T. popeiorum is the only Popeia species known from China. During the past two years, five adult Popeia specimens(4 males, 1 female) were collected from Yingjiang County, Southern Yunnan, China. Molecular, morphological and ecological data show distinct differences from known species, herein we describe these specimens as a new species Trimeresurus yingjiangensis sp. nov Chen, Ding, Shi and Zhang, 2018. Morphologically, the new species distinct from other Popeia species by a combination of following characters:(1) dorsal body olive drab,without cross bands on the scales;(2) a conspicuous bicolor ventrolateral stripe present on each side of males, first row of dorsal scales firebrick with a white ellipse dot on posterior upper part in male, these strips absent in females;(3) eyes firebrick in both gender;(4) suboculars separated from 3rd upper labial by one scale on each side;(5) ventrals 164–168(n = 5);(6) MSR 21.展开更多

However, the spatial distribution o...Objective: Hypoxia is a significant feature of solid tumors, including pancreatic ductal adenocarcinoma(PDAC). It is associated with tumor invasion, metastasis, and drug resistance. However, the spatial distribution of hypoxia-related heterogeneity in PDAC remains unclear.Methods: Spatial transcriptomics(STs), a new technique, was used to investigate the ST features of engrafted human PDAC in the ischemic hind limbs of nude mice. Transcriptomes from ST spots in the hypoxic tumor and the control were clustered using differentially-expressed genes. These data were compared to determine the spatial organization of hypoxia-induced heterogeneity in PDAC. Clinical relevance was validated using the Tumor Cancer Genome Atlas and KM-plotter databases. The CMAP website was used to identify molecules that may serve as therapeutic targets for PDAC.Results: ST showed that the tumor cell subgroups decreased to 7 subgroups in the hypoxia group, compared to 9 subgroups in the control group. Different subgroups showed positional characteristics and different gene signatures. Subgroup 6 located at the invasive front showed a higher proliferative ability under hypoxia. Subgroup 6 had active functions including cell proliferation, invasion, and response to stress. Expressions of hypoxia-related genes, LDHA, TPI1, and ENO1, induced changes. CMAP analysis indicated that ADZ-6482, a PI3 K inhibitor, was targeted by the invasive subgroup in hypoxic tumors.Conclusions: This study is the first to describe hypoxic microenvironment-induced spatial transcriptome changes in PDAC, and to identify potential treatment targets for PDAC. These data will provide the basis for further investigations of the prognoses and treatments of hypoxic tumors.展开更多

Previously, we found that hypoxia promotes ENO1 expression and PDAC invasion. However, the underlying molecular mechanism was remains unclear.Methods: The relationship between ENO1 expression and clinicopathological characteristics was analyzed in 84 patients with PADC. The effects of CoCl2-induced hypoxia and ENO1 downregulation on the apoptosis, invasion, and proliferation of PDAC cells were evaluated in vitro and in vivo. Hypoxia-and ENO1-induced gene expression was analyzed by transcriptomic sequencing.Results: The prognosis of PDAC with high ENO1 expression was poor(P < 0.05). High ENO1 expression was closely associated with histological differentiation and tumor invasion in 84 PDAC cases(P < 0.05). Hypoxia increased ENO1 expression in PDAC and promoted its migration and invasion. Apoptotic cells and the apoptosis marker caspase-3 in the CoCl_(2)-treated ENO1-sh group were significantly elevated(P < 0.05). Transcriptomic sequencing indicated that CoCl_(2)-induced PDAC cells initiated MAPK signaling. Under hypoxic conditions, PDAC cells upregulated ENO1 expression, thereby accelerating ERK phosphorylation and inhibiting apoptosis(P < 0.05). Consistent results were also observed in a PDAC-bearing mouse hindlimb ischemia model.Conclusions: Hypoxia-induced ENO1 expression promotes ERK phosphorylation and inhibits apoptosis, thus leading to PDAC survival and invasion. We r...Extragonadal primary yolk sac tumor of the intestinal tract origin is exceedingly rare. Through a multiple disciplinary team, the diagnosis and treatment of primary intestinal yolk sac tumor were further defined. We report 2 such cases with detailed histologic and immunohistochemical analysis. The two patients were a 7-year-old girl and a 29-year-old woman. Both of them preoperatively had an elevated serum alpha fetoprotein(AFP) level(≥ 1,210 ng/mL). The tumors are located in the intestine and imaging examination indicated the rectum as the primary site. Grossly the mass was grey-white and crisp texture. Microscopic examination featured reticular, microcystic, macrocystic, papillary, solid, and some glandular patterns. Immunohistochemically,tumor cells of both cases were positive for SALL4, AFP, pan-cytokeratin(AE1/AE3), and glypican-3. Simultaneously, a stain for EMA, OCT4, CD30, HCG, vimentin and CK20 were negative in all 2 neoplasms. The features of morphology,immunohistochemistry, laboratory examinations and imaging studies consist of the diagnosis of primary yolk sac tumor of the intestine.展开更多
Objective:The immunoscore,which is used to quantify immune infiltrates,has greater relative prognostic value than tumor,node,and metastasis(TNM)stage and might serve as a new system for classification of colorectal ca...Objective:The immunoscore,which is used to quantify immune infiltrates,has greater relative prognostic value than tumor,node,and metastasis(TNM)stage and might serve as a new system for classification of colorectal cancer.However,a comparable immunoscore for predicting lung adenocarcinoma(LUAD)prognosis is currently lacking.Methods:We analyzed the expression of 18 immune features by immunohistochemistry in 171 specimens.The relationship of immune marker expression and clinicopathologic factors to the overall survival(OS)was analyzed with the Kaplan-Meier method.A nomogram was developed by using the optimal features selected by least absolute shrinkage and selection operator(LASSO)regression in the training cohort(n=111)and evaluated in the validation cohort(n=60).Results:The indicators integrated in the nomogram were TNM stage,neuron-specific enolase,carcino-embryonic antigen,CD8 center of tumor(CT),CD8 invasive margin(IM),Fox P3 CT,and CD45 ROCT.The calibration curve showed prominent agreement between the observed 2-and 5-year OS and that predicted by the nomogram.To simplify the nomogram,we developed a new immune-serum scoring system(I-SSS)based on the points awarded for each factor in the nomogram.Our I-SSS was able to stratify same-stage patients into different risk subgroups.The combination of I-SSS and TNM stage had better prognostic value than the TNM stage alone.Conclusions:Our new I-SSS can accurately and individually predict LUAD prognosis and may be used to supplement prognostication based on the TNM stage.展开更多
Lithium-sulfur batteries have attracted attention because of their high energy density.However,the "shuttle effect" caused by the dissolving of polysulfide in the electrolyte has greatly hindered the widespr...Lithium-sulfur batteries have attracted attention because of their high energy density.However,the "shuttle effect" caused by the dissolving of polysulfide in the electrolyte has greatly hindered the widespread commercial use of lithiumsulfur batteries.In this paper,a novel two-dimensional TiS2/graphene heterostructure is theoretically designed as the anchoring material for lithium-sulfur batteries to suppress the shuttle effect.This heterostructure formed by the stacking of graphene and TiS2 monolayer is the van der Waals type,which retains the intrinsic metallic electronic structure of graphene and TiS2 monolayer.Graphene improves the electronic conductivity of the sulfur cathode,and the transferred electrons from graphene enhance the polarity of the TiS2 monolayer.Simulations of the polysulfide adsorption show that the TiS2/graphene hetero structure can maintain good metallic properties and the appropriate adsorption energies of 0.98-3.72 eV,which can effectively anchor polysulfides.Charge transfer analysis suggests that further enhancement of polarity is beneficial to reduce the high proportion of van der Waals(vdW) force in the adsorption energy,thereby further enhancing the anchoring ability.Low Li2 S decomposition barrier and Li-ion migration barrier imply that the heterostructure has the ability to catalyze fast electrochemical kinetic processes.Therefore,TiS2/graphene heterostructure could be an important candidate for ideal anchoring materials of lithium-sulfur batteries.展开更多
As the demand for the development of cloud computing grows, more and more organizations have outsourced their data and query services to the cloud for cost-saving and flexibility. Suppose an organization that has a gr...As the demand for the development of cloud computing grows, more and more organizations have outsourced their data and query services to the cloud for cost-saving and flexibility. Suppose an organization that has a great number of users querying the cloud-deployed multiple proxy servers to achieve cost efficiency and load balancing. Given n queries, each of which is expressed as several keywords, and k proxy servers, the problem to be solved is how to classify n queries into k groups, in order to minimize the difference between each group and the number of distinct keywords in all groups. Since this problem is NP-hard, it is solved in mathematic and heuristic ways. Mathematic grouping uses a local optimization method, and heuristic grouping is based on k-means. Specifically, two extensions are provided: the first one focuses on robustness, i.e., each user obtains search results even if some proxy servers fail; the second one focuses on benefit, i.e., each user can retrieve as many files as possible that may be of interest without increasing the sum. Extensive evaluations have been conducted on both a synthetic dataset and real query traces to verify the effectiveness of our strategies.展开更多
