BACKGROUND Difficulty in obtaining tetracycline,increased adverse reactions,and relatively complicated medication methods have limited the clinical application of the classic bismuth quadruple therapy.Therefore,the se...BACKGROUND Difficulty in obtaining tetracycline,increased adverse reactions,and relatively complicated medication methods have limited the clinical application of the classic bismuth quadruple therapy.Therefore,the search for new alternative drugs has become one of the research hotspots.In recent years,minocycline,as a semisynthetic tetracycline,has demonstrated good potential for eradicating Helicobacter pylori(H.pylori)infection,but the systematic evaluation of its role remains lacking.AIM To explore the efficacy,safety,and compliance of minocycline in eradicating H.pylori infection.METHODS We comprehensively retrieved the electronic databases of PubMed,Embase,Web of Science,China National Knowledge Infrastructure,SinoMed,and Wanfang database as of October 30,2023,and finally included 22 research reports on H.pylori eradication with minocycline-containing regimens as per the inclusion and exclusion criteria.The eradication rates of H.pylori were calculated using a fixed or a random effect model,and the heterogeneity and publication bias of the studies were measured.RESULTS The single-arm meta-analysis revealed that the minocycline-containing regimens achieved good overall H.pylori eradication rates,reaching 82.3%[95%confidence interval(CI):79.7%-85.1%]in the intention-to-treat analysis and 90.0%(95%CI:87.7%-92.4%)in the per-protocol analysis.The overall safety and compliance of the minocycline-containing regimens were good,demonstrating an overall incidence of adverse reactions of 36.5%(95%CI:31.5%-42.2%).Further by traditional meta-analysis,the results showed that the minocycline-containing regimens were not statistically different from other commonly used eradication regimens in eradication rate and incidence of adverse effects.Most of the adverse reactions were mild to moderate and well-tolerated,and dizziness was relatively prominent in the minocycline-containing regimens(16%).CONCLUSION The minocycline-containing regimens demonstrated good efficacy,safety,and compliance in H.pylori eradication.Minocycline has good potential to replace tetracycline for eradicating H.pylori infection.展开更多
Time-frequency analysis is a successfully used tool for analyzing the local features of seismic data.However,it suffers from several inevitable limitations,such as the restricted time-frequency resolution,the difficul...Time-frequency analysis is a successfully used tool for analyzing the local features of seismic data.However,it suffers from several inevitable limitations,such as the restricted time-frequency resolution,the difficulty in selecting parameters,and the low computational efficiency.Inspired by deep learning,we suggest a deep learning-based workflow for seismic time-frequency analysis.The sparse S transform network(SSTNet)is first built to map the relationship between synthetic traces and sparse S transform spectra,which can be easily pre-trained by using synthetic traces and training labels.Next,we introduce knowledge distillation(KD)based transfer learning to re-train SSTNet by using a field data set without training labels,which is named the sparse S transform network with knowledge distillation(KD-SSTNet).In this way,we can effectively calculate the sparse time-frequency spectra of field data and avoid the use of field training labels.To test the availability of the suggested KD-SSTNet,we apply it to field data to estimate seismic attenuation for reservoir characterization and make detailed comparisons with the traditional time-frequency analysis methods.展开更多

To effectively replace the isotope radiation source in litho-density logging,this study presents a method for measuring the formation density and photoelectric absorption index(Pe)using a switchable X-ray tube.First,t...To effectively replace the isotope radiation source in litho-density logging,this study presents a method for measuring the formation density and photoelectric absorption index(Pe)using a switchable X-ray tube.First,the gamma-ray litho-density logging(GLD)method for measuring formation density and Pe using chemical sources is introduced.Then,a benchmark verification based on the X-ray litho-density logging tool prototype and data published by Simon(In:Paper presented at the SPWLA 59th annual logging symposium,London,UK,2018)was carried out using Monte Carlo numerical simulations.Second,the impacts of the photoelectric effect and detector statistical error on the GLD method were analyzed.Finally,based on a theoretical analysis,the formation density and Pe measurement algorithm(double energy window(DEW)method)was improved,which was found to be suitable for X-ray litho-density logging.Moreover,the results obtained using this algorithm were compared with those obtained using the GLD method.The results indicate that owing to the impact of photoelectric effect and detector statistical error on the density energy window,the accuracy of formation density and Pe measurement using the GLD method is relatively low,with the uncertainty in formation density and Pe measurement reaching 2.620±0.047 g/cm3 and 4.090±0.580 b/e,respectively.In comparison,the DEW method can improve the accuracy of density and Pe measurement to 0.006 g/cm3 and 0.065 b/e,respectively,as the photoelectric effect in the density window is corrected using the counts in the lithology window of the energy spectrum.This study aims to provide a new theoretical foundation for processing X-ray litho-density logs in the future.展开更多
Objective:To detect the CHRNA7 gene mutation and polymorphism in Southern Han Chinese patients with nocturnal frontal lobe epilepsy(NFLE).Methods:Blood samples were collected from 215 Southern Han Chinese patients wit...Objective:To detect the CHRNA7 gene mutation and polymorphism in Southern Han Chinese patients with nocturnal frontal lobe epilepsy(NFLE).Methods:Blood samples were collected from 215 Southern Han Chinese patients with NFLE and 200 healthy Southern Han Chinese control subjects.Genomic DNA was extracted,and CHRNA7 whole genome exons were amplified by the polymerase chain reaction and subjected to Sanger sequencing.Results:No CHRNA7 gene mutation was detected in all of the NFLE patients.However,five single nucleotide polymorphisms(SNPs)in sporadic cases were found,located in exons 5,6.and 7 of the CHRNA7 gene.Among them,c.690G>A and c.698A>G are known SNPs,while c.370G>A,c.654C>T,and c.497-498delTG were newly discovered SNPs.These SNPs were also found in some of the healthy controls.Conclusions:No CHRNA7 gene mutation was identified in Southern Han Chinese patients with NFLE.The CHRNA7 gene is probably not responsible for NFLE in this population.展开更多
The Shanghai high-repetition-rate XFEL and extreme light facility(SHINE)under construction is designed to be one of the most advanced free electron laser(FEL)facilities in the world.The main part of the SHINE facility...The Shanghai high-repetition-rate XFEL and extreme light facility(SHINE)under construction is designed to be one of the most advanced free electron laser(FEL)facilities in the world.The main part of the SHINE facility is an 8 GeV superconducting Linac operating in continuous wave(CW)mode.The Linac consists of seventy-five 1.3 GHz and two 3.9 GHz cryomodules.Based on the design and experience of 3.9 GHz cavities in the European X-ray free electron laser(E-XFEL)and the Linac coherent light source-II(LCLS-II)projects,we optimize the SHINE 3.9 GHz cavity design to adapt it for CW mode operation.In this paper,we present a particular redesign of the end group for the SHINE 3.9 GHz superconducting cavity that includes a redesign of the end cell and beam pipe to shift away the potentially troublesome lowest highorder modes(HOMs),a modification of the main coupler antenna,and a tuning of the HOM notch filter to meet the cavity requirements.RF losses calculations on the HOM coupler antennas show that the overheating on the inner conductor at the operating mode is diminished signifi-cantly.Furthermore,we have also studied the HOMs to ensure there are no dangerously trapped modes in the optimized cavity design.展开更多
CDGSH iron sulfur domain 2 can inhibit ferroptosis,which has been associated with cerebral ischemia/reperfusion,in individuals with head and neck cancer.Therefore,CDGSH iron sulfur domain 2 may be implicated in cerebr...CDGSH iron sulfur domain 2 can inhibit ferroptosis,which has been associated with cerebral ischemia/reperfusion,in individuals with head and neck cancer.Therefore,CDGSH iron sulfur domain 2 may be implicated in cerebral ischemia/reperfusion injury.To validate this hypothesis in the present study,we established mouse models of occlusion of the middle cerebral artery and HT22 cell models of oxygen-glucose deprivation and reoxygenation to mimic cerebral ischemia/reperfusion injury in vivo and in vitro,respectively.We found remarkably decreased CDGSH iron sulfur domain 2 expression in the mouse brain tissue and HT22 cells.When we used adeno-associated virus and plasmid to up-regulate CDGSH iron sulfur domain 2 expression in the brain tissue and HT22 cell models separately,mouse neurological dysfunction was greatly improved;the cerebral infarct volume was reduced;the survival rate of HT22 cells was increased;HT22 cell injury was alleviated;the expression of ferroptosis-related glutathione peroxidase 4,cystine-glutamate antiporter,and glutathione was increased;the levels of malondialdehyde,iron ions,and the expression of transferrin receptor 1 were decreased;and the expression of nuclear-factor E2-related factor 2/heme oxygenase 1 was increased.Inhibition of CDGSH iron sulfur domain 2 upregulation via the nuclear-factor E2-related factor 2 inhibitor ML385 in oxygen-glucose deprived and reoxygenated HT22 cells blocked the neuroprotective effects of CDGSH iron sulfur domain 2 up-regulation and the activation of the nuclear-factor E2-related factor 2/heme oxygenase 1 pathway.Our data indicate that the up-regulation of CDGSH iron sulfur domain 2 can attenuate cerebral ischemia/reperfusion injury,thus providing theoretical support from the perspectives of cytology and experimental zoology for the use of this protein as a therapeutic target in patients with cerebral ischemia/reperfusion injury.展开更多
BACKGROUND Diabetic gastroparesis(DGP)is a prevalent complication of diabetes that impairs people's quality of life and places a significant financial burden on them.The gastrointestinal symptoms of DGP patients c...BACKGROUND Diabetic gastroparesis(DGP)is a prevalent complication of diabetes that impairs people's quality of life and places a significant financial burden on them.The gastrointestinal symptoms of DGP patients can be improved by several Traditional Chinese Medicine(TCM)decoctions that have been shown to be effective in treating the disease.There are still many unanswered questions regarding the identification of appropriate therapeutic agents for the treatment of DGP in clinical practice.AIM To analyze the efficacy of several TCM decoctions in the treatment of DGP using Bayesian network meta-analysis for reference.METHODS PubMed,EMBASE,Cochrane Library,Web of Science,China National Knowledge Infrastructure,The China Biology Medicine DVD,Wanfang,and CQVIP were searched from inception to September 17,2022,to collect randomized controlled trials(RCTs)about TCM decoctions for DGP.Clinical effects and symptom scores were the primary outcomes.Additionally,we assessed motilin(MOT),somatostatin(SS),gastrin(GAS),gastric emptying rate,gastric emptying time,and adverse drug events as secondary outcomes.RESULTS A total of 67 eligible RCTs involving 4790 DGP patients and 7 TCM decoctions were included.The results of network meta-analysis(NMA)and surface under the cumulative ranking curve showed that with western medicine(WM)as a common control,the Banxia Xiexin Decoction(BXXD)+WM was most effective in clinical effects and enhancing early satiety scores;the Simo decoction(SMD)+WM was most effective in improving nausea and vomiting scores and anorexia scores,bloating scores;the Chaishao Liujunzi Decoction(CSLJD)was most effective in MOT,the Zhishi Xiaopi Decoction(ZSXPD)was most effective in SS and upgrading emptying rate;the Jianpi Xiaozhi Decoction was most effective in GAS;the CSLJD+WM was most effective in improving gastric emptying time.CONCLUSION These NMA results suggest that the BXXD+WM and SMD+WM may be one of the potential optimal treatments.Due to various limitations,further large-sample,double-blind,multi-center randomized RCTs are needed.展开更多
Objective:To explore the protective effects of ginseng mixture on myocardial fibrosis(MF)in rats.Methods:A total of 60 Wistar rats were randomly divided into control group without modeling operation,and another 4 grou...Objective:To explore the protective effects of ginseng mixture on myocardial fibrosis(MF)in rats.Methods:A total of 60 Wistar rats were randomly divided into control group without modeling operation,and another 4 groups using subcutaneous injections of isopropyl adrenaline for 10 d to set up the MF model:model group with saline lavage treatment after modeling,captopril group with captopril lavage,ginseng mixture group A and group B with low and high dose mixture treatment respectively.After treatment for 14 d,abdominal aorta and myocardial tissue were extracted to observe the pathological morphological changes and heart weight index in each group.Results:The left ventricular weight and heart heavy index of captopril group and group B were significantly lower than that of model group and group A(P<0.05);Model group and group A showed a higher hydroxyproline(Hyp)content in myocardial tissue than the control group and lower catalase(CAT)activity than Gontrol group(P<0.05);captopril group and group B showed a lower Hyp content and higher CAT activity compared with group A and model group(P<0.05),a significantly lower level of serum glutathione peroxidase(GSH-PX)and CAT and a higher level of serum creatine kinase,lactate dehydrogenase and H_2O_2 in model group and group A were observed compared with the control group(P<0.05).A higher level of GSH-PX and CAT and a lower level of creatine kinase,lactate dehydrogenase and H_2O_2 in captopril group and group B were observed compared with group A and model group(P<0.05);and histopathological examination showed that in captopril group and group B,secretion of collagen fiber was significantly inhibited and myocardial injury was significantly lighter than that of model group.Conclusions:Ginseng mixture plays a protective effect on myocardium by inhibiting antioxidant process of MF.展开更多
Objective:To observe the protection effect of rhTPO and granulocyte colony stimulating factor(G-CSFi on brain nerve after hypoxic ischemic brain damage(HIBD)in neonatal rats,exploring new ways for the laboratory basis...Objective:To observe the protection effect of rhTPO and granulocyte colony stimulating factor(G-CSFi on brain nerve after hypoxic ischemic brain damage(HIBD)in neonatal rats,exploring new ways for the laboratory basis of treatment for hypoxic ischemic encephalopathy,and provide for possible.Methods:A total of 120 newborn SD rats aging 7 d were randomly divided into control group,model group,TPO group and G-CSF group,using the method of blockingleft carotid artery to establish HIBD model.The left carotid artery was only seperated rather than blocked in the control group;after modeling,saline injection,rhTPO treatment and G-CSF treatment were adopted in the model group,TPO group and C-CSF group respectively.Then 10rats of 4 groups were executed at Day 3,7,14 after modeling,brain tissue was extracted to observe the brain damage:Immunohistochemical method was used to observe the histopathological changes of brain tissue and changes of nest protein(nestin)expression.Results:Injured brain mass of model group,TPO group and G-CSF group were significantly higher than that of control group at corresponding time point(P<0.05).Injured brain mass of TPO group and G-CSF group were significantly lower than that of model group(P<0.05),and with the increase of age,more significant increasing trend.At Day 3 after modeling,the expression of nestin positive cells in cerebral cortex of model group,TPO group and G-CSF gnmp increased significantly than that of control group{P<0.05);nestin positive cells of G-CSF group outnumbered TPO group significantly(P<0.05).Conclusions:The early TPO,G-CSF treatment of HIBD rats can improve brain function after hypoxia ischemia by neural protection.G-CSF can promote the differentiation of neural cells proliferation,and reduee degeneration and necrosis of nerve cells.展开更多
Objective The protein interacting with C kinase 1(PICK1)plays a critical role in vesicle trafficking,and its deficiency in sperm cells results in abnormal vesicle trafficking from Golgi to acrosome,which eventually di...Objective The protein interacting with C kinase 1(PICK1)plays a critical role in vesicle trafficking,and its deficiency in sperm cells results in abnormal vesicle trafficking from Golgi to acrosome,which eventually disrupts acrosome formation and leads to male infertility.Methods An azoospermia sample was filtered,and the laboratory detection and clinical phenotype indicated typical azoospermia in the patient.We sequenced all of the exons in the PICK1 gene and found that there was a novel homozygous variant in the PICK1 gene,c.364delA(p.Lys122SerfsX8),and this protein structure truncating variant seriously affected the biological function.Then we constructed a PICK1 knockout mouse model using clustered regularly interspaced short palindromic repeat cutting technology(CRISPRc).Results The sperm from PICK1 knockout mice showed acrosome and nucleus abnormalities,as well as dysfunctional mitochondrial sheath formation.Both the total sperm and motility sperm counts were decreased in the PICK1 knockout mice compared to wild-type mice.Moreover,the mitochondrial dysfunction was verified in the mice.These defects in the male PICK1 knockout mice may have eventually led to complete infertility.Conclusion The c.364delA novel variant in the PICK1 gene associated with clinical infertility,and pathogenic variants in the PICK1 may cause azoospermia or asthenospermia by impairing mitochondrial function in both mice and humans.展开更多

And 26 core components were obtained, including quercetin, luteolin, kaempferol, naringenin, and baicalein;The GO enrichment results showed 224 biological processes, 15 molecular functions and 33 cell components related to inflammation;There were 5 inflammatory signaling pathways in KEGG enrichment results, including TNF signaling pathway, Nod-like receptor signaling pathway, Toll-like receptor signaling pathway, MAPK signaling pathway and cytokine receptor interaction. Objective:To explore the changes of cannabinoid receptor(CB1)and AMPA receptor subunit GluA1 in the medial prefrontal cortex(mPFC)of depression model rats induced by chronic unpredictable mild stress(CUMS).Methods:30 ...Objective:To explore the changes of cannabinoid receptor(CB1)and AMPA receptor subunit GluA1 in the medial prefrontal cortex(mPFC)of depression model rats induced by chronic unpredictable mild stress(CUMS).Methods:30 rats were randomly divided into three groups.The three-week model group was given a three-week CUMS model,the four-week model group was given a four-week CUMS model,and the control group was given no treatment.The behavior of rats were detected in each group,and the expression of CB1 and GluA1 protein in synaptosomes in mPFC brain region was detected by Western blot.Results:Compared with the control group,the surcose preference decreased,the feeding latency increased in the novel inhibition feeding test,and the immobility time increased in the forced swimming test in the four-week model group.However,after three weeks of CUMS,there was no obvious change in the behavior of rats compared with the control group,suggesting that four-week model of CUMS was successful.CUMS can reduce the expression level of CB1 and GluA1 protein in synaptosomes of mPFC brain area in four-week model group(P<0.05),but there was no significant difference between three-week model group and control group.Conclusion:Four-week CUMS was more likely to lead to depressive-like behavior in rats,which may be closely related to the expression of CB1 and GluA1 in mPFC.展开更多
The coastal wetlands of the Yellow River Delta(YRD)in China are crucial for their valuable resources,environmental significance,and economic contributions.However,these wetlands are also vulnerable to the dual threats...The coastal wetlands of the Yellow River Delta(YRD)in China are crucial for their valuable resources,environmental significance,and economic contributions.However,these wetlands are also vulnerable to the dual threats of climate change and human disturbances.Despite substantial attention to the historical shifts in YRD's coastal wetlands,uncertainties remain regarding their future trajectory in the face of compound risks from climate change and anthropogenic activities.Based on a range of remote sensing data sources,this study undertakes a comprehensive investigation into the evolution of YRD's coastal wetlands between 2000 and 2020.Subsequently,the potential fate of coastal wetlands is thoroughly analyzed through the Land Use/Cover Change(LUCC)simulation using System Dynamic-Future Land Use Simulation(SD-FLUS)model and the extreme water levels projection integrated future sea-level rise,storm surge,and astronomical high tide in 2030,2050,and 2100 under scenarios of SSP1-2.6,SSP2-4.5,and SSP5-8.5.Results revealed that YRD's coastal wetlands underwent a marked reduction,shrinking by 1688.72 km²from 2000 to 2020.This decline was mostly attributed to the substantial expansion in the areas of artificial wetlands(increasing by 823.78 km2),construction land(increasing by 767.71 km²),and shallow water(increasing by 274.58 km²).Looking ahead to 2030-2100,the fate of coastal wetlands appears to diverge based on different scenarios.Under the SSP1-2.6 scenario,the area of coastal wetland is projected to experience considerable growth.In contrast,the SSP5-8.5 scenario anticipates a notable decrease in coastal wetlands.Relative to the inundated area suffered from the current extreme water levels,the study projects a decrease of 6.8%-10.6%in submerged coastal wetlands by 2030 and 9.4%-18.2%by 2050 across all scenarios.In 2100,these percentages are projected to decrease by 0.4%(SSP2-4.5)and 27.1%(SSP5-8.5),but increase by 35.7%(SSP1-2.6).Results suggest that coastal wetlands in the YRD will face a serious compound risk from climate change and intensified human activities in the future,with climate change being the dominant factor.More effcient and forward-looking measures must be implemented to prioritize the conservation and management of coastal wetland ecosystems to address the challenges,especially those posed by climate change.展开更多

文摘BACKGROUND Difficulty in obtaining tetracycline,increased adverse reactions,and relatively complicated medication methods have limited the clinical application of the classic bismuth quadruple therapy.Therefore,the search for new alternative drugs has become one of the research hotspots.In recent years,minocycline,as a semisynthetic tetracycline,has demonstrated good potential for eradicating Helicobacter pylori(H.pylori)infection,but the systematic evaluation of its role remains lacking.AIM To explore the efficacy,safety,and compliance of minocycline in eradicating H.pylori infection.METHODS We comprehensively retrieved the electronic databases of PubMed,Embase,Web of Science,China National Knowledge Infrastructure,SinoMed,and Wanfang database as of October 30,2023,and finally included 22 research reports on H.pylori eradication with minocycline-containing regimens as per the inclusion and exclusion criteria.The eradication rates of H.pylori were calculated using a fixed or a random effect model,and the heterogeneity and publication bias of the studies were measured.RESULTS The single-arm meta-analysis revealed that the minocycline-containing regimens achieved good overall H.pylori eradication rates,reaching 82.3%[95%confidence interval(CI):79.7%-85.1%]in the intention-to-treat analysis and 90.0%(95%CI:87.7%-92.4%)in the per-protocol analysis.The overall safety and compliance of the minocycline-containing regimens were good,demonstrating an overall incidence of adverse reactions of 36.5%(95%CI:31.5%-42.2%).Further by traditional meta-analysis,the results showed that the minocycline-containing regimens were not statistically different from other commonly used eradication regimens in eradication rate and incidence of adverse effects.Most of the adverse reactions were mild to moderate and well-tolerated,and dizziness was relatively prominent in the minocycline-containing regimens(16%).CONCLUSION The minocycline-containing regimens demonstrated good efficacy,safety,and compliance in H.pylori eradication.Minocycline has good potential to replace tetracycline for eradicating H.pylori infection.

文摘Time-frequency analysis is a successfully used tool for analyzing the local features of seismic data.However,it suffers from several inevitable limitations,such as the restricted time-frequency resolution,the difficulty in selecting parameters,and the low computational efficiency.Inspired by deep learning,we suggest a deep learning-based workflow for seismic time-frequency analysis.The sparse S transform network(SSTNet)is first built to map the relationship between synthetic traces and sparse S transform spectra,which can be easily pre-trained by using synthetic traces and training labels.Next,we introduce knowledge distillation(KD)based transfer learning to re-train SSTNet by using a field data set without training labels,which is named the sparse S transform network with knowledge distillation(KD-SSTNet).In this way,we can effectively calculate the sparse time-frequency spectra of field data and avoid the use of field training labels.To test the availability of the suggested KD-SSTNet,we apply it to field data to estimate seismic attenuation for reservoir characterization and make detailed comparisons with the traditional time-frequency analysis methods.

文摘In this editorial,we comment on the article by Mei et al.Nonalcoholic steatohep-atitis(NASH)is a severe inflammatory subtype of nonalcoholic fatty liver disease(NAFLD)with pathological features including steatosis,hepatocellular damage,and varying degrees of fibrosis.With the epidemic of metabolic diseases and obesity,the prevalence of NAFLD in China has increased,and it is now similar to that in developed countries;thus,NAFLD has become a major chronic liver disease in China.Human epidemiological data suggest that estrogen has a protective effect on NASH in premenopausal women and that sex hormones influence the development of liver disease.This review focuses on the path-ogenesis,treatment,and relationship between NASH and other diseases as well as on the relationship between NASH and sex hormone metabolism,with the aim of providing new strategies for the treatment of NASH.

文摘Objective:To investigate the preventive and therapeutic effect and mechanism of simvastatin on secondary inflammatory damage of rats with cerebral hemorrhage.Methods:Sixty SD rat aged 9-12 weeks were chosen and divided into the control group,model group and simvastatintreated group randomly with 20 rats in each group.Rats in the model group and simvastatintreated group were infused with autologous fresh uncoagulated blood to the right brain tissue of the basal ganglia to build the cerebral hemorrhage model,while rats in the control group were treated with the same amount of normal saline.Then,rats in the simvastatin-treated group were given a gavage of 3 mg/kg of simvastatin once a day after modeling.Rats in the three groups were given nerve dysfunction score(NDS) and wet-dry weighting method was used to detect the brain water content(BWC) of brain tissues around the lesion of the rats.Then Nissl staining was conducted and the undamaged neurons were counted.Immunohistochemical SP method was applied to count the number of NF-d the immuno fluorκB,TLR4 and IL-1escence method wasβ positive cells in brain tissues around the lesions,an employed to determine the expression levels of NF-κB,TLR4 and IL-1me points were aβ proteins.Results:The NDS results of the simvastatin-treated group at all till significantly higher than those of the model group(P < 0.05);the BWC values of the simvastatin-treated group at all time points were all significantly lower than those of the model group at the same periods(P < 0.05);the number of the undamaged neurons around the lesions of the simvastatin-treated group at all time points were all significantly higher than those of the model group(P < 0.05);seven days after treatment,the number of the NF-κB,TLR4 and IL-1β positive cells in brain tissues around the lesions of the simvastatin-treated group were all significantly lower than those of the model group(P < 0.05),and its expression levels of NF-ower than those of the model group(κB,TLR4 and IL-1P < 0.05).Conclusioβ protein were also significantly lns:Simvastatin can inhibit the expressions of NF-κB,TLR4 and IL-1β proteins in rats with cerebral hemorrhage,and protect neurons and reduce secondary inflammatory damages by down-regulating the above protein-mediated inflammatory responses.

文摘To effectively replace the isotope radiation source in litho-density logging,this study presents a method for measuring the formation density and photoelectric absorption index(Pe)using a switchable X-ray tube.First,the gamma-ray litho-density logging(GLD)method for measuring formation density and Pe using chemical sources is introduced.Then,a benchmark verification based on the X-ray litho-density logging tool prototype and data published by Simon(In:Paper presented at the SPWLA 59th annual logging symposium,London,UK,2018)was carried out using Monte Carlo numerical simulations.Second,the impacts of the photoelectric effect and detector statistical error on the GLD method were analyzed.Finally,based on a theoretical analysis,the formation density and Pe measurement algorithm(double energy window(DEW)method)was improved,which was found to be suitable for X-ray litho-density logging.Moreover,the results obtained using this algorithm were compared with those obtained using the GLD method.The results indicate that owing to the impact of photoelectric effect and detector statistical error on the density energy window,the accuracy of formation density and Pe measurement using the GLD method is relatively low,with the uncertainty in formation density and Pe measurement reaching 2.620±0.047 g/cm3 and 4.090±0.580 b/e,respectively.In comparison,the DEW method can improve the accuracy of density and Pe measurement to 0.006 g/cm3 and 0.065 b/e,respectively,as the photoelectric effect in the density window is corrected using the counts in the lithology window of the energy spectrum.This study aims to provide a new theoretical foundation for processing X-ray litho-density logs in the future.

文摘Objective:To detect the CHRNA7 gene mutation and polymorphism in Southern Han Chinese patients with nocturnal frontal lobe epilepsy(NFLE).Methods:Blood samples were collected from 215 Southern Han Chinese patients with NFLE and 200 healthy Southern Han Chinese control subjects.Genomic DNA was extracted,and CHRNA7 whole genome exons were amplified by the polymerase chain reaction and subjected to Sanger sequencing.Results:No CHRNA7 gene mutation was detected in all of the NFLE patients.However,five single nucleotide polymorphisms(SNPs)in sporadic cases were found,located in exons 5,6.and 7 of the CHRNA7 gene.Among them,c.690G>A and c.698A>G are known SNPs,while c.370G>A,c.654C>T,and c.497-498delTG were newly discovered SNPs.These SNPs were also found in some of the healthy controls.Conclusions:No CHRNA7 gene mutation was identified in Southern Han Chinese patients with NFLE.The CHRNA7 gene is probably not responsible for NFLE in this population.

文摘The Shanghai high-repetition-rate XFEL and extreme light facility(SHINE)under construction is designed to be one of the most advanced free electron laser(FEL)facilities in the world.The main part of the SHINE facility is an 8 GeV superconducting Linac operating in continuous wave(CW)mode.The Linac consists of seventy-five 1.3 GHz and two 3.9 GHz cryomodules.Based on the design and experience of 3.9 GHz cavities in the European X-ray free electron laser(E-XFEL)and the Linac coherent light source-II(LCLS-II)projects,we optimize the SHINE 3.9 GHz cavity design to adapt it for CW mode operation.In this paper,we present a particular redesign of the end group for the SHINE 3.9 GHz superconducting cavity that includes a redesign of the end cell and beam pipe to shift away the potentially troublesome lowest highorder modes(HOMs),a modification of the main coupler antenna,and a tuning of the HOM notch filter to meet the cavity requirements.RF losses calculations on the HOM coupler antennas show that the overheating on the inner conductor at the operating mode is diminished signifi-cantly.Furthermore,we have also studied the HOMs to ensure there are no dangerously trapped modes in the optimized cavity design.

文摘CDGSH iron sulfur domain 2 can inhibit ferroptosis,which has been associated with cerebral ischemia/reperfusion,in individuals with head and neck cancer.Therefore,CDGSH iron sulfur domain 2 may be implicated in cerebral ischemia/reperfusion injury.To validate this hypothesis in the present study,we established mouse models of occlusion of the middle cerebral artery and HT22 cell models of oxygen-glucose deprivation and reoxygenation to mimic cerebral ischemia/reperfusion injury in vivo and in vitro,respectively.We found remarkably decreased CDGSH iron sulfur domain 2 expression in the mouse brain tissue and HT22 cells.When we used adeno-associated virus and plasmid to up-regulate CDGSH iron sulfur domain 2 expression in the brain tissue and HT22 cell models separately,mouse neurological dysfunction was greatly improved;the cerebral infarct volume was reduced;the survival rate of HT22 cells was increased;HT22 cell injury was alleviated;the expression of ferroptosis-related glutathione peroxidase 4,cystine-glutamate antiporter,and glutathione was increased;the levels of malondialdehyde,iron ions,and the expression of transferrin receptor 1 were decreased;and the expression of nuclear-factor E2-related factor 2/heme oxygenase 1 was increased.Inhibition of CDGSH iron sulfur domain 2 upregulation via the nuclear-factor E2-related factor 2 inhibitor ML385 in oxygen-glucose deprived and reoxygenated HT22 cells blocked the neuroprotective effects of CDGSH iron sulfur domain 2 up-regulation and the activation of the nuclear-factor E2-related factor 2/heme oxygenase 1 pathway.Our data indicate that the up-regulation of CDGSH iron sulfur domain 2 can attenuate cerebral ischemia/reperfusion injury,thus providing theoretical support from the perspectives of cytology and experimental zoology for the use of this protein as a therapeutic target in patients with cerebral ischemia/reperfusion injury.

文摘BACKGROUND Diabetic gastroparesis(DGP)is a prevalent complication of diabetes that impairs people's quality of life and places a significant financial burden on them.The gastrointestinal symptoms of DGP patients can be improved by several Traditional Chinese Medicine(TCM)decoctions that have been shown to be effective in treating the disease.There are still many unanswered questions regarding the identification of appropriate therapeutic agents for the treatment of DGP in clinical practice.AIM To analyze the efficacy of several TCM decoctions in the treatment of DGP using Bayesian network meta-analysis for reference.METHODS PubMed,EMBASE,Cochrane Library,Web of Science,China National Knowledge Infrastructure,The China Biology Medicine DVD,Wanfang,and CQVIP were searched from inception to September 17,2022,to collect randomized controlled trials(RCTs)about TCM decoctions for DGP.Clinical effects and symptom scores were the primary outcomes.Additionally,we assessed motilin(MOT),somatostatin(SS),gastrin(GAS),gastric emptying rate,gastric emptying time,and adverse drug events as secondary outcomes.RESULTS A total of 67 eligible RCTs involving 4790 DGP patients and 7 TCM decoctions were included.The results of network meta-analysis(NMA)and surface under the cumulative ranking curve showed that with western medicine(WM)as a common control,the Banxia Xiexin Decoction(BXXD)+WM was most effective in clinical effects and enhancing early satiety scores;the Simo decoction(SMD)+WM was most effective in improving nausea and vomiting scores and anorexia scores,bloating scores;the Chaishao Liujunzi Decoction(CSLJD)was most effective in MOT,the Zhishi Xiaopi Decoction(ZSXPD)was most effective in SS and upgrading emptying rate;the Jianpi Xiaozhi Decoction was most effective in GAS;the CSLJD+WM was most effective in improving gastric emptying time.CONCLUSION These NMA results suggest that the BXXD+WM and SMD+WM may be one of the potential optimal treatments.Due to various limitations,further large-sample,double-blind,multi-center randomized RCTs are needed.

文摘Objective:To explore the protective effects of ginseng mixture on myocardial fibrosis(MF)in rats.Methods:A total of 60 Wistar rats were randomly divided into control group without modeling operation,and another 4 groups using subcutaneous injections of isopropyl adrenaline for 10 d to set up the MF model:model group with saline lavage treatment after modeling,captopril group with captopril lavage,ginseng mixture group A and group B with low and high dose mixture treatment respectively.After treatment for 14 d,abdominal aorta and myocardial tissue were extracted to observe the pathological morphological changes and heart weight index in each group.Results:The left ventricular weight and heart heavy index of captopril group and group B were significantly lower than that of model group and group A(P<0.05);Model group and group A showed a higher hydroxyproline(Hyp)content in myocardial tissue than the control group and lower catalase(CAT)activity than Gontrol group(P<0.05);captopril group and group B showed a lower Hyp content and higher CAT activity compared with group A and model group(P<0.05),a significantly lower level of serum glutathione peroxidase(GSH-PX)and CAT and a higher level of serum creatine kinase,lactate dehydrogenase and H_2O_2 in model group and group A were observed compared with the control group(P<0.05).A higher level of GSH-PX and CAT and a lower level of creatine kinase,lactate dehydrogenase and H_2O_2 in captopril group and group B were observed compared with group A and model group(P<0.05);and histopathological examination showed that in captopril group and group B,secretion of collagen fiber was significantly inhibited and myocardial injury was significantly lighter than that of model group.Conclusions:Ginseng mixture plays a protective effect on myocardium by inhibiting antioxidant process of MF.

文摘Objective:To observe the protection effect of rhTPO and granulocyte colony stimulating factor(G-CSFi on brain nerve after hypoxic ischemic brain damage(HIBD)in neonatal rats,exploring new ways for the laboratory basis of treatment for hypoxic ischemic encephalopathy,and provide for possible.Methods:A total of 120 newborn SD rats aging 7 d were randomly divided into control group,model group,TPO group and G-CSF group,using the method of blockingleft carotid artery to establish HIBD model.The left carotid artery was only seperated rather than blocked in the control group;after modeling,saline injection,rhTPO treatment and G-CSF treatment were adopted in the model group,TPO group and C-CSF group respectively.Then 10rats of 4 groups were executed at Day 3,7,14 after modeling,brain tissue was extracted to observe the brain damage:Immunohistochemical method was used to observe the histopathological changes of brain tissue and changes of nest protein(nestin)expression.Results:Injured brain mass of model group,TPO group and G-CSF group were significantly higher than that of control group at corresponding time point(P<0.05).Injured brain mass of TPO group and G-CSF group were significantly lower than that of model group(P<0.05),and with the increase of age,more significant increasing trend.At Day 3 after modeling,the expression of nestin positive cells in cerebral cortex of model group,TPO group and G-CSF gnmp increased significantly than that of control group{P<0.05);nestin positive cells of G-CSF group outnumbered TPO group significantly(P<0.05).Conclusions:The early TPO,G-CSF treatment of HIBD rats can improve brain function after hypoxia ischemia by neural protection.G-CSF can promote the differentiation of neural cells proliferation,and reduee degeneration and necrosis of nerve cells.

文摘Objective The protein interacting with C kinase 1(PICK1)plays a critical role in vesicle trafficking,and its deficiency in sperm cells results in abnormal vesicle trafficking from Golgi to acrosome,which eventually disrupts acrosome formation and leads to male infertility.Methods An azoospermia sample was filtered,and the laboratory detection and clinical phenotype indicated typical azoospermia in the patient.We sequenced all of the exons in the PICK1 gene and found that there was a novel homozygous variant in the PICK1 gene,c.364delA(p.Lys122SerfsX8),and this protein structure truncating variant seriously affected the biological function.Then we constructed a PICK1 knockout mouse model using clustered regularly interspaced short palindromic repeat cutting technology(CRISPRc).Results The sperm from PICK1 knockout mice showed acrosome and nucleus abnormalities,as well as dysfunctional mitochondrial sheath formation.Both the total sperm and motility sperm counts were decreased in the PICK1 knockout mice compared to wild-type mice.Moreover,the mitochondrial dysfunction was verified in the mice.These defects in the male PICK1 knockout mice may have eventually led to complete infertility.Conclusion The c.364delA novel variant in the PICK1 gene associated with clinical infertility,and pathogenic variants in the PICK1 may cause azoospermia or asthenospermia by impairing mitochondrial function in both mice and humans.

文摘Objective:To establish the quality standard of Danmu exract syrup.Methods:TLC method was used to qualitatively identify Nauclea officinalis in Danmu exract syrup.The content of strictosamide and chlorogenic acid in Danmu exract syrup was determined by high efficiency axillary chromatography.Results:The spots were clear and there was no negative interference.The linear relationship of strictosamide was good in the range of 100~300ug/ml(R^(2)=0.9997).The average recovery was 99.1%and RSD was 1.16%(n=9).Chlorogenic acid had a good linear relationship in the range of 6.7~20.2ug/ml(R^(2)=0.9991).The average recovery was 99.9%and RSD was 1.10%(n=9).Conclusion:The standard is safe and reasonable,the method is simple,accurate and reproducible,and it can be used for the quality control of Danmu exract syrup.

文摘Objective:Based on the quality analysis of Nauclea officinalis in different growth years and different months in the same year,the most suitable harvest year and the specific month in the same year were determined according to the evaluation of the active components in the stem of medicinal parts of Nauclea officinalis.Methods:LC-MS/MS method was used to investigate the quality of Nauclea officinalis from different growth years and different months in the same year,mainly including the content determination of the active ingredients in the medicine.Results:With the prolongation of production life,the content of active ingredients in stem of Nauclea officinalis increased continuously.After reaching a certain number of years,the content of active ingredients increased little;In the middle of the same year,from mid-October to mid-June of the next year,the content of iso-vincin lactam was relatively high,especially between the end of March and the end of May.The content was high and stable,and the highest point was the stem sample collected in May.Conclusion:The content of each component in the stem of Nauclea officinalis with a growth life of more than 5 years has reached a relatively high and stable level.Nauclea officinalis with a growth life of more than 5 years should be collected as raw materials for medicinal materials.The month with high and stable content of isocyanoside lactam,namely May of each year,was taken as the best harvest time of gallbladder in one year.The best harvest time was from mid-October to mid-June of the next year,or from March to May of each year.

文摘Objective:To investigate the effects of chronic unpredictable mild stress(CUMS)on perineuronal nets(PNNs)andγ-aminobutyric acid(GABA)-ergic neurons in the medial prefrontal cortex(mPFC)of adult rats.Methods:28 rats were randomly divided into two groups.The model group adopted CUMS to establish a depression model,and the control group did not give any treatment.The density of PNNs and the percentage of PNNs positive(PNNs^(+))and parvalbumin positive(PV^(+))neurons in total PV^(+)neurons in the mPFC were detected by immunofluorescence.The protein expression of main components of PNNs,Aggrecan and Brevican,and GABA main synthase glutamic acid decarboxylase 67(GAD67)in the mPFC were detected by Western blot.Results:The density of PNNs and the percentage of PNNs^(+)and PV^(+)neurons in total PV^(+)neurons in the mPFC in the model group were decreased compared with the control group(P<0.05);The expression levels of PNNs component proteins Aggrecan and Brevican,and GABA main synthase GAD67 were also decreased in the model group(P<0.01).Conclusion:The levels of PNNs and GABA synthase GAD67 in the mPFC of rats were decreased after chronic unpredictable mild stress.

文摘Objective:To explore the possible mechanism of Qingfei Paibu Decoction in the treatment of COVID-19 from the perspective of cytokine storm by network pharmacology.Methods: The TCMSP and SymMap databases were used to screen out the active components and targets of Qingfei Paibu Decoction;The GeneCards database was used to screen the predicted targets of COVID-19;Two targets were mapped;The STRING database and Cytoscape3.7.2 software were used to construct the network diagram of active drag-ingredient-target and screen out the core components and targets;The GO enrichment analysis and KEGG pathway enrichment analysis were carried out by OmicShare cloud platform and David respectively.Results:A total of 52 potential targets of Qingfei Paibu Decoction for the treatment of COVID-19 were obtained, among which 17 were core targets related to inflammation, mainly including cytokines such as IL, IFN, TNF and chemokines. And 26 core components were obtained, including quercetin, luteolin, kaempferol, naringenin, and baicalein;The GO enrichment results showed 224 biological processes, 15 molecular functions and 33 cell components related to inflammation;There were 5 inflammatory signaling pathways in KEGG enrichment results, including TNF signaling pathway, Nod-like receptor signaling pathway, Toll-like receptor signaling pathway, MAPK signaling pathway and cytokine receptor interaction. Conclusion: Qingfei Paibu Decoction can inhibit cytokine storms by acting on multiple targets and pathways with multiple components and thus treat COVID-19.

文摘Objective:To explore the changes of cannabinoid receptor(CB1)and AMPA receptor subunit GluA1 in the medial prefrontal cortex(mPFC)of depression model rats induced by chronic unpredictable mild stress(CUMS).Methods:30 rats were randomly divided into three groups.The three-week model group was given a three-week CUMS model,the four-week model group was given a four-week CUMS model,and the control group was given no treatment.The behavior of rats were detected in each group,and the expression of CB1 and GluA1 protein in synaptosomes in mPFC brain region was detected by Western blot.Results:Compared with the control group,the surcose preference decreased,the feeding latency increased in the novel inhibition feeding test,and the immobility time increased in the forced swimming test in the four-week model group.However,after three weeks of CUMS,there was no obvious change in the behavior of rats compared with the control group,suggesting that four-week model of CUMS was successful.CUMS can reduce the expression level of CB1 and GluA1 protein in synaptosomes of mPFC brain area in four-week model group(P<0.05),but there was no significant difference between three-week model group and control group.Conclusion:Four-week CUMS was more likely to lead to depressive-like behavior in rats,which may be closely related to the expression of CB1 and GluA1 in mPFC.

文摘The coastal wetlands of the Yellow River Delta(YRD)in China are crucial for their valuable resources,environmental significance,and economic contributions.However,these wetlands are also vulnerable to the dual threats of climate change and human disturbances.Despite substantial attention to the historical shifts in YRD's coastal wetlands,uncertainties remain regarding their future trajectory in the face of compound risks from climate change and anthropogenic activities.Based on a range of remote sensing data sources,this study undertakes a comprehensive investigation into the evolution of YRD's coastal wetlands between 2000 and 2020.Subsequently,the potential fate of coastal wetlands is thoroughly analyzed through the Land Use/Cover Change(LUCC)simulation using System Dynamic-Future Land Use Simulation(SD-FLUS)model and the extreme water levels projection integrated future sea-level rise,storm surge,and astronomical high tide in 2030,2050,and 2100 under scenarios of SSP1-2.6,SSP2-4.5,and SSP5-8.5.Results revealed that YRD's coastal wetlands underwent a marked reduction,shrinking by 1688.72 km²from 2000 to 2020.This decline was mostly attributed to the substantial expansion in the areas of artificial wetlands(increasing by 823.78 km2),construction land(increasing by 767.71 km²),and shallow water(increasing by 274.58 km²).Looking ahead to 2030-2100,the fate of coastal wetlands appears to diverge based on different scenarios.Under the SSP1-2.6 scenario,the area of coastal wetland is projected to experience considerable growth.In contrast,the SSP5-8.5 scenario anticipates a notable decrease in coastal wetlands.Relative to the inundated area suffered from the current extreme water levels,the study projects a decrease of 6.8%-10.6%in submerged coastal wetlands by 2030 and 9.4%-18.2%by 2050 across all scenarios.In 2100,these percentages are projected to decrease by 0.4%(SSP2-4.5)and 27.1%(SSP5-8.5),but increase by 35.7%(SSP1-2.6).Results suggest that coastal wetlands in the YRD will face a serious compound risk from climate change and intensified human activities in the future,with climate change being the dominant factor.More effcient and forward-looking measures must be implemented to prioritize the conservation and management of coastal wetland ecosystems to address the challenges,especially those posed by climate change.
