The extraction process was a pressurized microwave-assisted extraction(PMAE). The seven ginsenosides were separated and determined by high-performance liquid chromatography(HPLC) with a ultraviolet(UV) detector, at 203 nm. The experiment results showed significant variations in the individual ginsenoside contents of the American ginseng in different parts and ages of the plant. The results demonstrated that the leaves, root hairs, and rhizomes of Panax quinquefolius L. contained higher ginsenoside contents, followed by the main roots and stems. The leaves contained dramatically higher levels of ginsenoside Rg1 Rb3, and Rd than the other four parts. Higher contents of Rb1 and Re were present in the main roots, root hairs, and rhizomes. The amount of ginsenoside content in the stems was the lowest. The total content of the seven ginsenosides in main roots, root hairs and rhizomes increased with the age of the plant. In contrast, the ginsenoside contents in the leaves and stems decreased with a year of growth.展开更多

Two kinds of solid microwave absorption media[carbonyl iron powder(CIP) and graphite powder(GP)] were used. When ISFME was applied, the heating rate was enhanced and the extraction time was obviously shorter than that consumed in conventional solvent-free microwave extraction(CSFME). Twenty-eight kinds of compounds in the essential oil were identified, and the total content fractions of the compounds obtained by CIP-ISFME, GP-ISFME, CSFME, and hydrodistillation(HD) were 96.65%, 96.06%, 97.22%, and 96.29%, respectively. The compounds in the essential oil obtained from Cinnamomum cassia Presl. by ISFME, CSFME, and HD were compared and the quantity of the essential oil was almost the same. The ISFME has been proved to be a feasible way to extract essential oil from dried samples, and there are several obvious advantages in ISFME over those of HD and CSFME, for example, shorter extraction time(30 min) and lower energy consumption.展开更多

The sandwich immunoreaction among goat-anti-human IgG, human IgG and goat-anti-human IgG labeled with colloidal gold nanoparticles was applied in this experiment. First, a sandwich im- munocomplex was formed on the surface of 96 well clear polystyrene high bind stripwellTM microplate. After the formation of the sandwich immunocomplex, a solution was added to dissociate the immunocomplex at room temperature. Then the solution of each well was transferred into the corresponding test tube. Thirdly, the rhodamine B solution was injected into each test tube. The rhodamine B chloraurate was extracted into ether that was measured with a spectrofluorometer. The experimental results indicate that the fluorescence intensity increased with the increase of human IgG concentration. The fluorescence intensity of rhodamine B chloraurate at 570 nm was proportional to the logarithm of human IgG concentration in a range from 10 to 5 × 10^5 ng/mL. It was shown that the determination of human IgG was easily made with the proposed fluorospectrophotometry.展开更多

Waters OASIS HLB was used for concentra...A rapid and effective method, solid phase extraction coupled with high performance liquid chromatography(SPE-HPLC), was applied to the separation and analysis of ginsenosides. Waters OASIS HLB was used for concentrating and purifying samples and Alltima C18(53 mm×7 mm, 3 μm) chromatography column was used for separating ginsenoside Rg1, Re, Rb1, Rc, Rb2 and Rd. These ginsenosides were analyzed within 20 min in our gradient elution process and the equilibrium time of the chromatography column cost only 5 min. Moreover, there was no obvious baseline drift in our experiment. This method was used to analyze the contents of ginsenosides in different ginseng products for quality control. Four ginseng products were studied, including two kinds of capsules, one kind of tablet and one kind of injection. The results show that the method developed in this paper had good accuracy, linearity and precision. Therefore, this method could be applied in quality control of ginseng products.展开更多

Although RFP was k...Ret finger protein(RFP) is a member of the tripartite motif family, which is characterized by a conserved RING finger of motif, a B-box, and a coiled-coil domain(they are called RBCC generally). Although RFP was known to be an oncogene when its RBCC moiety was connected with a tyrosine kinase domain by DNA rearrangement, its biological function was not well defined. In this study, by using real-time RT-PCR, the RFP expressions in human and mouse normal tissues, and in the cervical squamous cell carcinoma, endometrial adenocarcinoma, gastric adenocarcinoma, esophageal squamous cell carcinoma, and brain cancer tissues were analyzed. The result of the study proved that the highest level of mRNA reverse transcription appeared in the normal testical tissue, whereas that in other normal tissues of human and mice were low. The mRNA reverse transcription level of RFP was higher in the endometrial adenocarcinoma tissue than in the cervical squamous cell carcinoma tissue; the mRNA reverse transcription level of RFP in the gastric adenocarcinoma tissue was significantly higher than that in the esophageal squamous cell carcinoma tissue. It was also found that the mRNA reverse transcription level of RFP in the brain cancer tissue was higher than that in the normal brain tissue. These results suggested that RFP could possibly be a useful molecular target for the development of new therapeutics for malignant tumors.展开更多

We investigated the distrib...CT9 is a recently cloned cancer-testis antigen, which is a member of the bromodomain and extraterminal family. Each member of this protein family contains two N-terminal bromodomain motifs. We investigated the distribution of CT9 in different tissues and the possibility for it to be used as a potential therapeutic target in cancer treament. By using the real-time RT-PCR method and 18SrRNA as an internal standard, we analyzed the CT9 expression in several normal human tissues and in the tissues of patients suffering from cancer. The result of this study shows that the highest level of mRNA is only present in testis tissue because the CT9 expression has not been detected in other normal tissues. In 6 of 10 cases of gastric adenocarcinoma, in 3 of 10 cases of esophageal squamous cell carcinoma, in 2 of 9 cases of endometrial carcinoma and only in 1 of 12 cases of brain cancer, the low level expression of CT9 was detected. In none of the 12 cases of cervical squamous cell carcinoma, the expression of CT9 was detected. Since the high level expression of CT9 is only found in the normal testis tissue, but the low expression in cancer tissues, for example tissues of cervical squamous cell carcinoma, brain cancer, endometfial adenocarcinoma, esophageal squamous cell carcinoma, we conclude that CT9 cannot be used as a cancer therapeutic target molecule for cervical squamous cell carcinoma, brain cancer, endometrial adenocarcinoma, esophageal squamous cell carcinoma.展开更多
