BACKGROUND Currently,endoscopic retrograde cholangiopancreatography(ERCP)plus laparoscopic cholecystectomy(LC)is the main treatment for cholecystolithiasis combined with choledocholithiasis.However,the treatment is un...BACKGROUND Currently,endoscopic retrograde cholangiopancreatography(ERCP)plus laparoscopic cholecystectomy(LC)is the main treatment for cholecystolithiasis combined with choledocholithiasis.However,the treatment is unsatisfactory,and the development of better therapies is needed.AIM To determine the clinical efficacy of LC plus cholangioscopy for cholecystolithiasis combined with choledocholithiasis.METHODS Patients(n=243)with cholecystolithiasis and choledocholithiasis admitted to The Affiliated Haixia Hospital of Huaqiao University(910th Hospital of Joint Logistic Support Force)between January 2019 and December 2023 were included in the study;111 patients(control group)underwent ERCP+LC and 132 patients(observation group)underwent LC+laparoscopic common bile duct exploration(LCBDE).Surgical success rates,residual stone rates,complications(pancreatitis,hyperamylasemia,biliary tract infection,and bile leakage),surgical indicators[intraoperative blood loss(IBL)and operation time(OT)],recovery indices(postoperative exhaust/defecation time and hospital stay),and serum inflammatory markers[C-reactive protein(CRP)],tumor necrosis factor-α(TNF-α),and interleukin-6(IL-6)were compared.RESULTS No significant differences in surgical success rates and residual stone rates were detected between the observation and control groups.However,the complication rate,IBL,OT,postoperative exhaust/defecation time,and hospital stays were significantly reduced in the observation group compared with the control group.Furthermore,CRP,TNF-α,and IL-6 Levels after treatment were reduced in the observation group compared with the levels in the control group.CONCLUSION These results indicate that LC+LCBDE is safer than ERCP+LC for the treatment of cholecystolithiasis combined with choledocholithiasis.The surgical risks and postoperative complications were lower in the observation group compared with the control group.Thus,patients may recover quickly with less inflammation after LCBDE.展开更多

Th...In order to effectively predict the fracture of AA7075-T6 sheet, the forming limit curves of AA7075-T6 high-strength sheet were drawn according to Morciniak Kuczyski (M K) model and Lou Huh criterion, respectively. The errors between the predicted values of the two theoretical prediction models and experimental values were calculated by error analysis. The forming limit curves were verified by the punch stretch test to evaluate the prediction accuracy of M K model and Lou Huh criterion. The error analysis results show that the mean error of Lou Huh criterion with the optimal parameters for all tensile specimens is 25.04%, while the mean error of M K model for all tensile specimens is 74.24%. The prediction accuracy of Lou Huh criterion in predicting the fracture of AA7075-T6 sheet is higher. The punch stretch test results show that the forming limit curve drawn by Lou Huh criterion can effectively predict the fracture of AA7075-T6 sheet, but the prediction accuracy of M K model is relatively poor.展开更多

In the IRAC images, this region shows strong emissions of polycyclic aromatic hydrocarbons in channel 4 and emis- sions of H2 in channel 2. Many of the H2 features are aligned to form jet-like struc- tures. Three bipolar jets in the NHa core region and a couple of jets northwest of the core have been identified. We identify the possible driving agents of the bipolar jets and show them to be very young. An embedded cluster has been detected in the NH3 core; many members in the cluster have spectral energy distributions that increase from JHK bands toward longer wavelengths, indicative of their early evolutionary stages. Millimeter and submillimeter continuum emissions in the NH3 core and the northwest subregion are found to coincide spatially with these presumable Class0/I sources. The existence of H2 bipolar jets and very young stellar objects suggests that star formation is continuing at the present epoch in these subregions. Combining in- formation from previous studies, we propose a sequential star formation scenario in the whole AFGL 5157 region.展开更多
Gravitational accretion accumulates the original mass.This process is crucial for us to understand the initial phases of star formation.Using the specific infall profiles in optically thick and thin lines,we searched ...Gravitational accretion accumulates the original mass.This process is crucial for us to understand the initial phases of star formation.Using the specific infall profiles in optically thick and thin lines,we searched the clumps with infall motion from the Milky Way Imaging Scroll Painting(MWISP)CO data in previous work.In this study,we selected 133 sources as a sub-sample for further research and identification.The excitation temperatures of these sources are between 7.0 and 38.5 K,while the H2 column densities are between 10^21 and 10^23 cm^-2.We have observed optically thick lines HCO+(1-0)and HCN(1-0)using the DLH 13.7-m telescope,and found 56 sources with a blue profile and no red profile in these two lines,which are likely to have infall motions,with a detection rate of 42%.This suggests that using CO data to restrict the sample can effectively improve the infall detection rate.Among these confirmed infall sources are 43 associated with Class O/I young stellar objects(YSOs),and 13 which are not.These 13 sources are probably associated with the sources in the earlier evolutionary stage.In comparison,the confirmed sources that are associated with Class O/I YSOs have higher excitation temperatures and column densities,while the other sources are colder and have lower column densities.Most infall velocities of the sources that we confirmed are between 10^-1 to 10^0 km s^-1,which is consistent with previous studies.展开更多
Objective:Nucleotide binding oligonucleoid-like receptor 3(NLRP3),caspase-1(caspase-1)and interleukin-1β(IL-1β)levels in the serum of patients with chronic heart failure were detected to study the changes of pyropto...Objective:Nucleotide binding oligonucleoid-like receptor 3(NLRP3),caspase-1(caspase-1)and interleukin-1β(IL-1β)levels in the serum of patients with chronic heart failure were detected to study the changes of pyroptosis in heart failure and its significance.Methods:180 patients with chronic heart failure(chronic heart failure group)and 40 healthy patients(normal control group)diagnosed in our hospital from February 2018 to October 2019 were selected.According to the New York heart association(NYHA)classification standard,includingⅡlevel 48 cases,Ⅲlevel 60 cases,Ⅳlevel in 72 patients.After admission,the n-terminal brain natriuretic peptide precursor(NT-proBNP),the left ventricular end-diastolic diameter(LVEDD)、the left ventricular end-diastolic volume(LVEDV)and the left ventricular ejection fraction(LVEF)were improved by ultrasonic morphological examination.ELISA was used to detect the expression levels of NLRP3,Caspase-1 and IL-1βin serum of all samples,and the association of NLRP3,Caspase-1,IL-1βand related indicators in patients with chronic heart failure of different grades was statistically analyzed,so as to investigate the relationship between pyroptosis and the occurrence and development of chronic heart failure.Results:chronic heart failure group NLRP3,Caspase-1,IL-1β,NT-proBNP and LVEDD、LVEDV were higher than normal control group,and LVEF was lower than normal control group,the difference was statistically significant(P<0.05).Levels of NLRP3,Caspase-1,IL-1β,NT-proBNP and LVEDD、LVEDV increased with cardiac function grading,while LVEF decreased with cardiac function grading.(P<0.05).Pearson analysis showed that NLRP3,Caspase-1 and IL-1βwere positively correlated with NT-proBNP and LVEDD、LVEDV(r=0.398,0.396,0.398,0.506,0.499,0.505,0.449,0.422,0.424,P<0.01)and negatively correlated with LVEF(r=-0.400,-0.555,-0.438,P<0.01).The area under the ROC curve of NLRP3,Caspase-1 and IL-1βfor diagnosing heart failure was 0.768,0.805 and 0.799,respectively,with statistically significant differences(P<0.01).Conclusion:Serum levels of NLRP3,caspase-1,and IL-1βwere higher in patients with chronic heart failure than in the control group,and continued to increase as the heart function deteriorated.Pyroptosis has certain guiding significance for the occurrence and development of chronic heart failure.展开更多

Ba...We carried out observations toward the giant molecular cloud W 37 with the J = 1 - 0 transitions of 12CO, 13CO and C180 using the 13.7 m single-dish telescope at the Delingha station of Purple Mountain Observatory. Based on these CO lines, we calculated the column densities and cloud masses for molecular clouds with radial velocities around +20 km s-1. The gas mass of W 37, calculated from 13CO emission, is 1.7 × 10^5 M, above the criterion to be considered a giant molecular cloud. The dense ridge of W 37 is a dense filament, which is supercritical in terms of linear mass ratio. Dense clumps found by C180 emission are aligned along the dense ridge at regular intervals of about 2.8 pc, similar to the clump separation caused by large-scale 'sausage instability'. We confirm the identification of the giant molecular filament (GMF) G 18.0-16.8 and find a new giant filament, G 16.5-15.8, located ~ 0.7° to the west of G 18.0-16.8. Both GMFs are not gravitationally bound, as indicated by their low linear mass ratio (- 80 M pc-l). We compared the gas temperature map with the dust temperature map from Herschel images, and found similar structures. The spatial distributions of class I objects and the dense clumps are reminiscent of triggered star formation occurring in the northwestern part of W 37, which is close to NGC 6611.展开更多

The control group was given routine treatment, while the treatment group was treated with comprehensive rehabilitationon the basis of the control group. The course of treatment for both groups was 10 d. The changes of body weight, milk consumption, NBNA score, GMFM score and Gesell score were observed. Results:After treatment, the body weight and milk consumption of the treatment group increased significantly compared with those before treatment (P<0.01), which was significantly better than that of the control group (P<0.01);the NBNA score of the treatment group was higher than that of the control group at 40 weeks of gestational age (P<0.05), and the abnormal rate of NBNA was lower than that of the control group (P<0.01). The GMFM scores of A-energy and B-energy areas and the five dimensions of social adaptation, big exercise, fine exercise, language and personal social interaction in the treatment group increased significantly in 1 month, 2 months and 3 months after birth,which were significantly better than those in the control group (P<0.05 or P<0.01). Conclusion: Super-early comprehensive rehabilitation can improve the physical quality of high-risk infants and promote the development of the nervous system, with remarkable effect.展开更多

This study assessed the efficacy and safety of tofacitinib in Chinese patients with RA enrolled in Phase 3 and long?term extension(LTE) studies.Methods: ORAL Sync was a 1?year, randomized, placebo?controlled, Phase 3 trial. Patients received tofacitinib 5 or 10 mg twice daily(BID) or placebo advanced to tofacitinib 5 or 10 mg BID at 3 or 6 months. All patients remained on ≥1 background conventional synthetic disease?modifying antirheumatic drug. ORAL Sequel is an open?label LTE study(data?cut: March 2015; data collection and analyses were ongoing, and study database was not locked at the time of analysis; study was closed in 2017). Efficacy outcomes: American College of Rheumatology(ACR) 20/50/70 response rates and Disease Activity Score in 28 joints using erythrocyte sedimentation rate(DAS28?4 [ESR]). Patient? and physician?reported outcomes: Health Assessment Questionnaire?Disability Index(HAQ?DI), Patient and Physician Global Assessment of Arthritis, and pain(visual analog scale). Safety was assessed throughout.Results: ORAL Sync included 218 patients; 192 were subsequently enrolled into ORAL Sequel. In ORAL Sync, more patients achieved ACR20(tofacitinib 5 mg BID, 67.4%; 10 mg BID, 70.6%; placebo, 34.1%) and DAS28?4(ESR) <2.6(tofacitinib 5 mg BID, 7.1%;10 mg BID, 13.1%; placebo, 2.3%) with tofacitinib versus placebo at Month 6. Mean changes from baseline in HAQ?DI were greater with tofacitinib versus placebo at Month 6. In ORAL Sequel, efficacy was consistent to Month 48. Incidence rates for adverse events of special interest in tofacitinib?treated patients were similar to the global population.Conclusions: Tofacitinib significantly reduced signs/symptoms and improved physical function and quality of life in Chinese patients with moderate?to?severely active RA up to Month 48. Background and Aims:Recently,the World Health Organization adopted the first-ever global hepatitis strategy with the dream of eliminating viral hepatitis as a public health threat by 2030.However,the epidemiology and ...Background and Aims:Recently,the World Health Organization adopted the first-ever global hepatitis strategy with the dream of eliminating viral hepatitis as a public health threat by 2030.However,the epidemiology and treatment rates of hepatitis C virus(HCV)infection in Western China are still unknown.Methods:A total of 111,916 adult individuals(15-96 years)who underwent the HCV-antibody(HCV-Ab)test in the Second Affiliated Hospital of Chongqing Medical University between 2013 and 2015 were included in this study.We retrospectively analyzed the electronic medical records'data for each,and the positivity of HCV-Ab and the treatment of HCV RNA-positive patients were evaluated.Results:During 2013-2015,the crude prevalence of HCVAb was 1.4%(95%CI:1.4-1.5;1,611/111,916)and the adjusted prevalence of HCV-Ab was 1.7%(95%CI:1.6-1.8),which was higher than in the 2006 national study(0.43%).The prevalence was 2-times higher in males than females(2.0%vs.1.1%,p<0.01).Notably,only 46%(434/951)of the HCV RNA-positive patients received standard peginterferon plus ribavirin treatment,with 370(82%)that completed treatment,of whom 272(74%)achieved sustained virologic response(SVR).Particularly,11%(32/292)of HCV RNA-positive patients were HBsAg-positive,and the SVR rate for them was lower than for the HBsAg-negative patients,but no significant difference was observed.Conclusions:HCV infection may have increased since 2006 in Western China.The SVR rate of peg-interferon plus ribavirin treatment was high,but the proportion of untreated HCV patients was large.Thus,more efforts need to be made by the government to create a scientific-based policy for HCV treatment and prevention.展开更多

Background:Zinc finger and BTB domain-containing protein 46(Zbtb46)is a transcription factor identified in classical dendritic cells,and maintains dendritic cell quiescence in a steady state.Zbtb46 has been reported t...Background:Zinc finger and BTB domain-containing protein 46(Zbtb46)is a transcription factor identified in classical dendritic cells,and maintains dendritic cell quiescence in a steady state.Zbtb46 has been reported to be a negative indicator of acute myeloid leukemia(AML).We found that Zbtb46 was expressed at a relatively higher level in hematopoietic stem and progenitor cells(HSPCs)compared to mature cells,and higher in AML cells compared to normal bone marrow(BM)cells.However,the role of Zbtb46in HSPCs and AML cells remains unclear.Therefore,we sought to elucidate the effect of Zbtb46 in normal hematopoiesis and AML cells.Methods:We generated Zbtb46^fl/fl and Zbtb46^fl/fl Mx1-Cre mice.The deletion of Zbtb46 in Zbtb46^fl/fl Mx1-Cre mice was induced by intraperitoneal injection of double-stranded poly(I).poly(C)(poly(I:C)),and referred as Zbtb46 cKO.After confirming the deletion of Zbtb46,the frequency and numbers of HSPCs and mature blood cells were analyzed by flow cytometry.Serial intraperitoneal injection of 5-fluorouracil was administrated to determine the repopulation ability of HSCs from Zbtb46^fl/fl and Zbtb46 cKO mice.The correlation between Zbtb46 expression and prognosis was analyzed using the data from the Cancer Genome Atlas.To investigate the role of Zbtb46 in AML cells,we knocked down the expression of Zbtb46 in THP-1 cells using lentiviral vectors expressing small hairpin RNAs targeting Zbtb46.Cell proliferation rate was determined by cell count assay.Cell apoptosis and bromodeoxyuridine incorporation were determined by flow cytometry.Results:The percentages and absolute numbers of HSPCs and mature blood cells were comparable in Zbtb46 cKO mice and its Zbtb46^fl/fl littermates(Zbtb46^fl/fl vs.Zbtb46 cKO,HPC:801,310±84,282 vs.907,202±97,403,t=0.82,P=0.46;LSK:86,895±7802 vs.102,210±5025,t=1.65,P=0.17;HSC:19,753±3116 vs.17,608±3508,t=0.46,P=0.67).The repopulation ability of HSCs from Zbtb46^fl/fl Mx1-Cre mice was similar to those from Zbtb46^fl/fl control(P=0.26).Zbtb46 had elevated expression in AML cells compared to total BM cells from normal control.Knockdown of Zbtb46 in THP-1 cells led to a significant increase in cell apoptosis and reduced cell growth and proliferation.Conclusion:Collectively,our data indicate that Zbtb46 is essential for survival and proliferation of AML cells,but dispensable for normal hematopoiesis.展开更多

An embedded cluster has been detected in the NH3 core; many members in the cluster have spectral energy distributions that increase from JHK bands toward longer wavelengths, indicative of their early evolutionary stages. Millimeter and submillimeter continuum emissions in the NH3 core and the northwest subregion are found to coincide spatially with these presumable Class0/I sources. The existence of H2 bipolar jets and very young stellar objects suggests that star formation is continuing at the present epoch in these subregions. Combining in- formation from previous studies, we propose a sequential star formation scenario in the whole AFGL 5157 region.

文摘Gravitational accretion accumulates the original mass.This process is crucial for us to understand the initial phases of star formation.Using the specific infall profiles in optically thick and thin lines,we searched the clumps with infall motion from the Milky Way Imaging Scroll Painting(MWISP)CO data in previous work.In this study,we selected 133 sources as a sub-sample for further research and identification.The excitation temperatures of these sources are between 7.0 and 38.5 K,while the H2 column densities are between 10^21 and 10^23 cm^-2.We have observed optically thick lines HCO+(1-0)and HCN(1-0)using the DLH 13.7-m telescope,and found 56 sources with a blue profile and no red profile in these two lines,which are likely to have infall motions,with a detection rate of 42%.This suggests that using CO data to restrict the sample can effectively improve the infall detection rate.Among these confirmed infall sources are 43 associated with Class O/I young stellar objects(YSOs),and 13 which are not.These 13 sources are probably associated with the sources in the earlier evolutionary stage.In comparison,the confirmed sources that are associated with Class O/I YSOs have higher excitation temperatures and column densities,while the other sources are colder and have lower column densities.Most infall velocities of the sources that we confirmed are between 10^-1 to 10^0 km s^-1,which is consistent with previous studies.

文摘Objective:Nucleotide binding oligonucleoid-like receptor 3(NLRP3),caspase-1(caspase-1)and interleukin-1β(IL-1β)levels in the serum of patients with chronic heart failure were detected to study the changes of pyroptosis in heart failure and its significance.Methods:180 patients with chronic heart failure(chronic heart failure group)and 40 healthy patients(normal control group)diagnosed in our hospital from February 2018 to October 2019 were selected.According to the New York heart association(NYHA)classification standard,includingⅡlevel 48 cases,Ⅲlevel 60 cases,Ⅳlevel in 72 patients.After admission,the n-terminal brain natriuretic peptide precursor(NT-proBNP),the left ventricular end-diastolic diameter(LVEDD)、the left ventricular end-diastolic volume(LVEDV)and the left ventricular ejection fraction(LVEF)were improved by ultrasonic morphological examination.ELISA was used to detect the expression levels of NLRP3,Caspase-1 and IL-1βin serum of all samples,and the association of NLRP3,Caspase-1,IL-1βand related indicators in patients with chronic heart failure of different grades was statistically analyzed,so as to investigate the relationship between pyroptosis and the occurrence and development of chronic heart failure.Results:chronic heart failure group NLRP3,Caspase-1,IL-1β,NT-proBNP and LVEDD、LVEDV were higher than normal control group,and LVEF was lower than normal control group,the difference was statistically significant(P<0.05).Levels of NLRP3,Caspase-1,IL-1β,NT-proBNP and LVEDD、LVEDV increased with cardiac function grading,while LVEF decreased with cardiac function grading.(P<0.05).Pearson analysis showed that NLRP3,Caspase-1 and IL-1βwere positively correlated with NT-proBNP and LVEDD、LVEDV(r=0.398,0.396,0.398,0.506,0.499,0.505,0.449,0.422,0.424,P<0.01)and negatively correlated with LVEF(r=-0.400,-0.555,-0.438,P<0.01).The area under the ROC curve of NLRP3,Caspase-1 and IL-1βfor diagnosing heart failure was 0.768,0.805 and 0.799,respectively,with statistically significant differences(P<0.01).Conclusion:Serum levels of NLRP3,caspase-1,and IL-1βwere higher in patients with chronic heart failure than in the control group,and continued to increase as the heart function deteriorated.Pyroptosis has certain guiding significance for the occurrence and development of chronic heart failure.

文摘We carried out observations toward the giant molecular cloud W 37 with the J = 1 - 0 transitions of 12CO, 13CO and C180 using the 13.7 m single-dish telescope at the Delingha station of Purple Mountain Observatory. Based on these CO lines, we calculated the column densities and cloud masses for molecular clouds with radial velocities around +20 km s-1. The gas mass of W 37, calculated from 13CO emission, is 1.7 × 10^5 M, above the criterion to be considered a giant molecular cloud. The dense ridge of W 37 is a dense filament, which is supercritical in terms of linear mass ratio. Dense clumps found by C180 emission are aligned along the dense ridge at regular intervals of about 2.8 pc, similar to the clump separation caused by large-scale 'sausage instability'. We confirm the identification of the giant molecular filament (GMF) G 18.0-16.8 and find a new giant filament, G 16.5-15.8, located ~ 0.7° to the west of G 18.0-16.8. Both GMFs are not gravitationally bound, as indicated by their low linear mass ratio (- 80 M pc-l). We compared the gas temperature map with the dust temperature map from Herschel images, and found similar structures. The spatial distributions of class I objects and the dense clumps are reminiscent of triggered star formation occurring in the northwestern part of W 37, which is close to NGC 6611.
文摘Objective: To observe the effect of super-early comprehensive rehabilitation intervention on neurodevelopmental prognosis of premature high-risk infants;Methods: Premature high-risk infantsmet the inclusion criteria were divided into control group (n=30) and treatment group (n=30) according to parental willingness. The control group was given routine treatment, while the treatment group was treated with comprehensive rehabilitationon the basis of the control group. The course of treatment for both groups was 10 d. The changes of body weight, milk consumption, NBNA score, GMFM score and Gesell score were observed. Results:After treatment, the body weight and milk consumption of the treatment group increased significantly compared with those before treatment (P<0.01), which was significantly better than that of the control group (P<0.01);the NBNA score of the treatment group was higher than that of the control group at 40 weeks of gestational age (P<0.05), and the abnormal rate of NBNA was lower than that of the control group (P<0.01). The GMFM scores of A-energy and B-energy areas and the five dimensions of social adaptation, big exercise, fine exercise, language and personal social interaction in the treatment group increased significantly in 1 month, 2 months and 3 months after birth,which were significantly better than those in the control group (P<0.05 or P<0.01). Conclusion: Super-early comprehensive rehabilitation can improve the physical quality of high-risk infants and promote the development of the nervous system, with remarkable effect.
文摘Background: Tofacitinib is an oral Janus kinase inhibitor for the treatment of rheumatoid arthritis(RA). This study assessed the efficacy and safety of tofacitinib in Chinese patients with RA enrolled in Phase 3 and long?term extension(LTE) studies.Methods: ORAL Sync was a 1?year, randomized, placebo?controlled, Phase 3 trial. Patients received tofacitinib 5 or 10 mg twice daily(BID) or placebo advanced to tofacitinib 5 or 10 mg BID at 3 or 6 months. All patients remained on ≥1 background conventional synthetic disease?modifying antirheumatic drug. ORAL Sequel is an open?label LTE study(data?cut: March 2015; data collection and analyses were ongoing, and study database was not locked at the time of analysis; study was closed in 2017). Efficacy outcomes: American College of Rheumatology(ACR) 20/50/70 response rates and Disease Activity Score in 28 joints using erythrocyte sedimentation rate(DAS28?4 [ESR]). Patient? and physician?reported outcomes: Health Assessment Questionnaire?Disability Index(HAQ?DI), Patient and Physician Global Assessment of Arthritis, and pain(visual analog scale). Safety was assessed throughout.Results: ORAL Sync included 218 patients; 192 were subsequently enrolled into ORAL Sequel. In ORAL Sync, more patients achieved ACR20(tofacitinib 5 mg BID, 67.4%; 10 mg BID, 70.6%; placebo, 34.1%) and DAS28?4(ESR) <2.6(tofacitinib 5 mg BID, 7.1%;10 mg BID, 13.1%; placebo, 2.3%) with tofacitinib versus placebo at Month 6. Mean changes from baseline in HAQ?DI were greater with tofacitinib versus placebo at Month 6. In ORAL Sequel, efficacy was consistent to Month 48. Incidence rates for adverse events of special interest in tofacitinib?treated patients were similar to the global population.Conclusions: Tofacitinib significantly reduced signs/symptoms and improved physical function and quality of life in Chinese patients with moderate?to?severely active RA up to Month 48. 文摘Background and Aims:Recently,the World Health Organization adopted the first-ever global hepatitis strategy with the dream of eliminating viral hepatitis as a public health threat by 2030.However,the epidemiology and treatment rates of hepatitis C virus(HCV)infection in Western China are still unknown.Methods:A total of 111,916 adult individuals(15-96 years)who underwent the HCV-antibody(HCV-Ab)test in the Second Affiliated Hospital of Chongqing Medical University between 2013 and 2015 were included in this study.We retrospectively analyzed the electronic medical records'data for each,and the positivity of HCV-Ab and the treatment of HCV RNA-positive patients were evaluated.Results:During 2013-2015,the crude prevalence of HCVAb was 1.4%(95%CI:1.4-1.5;1,611/111,916)and the adjusted prevalence of HCV-Ab was 1.7%(95%CI:1.6-1.8),which was higher than in the 2006 national study(0.43%).The prevalence was 2-times higher in males than females(2.0%vs.1.1%,p<0.01).Notably,only 46%(434/951)of the HCV RNA-positive patients received standard peginterferon plus ribavirin treatment,with 370(82%)that completed treatment,of whom 272(74%)achieved sustained virologic response(SVR).Particularly,11%(32/292)of HCV RNA-positive patients were HBsAg-positive,and the SVR rate for them was lower than for the HBsAg-negative patients,but no significant difference was observed.Conclusions:HCV infection may have increased since 2006 in Western China.The SVR rate of peg-interferon plus ribavirin treatment was high,but the proportion of untreated HCV patients was large.Thus,more efforts need to be made by the government to create a scientific-based policy for HCV treatment and prevention.

文摘Background:Zinc finger and BTB domain-containing protein 46(Zbtb46)is a transcription factor identified in classical dendritic cells,and maintains dendritic cell quiescence in a steady state.Zbtb46 has been reported to be a negative indicator of acute myeloid leukemia(AML).We found that Zbtb46 was expressed at a relatively higher level in hematopoietic stem and progenitor cells(HSPCs)compared to mature cells,and higher in AML cells compared to normal bone marrow(BM)cells.However,the role of Zbtb46in HSPCs and AML cells remains unclear.Therefore,we sought to elucidate the effect of Zbtb46 in normal hematopoiesis and AML cells.Methods:We generated Zbtb46^fl/fl and Zbtb46^fl/fl Mx1-Cre mice.The deletion of Zbtb46 in Zbtb46^fl/fl Mx1-Cre mice was induced by intraperitoneal injection of double-stranded poly(I).poly(C)(poly(I:C)),and referred as Zbtb46 cKO.After confirming the deletion of Zbtb46,the frequency and numbers of HSPCs and mature blood cells were analyzed by flow cytometry.Serial intraperitoneal injection of 5-fluorouracil was administrated to determine the repopulation ability of HSCs from Zbtb46^fl/fl and Zbtb46 cKO mice.The correlation between Zbtb46 expression and prognosis was analyzed using the data from the Cancer Genome Atlas.To investigate the role of Zbtb46 in AML cells,we knocked down the expression of Zbtb46 in THP-1 cells using lentiviral vectors expressing small hairpin RNAs targeting Zbtb46.Cell proliferation rate was determined by cell count assay.Cell apoptosis and bromodeoxyuridine incorporation were determined by flow cytometry.Results:The percentages and absolute numbers of HSPCs and mature blood cells were comparable in Zbtb46 cKO mice and its Zbtb46^fl/fl littermates(Zbtb46^fl/fl vs.Zbtb46 cKO,HPC:801,310±84,282 vs.907,202±97,403,t=0.82,P=0.46;LSK:86,895±7802 vs.102,210±5025,t=1.65,P=0.17;HSC:19,753±3116 vs.17,608±3508,t=0.46,P=0.67).The repopulation ability of HSCs from Zbtb46^fl/fl Mx1-Cre mice was similar to those from Zbtb46^fl/fl control(P=0.26).Zbtb46 had elevated expression in AML cells compared to total BM cells from normal control.Knockdown of Zbtb46 in THP-1 cells led to a significant increase in cell apoptosis and reduced cell growth and proliferation.Conclusion:Collectively,our data indicate that Zbtb46 is essential for survival and proliferation of AML cells,but dispensable for normal hematopoiesis.
