Multidrug resistance gene-1 (MDR1) gene expression was assayed by semi-quantitative reverse transcriptionpolymerase chain reaction and the expression of p53 protein was examined by Western blotting.RESULTS: The reversal activity after treatment with wide type p53 gene was increased up to 4.982 fold at 48 h. The expression of MDR1 gene decreased significantly after treatment with wide-type p53 gene, and the expression of p53 protein lasted for about 5 d, with a peak at 48 h, and began to decrease at 72 h.CONCLUSION: Wide-type p53 gene has a remarkable reversal activity for the high expression of MDR1 gene in colorectal cancers. The reversal effects seem to be in a time dependent manner. It might have good prospects in clinical application.展开更多
AIM: To investigate the antitumor activity of adriamycin (ADR) encapsulated in nanoparticles (NADR) and injected into the hepatic artery of hepatoma-bearing rats.METHODS: NADR was prepared by the interfacial polymeriz...AIM: To investigate the antitumor activity of adriamycin (ADR) encapsulated in nanoparticles (NADR) and injected into the hepatic artery of hepatoma-bearing rats.METHODS: NADR was prepared by the interfacial polymerization method. Walker-256 carcinosarcomas were surgically implanted into the left liver lobes of 60 male Wistar rats, which were divided into 4 groups at random (15 rats per group). On the 7th day after implantation, normal saline (NS), free ADR (FADR), NADR, or ADR mixed with unloaded nanoparticles (ADR+NP) was respectively injected via the hepatic artery (i.a.) of rats in different groups. The dose of ADR in each formulation was 2.0 mg/kg body weight and the concentration was 1.0 mg/mL. Survival time, tumor enlargement ratio, and tumor necrosis degree were compared between each group.RESULTS: Compared with the rats that received NS i.a.,the rats that received FADR or ADR+NP acquired apparent inhibition on tumor growth, as well as prolonged their life span. Further significant anticancer efficacy was observed in rats that received i.a. administration of NADR. Statistics indicated that NADR brought on a more significant tumor inhibition and more extensive tumor necrosis, as compared to FADR or ADR+NP. The mean tumor enlargement ratio on the 7th day after NADR i.a. was 1.106. The mean tumorbearing survival time was 39.50 days. Prolonged life span ratio was 109.22% as compared with rats that accepted NS.CONCLUSION: Therapeutic effect of ADR on liver malignancy can be significantly enhanced by its nanopaticle formulation and administration via hepatic artery.展开更多

Cytogenetic study was conducted by using flow cytometry and chromosome analysis. Expressions of tumor markers such as α-fetoprotein (AFP), cytokeratin (CK) and hepatoma metastasis-associated factor HAb18G/CD147 on the FHCC-98 cells were detected by immunocytochemistry or Western blotting. Lactic dehydrogenase (LDH) isoenzymes were detected by polyacrylamide gel electrophoresis (PAGE).Xenograft was performed by inoculating FHCC-98 cells into the flanks of nude mice.RESULTS: Morphology of FHCC-98 cells was the same as that of other malignant cells. The expressions of the cells were positive for HAb18G/CD147 and CK, and negative for AFP. Its population doubling time was 21.4 h. The cell DNA was tetraploid and the major chromosomes were triploid by cytogenetics analysis. The tumorigenicity in nude mice was 100%. PAGE showed four bands representing LDH2, LDH3,LDH4 and LDH5.CONCLUSION: FHCC-98 is a novel HCC cell line and an ideal cell model for further exploring the mechanism of hepatocellular carcinoma invasion and metastasis.展开更多

Large crystal fused magnesia, natural flake graphite with different particle sizes and anti-oxidant were adopted as raw material for preparation of specimens. However, the results show that the physical properties, oxidation resistance and thermal shock resistance of low carbon MgO-C materials with content of 4.0wt% graphite are improved obviously through the use of special and suitable size graphite. The excellent performance achieved was considered as a result of microstructure modification of MgO-C materials. Therefore, it is suggested that both fine and micro grade natural flake graphite used for production of low carbon MgO-C bricks.展开更多

With the increase of Cd addition the strength of alloy rises at first...The present work has investigated the effect of trace elements Cd and Sn on the microstructure and mechanical properties of Al-Si-Cu-Mg cast alloy. With the increase of Cd addition the strength of alloy rises at first and then drops. The optimal amount of Cd and Sn addition for Al-Si-Cu-Mg alloy is about 0.27% and 0.1% respectively. Due to the formation of some coarse Cd-rich phases and pure Cd particles the mechanical properties of alloy decrease when Cd amount exceeds 0.27%. When more than 0.1% Sn added, some Sn atoms form low-melting eutectic compound at grain boundary, and then cause over-burning in alloy when solution treated, which may deteriorate properties of alloy, especially ductility of alloy. On the other hand, the addition of Cd and Sn remarkably increases the peak hardness and reduces the time to reach aging peak in Al-Si-Cu-Mg alloy. The action of Cd /Sn in quaternary Al-Si-Cu-Mg alloy is effectively the same as that occur in binary Al-Cu alloy that the enhanced hardening associated with Cd / Sn addition is due to the promotion of the 6’ phase.展开更多

Background Central venous pressure (CVP) and intrathoracic blood volume index (ITBVI) were used to assess the fluid status. It has previously been shown that CVP is not as accurate as ITBVI for all the shock patie...Background Central venous pressure (CVP) and intrathoracic blood volume index (ITBVI) were used to assess the fluid status. It has previously been shown that CVP is not as accurate as ITBVI for all the shock patients. We therefore hypothesized that the change of CVP has the ability to predict fluid responsiveness in some clinical cases of shock. Methods From September 1st 2009 to September 1st 2011, sixty-three patients with shock from different Intensive Care Unit (ICU) were collected into this retrospective study. All the patients received fluid challenge strategy (infusing 300 ml hydroxyethyl starch in 20 minutes), were monitored with CVP and pulse-indicated continuous cardiac output (PICCO). The correlation between changes in cardiac index (ACI), CVP (ACVP) and ITBVI (AITBVI) were analyzed. Fluid responsiveness was defined as an increase in CI 〉10%. Receiver operating characteristic (ROC) curves were generated for ACVP and AITBVI. Results For all the patients, there was no correlation between ACI and ACVP (P=0.073), but in the subgroup analysis, the correlation between ACI and ACVP was significant in those younger than 60 years old (P=0.018) and those with hypovolemic shock (P=0.001). The difference of areas under the ROC curves of ACVP and AITBVI were not statistically significant in the group younger than 60 years old or hypovolemic shock group (P 〉0.05, respectively). However, no similar results can be found in the group older than 60 years old and the other two shock type groups from ROC curves of ACVP and AITBVI. Conclusions ACVP is not suitable for evaluating the volume status of the shock patients with fluid resuscitation regardless of their condition. However, in some ways, ACVP have the ability to predict fluid responsiveness in the younger shock patients or in the hypovolemic shock patients.展开更多
