花期低温对草莓叶片光系统荧光特性及保护酶活性的影响

以草莓品种“赛娃”(Fragaria ananassa cv.Selva)为试材,于2022年11-12月在南京信息工程大学开展人工环境控制试验。11月中旬草莓苗进入开花期,挑选长势一致的草莓苗放入人工气候室进行低温处理。温度设置3个水平,即5℃(日最低气温)/15... 以草莓品种“赛娃”(Fragaria ananassa cv.Selva)为试材,于2022年11-12月在南京信息工程大学开展人工环境控制试验。11月中旬草莓苗进入开花期,挑选长势一致的草莓苗放入人工气候室进行低温处理。温度设置3个水平,即5℃(日最低气温)/15℃(日最高气温)、10℃/20℃和15℃/25℃(CK),处理持续时间为3d、6d和9d。测定不同低温持续天数下草莓叶片光系统荧光诱导动力学参数和保护酶活性等指标,以期了解低温对草莓叶片光系统荧光特性及保护酶活性的影响。结果表明:(1)在日最低温5~15℃范围内,随着温度的降低,持续时间的延长,草莓叶片单位反应中心吸收的光能(ABS/RC)、单位反应中心耗散掉的能量(DIo/RC)和单位反应中心捕获的用于还原QA的能量(TRo/RC)均有所升高,但单位反应中心捕获的用于电子传递的能量(ETo/RC)下降。日最低温5℃持续9d处理的ABS/RC、DIo/RC分别为CK的1.367倍和1.718倍。日最低温10℃持续3d、6d、9d和日最低温5℃持续9d的TRo/RC分别较CK增加了7.42%、14.66%、25.27%和30.48%,而ETo/RC分别较CK下降了1.71%、20.39%、26.65%和28.32%。用于热耗散的量子比率(φDo)随着低温持续时间的延长而显著增大,日最低温5℃持续3d和9d时分别较CK升高了12.34%和25.32%。然而用于电子传递的量子产额(φEo)随低温持续时间的延长而降低,日最低温5℃持续3d和9d分别较CK下降了27.92%和43.02%。(2)随着温度的降低和低温持续时间的延长,草莓叶片超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化物酶(POD)的活性和丙二醛(MDA)含量逐渐增加。日最低温5℃持续6d时SOD和POD的活性达到最大,分别为CK的1.39倍和2.03倍,CAT受低温影响变化幅度较小。日最低气温5℃持续9d的MDA含量最高,为CK的2.10倍。研究显示低温会破坏草莓叶片光系统的结构和功能,光化学效率下降,以热量散失的能量增加,导致PSⅡ供体侧提供电子的能力下降,电子传递效率降低,草莓叶片会激发体内的抗氧化系统,对环境产生一定的抵抗能力,表现为SOD、POD、CAT的活性较CK相比均有不同程度的升高,但细胞膜结构仍有一定损伤,膜透性增加。 展开更多

以四季草莓品种‘赛娃’(Fragaria ananassa cv.Selva)为试材,于2022年在南京信息工程大学Venlo型温室及人工气候室内设计环境控制试验。试验设置昼温/夜温15℃/5℃(T1)、20℃/10℃(T2)两个低温水平,低温持续天数设置3d、6d和9d,以25℃... 以四季草莓品种‘赛娃’(Fragaria ananassa cv.Selva)为试材,于2022年在南京信息工程大学Venlo型温室及人工气候室内设计环境控制试验。试验设置昼温/夜温15℃/5℃(T1)、20℃/10℃(T2)两个低温水平,低温持续天数设置3d、6d和9d,以25℃/15℃温室环境下生长的植株为对照(CK),系统测定叶片光合参数和气孔指标。结果表明:(1)花期低温明显抑制草莓植株叶片的光合能力,随着胁迫程度的加强,草莓花期叶片的最大净光合速率(Pnmax)、光饱和点(LSP)、气孔导度(Gs)和蒸腾速率(Tr)均呈降低趋势;光补偿点(LCP)、暗呼吸速率(Rd)、胞间CO_(2)浓度(Ci)、气孔限制值(Ls)和水分利用效率(WUE)均呈升高趋势,引起光合速率降低的主要原因是气孔限制。(2)低温使得草莓花期叶片叶绿素a、叶绿素b、叶绿素总量(a+b)及类胡萝卜素含量降低,随低温胁迫时间增加,光合色素含量略微回升,但仍显著低于CK。(3)低温胁迫对叶片气孔特性的影响尤为显著,气孔长度、气孔宽度、气孔开度、气孔周长、气孔面积和气孔开张比均随温度降低及胁迫时间增加显著减小,气孔密度有减小趋势但差异不显著。试验结果显示花期低温可降低草莓花期叶片气孔尺寸和叶绿素含量,抑制叶片光合能力。 展开更多

北方寒区日光温室冬季生产基本无通风,为了探寻室内弱光、高湿、低温及低风速环境下的番茄蒸腾量模拟模型,基于Penman-Monteith(P-M)方程及适应此特定环境的边界层空气动力学阻力、气孔平均阻力、土壤热通量等参数模拟了温室长季节栽培... 北方寒区日光温室冬季生产基本无通风,为了探寻室内弱光、高湿、低温及低风速环境下的番茄蒸腾量模拟模型,基于Penman-Monteith(P-M)方程及适应此特定环境的边界层空气动力学阻力、气孔平均阻力、土壤热通量等参数模拟了温室长季节栽培番茄( Lycopersicon esculentum Mill)单株的蒸腾速率并进行了试验验证,揭示了蒸腾速率与净辐射、饱和水汽压差的日变化规律,确定了蒸腾速率与植株上方净辐射的定量关系,检验了土壤热通量取值对蒸腾速率的影响。结果显示:2017-12-11—2018-01-03室内太阳总辐射最大值367 W·m ^-2 、夜晚及阴天相对湿度接近100%、室内风速接近0 m·s^-1 的情况下,单株植株边界层空气动力学阻力变化范围晴天为147~438 s·m^-1 ,阴天为 211~365 s·m^-1;气孔平均阻力晴天69~1 506 s·m^-1 ,阴天132~1 151 s·m^-1;P-M方程模拟的单株番茄逐时蒸腾速率在晴天、阴天中午的平均值分别为0.06、0.02 mm·h ^-1 ,模拟值与实测值比较,平均相对误差约为10%。研究还表明,单株番茄上方净辐射量的43.5%通过蒸腾作用转化为潜热;试验环境下,土壤热通量的取值变化对蒸腾速率影响不大。研究确定的蒸腾速率估算模型可为北方寒区冬季日光温室基质袋培番茄蒸腾量估算以及水分管理提供参考。 展开更多

生态系统呼吸是陆地生态系统向大气释放CO_2的最大分量,与温度升高存在着复杂的相互作用。揭示温度升高对生态系统呼吸的影响有助于阐明陆地生态系统碳循环与气候间的反馈作用。在总结生态系统呼吸定义及观测方法的基础上,重点就温度升... 生态系统呼吸是陆地生态系统向大气释放CO_2的最大分量,与温度升高存在着复杂的相互作用。揭示温度升高对生态系统呼吸的影响有助于阐明陆地生态系统碳循环与气候间的反馈作用。在总结生态系统呼吸定义及观测方法的基础上,重点就温度升高影响陆地生态系统呼吸的研究结果进行综述,从研究对象、增温方式、增温梯度、增温幅度、影响效果及影响机理等方面进行归纳。结果表明,已有研究主要以极地、青藏高原等地的陆地生态系统为研究对象,增温手段以开顶箱(OTCs)式增温为主,并逐渐向开放式红外增温扩展。增温梯度大多设置增温和对照两个梯度且增温幅度多数介于1~3℃。已有研究发现增温显著促进了多数站点的生态系统呼吸,但在青藏高原等水分限制的陆地生态系统,增温可能导致生态系统呼吸没有发生显著变化甚至显著降低。增温影响陆地生态系统呼吸的机理大多从生态系统呼吸的温度敏感性及生态系统呼吸组分所占比例的变化等着手来探讨。进而提出未来需要重视箱式法在陆地生态系统呼吸及其组分观测中的作用,基于陆地生态系统呼吸组分的变化阐明生态系统呼吸对外界环境变化的响应,并积极开展梯度增温对农田生态系统呼吸的影响研究。 展开更多

目的:分析CO2激光联合重组人干扰素α-2b凝胶治疗面部扁平疣患者的疗效及对患者皮肤病生活质量评分的影响。方法:选取2018年7月-2019年12月笔者科室收治的62例面部扁平疣患者。根据随机对照实验将患者分为研究组(31例)与对照组(31例)。... 目的:分析CO2激光联合重组人干扰素α-2b凝胶治疗面部扁平疣患者的疗效及对患者皮肤病生活质量评分的影响。方法:选取2018年7月-2019年12月笔者科室收治的62例面部扁平疣患者。根据随机对照实验将患者分为研究组(31例)与对照组(31例)。对照组采用CO2激光治疗,研究组给予重组人干扰素α-2b凝胶辅助CO2激光治疗。观察两组患者治疗效果、皮肤病生活质量指数量表(DLQI)评分、不良反应率及复发率。结果:治疗后,研究组治疗有效率达93.55%,高于对照组的70.97%,两组比较有显著性差异(P<0.05);两组患者DLQI评分均降低,研究组低于对照组,具有统计学意义(P<0.05);研究组不良反应率及复发率均低于对照组,具有统计学意义(P<0.05)。结论:采用重组人干扰素α-2b凝胶辅助CO2激光治疗面部扁平疣,能有效提高机体免疫能力,清除体内HPV病毒,降低并发症及复发率,临床疗效佳。 展开更多

The seven ginsenoside... Ginsenosides Rgl, Re, Rb1, Rc, Rb2, Rb3, and Rd in different parts of the American ginseng plant were investigated. The extraction process was a pressurized microwave-assisted extraction(PMAE). The seven ginsenosides were separated and determined by high-performance liquid chromatography(HPLC) with a ultraviolet(UV) detector, at 203 nm. The experiment results showed significant variations in the individual ginsenoside contents of the American ginseng in different parts and ages of the plant. The results demonstrated that the leaves, root hairs, and rhizomes of Panax quinquefolius L. contained higher ginsenoside contents, followed by the main roots and stems. The leaves contained dramatically higher levels of ginsenoside Rg1 Rb3, and Rd than the other four parts. Higher contents of Rb1 and Re were present in the main roots, root hairs, and rhizomes. The amount of ginsenoside content in the stems was the lowest. The total content of the seven ginsenosides in main roots, root hairs and rhizomes increased with the age of the plant. In contrast, the ginsenoside contents in the leaves and stems decreased with a year of growth. 展开更多

An improved solvent-free microwave extraction(ISFME) was used for the extraction of essential oil from dried Cinnamomum cassia Presl. Two kinds of solid microwave absorption media[carbonyl iron powder(CIP) and graphite powder(GP)] were used. When ISFME was applied, the heating rate was enhanced and the extraction time was obviously shorter than that consumed in conventional solvent-free microwave extraction(CSFME). Twenty-eight kinds of compounds in the essential oil were identified, and the total content fractions of the compounds obtained by CIP-ISFME, GP-ISFME, CSFME, and hydrodistillation(HD) were 96.65%, 96.06%, 97.22%, and 96.29%, respectively. The compounds in the essential oil obtained from Cinnamomum cassia Presl. by ISFME, CSFME, and HD were compared and the quantity of the essential oil was almost the same. The ISFME has been proved to be a feasible way to extract essential oil from dried samples, and there are several obvious advantages in ISFME over those of HD and CSFME, for example, shorter extraction time(30 min) and lower energy consumption. 展开更多

A method for the determination of human immunoglobulin G(IgG) based on a colloidal gold label by fluorospectrophotometry was developed. The sandwich immunoreaction among goat-anti-human IgG, human IgG and goat-anti-human IgG labeled with colloidal gold nanoparticles was applied in this experiment. First, a sandwich im- munocomplex was formed on the surface of 96 well clear polystyrene high bind stripwellTM microplate. After the formation of the sandwich immunocomplex, a solution was added to dissociate the immunocomplex at room temperature. Then the solution of each well was transferred into the corresponding test tube. Thirdly, the rhodamine B solution was injected into each test tube. The rhodamine B chloraurate was extracted into ether that was measured with a spectrofluorometer. The experimental results indicate that the fluorescence intensity increased with the increase of human IgG concentration. The fluorescence intensity of rhodamine B chloraurate at 570 nm was proportional to the logarithm of human IgG concentration in a range from 10 to 5 × 10^5 ng/mL. It was shown that the determination of human IgG was easily made with the proposed fluorospectrophotometry. 展开更多

Eighteen sex hormones in antler velvet were determined by high performance liquid chromatography tandem mass spectrometry.The solid phase extraction was applied to eliminating the matrix effect.The experimental conditions were examined and optimized.Under the optimal conditions,the proposed method provides the good linearities and determination limits(0.2―1.0 μg/kg) of the analytes investigated.The recoveries ranging from 72.3% to 149.5% were obtained for the target analytes at two concentration levels.This method was applied to the determination of eighteen sex hormones in different kinds of antler velvet samples and the obtained results are satisfactory.The results indicate that the proposed method is suitable for the determination of sex hormones in antler velvet samples. 展开更多

为解决中国东北地区农业基础设施建设不完善,农业生产效率不高等诸多问题,本研究根据水足迹理论测算东北地区2009—2018年主要农作物水足迹总量,运用对数均值指数分解法,将影响水足迹总量驱动因素分解为产量因素、种植规模因素、虚拟水... 为解决中国东北地区农业基础设施建设不完善,农业生产效率不高等诸多问题,本研究根据水足迹理论测算东北地区2009—2018年主要农作物水足迹总量,运用对数均值指数分解法,将影响水足迹总量驱动因素分解为产量因素、种植规模因素、虚拟水含量因素和作物结构因素,从时间和空间格局双重视角下进行驱动因素贡献情况的分析。结果表明,农业生产水足迹总量总体呈现上升趋势。从时间角度看,作物产量和种植面积占主导地位,作物结构因素的贡献相对较小,虚拟水因素从抑制作用逐渐转变为促进作用;在空间格局上,东北地区水足迹总量存在由南向北的递增趋势。前期各省主要的水足迹总量增长由产量和面积因素贡献,后期种植结构变化以及作物自然因素的不同需求,导致虚拟水因素加剧了各省之间水足迹的差异。从优化空间布局和作物结构,提高农作物生产效率及灌溉用水资源利用率,推动绿色农业发展等方面提出建议。 展开更多

Be able to form large particles with protein via electrostatic force to enhance the intensity of resonance light scattering(RLS),(NH4)2SO4 was used as a RLS probe to determine bovine serum albumin(BSA),human serum albumin(HSA) and ovum albumin(OVA).The experimental conditions were investigated,including the acidity and saturation degree of (NH4)2SO4.Under the optimum conditions,the enhanced RLS intensity is proportional to the concentration of BSA,HSA and OVA in the ranges of 5×10-8―1×10-6,2.5×10-8―8×10-7 and 5×10-8―1.5×10-6 mol/L,respectively.The detection limits for BSA,HAS and OVA are 6.6×10-9,3.8×10-9 and 7.4×10-9 mol/L,respectively.The effects of foreign substances were also examined.The practical and synthetic samples were analyzed with satisfactory results. 展开更多

A rapid and effective method, solid phase extraction coupled with high performance liquid chromatography(SPE-HPLC), was applied to the separation and analysis of ginsenosides. Waters OASIS HLB was used for concentrating and purifying samples and Alltima C18(53 mm×7 mm, 3 μm) chromatography column was used for separating ginsenoside Rg1, Re, Rb1, Rc, Rb2 and Rd. These ginsenosides were analyzed within 20 min in our gradient elution process and the equilibrium time of the chromatography column cost only 5 min. Moreover, there was no obvious baseline drift in our experiment. This method was used to analyze the contents of ginsenosides in different ginseng products for quality control. Four ginseng products were studied, including two kinds of capsules, one kind of tablet and one kind of injection. The results show that the method developed in this paper had good accuracy, linearity and precision. Therefore, this method could be applied in quality control of ginseng products. 展开更多

Expression Levels of RFP in Normal and Cancer Human Tissues via Real-time RT-PCR Detection

Ret finger protein(RFP) is a member of the tripartite motif family, which is characterized by a conserved RING finger of motif, a B-box, and a coiled-coil domain(they are called RBCC generally). Although RFP was known to be an oncogene when its RBCC moiety was connected with a tyrosine kinase domain by DNA rearrangement, its biological function was not well defined. In this study, by using real-time RT-PCR, the RFP expressions in human and mouse normal tissues, and in the cervical squamous cell carcinoma, endometrial adenocarcinoma, gastric adenocarcinoma, esophageal squamous cell carcinoma, and brain cancer tissues were analyzed. The result of the study proved that the highest level of mRNA reverse transcription appeared in the normal testical tissue, whereas that in other normal tissues of human and mice were low. The mRNA reverse transcription level of RFP was higher in the endometrial adenocarcinoma tissue than in the cervical squamous cell carcinoma tissue; the mRNA reverse transcription level of RFP in the gastric adenocarcinoma tissue was significantly higher than that in the esophageal squamous cell carcinoma tissue. It was also found that the mRNA reverse transcription level of RFP in the brain cancer tissue was higher than that in the normal brain tissue. These results suggested that RFP could possibly be a useful molecular target for the development of new therapeutics for malignant tumors. 展开更多

We investigated the distrib... CT9 is a recently cloned cancer-testis antigen, which is a member of the bromodomain and extraterminal family. Each member of this protein family contains two N-terminal bromodomain motifs. We investigated the distribution of CT9 in different tissues and the possibility for it to be used as a potential therapeutic target in cancer treament. By using the real-time RT-PCR method and 18SrRNA as an internal standard, we analyzed the CT9 expression in several normal human tissues and in the tissues of patients suffering from cancer. The result of this study shows that the highest level of mRNA is only present in testis tissue because the CT9 expression has not been detected in other normal tissues. In 6 of 10 cases of gastric adenocarcinoma, in 3 of 10 cases of esophageal squamous cell carcinoma, in 2 of 9 cases of endometrial carcinoma and only in 1 of 12 cases of brain cancer, the low level expression of CT9 was detected. In none of the 12 cases of cervical squamous cell carcinoma, the expression of CT9 was detected. Since the high level expression of CT9 is only found in the normal testis tissue, but the low expression in cancer tissues, for example tissues of cervical squamous cell carcinoma, brain cancer, endometfial adenocarcinoma, esophageal squamous cell carcinoma, we conclude that CT9 cannot be used as a cancer therapeutic target molecule for cervical squamous cell carcinoma, brain cancer, endometrial adenocarcinoma, esophageal squamous cell carcinoma. 展开更多
