Enhancing photosynthetic efficiency is a major goal for improving crop yields under agricultural field conditions and is associated with chloroplast biosynthesis and development.In this study,we demonstrate that Golde...Enhancing photosynthetic efficiency is a major goal for improving crop yields under agricultural field conditions and is associated with chloroplast biosynthesis and development.In this study,we demonstrate that Golden2-like 1a(BnGLK1a)plays an important role in regulating chloroplast development and photosynthetic efficiency.Overexpressing BnGLK1a resulted in significant increases in chlorophyll content,the number of thylakoid membrane layers and photosynthetic efficiency in Brassica napus,while knocking down BnGLK1a transcript levels through RNA interference(RNAi)had the opposite effects.A yeast two-hybrid screen revealed that BnGLK1a interacts with the abscisic acid receptor PYRABACTIN RESISTANCE 1-LIKE 1-2(BnPYL1-2)and CONSTITUTIVE PHOTOMORPHOGENIC 9 SIGNALOSOME 5A subunit(BnCSN5A),which play essential roles in regulating chloroplast development and photosynthesis.Consistent with this,BnGLK1a-RNAi lines of B.napus display hypersensitivity to the abscisic acid(ABA)response.Importantly,overexpression of BnGLK1a resulted in a 10%increase in thousand-seed weight,whereas seeds from BnGLK1a-RNAi lines were 16%lighter than wild type.We propose that BnGLK1a could be a potential target in breeding for improving rapeseed productivity.Our results not only provide insights into the mechanisms of BnGLK1a function,but also offer a potential approach for improving the productivity of Brassica species.展开更多

Some of the earliest bio-sedimentary records of life on Earth are represented by microbial carbonates,which are also critical geochemical archives of ancient seawater chemistry and the environmental circumstances in w...Some of the earliest bio-sedimentary records of life on Earth are represented by microbial carbonates,which are also critical geochemical archives of ancient seawater chemistry and the environmental circumstances in which they precipitated.Reconstructing paleo-microbial environments on Earth and potentially other planets requires precise determination of the depositional ages of these materials.The(abiogenic)carbonate geochemistry communities can now use developments in in-situ laser ablation U-Pb dating using inductively coupled plasma mass spectrometry(LA-ICP-MS).Due to the effects of impurity mixing and diagenesis,microbial carbonates have received little geochronological study despite their broad relevance for understanding ancient seawater’s environmental conditions and geochemical compositions.This study demonstrates using time-of-flight mass spectrometry(TOF-MS)to perform quick,quantitative elemental mapping before U-Pb spot dating to improve experiment success rates and data reliability and offers four practical application examples.展开更多

Apical actin filaments were thought to be involved in the regu- lati...Pollen tube growth is an essential step during flowering plant reproduction, whose growth depends on a population of dynamic apical actin filaments. Apical actin filaments were thought to be involved in the regu- lation of vesicle fusion and targeting in the pollen tube. However, the molecular mechanisms that regulate the construction of apical actin structures in the pollen tube remain largely unclear. Here, we identify profilin as an important player in the regulation of actin polymerization at the apical membrane in the pollen tube. Downregulation of profilin decreased the amount of filamentous actin and induced disorganization of apical actin filaments, and reduced tip-directed vesicle transport and accumulation in the pollen tube. Direct visualization of actin dynamics revealed that the elongation of actin filaments originating at the apical membrane decreased in profilin mutant pollen tubes. Mutant profilin that is defective in binding poly-L-proline only partially rescues the actin polymerization defect in profilin mutant pollen tubes, although it fully rescues the actin turnover phenotype. We propose that profilin controls the construction of actin structures at the pollen tube tip, presumably by favoring formin-mediated actin polymerization at the apical membrane.展开更多
A series of biodegradable amphoteric chitosan-based flocculants (3-chloro-2-hydroxypropyl trimethyl ammonium chloride (CTA) modified carboxymethyl chitosan, denoted as CMC-CTA) with different substitution degrees ...A series of biodegradable amphoteric chitosan-based flocculants (3-chloro-2-hydroxypropyl trimethyl ammonium chloride (CTA) modified carboxymethyl chitosan, denoted as CMC-CTA) with different substitution degrees of CTA were prepared successfully. The content of carboxymethyl groups in each CMC-CTA sample was kept almost constant. The solubility of the various flocculants showed that, higher cationic content of flocculants caused a better solubility. The flocculation experiments using kaolin suspension as synthetic water at the laboratory scale indicated that the substitution degree of CTA was one of the key factors for the flocculation properties. With the increase of cationic content, the flocculants were demonstrated better flocculation performance and lower dosage requirement. Flocculation kinetics model of particles collisions combining zeta potential and turbidity measurements was employed to investigate the effects of the cationic content of the flocculants on the flocculation properties from the viewpoint of flocculation mechanism in detail. Furthermore, flocculation performance using raw water from Zhenjiang part of Yangtze River at the pilot scale showed the similar effects to those at the laboratory scale.展开更多
