不同介质对低渗诱导葡萄牙牡蛎三倍体的影响 被引量:2

以葡萄牙牡蛎(Crassostrea angulata)为研究对象,分别使用6种介质(NaCl、KCl、CaCl2、MgCl2、蔗糖、海水)配制的梯度低渗溶液,在40%~50%受精卵出现第一极体(PB1)时处理15 min,进行三倍体诱导。研究表明,对于同种介质,卵裂率和孵化率均... 以葡萄牙牡蛎(Crassostrea angulata)为研究对象,分别使用6种介质(NaCl、KCl、CaCl2、MgCl2、蔗糖、海水)配制的梯度低渗溶液,在40%~50%受精卵出现第一极体(PB1)时处理15 min,进行三倍体诱导。研究表明,对于同种介质,卵裂率和孵化率均随渗透压升高而增大,三倍体诱导率则先增后减,在渗透压为P8(盐度为8的海水渗透压)时达到最高。在同一渗透压P8条件下,除CaCl2组外,各组卵裂率、孵化率、诱导率与低渗海水组相比无显著差异(P>0.05),CaCl2组卵裂率、孵化率降低,而诱导率显著较高(P<0.05)。各低渗组卵裂率、孵化率均低于自然海水组。研究结果表明,低渗海水法诱导贝类三倍体时物理的低渗因素发挥主要作用,Ca2+可能起到辅助作用。 展开更多

为探究低盐诱导对长牡蛎(Crassostrea gigas)胚胎发育和发育初期的氧化应激、能量供应及渗透调节的影响。实验观察了低盐诱导组和对照组胚胎到达不同发育时期所用的时间以及发育初期部分酶活力的变化。研究显示,在40%~50%受精卵出现第... 为探究低盐诱导对长牡蛎(Crassostrea gigas)胚胎发育和发育初期的氧化应激、能量供应及渗透调节的影响。实验观察了低盐诱导组和对照组胚胎到达不同发育时期所用的时间以及发育初期部分酶活力的变化。研究显示,在40%~50%受精卵出现第一极体时,用盐度4~12的低盐海水处理长牡蛎受精卵15 min会导致其胚胎发育明显迟缓,且诱导盐度越低,发育速度越慢,最慢(盐度4)比对照组(盐度32)到达D形幼虫的时间滞后了4 h以上。同时,用盐度8处理15 min后,受精卵的丙酮酸激酶(Pyruvate kinase,PK)和Na^+-K+-ATP酶水平显著降低,总抗氧化能力(Total-antioxidant capacity,T-AOC)水平显著升高(P<0.05),谷草转氨酶(Glutamic oxalacetic transaminase,GOT)水平无显著变化(P>0.05);转入自然海水中恢复1.5 h后,四细胞期胚胎丙酮酸激酶、Na^+-K^+-ATP酶水平恢复到与对照组无显著差异,T-AOC水平回落至接近对照组水平,而谷草转氨酶水平无明显改变。说明低盐胁迫可能会导致长牡蛎受精卵糖代谢受阻,渗透调节紊乱,同时受到较严重的氧化胁迫,且这些改变在转入自然海水一定时间后即可恢复,不会对胚胎发育产生持续性影响,而GOT表达水平则始终无明显变化。研究结果表明,低盐诱导不仅会导致牡蛎受精卵胚胎发育迟缓,还可能会使其受到严重的氧化胁迫,同时阻碍其代谢功能和渗透调节。 展开更多

气体泄漏对气体绝缘全封闭组合电器(gas insulated substation,GIS)局部放电的影响有待研究,但缺少相关研究手段。为此,首先提出等效相对介电常数的概念,推导等效相对介电常数与气室压强、温度、分子数等的关系,从极化原理上分析其表征... 气体泄漏对气体绝缘全封闭组合电器(gas insulated substation,GIS)局部放电的影响有待研究,但缺少相关研究手段。为此,首先提出等效相对介电常数的概念,推导等效相对介电常数与气室压强、温度、分子数等的关系,从极化原理上分析其表征气体泄露程度的可行性;然后利用XFdtd三维电磁场仿真软件建立同轴波导结构自由金属微粒缺陷下的局部放电(partial discharge,PD)模型,仿真分析发生气体泄露后的局部放电情况;通过对特高频(ultra high frequency,UHF)电磁波时域信号与放电频谱的分析,探究气体泄漏的影响。仿真结果表明,基于等效相对介电常数气体泄漏对GIS局部放电影响的分析得以实现,同时揭示了脉冲宽度、主频、信号幅值等特征量的变化规律。 展开更多

A feasible approach to selectively recover iron and rare earth elements(REEs)from red mud through acid leaching-coordination-solvent extraction was proposed.The leaching efficiencies of Fe,Al,Ti,Sc,La,Ce,Nd and Y can reach up to 95.9%,82.1%,68.3%,93.3%,82.3%,96.9%,98.3%and 95.6%,respectively,under the optimal condition in the leaching process.Aliquat 336 showed excellent extraction performance of iron in chloride-rich solution,and the maximum extraction efficiency can reach over 96%in one time extraction while the loss of other metals was less than 10%,under the condition of Aliquat 336 concentration(v/v)of 30%,aqueous-organic ratio of 1.0 and extraction time of 20 min.Furthermore,P204 can effectively extract the scandium while Al and most other REEs remain in the aqueous phase.This approach may provide a new insight for the recovery of valuable resources from red mud. 展开更多

目前我国大型冶炼企业产生的污酸均被当做一种高浓度重金属废水来处理,不仅需要高额的废水处理费用,而且还会产生大量的废水处理渣.结合污酸及氧化锌烟灰的主要成份,采用循环浸出工艺,利用污酸对氧化锌烟灰进行浸出,浸出完全后,综合回... 目前我国大型冶炼企业产生的污酸均被当做一种高浓度重金属废水来处理,不仅需要高额的废水处理费用,而且还会产生大量的废水处理渣.结合污酸及氧化锌烟灰的主要成份,采用循环浸出工艺,利用污酸对氧化锌烟灰进行浸出,浸出完全后,综合回收浸出液中的Cu、Zn、As.实验研究了终点pH、浸出温度、浸出时间对污酸一次浸出和二次循环浸出的影响,以及双氧水加入量、温度、时间对一次除As的影响和硫化钠加入量、温度、时间对二次除As的影响.实验表明:最佳一次浸出条件为终点pH值为1.5、反应温度为85℃、反应时间为5 h;最佳二次循环浸出条件为终点pH值为4、反应温度为85℃;最佳一次除As条件为每毫升二次循环浸出液添加0.067 mL双氧水、反应温度为40℃、反应时间为1.5 h;最佳二次除As条件为每毫升一次除As后液添加0.02 g硫化钠、反应温度为35℃、反应时间为2 h.污酸综合利用后,原来的高浓度重金属废水变成了中性废水,其中的重金属(As、Cu、Zn)质量浓度分别降至3.26、2.63和50.63 mg·L^(−1),稍加处理即可达到污水综合排放标准.此工艺既综合回收了污酸和氧化锌烟灰中的有价成份,又集中处理了有害元素As,消减了危险废物的产生量,达到了节能减排的目的. 展开更多

近年来,学者们主要关注如何使用内部资源推进健康中国建设,很少将研究视角拓展至外部资源。为此,文章将外商直接投资数据与中国综合社会调查数据(2012~2017)进行匹配,详细分析外资企业的涌入与中国居民健康状况之间的关系。结果发现,外... 近年来,学者们主要关注如何使用内部资源推进健康中国建设,很少将研究视角拓展至外部资源。为此,文章将外商直接投资数据与中国综合社会调查数据(2012~2017)进行匹配,详细分析外资企业的涌入与中国居民健康状况之间的关系。结果发现,外商直接投资规模的增长可以显著提高中国居民的健康水平,且开展稳健性检验和安慰剂检验后,结论保持不变。机制分析表明,外商直接投资主要依托经济渠道而非环境渠道推进健康中国建设。异质性分析发现,外商直接投资对低收入群体和低学历群体健康水平提升的助推作用显著强于高收入群体和高学历群体。此外,政策支持力度越大或人才对接能力越强,引进外资就越有利于当地居民健康状况的改善。 展开更多

Sequential Therapy Based on Evolvement of Patterns: A New Model for Treatment of Alzheimer's Disease 被引量:16

In order to solve the problem of long-term (>9 months) efficacy in the treatment of Alzheimer's disease (AD) by conventional therapy (CT), a staged and multiply-targeted sequential therapy based on the evolvement of patterns (STEP) was developed. Its main innovations include:(1) the time order of evolution of patterns defined by Chinese medicine (CM) in AD was found, that is,"the orderly pattern evolution starting from Shen (Kidney) deficiency, progressing to phlegm, stasis and fire, and worsening to severe toxin as well as functional collapse";(2) the cascade hypothesis of Shen deficiency in AD and its sequential therapy based on Shen-reinforcing was proposed, that is,"reinforcing Shen in the early stage and throughout the whole process, resolving phlegm, activating blood and purging fire in the middle stage, detoxifying and replenishing vitality to stop the collapse in the advanced stage", and through meta-analysis, clinical drug use was optimized, thus the leap from "inferential selection" to "evidence-based selection" was realized;(3) the STEP regimen combined with CT maintained cognitive and behavioral stability in AD patients for at least 12 months, with cognitive enhancement and behavioral synergy after 9 months, and cognitive benefit was superior to CT at 9, 12, 15, 18, 21, and 24 months, respectively. The 2-year cognitive improvement rate was increased by 25.64%(P=0.020) and the cognitive deterioration rate was decreased by 48.71%(P=0.000). Among them, the cognitive and functional benefits of Shen-reinforcing therapy for very early AD (350 cases) for 1 year were better than the placebo (P<0.001), and the dementia conversion rate was reduced by 8.85%(P=0.002). The behavioral symptomatic relief of patients with vascular dementia received fire-purging therapy (540 cases) was superior to those received CT (P=0.016). These data suggested that the STEP regimen has synergistic effects on CTs at least in terms of cognitive benefit, and the earlier the use, the greater the benefit will have. Therefore, the STEP regimen should be considered as one of the clinical options, particularly for the dearth of effective pharmaceutical or immunological interventions that are currently available for AD. 展开更多

Objective:To investigate the effect of GAPT,an extract mixture from Radix Ginseng,Rhizoma Acor tatarinowii,Radix Polygalae and Radix Curcuma(containing ingredient of turmeric),etc.on expression of tau protein and its phosphorylation related enzyme in hippocampal neurons of APPV717I transgenic mice.Methods:Sixty three-month-old APPV717I transgenic mice were randomly divided into model group,donepezil group[0.92 mg/(kg·d)],the low,medium and high dosage of GAPT groups[0.075,0.15,0.30 g/(kg·d),12 in each group],and 12 three-month-old C57BL/6J mice were set as a normal control group,treatments were administered orally once a day respectively,and both the normal group and model group were given 0.5%sodium carboxymethyl cellulose solution.Immunohistochemistry(IHC)and Western blot analysis were used to detect the expression of total tau protein(Tau-5),cyclin-dependent kinase 5(CDK5)and protein phosphatase 2A(PP2A)in hippocampal neurons of experimental mice after 8-month drug administration(11 months old).Results:In the model group,the expression of Tau-5 and CDK5 were increased,whereas the expression of PP2A was decreased in hippocampal neurons,which were significantly different compared with that in the normal group(all P〈0.01).IHC test indicated the number and area of either Tau-5 or CDK5 positive cells were decreased with a dose-depended way in GAPT groups,and an increase of PP2A.Compared with the model group,the changes were significant in GAPT groups(P〈0.05 or P〈0.01).Similar results were shown by Western blot.Conclusion:GAPT could attenuate abnormal hyperphosphorylation of tau protein in hippocampal neurons of APPV717I transgenic mice via inhibiting the expression of CDK5 and activating the expression of PP2A. 展开更多
