为研究航天相关神经眼综合征(SANS)相关特征参数与微重力的相关性,以Sprague-Dawley(SD)大鼠为悬尾动物模型,构建了超长扫描深度的光学相干断层扫描(ULOCT)装置,测量大鼠眼部特征参数(角膜、前房、晶状体厚度和玻璃体腔深度及眼轴长度)... 为研究航天相关神经眼综合征(SANS)相关特征参数与微重力的相关性,以Sprague-Dawley(SD)大鼠为悬尾动物模型,构建了超长扫描深度的光学相干断层扫描(ULOCT)装置,测量大鼠眼部特征参数(角膜、前房、晶状体厚度和玻璃体腔深度及眼轴长度),以及视网膜超高分辨率(UHROCT)测量装置,测量大鼠眼底亚层结构参数(视神经层、视网膜内层、视网膜外层、脉络膜和巩膜的厚度)。结果表明大鼠悬尾1月后,眼底彩照未见明显视盘水肿等症状,但OCT结果显示大鼠眼结构发生明显改变:角膜变薄(p<0.01),眼轴缩短(p<0.05),视网膜外层变薄(p<0.01)。这些特征参数变化(尤其是眼轴明显缩短)展现了SANS亚临床前期的症状。本文为后续微观层面的SANS分子机制研究提供了宏观动物模型以及在体眼部测量新方法。 展开更多

目的探究microRNA-29b(miR-29b)在肝纤维化过程中的作用,以及对肝星状细胞增殖和凋亡的影响。方法将大鼠随机分为对照组和模型组,每组10只。模型组大鼠经腹部注射60%3 ml/kg CCl4复制肝纤维化模型;对照组注射等量生理盐水。分别观察两... 目的探究microRNA-29b(miR-29b)在肝纤维化过程中的作用,以及对肝星状细胞增殖和凋亡的影响。方法将大鼠随机分为对照组和模型组,每组10只。模型组大鼠经腹部注射60%3 ml/kg CCl4复制肝纤维化模型;对照组注射等量生理盐水。分别观察两组大鼠肝纤维化情况,检测肝组织中miR-29b、TGF-β1、SAMD3基因及TGF-β1、SAMD3蛋白的表达。体外培养肝星状细胞HSC-T6、LX-2,转染miR-29b siRNA,检测miR-29b对HSC-T6、LX-2细胞增殖、凋亡的影响,以及对TGF-β1、SAMD3表达的影响。结果与对照组相比,模型组心肌纤维比例增加,随着时间延长,第6和12周心肌纤维比例逐渐升高(P <0.05)。与对照组相比,模型组miR-29b基因相对表达量降低,随着时间延长,模型组miR-29b基因相对表达量逐渐降低(P<0.05)。与对照组相比,模型组TGF-β1、SAMD3基因和蛋白相对表达量升高;随着时间延长,TGF-β1、SAMD3基因和蛋白相对表达量逐渐升高(P <0.05)。在HSC-T6、LX-2细胞中,miR-29b转染组G1期细胞比例高于空白对照组、阴性转染组,S期细胞比例低于空白对照组、阴性转染组(P<0.05)。miR-29b转染组HSC-T6、LX-2细胞凋亡率高于空白对照组、阴性转染组(P<0.05)。与空白对照组、阴性转染组相比,HSC-T6、LX-2细胞中TGF-β1、SAMD3基因和蛋白相对表达量升高,miR-29b基因相对表达量降低(P<0.05)。结论 miR-29b在肝纤维化过程中表达降低,miR-29b可抑制肝星状细胞增殖,诱导其凋亡。 展开更多

Full grown oocytes derived from Bufo Bufo gargarizans rearing at high temperature environment (24℃),never underwent GVBD after progesterone treatment. No p34cdc2, H1 kinase activity was detected in the oocytes after progesterone stimulation or OA microinjection; Western blotting analysis showed that the level of p3cdc2, and p33 in the oocytes are significantly lower than those in the oocytes derived from the hibernating toads (below 10 ℃). 25S-Met incorporation analysis showed that when the oocytes were incubated at 6℃, synthesis of about thirty defferent polypeptides was promoted or induced, including p34cdc2 and some other p13suc1-binding proteins. All these results indicated that a low temperature environment is essential for the oocytes of Bufo Bufo gargarizans to express and store some cell cycle drivers and its regulators, and to gain the maturation competence. These results will also provide a new clue for explaining the molecular mechanisms why gametogenesis of some organisms depends on a relative low temperature and how to maintain the geographical distribution of some animals. 展开更多

Objective To establish C57BL/6J embryonic stem (ES) cell lines with potential germ- line contribution Methods ES cells were isolated from blastocyst inner cell mass of C5 7BL/6J mice, and cultured for 15 passages, and then injected into blastococels of ICR mice blastocysts to establish chimeric mice. Results Three ES cell lines (mC57ES1,mC57ES3, mC57ES7) derived from the inner cell mass of C57BL/6J mice blastocysts were established. They were characteristic of undifferentiated state, including normal XY karyotype, expression of a specific cell surface marker “stage-specific embryonic antigen-I” and alkaline phosphatase in continuous passage. When injected into immunodeficient mice, mC57ES1 cells consistently differentiated into derivatives of all three embryonic germ layers. When mC57ES1 cells were transferred into ICR mice blastocysts, 4 chimeric mice have been obtained. One male of them revealed successful germ-line transmission. Conclussion We have obtained C57BL/6J ES cell lines with a potential germ-line contribution, which can be used to generate transgenic and gene knock-out mice. 展开更多

It is also well known that, in scrotal mammals, spermatogenesis depends greatly on the maintenance of comparatively low temperature in the scrotum. To investigate whether the expression of cdc2 and Cyclin B1 in spermatogenic cells during spermatogenesis is actually a temperature dependent event, in situ hybridization, Western blotting and immunohistochemistry analysis were used to study the expression of cdc2 and Cyclin B1 in normal and cryptorchid testis. Results showed that the abdominal temperature had no significant influence on the transcription of cdc2 and Cyclin B1 in the spermatogonia and pachytene/diplotene primary spermatocytes, but it blocked the translation of them. Due to the deficiency of p34cdc2 and Cyclin B1, the spermatogonia and pachytene/diplotene primary spermatocytes were unable to form MPF, hence, they couldn’t undergo karyokinesis. The development of primary spermatocytes was arrested at the G2 to M phase transition. We also found that testosterone could regulate the Cyclin B1 expression in spermatogenic cells. Muscular injection of testosterone could recover spermatogenesis in the unilateral scrotal testis which was influenced by the contralateral cryptorchid testis, but it could not salvage the spermatogenesis block in the cryptorchid testis. 展开更多

Chang’e-5 explorer successfully acquired lunar regolith core samples from depths of greater than 1 m of lunar surface.This study analyzed the lunar core drilling process based on the telemetry data,image information,and returned samples to optimize the sampling device design and enhance the understanding of the lunar regolith.In particular,a prediction method for the projected drilling path and local terrain fitting of drilling dip angle was proposed based on the flight events recorded during the core drilling process and the image information acquired before,during,and after sampling.The results revealed that the drilling dip angle of Chang’e-5 was approximately2.3.,and the deviation of the drilling length and depth was less than 2 mm.For continuous drilling,a fusion method based on telemetry data and image information was applied to determine the demarcation point of drilling with and without the lunar soil.The position of the demarcation point implied that the drilling point remained at approximately 6 mm loose soil,thereby lagging the action of the force response.Additionally,a characteristic parameter comparison method was proposed for the lunar and ground drilling to analyze the status of the lunar soil.Furthermore,the analysis results revealed that the majority of the Chang’e-5 drilling samples were derived from 0–73.8 cm below the lunar surface and few samples were extracted below 73.8 cm,as the drilling encountered several rocky regions.Moreover,the drilling point exhibited two prominent stratification variations at~28.7 cm and~70 cm below the lunar surface.Ultimately,the preliminary relationship between sample dissected position in soft tube and drilling displacement was analyzed.The segmented estimation results can support research on subsurface lunar soil. 展开更多

视网膜血管形态学特征与急性轻症脑卒中患者脑侧支循环和预后的相关性研究

目的:分析视网膜血管形态特征与软脑膜侧支循环功能的相关性,探索急性缺血性卒中患者侧支循环功能及预后的预测模型。方法:纳入53例急性缺血性卒中患者,利用头颈部CT血管造影(CT angiography,CTA)检查评估侧支循环评分,分为良好侧支组... 目的:分析视网膜血管形态特征与软脑膜侧支循环功能的相关性,探索急性缺血性卒中患者侧支循环功能及预后的预测模型。方法:纳入53例急性缺血性卒中患者,利用头颈部CT血管造影(CT angiography,CTA)检查评估侧支循环评分,分为良好侧支组及不良侧支组;以90天改良Rankin评分作为预后指标。分析眼底彩色照片,提取视网膜血管形态参数,包括:视网膜各象限的动、静脉血管主干、分支管径、弯曲度、均衡度、血管树分型维数、视杯及视盘几何尺寸等。组间差异显著的参数,与侧支循环评分、卒中预后评分进行相关性分析,并建立回归模型进一步验证。结果:本研究纳入的病例中,良好侧支组的良好预后率显著高于不良侧支组。视网膜血管形态参数中,中央视网膜动脉当量[(43.29±5.02)vs(39.05±4.21),P=0.002)]和动脉平均管径[(51.49(5.35)vs(49.07±4.29),P=0.040]在组间差异显著。其余参数无显著差异。中央视网膜动脉当量和动脉平均管径与软脑膜侧支循环评分显著相关,相关系数分别为0.418(P=0.002)和0.290(P=0.035),其余参数无显著相关性。对侧支循环评分进行Logistic回归提示,CRAE与侧支评分相关(OR=1.349,95%CI 1.101~1.653,P=0.004),CRAE预测侧支循环评分的ROC曲线下面积为0.747(95%CI 0.615~0.879,P=0.002);动脉平均管径在侧支循环评分的回归分析中不具有统计学意义(OR=1.086,95%CI 0.976~1.209,P=0.129)。各视网膜血管参数在90天改良Rankin评分的Logistic回归中均无统计学意义。结论:在轻型卒中患者中,中央视网膜动脉当量与软脑膜侧支循环功能显著相关,可反映软脑膜侧支循环功能状态。本研究检验的视网膜血管形态参数均不能预测缺血性脑卒中预后。 展开更多

Three organo-montmorillonites were prepared using surfactants, and their adsorption behaviors toward sulfamethoxazole(SMX) were investigated. The surfactants used were cetyltrimethyl ammonium bromide(CTMAB), 3-(N,N-dimethylhexadecylammonio) propane sulfonate(HDAPS) and 1,3-bis(hexadecyldimethylammonio)-propane dibromide(BHDAP). The properties of the organo-montmorillonites were characterized by X-ray diffraction, scanning electron microscopy and N2adsorption–desorption isotherm measurements. Results showed that the interlayer spacing of montmorillonite was increased and the surface area as well as the morphology were changed. Batch adsorption experiments showed that the surfactant loading amount had a great effect on the adsorption of SMX. The adsorption process was p H dependent and the maximum adsorption capacity was obtained at p H 3 for HDAPS-Mt, while CTMAB-Mt and BHDAP-Mt showed a high removal efficiency at 3–11. The adsorption capacity increased with the initial SMX concentration and contact time but decreased with increasing solution ionic strength.Kinetic data were best described by the pseudo second-order model. Equilibrium data were best represented by the Langmuir model, and the Freundlich constant(n) indicated a favorable adsorption process. The maximum adsorption capacity of SMX was 235.29 mg/g for CTMAB-Mt, 155.28 mg/g for HDAPS-Mt and 242.72 mg/g for BHDAP-Mt. Thermodynamic parameters were calculated to evaluate the spontaneity and endothermic or exothermic nature. The adsorption mechanism was found to be dominated by electrostatic interaction,while hydrophobic interaction played a secondary role. 展开更多
