT)administration led the suspense of neonatal heart regeneration after AR injury.We transplanted cardiac macrophages sorted from AR-injured neonatal mouse hearts into MI-injured adult mice and found that transfusion of neonatal mouse cardiac macrophages facilitated cardiac repair and enhanced cardiomyocyte proliferation effectively,which served a potential intervention to improve prognosis of patients suffering cardiac diseases. 展开更多

The RNA editing tool CRISPR-CasRx has provided a platform for a range of transcriptome analysis tools and therapeutic approaches with its broad efficacy and high specificity.To enable the application of CasRx in vivo,we established a Credependent CasRx knock-in mouse.Using these mice,we specifically knocked down the expression of Meis1 and Hoxb13 in cardiomyocytes,which induced cardiac regeneration after myocardial infarction.We also knocked down the lnc RNA Mhrt in cardiomyocytes with the CasRx knock-in mice,causing hypertrophic cardiomyopathy.In summary,we generated a Credependent CasRx knock-in mouse that can efficiently knock down coding gene and lnc RNA expression in specific somatic cells.This in vivo CRISPR-CasRx system is promising for gene function research and disease modeling. 展开更多
