A simple and accurate high-performance liquid chromatography(HPLC)coupled with diode array detector(DAD)and evaporative light scattering detector(ELSD)was established for the determination of six bioactive compo...A simple and accurate high-performance liquid chromatography(HPLC)coupled with diode array detector(DAD)and evaporative light scattering detector(ELSD)was established for the determination of six bioactive compounds in Zhenqi Fuzheng preparation(ZFP).The monitoring wavelengths were 254,275 and 328 nm.Under the optimum conditions,good separation was achieved,and the assay was fully validated in respect of precision,repeatability and accuracy.The proposed method was successfully applied to quantify the six ingredients in 31 batches of ZFP samples and evaluate the variation by hierarchical cluster analysis(HCA),which demonstrated significant variations on the content of these compounds in the samples from different manufacturers with different preparation procedures.The developed HPLC method can be used as a valid analytical method to evaluate the intrinsic quality of this preparation.展开更多
Objective:To study the mechanism of Yuanhuzhitong decoction for the treatment of osteoarthritis from the perspective of network pharmacology.Methods:The active ingredients of Yuanhuzhitong decoction were screened out ...Objective:To study the mechanism of Yuanhuzhitong decoction for the treatment of osteoarthritis from the perspective of network pharmacology.Methods:The active ingredients of Yuanhuzhitong decoction were screened out from the TCMSP database,and then the targets of the active ingredients were extracted.The drug-ingredient-target network was constructed using Cytoscape software.The genes of osteoarthritis were obtained by OMIM,GeneCards and DisGeNET databases.Drug targets and disease targets were mapped together,and the relationship of the key target protein-protein interaction(PPI)was obtained through the online STRING database.Key targets were analyzed by Clue GO plug-in for GO functional enrichment and KEGG signaling pathway enrichment.Results:A total of 71 active ingredients and 99 unique targets of alkaloids,coumarins,volatile oils,steroids,etc were obtained through the threshold of OB 30%and DL 0.18.45 common targets were obtained through Venn diagram,among which the core targets were IL6,VEGFA,CASP3,CCND1,ESR1,etc.GO enrichment analysis revealed that the biological processes of Yuanhuzhitong decoction for OA were mainly concentrated in cell migration,differentiation,nucleotide metabolism and synthesis,regulation of pri-miRNA transcription by RNA polymerase II,and apoptosis.KEGG analysis showed that Yuanhuzhitong decoction may act on HIF-1 signal pathway,TNF signal pathway,NF-κB signal pathway and other pathways.The results showed that the main active ingredients in Yuanhuzhitong decoction could play an important role for the treatment of osteoarthritis through"multi-component,multi-target and multi-pathway".Conclusion:This study analyzed the main active ingredients,key targets,signaling pathways and mechanism of Yuanhuzhitong decoction from the perspective of network pharmacology,laying a foundation for further exploration of its mechanism.展开更多

Methods: A total of 102 patients with craniocer...Objective: To investigate the effect of propofol anesthesia on oxidative stress, neurological function and inflammatory cytokines in patients with craniocerebral trauma. Methods: A total of 102 patients with craniocerebral trauma who underwent surgery in our hospital from December 2014 to January 2017 were randomly divided into control group and observation group, each contained 51 cases. The control group was given 1%-2% of sevoflurane and 0.1-0.2 μg/kg/min of remifentanil and 20-30 μg/kg/h of vecuronium for anesthesia maintenance. The observation group was given propofol 4-6 mg/kg/h, remifentanil 0.1-0.2 μg/kg/min and vecuronium 20-30 μg/kg/h for anesthesia maintenance. The levels of oxidative stress, neurological function, and inflammatory factors were assessed in both groups. Results:Compared with before treatment, the levels of SOD and HO-1 in the two groups were significantly increased and the levels of MDA were significantly decreased, the difference was significant, and the levels of SOD and HO-1 in the observation group were significantly higher than control group, the level of MDA was significantly lower than that of the control group, the difference was significant. Compared with before treatment, the levels of NSE, GFAP and Tau level were significantly decreased in the two groups after treatment, and level in observation group was lower than control group, the difference was statistically significant. Compared with before treatment, the levels of IL-6, TNF-α and CRP in the two groups after treatment were significantly lower than those in the control group, the difference was statistically significant. Conclusion: Propofol anesthesia can significantly reduce the oxidative stress injury, inhibit the inflammatory reaction and protect the neurological function of patients. The effect is better than isoflurane anesthesia, and it is worthy of clinical application.展开更多

Objective:To investigate the efficacy of TongLuoZhiTong Gel in the treatment of frozen shoulder model and the effects of frozen shoulder-related factors IL-1β,TNF-αandMMP-3 in rabbit.Methods:Twenty-four male healthy...Objective:To investigate the efficacy of TongLuoZhiTong Gel in the treatment of frozen shoulder model and the effects of frozen shoulder-related factors IL-1β,TNF-αandMMP-3 in rabbit.Methods:Twenty-four male healthy adult rabbits were randomly divided into the following three groups:blank group(group A),model control group(group B),and TongLuoZhiTong Gel treatment group(group C).Except for the blank group,the rabbits were subjected to a frozen shoulder model by continuous mechanical strain and ice application.After the model was completed,both groups A and B were not treated;group C was treated with external application of TongLuoZhiTong Gel,5 times a week for 2 weeks.Two weeks later,the synovial tissue was abstracted in the frozen shoulder joint,and then observed under the light microscope after HE staining.The Krenn grading of chronic synovitis was used to evaluated the degree of inflammation of the synovium.At the same time,the expression levels of IL-1β,TNF-αand MMP-3 in serum and periarticular tissues of each group were detected by enzyme-linked immunosorbent assay(ELISA).Results:Compared with the blank group,the Krenn score of the shoulder group was significantly increased after model establishment(P<0.05),and the local skin was red and swollen,suggesting successful modeling.Compared with the model group,after two weeks treatment of TongLuoZhiTong Gel,the inflammatory infiltration of the synovial tissue sections was significantly relieved,and the Krenn score was significantly decreased(P<0.05).In terms of fibrogenic chemical factors and inflammatory chemical factors,TongLuoZhiTong Gel treatment group can significantly reduce the expression of IL-1β,TNF-αand MMP-3 in rabbit serum and periarticular tissue homogenate(P<0.05).Conclusion:TongLuoZhiTong Gel can effectively alleviate the synovial inflammation of the frozen shoulder model of rabbits,and can significantly reduce the levels of IL-1β,TNF-αand MMP-3 in the serum and surrounding tissues of rabbits.展开更多
Objective:To evaluate the clinical efficacy of stone needle therapy based on blood stasis theory for different traditional Chinese medicine(TCM)syndrome types of patients with knee osteoarthritis in early and middle s...Objective:To evaluate the clinical efficacy of stone needle therapy based on blood stasis theory for different traditional Chinese medicine(TCM)syndrome types of patients with knee osteoarthritis in early and middle stage.Methods:Totally 36 patients with knee osteoarthritis were enrolled in the orthopaedic ward between March 2016 and December 2017,and diagnosed as qi stagnation and blood stasis syndrome,cold dampness and stasis syndrome,or phlegm and stasis syndrome according to TCM syndrome differentiation.After treatment,stone needle was used for ironing on the different meridian acupoints according to different syndrome.The efficacy of patients with the above three syndromes was respectively evaluated,and the visual analogue scale(VAS)score and western ontario and mcMaster universities arthritis Index(WOMAC)score of the three groups were calculated before treatment,2 weeks,and 4 weeks after treatment.Results:The effective rates of the three groups were 86.67%,81.82%,and 80.00%,respectively.After 2 weeks and 4 weeks of treatment,the VAS and WOMAC scores of the three groups decreased significantly compared to treatment before(P<0.05).Conclusion:Stone needle therapy for ironing on the acupoints around the knee joint has a good therapeutic effect on knee osteoarthritis of different syndrome types,which is worthy of further application.展开更多
