BACKGROUND Perivascular epithelioid cell tumor(PEComa)is a rare mesenchymal neoplasm that predominantly affects the kidney and uterus.The occurrence of this tumor in the liver,particularly with simultaneous involvement of the liver and kidney,is exceedingly uncommon.Pathological diagnosis is the gold standard.PEComas usually show positive immunohistochemical staining for melanocytic(HMB-45,Melan-A)and myoid(SMA,muscle-specific actin)markers.CASE SUMMARY We presented a noteworthy case of malignant PEComa affecting both the liver and kidney in a 53-year-old man with tuberous sclerosis complex(TSC).FAT2 and TP73 mutations in the kidney were identified and positive expression of diagnostic markers including HMB-45,Melan A,and TFE3 were detected.In addition,we demonstrated that hepatic artery perfusion chemotherapy was ineffective for hepatic PEComa,while surgery remained the most effective approach.Everolimus showed an excellent efficacy in the postoperative treatment of the tumor.CONCLUSION Surgical treatment is preferred for malignant PEComa affecting liver and kidney,especially with TSC;everolimus is effective postoperatively. 展开更多

As a unique probe,the precision measurement of pp solar neutrinos is important for studying the sun’s energy mechanism as it enables monitoring the thermodynamic equilibrium and studying neutrino oscillations in the vacuum-dominated region.For a large-scale liquid scintillator detector,a bottleneck for pp solar neutrino detection is the pile-up events of intrinsic14C decay.This paper presents a few approaches to discriminating between pp solar neutrinos and ^(14)C pile-up events by considering the differences in their time and spatial distributions.In this study,a Geant4-based Monte Carlo simulation is conducted.Multivariate analysis and deep learning technology are adopted to investigate the capability of ^(14)C pile-up reduction.The BDTG (boosted decision trees with gradient boosting) model and VGG network demonstrate good performance in discriminating pp solar neutrinos and ^(14)C double pile-up events.Under the ^(14)C concentration assumption of 5×10-18g/g,the signal significance can achieve 10.3 and 15.6 using the statistics of only one day.In this case,the signal efficiency for discrimination using the BDTG model while rejecting 99.18% ^(14)C double pile-up events is 51.1%,and that for the case where the VGG network is used while rejecting 99.81%of the ^(14)C double pile-up events is 42.7%. 展开更多

Flavonoid 3’-hydroxylase(F3’ H) and 3’,5’-hydroxylase(F3’5’ H) generate precursor molecules for the synthesis of cyanidin-based anthocyanins(red) and delphinidin-based anthocyanins(blue to purple) in Ribes nigrum L.(black currant). In this study, full-length 1780 and 165’-bp cDNA homologs of RnF3’5’ H1 and RnF3’ H1 from black currant were identified and cloned using a homologous cloning technique. Data revealed that Rn F3’5’ H1 and RnF3’ H1 are homologs that encode enzymes involved in anthocyanin synthesis from different plants,which phylogenetically cluster with the CYP75 B and CYP75 A families in the P45’ superfamily, respectively.The enzymes encoded by these two genes also shared a high homology with flavonoid hydroxylases identified from other plants. Furthermore, RnF3’5’ H1 and RnF3’ H1 levels were upregulated during fruit maturation. RnF3’5’ H1 levels were associated with both anthocyanin and soluble carbohydrate levels in blackcurrant, while RnF3’ H1 expression did not have such an association. The structure and expression patterns of RnF3’5’ H1 and RnF3’ H1 in blackcurrant were also characterized. Further studies should aid understanding of anthocyanin biosynthesis in black currant to develop molecular approaches and manipulate anthocyanin production in blackcurrant. 展开更多

The influence of ... Background:Gastric cancer (GC) is one of the most globally prevalent cancers in the world. The pathogenesis of GC has not been fully elucidated, and there still lacks effective targeted therapeutics. The influence of altered kinesin superfamily protein 22 (KIF22) expression in GC progression is still unclearly. The aim of this study was to investigate the KIF22 effects on GC and related mechanisms.Methods:Gastric carcinoma tissues and matching non-cancerous tissues were collected from patients with GC who have accepted a radical gastrectomy in Lanzhou University Second Hospital from May 2013 to December 2014. The expression of KIF22 was examined in GC of 67 patients and 20 para-carcinoma tissues by immunochemical staining. The relationship between the expression of KIF22 and clinicopathologic characteristics was next investigated in the remaining 52 patients except for 15 patients who did not complete follow-up for 5 years. Cell viability was performed via 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) test and colony formation assay in the MGC-803 and BGC-823 GC cells. Cell scratch and trans-well invasion assay was performed to assess migration ability in the MGC-803 and BGC-823 GC cells. Gene set enrichment analysis (GSEA) pathway enrichment analysis was performed to explore the potential functions. Cell cycle was detected by flow cytometry. In addition, the two GC cell lines were used to elucidate the underlying mechanism of KIF22 in GC in vitro via assessing the effects on mitogenactivated protein kinase and extracellular regulated protein kinases (MAPK/ERK) signal transduction pathway-related expressions by Western blotting assays. The differences were compared by t tests, one-way analysis of variance, and Chi-squared tests. Results:The study showed that KIF22 was up-regulated in GC, and KIF22 high expression was significantly related to differentiation degree ( χ2 = 12.842, P = 0.002) and poorly overall survivals. GSEA pathway enrichment analysis showed that KIF22 was correlated with the cell cycle. Silence of KIF22 decreased the ability of the proliferation and migration in gastric cells, induced G1/S phase cell cycle arrest via regulating the MAPK-ERK pathways. Conclusions:KIF22 protein level was negatively correlated with prognosis. KIF22 knockdown might inhibit proliferation and metastasis of GC cells via the MAPK-ERK signaling pathway. 展开更多

Background We propose to measure the coherent elastic neutrino-nucleus scattering(CEvNS)using a dual-phase liquid argon time projection chamber(TPC)with 200 kg fiducial mass.The detector is expected to be adjacent to the JUNOTAO experiment and to be about 35 m from a reactor core with 4.6 GW thermal power at Taishan.The antineutrino flux is approximately 6×10^(12)cm^(−1)s^(−1) at this location,leading to more than 11,000 coherent scattering events per day in the fiducial mass.Motivation The nuclear recoil energies concentrate in the sub-keV region,corresponding to less than ten ionization electrons in the liquid argon.The key question is how to veto and shield the background in the hall where the vertical overburden is about 5 m.w.e.And what is the signal count rate and the background rate.In addition,what physical parameters can be measured what is the sensitivity.Methods We used the Geant4 to simulate the backgrounds from cosmic ray muons and ambient radioactivity decays.And a veto and shielding design is presented.Then a χ^(2) function is constructed and the sensitivity calculate package built to calculate the sensitivity of physical parameters.Results The detection of several ionization electrons can be achieved in the dual-phase TPC due to the large amplification in the gas region.With a feasible detection threshold of four ionization electrons,the signal rate is 955 per day.The detector is designed to be shielded well from cosmogenic backgrounds and ambient radioactivities to reach a 16%background-tosignal ratio in the energy region of interest.With the large CEvNS sample,the expected sensitivity of measuring the weak mixing angle sin^(2)θ_(w),and of limiting the neutrino magnetic moment are discussed.In addition,a synergy between the reactor antineutrino CEvNS experiment and the dark matter experiment is foreseen. 展开更多
