Transcriptomic analysis reveals the promotion of lymph node metastasis by Helicobacter pylori infection via upregulating chemokine (C-X-C motif) receptor 2 expression in gastric carcinoma

Gastric carcinoma (GC) progression is mainly caused by local aggression and lymph node metastasis. However, some patients with early T-stage disease have lymph node metastasis, whereas some patients with late T-stage disease do not have lymph node metastasis, which indicates that invasion and metastasis are not always sequential in some GC patients. In the present study, the data of 101 GC cases were acquired from TCGA and divided into T-late-N-negative and T-early-N-positive groups according to pathological stages. A total of 338 genes were identified as differential genes between the T-late-N-negative and T-early-N-positive groups. GSEA showed that epithelial cell signaling in the Helicobacter pylori (HP ) infection pathway was enriched in the T-early-N-positive group. MB staining indicated that the HP infection rate was 63% (39/62) in N-positive patients compared to 42% (16/38) in N-negative patients. To investigate the potential mechanism, we focused on the gene chemokine (C-X-C motif) receptor 2 (CXCR2), which was not only clustered in the gene set of epithelial cells signaling in the HP infection pathway but also significantly upregulated in T-early-N-positive GC by the analysis of the different genes based on the TCGA dataset. A meta-analysis showed that CXCR2 expression was positively correlated with N-stage but not with T-stage in GC. This study indicated that invasion and metastasis could be independent processes driven by different molecular mechanisms in some GC patients. HP infection was a potential factor that promoted lymph node metastasis by upregulating CXCR2 expression. 展开更多

In recent years,as living standards have continued to improve,the number of diabetes patients in China,along with the incidence of complications associated with the disease,has been increasing.Among these complications,diabetic foot disease is one of the main causes of disability and death in diabetic patients.Due to the differences in economy,culture,religion and level of medical care available across different regions,preventive and treatment methods and curative results for diabetic foot vary greatly.In multidisciplinary models built around diabetic foot,the timely assessment and diagnosis of wounds and appropriate methods of prevention and treatment with internal and external surgery are key to clinical practice for this pathology.In 2019,under the leadership of the Jiangsu Medical Association and Chinese Diabetes Society,the writing group for the Guidelines on multidisciplinary approaches for the prevention and management of diabetic foot disease(2020 edition)was established with the participation of scholars from the specialist areas of endocrinology,burn injury,vascular surgery,orthopedics,foot and ankle surgery and cardiology.Drawing lessons from diabetic foot guidelines from other countries,this guide analyses clinical practices for diabetic foot,queries the theoretical basis and grades and gives recommendations based on the characteristics of the pathology in China.This paper begins with assessments and diagnoses of diabetic foot,then describes treatments for diabetic foot in detail,and ends with protections for high-risk feet and the prevention of ulcers.This manuscript covers the disciplines of internal medicine,surgical,nursing and rehabilitation and describes a total of 50 recommendations that we hope will provide procedures and protocols for clinicians dealing with diabetic foot. 展开更多

Temporal–spatial distribution and diastereoisomer pattern of hexabromocyclododecane in the vicinity of a chemical plant 被引量:4

Hexabromocyclododecane(HBCD) is an effective brominated flame-retardant additive, which is mainly produced in the coastal area of China. This study collected soil samples from a HBCD production plant and its surrounding area in Weifang, Shandong Province, China, and analyzed the temporal–spatial distribution of HBCD and its diastereoisomers in soil. The analysis results showed that the concentration of HBCD in soil near the plant was much higher than normal values, with an annual average concentration reaching 5405 ng/g. Soils 1,2 and 4 km away from the plant were also analyzed, showing that the concentration of HBCD in soil decreased accordingly with the distance from the pollution sources. In order to investigate the effect of the season on HBCD content, the soil samples were collected in all four seasons of the year 2017–2018. According to variations in the wind direction, the concentration of HBCD in soil was also changed. The distribution trend showed that the concentration of HBCD in soil in the downwind direction of the prevailing wind was higher than that in the upwind direction. In addition, this work analyzed the distribution of HBCD in vertical soil sections. It was found that the concentration of HBCD decreased with depth in the soil vertical profile. Finally, the various diastereoisomer patterns in the soil compartments were examined, finding that α-HBCD and γ-HBCD were the predominant diastereoisomers in the soil of the study area. 展开更多
