我国辣椒病毒病发生情况及发展趋势--基于2018年和2019年辣椒主产区的调查 被引量:20

为明确我国辣椒病毒病的种类和发展趋势,2018年和2019年在我国16个省(市、自治区)辣椒主产区采集具有病毒病疑似症状的样品105份,利用双抗体夹心酶联免疫吸附反应(DAS-ELISA)对12种辣椒主要病毒进行检测。结果表明,有70份样品为病毒病... 为明确我国辣椒病毒病的种类和发展趋势,2018年和2019年在我国16个省(市、自治区)辣椒主产区采集具有病毒病疑似症状的样品105份,利用双抗体夹心酶联免疫吸附反应(DAS-ELISA)对12种辣椒主要病毒进行检测。结果表明,有70份样品为病毒病感染样品,病毒检出率为66.67%。检出病毒种类共10种,其中辣椒轻斑驳病毒(Pepper mild mottle virus,PMMoV)检出率最高,为24.76%,其次为黄瓜花叶病毒(Cucumber mosaic virus,CMV)、蚕豆萎蔫病毒(Broad bean wilt virus,BBWV)、烟草蚀纹病毒(Tobacco etch virus,TEV)和番茄斑萎病毒(Tomato spotted wilt virus,TSWV),检出率分别为19.05%、12.38%、12.38%和10.48%。我国辣椒主产区多种病毒复合侵染发生较为普遍,病毒复合侵染率为40%,其中以2种病毒复合侵染比例最高,为28.57%。通过对两年辣椒病毒病检测结果进行比较,12种病毒中有7种检出率有上升趋势。总体而言,PMMoV和CMV为近两年危害我国辣椒生产较为严重的病毒病;此外,PMMoV、BBWV-1,2、TEV、TSWV具有流行风险,需要严加防范。 展开更多

为研究机动车道路行驶过程中轮胎磨损排放的颗粒物理化特性,利用轮胎轮廓仿真磨耗仪,对国内主流17种轮胎胎面进行仿真磨耗实验,获得颗粒物样品,提取并检测其中18种元素和20种多环芳烃(PAHs)的含量.结果显示,元素和PAHs含量因轮胎品牌和... 为研究机动车道路行驶过程中轮胎磨损排放的颗粒物理化特性,利用轮胎轮廓仿真磨耗仪,对国内主流17种轮胎胎面进行仿真磨耗实验,获得颗粒物样品,提取并检测其中18种元素和20种多环芳烃(PAHs)的含量.结果显示,元素和PAHs含量因轮胎品牌和速度等级的不同而差异显著.18种元素平均含量为(99.04±68.43)mg/g,占样品总重的9.90%,其中Si(88.97±67.85)mg/g、Zn(6.77±1.64)mg/g和Na(1.05±0.75)mg/g的平均含量均超过1mg/g,Cd的含量最低,为(0.43±0.31)μg/g.20种PAHs含量之和(∑20PAHs)在12.13~433.64μg/g,平均为(94.13±110.18)μg/g,PY的平均含量最高(30.98±31.27)μg/g,其次是CHR、BaP、FA、PHE和BghiP,平均含量最低的是AC(0.58±0.2)μg/g;从环数看,以4环PAHs为主(占∑20PAHs的45.03%~67.93%),其次为3环(平均含量为15.45%)和5环(平均含量为12.62%).总体来说,国外品牌轮胎样品中元素和PAHs含量略高于国内品牌,而主要PAHs环数略低于国内品牌. 展开更多

微生物诱导碳酸钙沉淀技术(microbially induced calcite precipitation,MICP),是近年来在国内兴起的一项多学科交叉的技术,工程技术人员将它引入到土木工程中,使得土体各项性能得以改良;黄河中下游的粉性土水稳定性差,毛细水作用大,干... 微生物诱导碳酸钙沉淀技术(microbially induced calcite precipitation,MICP),是近年来在国内兴起的一项多学科交叉的技术,工程技术人员将它引入到土木工程中,使得土体各项性能得以改良;黄河中下游的粉性土水稳定性差,毛细水作用大,干燥时强度高但潮湿时强度显著下降,利用MICP技术加固粉性土值得研究。以粉性土为研究对象,首先研究了不同糯米浆浓度下土样的力学性能,以最优糯米浆浓度作为改良材料对MICP技术进行改良;其次控制胶菌质量比为2∶1,研究了不同胶结液浓度下MICP技术的产碳酸钙量以及Ca2+转化率,得出MICP技术的最佳配比,结合最优糯米浆浓度形成改良MICP技术;最后研究了素土、MICP加固土、改良MICP加固土土样养护7 d后的力学性能。研究表明:土样最优糯米浆浓度为3%,此时土体无侧限抗压强度与内摩擦角达到最大;MICP技术最佳配比为2∶1胶菌质量比、0.5 mol/L胶结液浓度,此时产碳酸钙量最多且Ca2+转化率在80%以上,添加3%糯米浆浓度的改良MICP技术Ca^(2+)转化率在90%以上,比MICP技术高10%;通过土的直接快剪试验和无侧限抗压强度试验,MICP加固土的黏聚力提高了约30%,内摩擦角变化不大,改良MICP加固土的黏聚力提高了约50%,内摩擦角提高了约17%。 展开更多

【目的】研究辣椒炭疽病抗性基因的遗传规律。在5号连锁群中,定位辣椒炭疽病抗性主效基因AnRGO5,并开发分子标记。【方法】以高抗炭疽病品种PBC932(Capsicum chinense)为父本,以高感炭疽病的中早熟材料77013(Capsicum annuum)为母本杂... 【目的】研究辣椒炭疽病抗性基因的遗传规律。在5号连锁群中,定位辣椒炭疽病抗性主效基因AnRGO5,并开发分子标记。【方法】以高抗炭疽病品种PBC932(Capsicum chinense)为父本,以高感炭疽病的中早熟材料77013(Capsicum annuum)为母本杂交产生BC4S1、BC4S2群体,用于辣椒绿熟期抗炭疽病基因定位。选用尖孢炭疽菌为试验用菌,采用显微注射法对绿熟果接病,根据病斑直径进行表型分析。根据抗、感亲本重测序结果,开发与绿熟期抗炭疽病连锁的Kaspar标记。【结果】辣椒果实表面病斑直径呈连续分布,符合数量性状的遗传特点。通过连锁分析,将炭疽病抗性基因AnRGO5定位在5号连锁群的标记P5L-866与标记P5L-259之间,遗传距离2.9cM。开发的标记P5L-117与基因紧密连锁,标记准确率93.5%。【结论】在辣椒的BC3S1群体中,将果实绿熟期抗性基因定位于5号染色体的标记区间内。辣椒炭疽病抗性遗传为显性遗传,是由两对主效基因控制的。 展开更多

Effect of Low Light on the Characteristics of Photosynthesis and Chlorophyll a Fluorescence During Leaf Development of Sweet Pepper 被引量:23

Low light stress is one of the main limiting factors which influence the production of sweet pepper under protected cultivation in China. In this experiment, two genotypes of sweet pepper, ShY (low light-tolerant genotype) and 20078 (low light-sensitive genotype), were used to study the effects of low light (photosynthetic photon flux density, PPFD was 75- 100 umol m-2 s-1, control 450-500 umol m-2 s-1) on photosynthesis during leaf development. The result indicated that under low light chlorophyll content, net photosynthetic rate (PN), photosynthetic apparent quantum efficiency (Фi) and carboxylation efficiency (CE) of sweet pepper leaves increased gradually and decreased after reaching the maximum levels. The time to reach the peak values for all the above parameters was delayed, whereas the light compensation point (LCP) decreased gradually along with leaf expansion. The decrease in maximum quantum yield of PS II (Fv/Fm) was not observed at any stages of the leaf development under low light condition, but the actual PS II efficiency under irradiance (ФPS II) was lower accompanied by an increased non-photochemical quenching (NPQ) in young and/or old leaves compared with mature leaves. The antenna thermal dissipation (D) was a main way of heat dissipation when young leaves received excessive light energy, while the decline in photosynthetic function in senescence leaf was mostly owing to the decrease in carbon assimilation capacity, followed by a significantly increased allocation of excessive energy (Ex). Compared with 20078, ShY could maintain higher PN, ФPS II and lower QA reduction state for a longer time during leaf development. Thus, in ShY photosynthetic efficiency and the activity of electron transport of PS II were not significantly affected due to low light stress. 展开更多

Fruit set and development are affected by many phytohormones, including gibberellin. Little is known regarding molecular mechanism underlying gibberellin mediated fruit set and development especially in Capsicum. Three gibberellin recep- tors, CaGIDlb.1, CaGIDlb.2 and CaGIDlc, and a DELLA protein, CaGAI, have been identified in Capsicum annuum L. During the fruit development, the expression level of CaGIDlc was low, and the expression fold change is mild. However, CaGIDlb. 1 and CaGIDlb.2 were relatively higher and more acute, which indicates that CaGIDlb. 1 and CaGID1b.2 may play an important role in fruit pericarp, placenta and seed. Ectopic expressions of CaGIDlb. 1, CaGIDlb.2 and CaGIDlc in Arabidopsis double mutant gidla gidlc increased plant height, among which CaGIDlb.2 had the most significant effect; CaGAI reduced plant height in double mutant rga-24/gai-t6, having a similar function to AtGID1 and AtGAI in stem elon- gation. Yeast two-hybrid (Y2H) and bimolecular fluorescence complementation (BiFC) assays indicated that CaGIDlb.1 and CaGID1 b.2 interact with CaGAI in a GA-dependent manner, while CaGIDlc interacts with CaGAI in a GA-independent manner. Our study reveals the key elements during gibberellin signaling in Capsicum and supports the critical importance of gibberellin for Capsicum fruit set and development. 展开更多

Pepper hybrid seeds production using male sterility could lower cost by reducing time and labour, and increase the genetic purity of the F1 seeds. To investigate the genetics of fertility restoration of the Peterson cytoplasmic sterility in pepper, a doubled haploid population of 115 pepper lines obtained from anther culture of the F1 hybrid between Yolo Wonder (sterility maintainer line) and Perennial (fertility restorer line) and the parental lines were test-crossed by 77013A (a strict cytoplasmic-genic male sterile line). The fertility of the test-crossed lines was assessed in greenhouse and open field with the following three criteria: pollen index (PI, visual estimation of pollen amount per flower), pollen number (PN, pollen counting under microscope), and seed number (SN, the number of seeds per fruit in open pollination). Correlations between the each couple of criteria within, as well as between the cultivation methods ranged from 0.55 to 0.84. Analysis of variance showed that the genotype (DH line) and environment were the significant sources of variation of the fertility. Narrow sense of heritance of fertility restoration ranged from 0.38 to 0.92, depending on the criteria and environment. The distribution of the progeny was continuous between the parental genotypes indicating the quantitative inheritance of fertility restoration. Inferred from segregation according to Snape et al.(1984), the number of segregating genes was estimated to be that three to four genetic factors were involved in pollen traits (PI and PN) and five to eight genetic factors in seed production (SN). The heredity analysis of the CMS will be helpful for understanding of the genetic mechanism of the fertility restoration and the exploitation of the CMS in hybrid seed production. 展开更多

Now the pepper in China contributes one-third of the world's peppers production. Genetic diversity of the pepper germplasm of China is expected interesting to know. To explore the structure of genetic diversity in Chinese pepper germplasm resources and possible relationship with cultivar types or geographic origin, we sampled and compared 372 Gen Bank pepper accessions(local cultivars and landraces) from 31 provinces, autonomous regions and municipalities of China and 31 additional accessions from other countries. These accessions were genotyped using 28 simple sequence repeat(SSR) markers spanning the entire pepper genome. We then investigated the genetic structure of the sampled collection using model-based analysis in STRUCTURE v2.3.4 and examined genetic relationships by the unweighted pair-group method of mathematical averages(UPGMA) in MEGA. In addition to geographic origin, we evaluated eight plant and fruit traits. In total, 363 alleles were amplified using the 28 SSR primers. Gene diversity, polymorphism information content and heterozygosity of the 28 SSR loci were estimated as 0.09–0.92, 0.08–0.92 and 0.01–0.34, respectively. The UPGMA cluster analysis clearly distinguished Capsicum annuum L. from other cultivated pepper species. Population structure analysis of the 368 C. annuum accessions uncovered three genetic groups which also corresponded to distinct cultivar types with respect to the plant and fruit descriptors. The genetic structure was also related to the geographic origin of the landraces. Overall results indicate that genetic diversity of Chinese pepper landraces were structured by migration of genotypes followed by human selection for cultivar types in agreement with consumption modes and adaptation to the highly diversified agro-climatic conditions. 展开更多
