Acalypha australis L.extract inhibits B16 melanoma cell metastasis through PI3K/AKT signaling pathway

Background:Melanoma is a deadly skin tumor resulting from the malignant transformation of melanocytes.It is highly malignant and invasive,with the highest mortality rate among skin cancers.Acalypha australis L.(AAL),a plant with dual medicinal and culinary purposes,is commonly regarded as an edible wild vegetable in southern China.Additionally,AAL has a long history of medicinal use in China,often employed for its hemostatic,anti-diarrheal,and anti-inflammatory properties.Modern pharmacology has demonstrated that AAL possesses functions such as weight loss,antimicrobial activity,antiviral effects,and treatment for ulcerative colitis.However,there is currently no research available regarding its effectiveness and mechanisms of action on melanoma.Methods:In this investigation,we used methyl thiazolyl tetrazolium assay to detect cell viability,transwell assay to detect cell migration and invasion ability,and Western blot assay to detect relevant signaling pathways.Results:The present study reveals that 2 mg/mL AAL effectively suppresses the metastasis of B16 cells,while simultaneously triggering the expression of key apoptosis-related proteins,including Bcl-2,Bax,and cleaved caspased 3.Subsequent investigations demonstrate that AAL exerts this inhibitory effect via the PI3K/AKT signal transduction pathway,as evidenced by the observed deficits in Ras,AKT,p-AKT,and PI3K expression levels.Conclusion:These findings indicated that AAL could be a valuable therapeutic option for reducing the metastatic potential of B16 melanoma cells. 展开更多

Inflammatory myofibroblastic tumor(IMT) in the liver is an uncommon lesion of uncertain pathogenesis.In most cases,symptomatological imaging and clinical studies suggest malignancy.We report a case of liver IMT with imaging findings from positron emission tomography/computed tomography(PET/CT),contrastenhanced computed tomography(CECT) and contrastenhanced ultrasonography(CEUS).This report was the first to depict a PET/CT scan of a liver IMT that revealed an inhomogeneous,intense(fluorine 18)-fluoro-2-deoxy-D-glucose uptake.The CECT and CEUS images showed a hepatic artery supplying blood to the mass and necrosis.The characteristic histopathological features and the presence of spindle cells expressing smooth muscle actin,collagen fibers and lymphocytes allowed for the diagnosis of liver IMT.Recognizing such findings will help to achieve a correct diagnosis and may prevent inappropriate treatment. 展开更多

The accessory olfactory bulb(AOB), located at the posterior dorsal aspect of the main olfactory bulb(MOB), is the first brain relay of the accessory olfactory system(AOS), which can parallelly detect and process volat... The accessory olfactory bulb(AOB), located at the posterior dorsal aspect of the main olfactory bulb(MOB), is the first brain relay of the accessory olfactory system(AOS), which can parallelly detect and process volatile and nonvolatile social chemosignals and mediate different sexual and social behaviors with the main olfactory system(MOS). However, due to its anatomical location and absence of specific markers, there is a lack of research on the internal and external neural circuits of the AOB. This issue was addressed by singlecolor labeling and fluorescent double labeling using retrograde rAAVs injected into the bed nucleus of the stria terminalis(BST), anterior cortical amygdalar area(ACo), medial amygdaloid nucleus(MeA), and posteromedial cortical amygdaloid area(PMCo) in mice. We demonstrated the effectiveness of this AOB projection neuron labeling method and showed that the mitral cells of the AOB exhibited efferent projection dispersion characteristics similar to those of the MOB. Moreover, there were significant differences in the number of neurons projected to different brain regions, which indicated that each mitral cell in the AOB could project to a different number of neurons in different cortices. These results provide a circuitry basis to help understand the mechanism by which pheromone information is encoded and decoded in the AOS. 展开更多

This paper presents a new type of automotive braking actuator for a kind of brake-by-wire system called decentralized electro-hydraulic braking system( DEHB) to replace the traditional automobile braking system. The actuator of this system is driven by an electrical motor instead of the conventional vacuum booster to make the brake pressure be linearity controlled quickly. Therefore,the system has the advantages of quick response speed,good control performance and simple structure. Firstly,an overview of the actuator and the whole DEHB system is shown. Secondly,the possibility of this new kind of actuator working for the system is ensured based on some braking theories. Thirdly,the appropriate dynamic simulations are done to get some results to show the relations of different parameters and the effect of braking. Eventually,the proper parameters are determined to build a test bench which shows that DEHB system can achieve the maximum pressure of 13 MPa within 100 ms after parametric optimization,and meanwhile,the actuator is able to reduce pressure quickly after maintaining high pressure. All of the bench test results can meet with the design requirements and real demand of vehicle and this actuator may improve vehicle braking effect in the future. Besides,this actuator can be widely applied to the regenerative braking system because of its linear braking performance. 展开更多

Methods:Blood serum HBsAg biomarker was examined by immune colloidal gold dipstick for 6582 Hui ethnicities, descriptive analysis applied to explore the distribution of prevalence of HBsAg.Results: Total of 372 subjects were positive of HBsAg with a prevalence of 4.68%;There were no significant differences between male and female for prevalence of HBsAg, however, an significant differences was detected among age groups,and the differences age group in older women. ALT and AST level higher in HBsAg positive group than those HBsAg negative subjects.Conclusions: The Hui elderly had a relatively lower prevalence of HBsAg than general population, further study needed to explore possible factors contribute to the low prevalence. 展开更多

This paper studies estimation and serial correlation test of a semiparametric varying-coefficient partially linear EV model of the form Y = X^Tβ +Z^Tα(T) +ε,ξ = X + η with the identifying condition E[(ε,η^T)^T] =0, Cov[(ε,η^T)^T] = σ^2Ip+1. The estimators of interested regression parameters /3 , and the model error variance σ2, as well as the nonparametric components α(T), are constructed. Under some regular conditions, we show that the estimators of the unknown vector β and the unknown parameter σ2 are strongly consistent and asymptotically normal and that the estimator of α(T) achieves the optimal strong convergence rate of the usual nonparametric regression. Based on these estimators and asymptotic properties, we propose the VN,p test statistic and empirical log-likelihood ratio statistic for testing serial correlation in the model. The proposed statistics are shown to have asymptotic normal or chi-square distributions under the null hypothesis of no serial correlation. Some simulation studies are conducted to illustrate the finite sample performance of the proposed tests. 展开更多

Community structure is a common topological property of complex networks, which attracted much attention from various fields. Optimizing quality functions for community structures is a kind of popular strategy for community detection, such as Modularity optimization. Here, we introduce a general definition of Modularity, by which several classical(multi-resolution) Modularity can be derived, and then propose a kind of adaptive(multi-resolution) Modularity that can combine the advantages of different Modularity. By applying the Modularity to various synthetic and real-world networks, we study the behaviors of the methods, showing the validity and advantages of the multi-resolution Modularity in community detection. The adaptive Modularity, as a kind of multi-resolution method, can naturally solve the first-type limit of Modularity and detect communities at different scales;it can quicken the disconnecting of communities and delay the breakup of communities in heterogeneous networks;and thus it is expected to generate the stable community structures in networks more effectively and have stronger tolerance against the secondtype limit of Modularity. 展开更多
