Study on the Protective Mechanism of Acupuncture Regulating ICAM-5 mRNA Expression in Rats with Ischemic Stroke

<b>Objective:</b> The objective is to investigate whether acupuncture can protect brain by regulating the expression of ICAM-5 mRNA in brain tissue. <b>Methods:</b> Male SD rats were used to co... <b>Objective:</b> The objective is to investigate whether acupuncture can protect brain by regulating the expression of ICAM-5 mRNA in brain tissue. <b>Methods:</b> Male SD rats were used to construct a middle cerebral artery occlusion reperfusion injury (Middle cerebral artery occlusion, MCAO) model, and the rats were randomly divided into 4 groups, namely the normal group (group A) and the sham operation group (group B), MCAO group (group C), MCAO + acupuncture group (group D);real-time fluorescent quantitative PCR (RT-qPCR) was used to detect the expression of ICAM-5 mRNA in the brain tissue of the ischemic area, and HE staining and TUNEL were used to detect the level of neuronal apoptosis to evaluate the therapeutic effect. <b>Results:</b> Acupuncture can improve neuronal morphology and reduce neuronal apoptosis in rats with ischemic stroke. RT-qPCR: compared with group A and group B, the expression level of ICAM-5 mRNA in the brain tissues of group C and group D decreased, while compared with group C, The expression of ICAM-5 mRNA in the brain tissue of group D was significantly up-regulated (P < 0.05). There was no statistical difference between group A and group B (P > 0.05). <b>Conclusion:</b> The effectiveness of acupuncture in the treatment of ischemic stroke may achieve neuroprotection by up-regulating the expression level of ICAM-5 mRNA. 展开更多

Objective: To observe the effect of acupuncture on the expression of ERM/PI3K/Akt signaling pathway in the brain tissue around the infarct focus of ischemic stroke model rats. Methods: According to the block randomization method, 80 male SD rats were divided into normal group (n = 10), sham operation group (n = 24), model group (n = 23) and acupuncture group (n = 23). The normal group was bred routinely without any intervention. In the sham operation group, only the skin was cut to find and the nerves and blood vessels were stripped and sutured. The model group and the acupuncture group were sutured to prepare the MCAO model. After the model was successfully established, acupuncture of the acupuncture group rats was given once a day, and the needles were kept for 30 minutes each time. The rats in the sham operation group and the model group only imitated capture and return, and were treated with acupuncture for 2 consecutive courses, each course was 5 days, and the two courses were rested for 2 days. After that, Western blot was used to detect ERM, changes in the concentration of PI3K and Akt proteins. Results: Compared with the normal group, the expression levels of ERM, PI3K, and Akt proteins in the sham operation group were reduced. Compared with the sham operation group, the expression levels of ERM, PI3K, and Akt proteins in the model group were significantly down-regulated. Compared with the model group, the protein expression levels of ERM, PI3K and Akt were significantly up-regulated in the acupuncture group, and the difference was statistically significant (P < 0.05). Conclusion: Acupuncture can significantly up-regulate the expression of ERM, PI3K and Akt proteins, and its effect mechanism may be related to the transduction of ERM/PI3K/Akt signaling pathway in cells. 展开更多

Cd-bearing solid wastes are considered to be a serious threat to the environment,and effective strategies for their treatment are urgently needed.Ceramic sintering has been considered as a promising method for efficiently incorporating heavy metal-containing solid wastes into various ceramic products.Mineral-rich dredged river sediment,especially Al and Si-containing oxides,can be treated as alternative ceramic precursors rather than being disposed of as solid wastes.To examine the feasibility of using waste sediment for Cd stabilization and the phase transition mechanisms,this study conducted a sintering scheme for the mixtures of CdO and dredged river sediment with different(Al+Si):Cd mole ratios.Detailed investigations have been performed on phases transformation,Cd incorporation mechanisms,elemental distribution,and leaching behaviors of the sintered products.Results showed that Cd incorporation and transformation in the sintered products were influenced by the mole ratio of(Al+Si):Cd.Among the high-Cd series((Al+Si):Cd=6:1),CdSiO_(3),Cd_(2)SiO_(4),CdAl_(2)(SiO_(4))_(2) and Cd_(2)Al_(2)Si_(2)O_(9) were predominant Cd-containing product phases,while Cd2Al2Si2O9 was replaced by CdAl_(4)O_(7) when the mole ratio of(Al+Si):Cd was 12:1(low-Cd series).Cd was efficiently stabilized in both reaction series after being sintered at≥900℃,with<5%leached ratio even after a prolonged leaching time,indicating excellent long-term Cd stabilization.This study demonstrated that both Cd-containing phases and the amorphous Al-/Si-containing matrices all played critical roles in Cd stabilization.A promising strategy can be proposed to simultaneously reuse the solid waste as ceramic precursors and stabilize heavy metals in the ceramic products. 展开更多

Multifunctional switchable materials are attracting tremendous interest because of their great application potential in signal processing,information encryption,and smart devices.Here,we reported an organic-inorganic hybrid thermochromic ferroelastic crystal,[TMIm][CuCl_(4)](TMIm=1,1,3,3-tetramethylimidazolidinium),which undergoes two reversible phase transitions at 333 K and 419 K,respectively.Intriguingly,these three phases experience a remarkable ferroelastic-paraelastic-ferroelastic(2/m-mmm-2/m)transition,which remains relatively unexplored in ferroelastics.Moreover,the ferroelastic domains can be simultaneously switched under temperature and stress stimuli.Meanwhile,[TMIm][CuCl_(4)]exhibits thermochromic phenomenon,endowing it with extra spectral encryption possibilities during information processing.Combined with dielectric switching behavior,[TMIm][Cu Cl_(4)]are promising for practical applications in memory devices,next-generation sensors,and encryption technology. 展开更多

Chromium slag usually contains various heavy metals,making its safe treatment difficult.Glassceramic sintering has been applied to resolve this issue and emerged as an effective method for metal immobilization by incorporating heavy metals into stable crystal structures.Currently,there is limited knowledge about the reaction pathways adopted by multiple heavy metals and the co-stabilization functions of the crystal structure.To study the Cu/Cr co-stabilization mechanisms during thermal treatment,a simulated system was prepared using a mixture with a molar ratio of Al_(2)O_(3):Fe_(2)O_(3):Cr_(2)O_(3):CuO=1:1:1:3.The samples were sintered at temperatures 600–1300℃ followed by intensive analysis of phase constitutions and microstructure development.A spinel phase(CuFe_(x)Al_(y)Cr_(2–x–y)O_(4))started to generate at 700℃ and the incorporation of Cu/Cr into the spinel largely complete at 900℃,although the spinel peak intensity continued increasing slightly at temperatures above 900℃.Fe_(2)O_(3)/Cr_(2)O_(3) was more easily incorporated into the spinel at lower temperatures,while more Al_(2)O_(3) was gradually incorporated into the spinel at higher temperatures.Additionally,sintered sample microstructures became more condensed and smoother with increased sintering temperature.Cu/Cr leachability substantially decreased after Cu/Cr incorporation into the spinel phase at elevated temperatures.At 600℃,the leached ratios for Cu and Cr were 6.28%and 0.65%,respectively.When sintering temperature was increased to 1300℃,the leached ratios for all metal components in the system were below 0.2%.This study proposes a sustainable method for managing Cu/Cr co-exist slag at reasonable temperatures. 展开更多

Fisher判别法可综合考虑事物的多重属性特征,进而分辨事物类型。若能将其应用于对干旱的识别,或将成为一种准确而有效的干旱识别方法。本研究以高寒草甸生态系统为研究对象,基于9年碳通量和小气候观测数据,以土壤含水量(soil water content,SWC)和饱和水汽压差(vapor pressure deficit,VPD)作为判别因子,利用Fisher判别法识别干旱。Fisher判别法可用于对高寒草甸生态系统干旱的识别。因其综合考虑了土壤水分匮缺和大气水分匮缺,故可以更合理准确地判断干旱的开始和结束时间。基于干旱早样本和非干旱样本的特征,构建判别方程为:y=24.46SWC-4.60VPD。当y>1时,样本点位于临界线上方,若持续10天以上,则判定为发生干早。两次干早过程间隔2天以内,可认为是一次干旱过程。随着研究年限的增加和观测数据的积累,该临界线方程尚有优化空间,其对干旱识别的准确度可进一步提高。 展开更多

Drought plays a prominent role in affecting ecosystem stability and ecosystem productivity.Based on eddy covariance and climatic observations during 2012-2020,the Fisher discriminant analysis method was employed to accurately detect drought occurrences.Furthermore,the ecosystem water sensitivity and its resistance to drought were quantified to evaluate the ecosystem stability.The results showed that the alpine meadow suffered drought most frequently at the beginning of the growing seasons.However,drought during the peak growing seasons reduced the gross primary productivity(GPP)the most,by 30.5±15.2%.In the middle of the peak growing seasons,the ecosystem water sensitivity was weak,and thus,the resistance to drought was strong,which resulted in high ecosystem stability.At the beginning and end of the peak growing seasons,the ecosystem stability was relatively weak.Ecosystem stability was positively related to the corresponding multiyear average soil water content(SWC_(ave)).However,drought occurring during high SWC_(ave)periods led to larger reductions in GPP,which indicated that the inhibitory effects of drought on ecosystems were more dependent on the occurrence time of droughts than on ecosystem stability. 展开更多

Consequential fate of bisphenol-attached PVC microplastics in water and simulated intestinal fluids

The ever-increasing prevalence of microplastics and different bisphenols made the presence of bisphenol-attached microplastics a critical concern.In this study,experiments were performed to examine desorption behaviors and cytotoxicity performance of contaminated microplastics in aquatic surroundings and intestinal environment after ingestion by organisms(cold-/warm-blooded).The kinetic study shows that the rate of desorption for bisphenols can be enhanced threefold under simulated warm intestinal conditions.The Freundlich isotherms indicate multiple-layer desorption of the bisphenols on the heterogeneous surfaces of polyvinyl chloride(PVC)microplastics.Hysteresis was detected in the adsorption/desorption of bisphenols in a water environment,but no adsorption/desorption hysteresis was observed in the simulated intestinal conditions of warm-blooded organisms.Due to enhanced bioaccessibility,the desorption results imply that the environmental risk of contaminated PVC microplastics may be significantly increased after ingestion at a high bisphenols dosage.Although with different IC_(50),the five bisphenols released under the intestinal conditions of warm-blooded organisms can cause higher proliferation reduction in fish and human cell lines than the bisphenols released in water.This study helps elucidate the consequential fate and potential cytotoxicity of contaminated microplastics and the possible implications of the microplastics as a critical vector for bisphenols to increase the potential health risks. 展开更多
