目的:研究抗虫抗草甘膦玉米转化体ZD12-6外源基因的分子特征,对ZD12-6的抗虫和抗草甘膦性状进行综合评价,为ZD12-6产业化提供基础信息。创新点:抗虫抗草甘膦转基因玉米ZD12-6是聚合了双抗虫基因和抗草甘膦基因的转化体,多基因聚合有利... 目的:研究抗虫抗草甘膦玉米转化体ZD12-6外源基因的分子特征,对ZD12-6的抗虫和抗草甘膦性状进行综合评价,为ZD12-6产业化提供基础信息。创新点:抗虫抗草甘膦转基因玉米ZD12-6是聚合了双抗虫基因和抗草甘膦基因的转化体,多基因聚合有利于后期品种转育和多性状叠加。对该转化体的分子特征信息分析和功能评价是评判其产业化价值的重要依据。方法:利用DNA印迹法(Southern blot)研究外源基因插入拷贝数;利用高效热不对称交错聚合酶链反应(hiTAIL-PCR)方法对外源基因的插入位点进行定位;通过蛋白质印迹法(Western blot)对外源蛋白进行定性分析;利用酶联免疫吸附测定(ELISA)分析外源蛋白表达量;通过生物测定对抗虫和草甘膦的抗性水平进行评估。结论:抗虫抗草甘膦玉米转化体ZD12-6中外源基因单拷贝插入,插入位点位于玉米1号染色体。ZD12-6对玉米上的主要害虫具有良好抗性,同时对草甘膦具有较强耐受性,具有产业化推广潜力。 展开更多

Human serum albumin(HSA) is widely utilized for medical purposes and biochemical research.Transgenic rice has proved to be an attractive bioreactor for mass production of recombinant HSA(rHSA).However,transgene spread is a major environmental and food safety concern for transgenic rice expressing proteins of medical value.This study aimed to develop a selectively terminable transgenic rice line expressing HSA in rice seeds,and a simple process for recovery and purification of rHSA for economical manufacture.An HSA expression cassette was inserted into a T-DNA vector encoding an RNA interference(RNAi) cassette suppressing the CYP81A6 gene.This gene detoxifies the herbicide bentazon and is linked to the 5-enolpyruvylshikimate-3-phosphate synthase(EPSPS) cassette which confers glyphosate tolerance.ANX Sepharose Fast Flow(ANX FF) anion exchange chromatography coupled with Butyl Sepharose High Performance(Butyl HP) hydrophobic interaction chromatography was used to purify rHSA.A transgenic rice line,HSA-84,was obtained with stable expression of rHSA of up to 0.72% of the total dry weight of the dehusked rice seeds.This line also demonstrated high sensitivity to bentazon,and thus could be killed selectively by a spray of bentazon.A two-step chromatography purification scheme was established to purify the rHSA from rice seeds to a purity of 99% with a recovery of 62.4%.Results from mass spectrometry and N-terminus sequencing suggested that the purified rHSA was identical to natural plasma-derived HSA.This study provides an alternative strategy for large-scale production of HSA with a built-in transgene safety control mechanism. 展开更多

A female-specific protein, vitellogenin (Vg), and its corresponding egg vitellin (Vt) are identified in the ectoparasitic wasp Nasonia vitripennis. Both native Vt and Vg have a molecular mass of about 350 kDa, which is composed of two subunits of approximately 190 kDa and 165 kDa under reducing and denaturing conditions (sodium dodecyl sulfate-polyacrylamide gel electrophoresis). An indirect sandwich enzyme-linked immunosorbent assay developed with both monoclonal and polyclonal antibodies against N. vitripennis Vt. Vg was first detected in the hemolymph on the 10th day after parasitism, and was first observed in oocytes on the 12th day. In adults deprived of food, the highest hemolymph Vg level occurred at the time of adult eclosion and the highest level of Vt in ovaries was found at 30 h after eclosion. In contrast, feeding adults with 20% sucrose resulted in the reduction of Vt uptake by ovaries and the extension of life span, but had little effect on Vg production. Deprived of hosts, starvation of female wasps had no significant effects on ovariole growth and oocyte maturation before the wasps died. However, starvation of female wasps supplied with hosts accelerated the wasps laying progeny into hosts, but resulted in a decrease of total progeny production by comparison with wasps fed with 20% sucrose. 展开更多

目的:获得具有高水平表达Cry1Ac、Cry1Ig和EPSPS基因并且具有抗虫、抗草甘膦活性的转基因水稻株。创新点:Cry1Ig为新发现的苏云金杆菌(Bt)杀虫基因;Cry1Ig与Cry1Ac共同使用有利于延缓害虫产生对Bt杀虫蛋白抗性。方法:将Cry1Ac、Cry1Ig和... 目的:获得具有高水平表达Cry1Ac、Cry1Ig和EPSPS基因并且具有抗虫、抗草甘膦活性的转基因水稻株。创新点:Cry1Ig为新发现的苏云金杆菌(Bt)杀虫基因;Cry1Ig与Cry1Ac共同使用有利于延缓害虫产生对Bt杀虫蛋白抗性。方法:将Cry1Ac、Cry1Ig和EPSPS(G10)基因的表达框依次插入到同一转移DNA(T-DNA)中,再将此T-DNA利用农杆菌介导法转化到水稻中。从转基因后代中,利用蛋白质印迹法(Western blot)、DNA印迹法(Southern blot)和热不对称交错聚合酶链式反应(TAIL-PCR)技术,筛选出能够高水平表达上述三个基因以及T-DNA单拷贝插入并且未发生明显插入突变的转化株。对于筛选到的转化株,利用Western blot进一步分析Cry1Ac、Cry1Ig和G10基因的表达水平;使用酶联免疫吸附测定(ELISA)检测Cry1Ac的表达量;使用棉铃虫、二化螟和稻纵卷叶螟为对象,测定转基因水稻的抗虫活性;使用草甘膦喷施法,测定转基因水稻的抗草甘膦活性。最后,在大田试验中,考察转基因水稻与非转基因水稻的基本农艺性状,观察T-DNA的插入是否对转基因水稻的生长产生明显的影响。结论:本实验最终获得具有高水平表达Cry1Ac、Cry1Ig和G10基因以及T-DNA单拷贝插入并且未引起明显变化的转化株GAI-14。GAI-14对棉铃虫、二化螟、稻纵卷叶螟以及草甘膦均具有明显的抗性。田间试验表明GAI-14在基本农艺性状上与非转基因水稻无明显差异。 展开更多
