利福喷丁联合氟康唑对耐药白念珠菌感染小鼠的作用及免疫机制分析 被引量:5

目的:观察利福喷丁联合氟康唑对耐药白念珠菌感染小鼠的保护作用,并探讨其作用机制。方法:100只ICR小鼠随机分为5组:空白对照组、模型组、利福喷丁组(0. 9 mg·kg^(-1))、氟康唑组(0. 5 mg·kg^(-1))、利福喷丁+氟康唑组(0. 9 m... 目的:观察利福喷丁联合氟康唑对耐药白念珠菌感染小鼠的保护作用,并探讨其作用机制。方法:100只ICR小鼠随机分为5组:空白对照组、模型组、利福喷丁组(0. 9 mg·kg^(-1))、氟康唑组(0. 5 mg·kg^(-1))、利福喷丁+氟康唑组(0. 9 mg·kg^(-1)+0. 5 mg·kg^(-1))。除空白对照组外,给予各组小鼠尾静脉注射耐药白念珠菌100悬液处理,构建真菌感染模型。感染2h后,利福喷丁组、氟康唑组、利福喷丁+氟康唑组分别腹腔注射给药,空白对照组和模型组腹腔注射等体积生理盐水,1次/d,连续给药7 d。真菌感染后连续观察30 d,统计各组大鼠生存情况,检测Th1、Th17、Th2细胞比例,观察肾脏组织病变情况及肾脏载菌量。结果:与空白对照组比较,模型组小鼠平均存活时间明显缩短,肾脏组织现大量真菌菌丝、孢子和炎症性病变,肾脏载菌量明显增多,外周血Th1、Th17细胞比例明显降低,Th2细胞比例明显增高(P <0. 05)。与模型组比较,利福喷丁组小鼠平均存活时间、肾脏组织病变程度及肾脏载菌量差异均无统计学意义(P> 0. 05);氟康唑组及利福喷丁+氟康唑组小鼠平均存活时间显著延长,肾脏组织中菌丝和孢子减少,肾脏组织病变程度减轻,肾脏载菌量显著减少,外周血Th1、Th17细胞比例显著升高,Th2细胞比例显著降低(P <0. 05);且联合作用效果显著优于氟康唑单独作用(P <0. 05)。结论:利福喷丁联合氟康唑可增强氟康唑对耐药白念珠菌的抗菌作用,其机制可能与调节Th1、Th17、Th2细胞分化有关。 展开更多

Objective:Hereditary colorectal cancer(CRC)accounts for approximately 5%–10%of all CRC cases.The full profile of CRC-related germline mutations and the corresponding somatic mutational profile have not been fully determined in the Chinese population.Methods:We performed the first population study investigating the germline mutation status in more than 1,000(n=1,923)Chinese patients with CRC and examined their relationship with the somatic mutational landscape.Germline alterations were examined with a 58-gene next-generation sequencing panel,and somatic alterations were examined with a 605-gene panel.Results:A total of 92 pathogenic(P)mutations were identified in 85 patients,and 81 likely pathogenic(LP)germline mutations were identified in 62 patients,accounting for 7.6%(147/1,923)of all patients.MSH2 and APC was the most mutated gene in the Lynch syndrome and non-Lynch syndrome groups,respectively.Patients with P/LP mutations had a significantly higher ratio of microsatellite instability,highly deficient mismatch repair,family history of CRC,and lower age.The somatic mutational landscape revealed a significantly higher mutational frequency in the P group and a trend toward higher copy number variations in the non-P group.The Lynch syndrome group had a significantly higher mutational frequency and tumor mutational burden than the nonLynch syndrome group.Clustering analysis revealed that the Notch signaling pathway was uniquely clustered in the Lynch syndrome group,and the MAPK and cAMP signaling pathways were uniquely clustered in the non-Lynch syndrome group.Population risk analysis indicated that the overall odds ratio was 11.13(95%CI:8.289–15.44)for the P group and 20.68(95%CI:12.89–33.18)for the LP group.Conclusions:Distinct features were revealed in Chinese patients with CRC with germline mutations.The Notch signaling pathway was uniquely clustered in the Lynch syndrome group,and the MAPK and cAMP signaling pathways were uniquely clustered in the non-Lynch syndrome group.Patients with P/LP germline mutations exhibited higher CRC risk. 展开更多

Modeling the heterogeneous traffic flow considering the effect of self-stabilizing and autonomous vehicles

With the increasing maturity of automatic driving technology,the homogeneous traffic flow will gradually evolve into the heterogeneous traffic flow,which consists of human-driving and autonomous vehicles.To better study the characteristics of the heterogeneous traffic system,this paper proposes a new car-following model for autonomous vehicles and heterogeneous traffic flow,which considers the self-stabilizing effect of vehicles.Through linear and nonlinear methods,this paper deduces and analyzes the stability of such a car-following model with the self-stabilizing effect.Finally,the model is verified by numerical simulation.Numerical results show that the self-stabilizing effect can make the heterogeneous traffic flow more stable,and that increasing the self-stabilizing coefficient or historical time length can strengthen the stability of heterogeneous traffic flow and alleviate traffic congestion effectively.In addition,the heterogeneous traffic flow can also be stabilized with a higher proportion of autonomous vehicles. 展开更多

In this study, we investigated the effect of different types of light and MeJA treatment on the accumulation of betulin and oleanolic acid in various organs of white birch. Our results showed that betulin and oleanolic were accumulated mainly in the stalk skin. The content of both substances in the stalk skin was significantly affected by seasons with a peak accumulation in August. The content of oleanolic and betulin was significantly decreased in the stem skin treated with 4 types of light (red, yellow, blue and green) compared with the plant with normal illumination. In contrast, oleanolic acid in leaves was increased by 13.28 folds when the white birch was treated with green light. Betulin was increased by 1.959 folds in leaves of white birch treated with blue light. The highest content of betulin and oleanolic acid in various organs of birch with appropriate shading treatment (light transmittance: 50%) was increased by 45.09% and 30.50%, respectively, in comparison with those with non-shading treatment. Content of oleanolic acid and betulin can be significantly improved in various parts of birch after treatment with different concentration of MeJA. The study lays the foundation to metabolic regulation of oleanolic acid and betulin in birch. 展开更多

Biomarker detection is key to identifying health risks.However,designing sensitive and single-use biosensors for early diagnosis remains a major challenge.Here,we report submonolayer lasers on optical fibers as ultrasensitive and disposable biosensors.Telecom optical fibers serve as distributed optical microcavities with high Q-factor,great repeatability,and ultralow cost,which enables whispering-gallery laser emission to detect biomarkers.It is found that the sensing performance strongly depends on the number of gain molecules.The submonolayer lasers obtained a six-order-of-magnitude improvement in the lower limit of detection(LOD)when compared to saturated monolayer lasers.We further achieve an ultrasensitive immunoassay for a Parkinson's disease biomarker,alpha-synuclein(α-syn),with a lower LOD of 0.32 pM in serum,which is three orders of magnitude lower than theα-syn concentration in the serum of Parkinson's disease patients.Our demonstration of submonolayer biolaser offers great potentials in high-throughput clinical diagnosis with ultimate sensitivity. 展开更多

Acousto-optic interactions, employed in the ultrafast laser regulation,possess remarkable advantages for fast tuning performance in a wide spectral range. Here, we propose an ultrafast fiber laser whose wideband tunability is provided by an acousto-optic structure fabricated with an etched single-mode fiber. Because of the laser polarization conversion induced by the coupling between the core and cladding vector modes in the etched fiber,a band-pass characteristic of the acousto-optic interaction is achieved to effectively regulate the inner-cavity gain range. Cooperating with a saturable absorber based on single-wall carbon nanotubes (SWCNTs) with polarization robustness,a soliton operating state is achieved in the tunable erbium-doped fiber laser.By controlling the acoustical wave frequency from 1.039 to 1.069 MHz, this soliton laser can be conveniently tuned in a wide spectral range from 1571.52 to 1539.26nm.Meanwhile, the laser pulses have near-transform-limited durations stably maintaining less than 2 ps at different wavelength channels,owing to the broadband nonlinear absorption of SWCNTs. 展开更多

Few-shot Learning for Named Entity Recognition Based on BERT and Two-level Model Fusion

Currently,as a basic task of military document information extraction,Named Entity Recognition(NER)for military documents has received great attention.In 2020,China Conference on Knowledge Graph and Semantic Computing(CCKS)and System Engineering Research Institute of Academy of Military Sciences(AMS)issued the NER task for test evaluation,which requires the recognition of four types of entities including Test Elements(TE),Performance Indicators(PI),System Components(SC)and Task Scenarios(TS).Due to the particularity and confidentiality of the military field,only 400 items of annotated data are provided by the organizer.In this paper,the task is regarded as a few-shot learning problem for NER,and a method based on BERT and two-level model fusion is proposed.Firstly,the proposed method is based on several basic models fine tuned by BERT on the training data.Then,a two-level fusion strategy applied to the prediction results of multiple basic models is proposed to alleviate the over-fitting problem.Finally,the labeling errors are eliminated by post-processing.This method achieves F1 score of 0.7203 on the test set of the evaluation task. 展开更多
