Aim To investigate the effect of DAPT (γ-secretase inhibitor) on the growth of human tongue carcinoma cells and to determine the molecular mechanism to enable the potential application of DAPT to the treatment of tongue carcinoma. Methodology Human tongue carcinoma Tca8113 cells were cultured with DAPT. Cell growth was determined using Indigotic Reduction method. The cell cycle and apoptosis were analyzed by flow cytometry. Real-time PCR and Immuno-Fluorescence (IF) were employed to determine the intracellular expression levels. Results DAPT inhibited the growth of human tongue carcinoma Tca8113 cells by inducing G0-G1 cell cycle arrest and apoptosis, The mRNA levels of Hairy/Enhancer of Split-1 (Hes-1), a target of Notch activation, were reduced by DAPT in a dose-dependent manner. Coincident with this observation, DAPT induced a dose-dependent promotion of constitutive Caspase-3 in Tca8113 cells. Conclusion DAPT may have a therapeutic value for human tongue carcinoma. Moreover, the effects of DAPT in tumor inhibition may arise partly via the modulation of Notch- 1 and Caspase-3. 展开更多

Mg alloys possess biodegradability,suitable mechanical properties,and biocompatibility,which make them possible to be used as biodegradable implants.However,the uncontrollable degradation of Mg alloys limits their general applications.In addition to the factors from the metallic materials themselves,like alloy compositions,heat treatment process and microstructure,some external factors,relating to the test/service environment,also affect the degradation rate of Mg alloys,such as inorganic salts,bioorganic small molecules,bioorganic macromolecules.The influence of bioorganic molecules on Mg corrosion and its protection has attracted more and more attentions.In this work,the cutting-edge advances in the influence of bioorganic molecules(i.e.,protein,glucose,amino acids,vitamins and polypeptide)and their coupling effect on Mg degradation and the formation of protection coatings were reviewed.The research orientations of biomedical Mg alloys in exploring degradation mechanisms in vitro were proposed,and the impact of bioorganic molecules on the protective approaches were also explored. 展开更多

Effects of Zn content (0, 0.5%, 1.5% and 4.5%) on the hot tearing characteristics of Mg?2%Y alloy were studied in aconstrained rod casting (CRC) apparatus attached with a load cell and data acquisition system. The experimental results indicate thatthe hot tearing susceptibility (HTS) is affected by the content of Zn. The Zn-free base alloy shows the lowest HTS. The HTS ofMg?xZn?2Y alloys increases with increasing Zn content, reaches the maximum at 1.5% Zn, and then decreases with further Znaddition. The high HTS observed in the alloy with 1.5% Zn is attributed to its high force release rate and large force drop duringsolidification. The hot cracks of casting are initiated and propagate along the dendritic or grain boundaries. The predictions of HTS ofMg?xZn?2Y alloys using ProCAST software are in good agreement with the results obtained by experimental measurements. 展开更多
