Evaluation of Uncertainty for Determination of Stevioside Content in Fermented Milk by HPLC

[Objectives]The paper was to establish an evaluation method for the uncertainty of stevioside(including stevioside,rebaudioside A,rebaudioside B,rebaudioside C,rebaudioside F,Dulcoside A,rubusoside and steviolbioside)content determination in fermented milk based on HPLC.[Methods]The mathematical model of stevioside content and the propagation rate of uncertainty were established,and the sources of uncertainty were analyzed.[Results]The uncertainty mainly came from four main aspects,including standard uncertainty u(C)introduced by solution concentration C,standard uncertainty u(V)introduced by sample volume V,standard uncertainty u(m)introduced by sample mass m weighing and standard uncertainty u(f_(rep))introduced by measurement repeatability of stevioside content after sample dissolution and constant volume.The uncertainty estimation table and fishbone chart of stevioside content X determination were established.The relative synthetic standard uncertainty of stevioside content was obtained,and the standard uncertainty was extended to form the measurement result of stevioside content and its uncertainty report.[Conclusions]The evaluation results can be directly applied to the daily practical detection work.

Crack Width Comparison between ACI 318, Eurocode 2 and GB 50010 for Flexural RC Members

Comparative research on different countries’ structural design codes holds great importance and can gain valuable insights: Awareness of Design Levels, Identifying Code Deficiencies and Optimizing Designs. The crack width of concrete structure is an important design aspect of the civil design. The four highly recognized and widely used crack width theories are systematically summarized. Based on the mentioned theories and project practices, American code ACI system, Eurocode 2 1992-1 and Chinese code GB 50010 have different crack width control requirement and calculation methods. The crack width control method based on ACI system code has evolved from the Z-factor method to the steel bar spacing control method which is simple and easy to be adopted for engineering. Meanwhile, the ACI 224.1R also gives a direct crack width calculation method consistent with the steel bar spacing control method. The Eurocode 2 and GB 50010 based on the bond-slip & no-slip theory consider much more affecting factors than ACI for predicting crack width. Taking the crack width calculation of Tunnel 5 intake as an example, the crack widths of the structure are calculated according to ACI system code, Eurocode 2 and GB 50010 respectively, the results show that the crack width results in various codes are not much different. The EN 1992-1 and GB 50010 results are almost the same which are less than the ACI 224.1Rresults.

Fluorescence-based PCR Detection of Goat Milk Powder Mixed with Cow Milk

[Objectives]To amplify and identify the specific gene fragments of cattle and sheep by real-time fluorescence-based PCR,and quickly and accurately identify the adulteration problem of goat milk powder according to the differences of mitochondrial genes among different species.[Methods]The specificity,limit of detection and repeatability test were carried out by isolating somatic cells,extracting template DNA and determining the reaction conditions of real-time fluorescence-based PCR.[Results]The real-time fluorescence-based PCR method could be used to identify the bovine and ovine-derived materials of milk powder with high specificity,high sensitivity and good repeatability,and the limit of detection of milk components in goat milk powder could reach 0.01%.[Conclusions]The real-time fluorescence-based PCR method can be used to quickly and accurately detect the milk components in goat milk powder,and provide some technical support for the adulteration detection of dairy industry.

Application of Six Sigma DMAIC Method in Improving the Good Rate of Blank Value of Cytological Method for Dairy Products

In this paper,the six Aigma DMAIC method was used to systematically analyze and study the process affecting the good rate of blank value of cytological method for dairy products.Through data collection,analysis and verification in each link,the room for improvement in the experimental process was identified.C&E matrix and FMEA were used to analyze the factors affecting the good rate of blank value.Through the DOE experiment design method and risk analysis,the main influencing factors were optimized and improved.The optimized experimental conditions were:silica gel activation temperature(500±50)℃,activation time(23±1)h,cells adherent rate(95±1)%,and single-use of 1.5 mL conical flask.Under the above conditions,the good rate of blank value of cytological method was improved from 52.8%to 100%.The improvement of these conditions is of great significance to ensure the accuracy of results,save the testing cost and improve the testing efficiency.

Evaluation on Uncertainty of Detection Results of Aerobic Plate Count

[Objectives]To determine the aerobic plate count(APC)in the milk samples,evaluate the uncertainty of the test results,and to provide a scientific basis for the quality control of the testing process.[Methods]In compliance with the national food safety standard Food Microbiological Examination:Aerobic Plate Count(GB 4789.2-2016),the aerobic plate count in the milk samples was detected.The source of the uncertainty of the test result was analyzed and a mathematical model was established in accordance with Evaluation and Expression of Uncertainty in Measurement(JJF 1059.1-2012).Then,the introduced uncertainty components were evaluated to determine the uncertainty of the final combined aerobic plate count.[Results]The expanded uncertainty of the test result of the aerobic plate count in the milk samples was 0.0434,and the logarithmic value interval of the results was(3.924,4.010),and the antilogarithm was taken to get the aerobic plate count in the sample to be 8395-10233 CFU/mL.[Conclusions]This method can effectively evaluate the uncertainty of the aerobic plate count,and ensure the accurate and scientific laboratory test data.

Analysis and Evaluation of Uncertainty in the Determination of Organic Pollutants

[Objectives]To determine the content of organic pollutants by CALUX bioassay.[Methods]According to JJF 1059.1-2012 Technical Specification for Evaluation and Expression of Uncertainty in Measurement,the determination results of organic pollutants were analyzed,various sources of uncertainty that may be introduced in the detection process were analyzed,and the mathematical model of uncertainty was established.Type A and B evaluation methods were used to calculate the components of uncertainty and extended uncertainty.[Results]In the range of 95%confidence interval,the determination result of organic pollutants was 6.59 pg/g fat,and the extended uncertainty was 0.0764 pg/g fat,so the determination results of organic pollutants could be expressed as(6.59±0.0764)pg/g fat(k=2).[Conclusions]This study can provide a basis for more accurate determination of organic pollutants.

Establishment of Molecular Biological Method for Identification of Bacteria by 16S rDNA and gyrB Gene

[Objectives]The paper was to establish a molecular biological method for identification of bacterial strains.[Methods]The thalli of standard bacterial strains existing in the laboratory were collected and genomic DNA was extracted for amplification of 16S rDNA and gyrB gene.The 16S rDNA and gyrB gene sequences were obtained after sequencing.Sequences were aligned and analyzed via EzBioCloud and NCBI database,and phylogenetic trees were constructed to determine the species relationship of strains.Meantime,they were compared with known strains.[Results]This method could identify 5 standard strains accurately to the species level.The 16S rDNA and gyrB gene sequences were aligned and analyzed in EzBioCloud database and NCBI database.The strain with the max score was consistent with the known strain.And the query cover and ident were both above 99%.[Conclusions]The established molecular biological method for identification of bacterial strains by 16S rDNA and gyrB gene has good accuracy,which effectively solves the problem that the laboratory identification of bacteria relies on traditional methods and the accuracy can not be guaranteed,and further improves the identification ability of laboratory bacterial strains.

A Precise Calibration Method on Phase Center of Uplink Antenna Array Considering Its Actual Pointing

With regard to the inferior techniques and low accuracy of phase center calibration of an antenna array, this paper proposes a new calibration method considering the actual antenna pointing by introducing a precise engineering surveying technique to measure the real state of antennas. First, an industrial photogrammetric system is utilized to obtain the coordinates of points on antenna panels in different postures, and the actual pointing of the mechanical axis is obtained via least-squares fitting. Then, based on this, the coordinates of antenna rotation center are obtained by seeking the intersection of mechanical axes via using the matrix method. Finally, the mechanical axis in arbitrary postures is estimated based on the inverse-angle weighting interpolation method, and the reliable phase center is obtained by moving a fixed length from the projective center along the mechanical axis. An uplink antenna array including three ? 3 m antennas is taken as experimental object, and all photogrammetric coordinate systems are unified by the engineering control network, with each antenna phase center precisely calibrated via the proposed method. The results of electrical signal synthesis indicate that this method can effectively overcome the influence of gravity deformation and mechanical installation error, and enhance the synthetic signal magnitude of the uplink antenna array.

Mining Related Articles for Automatic Journal Cataloging

Purpose： This paper is an investigation of the effectiveness of the method of clustering biomedical journals through mining the content similarity of journal articles. Design/methodology/approach： 3,265 journals in Pub Med are analyzed based on article content similarity and Web usage, respectively. Comparisons of the two analysis approaches and a citation-based approach are given.Findings： Our results suggest that article content similarity is useful for clustering biomedical journals, and the content-similarity-based journal clustering method is more robust and less subject to human factors compared with the usage-based approach and the citation-based approach. Research limitations： Our paper currently focuses on clustering journals in the biomedical domain because there are a large volume of freely available resources such as Pub Med and Me SH in this field. Further investigation is needed to improve this approach to fit journals in other domains.Practical implications： Our results show that it is feasible to catalog biomedical journals by mining the article content similarity. This work is also significant in serving practical needs in research portfolio analysis.Originality/value： To the best of our knowledge, we are among the first to report on clustering journals in the biomedical field through mining the article content similarity. This method can be integrated with existing approaches to create a new paradigm for future studies of journal clustering.

Determination of Naringin Content in Rhizoma Drynariae by High Performance Liquid Chromatography

[Objectives]To accurately determine the naringin content in Rhizoma Drynariae.[Methods]The high performance liquid chromatography(HPLC)method was applied in the determination of the naringin content in Rhizoma Drynariae.The sample was sonicated at room temperature.The mobile phase was 0.1%phosphoric acid-acetonitrile(75∶25),detected by diode array detector at the wavelength of 284 nm,and quantified by external standard method.[Results]The linearity of naringin was good in the concentration range of 5-500μg/mL with a correlation coefficient of 0.9999.[Conclusions]This method has good linearity,easy operation,correctness and reproducibility as required,and is expected to provide a method for the determination of naringin content in Rhizoma Drynariae.

Simultaneous Determination of Content of VK_(1) and VK_(2) in Milk and Dairy Products by High Performance Liquid Chromatography

[Objectives]To make simultaneous determination of vitamin K_(1)(VK_(1))and vitamin K_(2)(VK_(2),mainly MK-4,MK-7 and MK-9)in milk and dairy products.[Methods]A high performance liquid chromatographic method was developed for the simultaneous determination of vitamin K_(1)(VK_(1))and vitamin K_(2)(VK_(2),mainly MK-4,MK-7 and MK-9)in milk and dairy products.After enzymatic digestion,the samples were extracted with hexane for VK_(1),MK-4,MK-7 and MK-9,subjected to gradient elution at excitation wavelength 243 nm and emission wavelength 430 nm,detected by high performance liquid chromatography with fluorescence detector and quantified by external standard method.[Results]The linearity of VK_(1),MK-4,MK-7 and MK-9 was good in the concentration range of 2.5-1000 ng/mL with the correlation coefficients greater than 0.999;The relative standard deviations(RSD)of VK_(1),MK-4,MK-7 and MK-9 in milk powder,liquid milk and yogurt were 1.32%-5.05%,1.10%-2.48% and 2.20%-3.47%,respectively;the recovery rates of VK_(1),MK-4,MK-7 and MK-9 at different levels in milk powder,liquid milk and yogurt were 81.1%-108%,81.8%-103%and 82.1%-99.2%,respectively.[Conclusions]The method is rapid,accurate,reproducible and capable of simultaneous determination of VK_(1),MK-4,MK-7 and MK-9.

[Objectives]To explore the inhibitory effect of lactic acid bacteria(LAB)on the growth of Salmonella.[Methods]The inhibitory effect of lactic acid bacteria on Salmonella was investigated by Oxford cup agar diffusion test from the aspects of lactic acid bacteria body,metabolites,pH,thermal stability and acid resistance.[Results]The growth of Salmonella was disturbed by the inhibition of lactic acid bacteria.The mechanism of inhibitory effect may be:simple low pH and lactic acid bacteria have no inhibitory effect on Salmonella;however,there are active substances in the metabolites of lactic acid bacteria,which have high thermal stability and a wide range of acid resistance,and are effective substances for inhibiting the growth of Salmonella.[Conclusions]This study is helpful for screening more natural preservative substances,reducing the contamination rate of Salmonella to food,and is expected to provide a theoretical basis for the preservation and preservation of food.

Taxonomy on crane flies in family Tipulidae and Limoniidae(Diptera: Tipuloidea) from Yaoluoping National Nature Reserve, Anhui, China

The males of Holorusia henana Yang, 1999 and Tipula（Vestiplex） jiangi Yang ＆ Yang, 1991 are redescribed and illustrated with new morphological characters. A key to all species is provided.

Intrapulse coherence degradation suppressing method of echo signal in coherent lidar

Aiming at coherence degradation during target detection,a suppressing method based on frequency-modulated continuous wave coherent lidar is proposed.Combined with a random iteration algorithm,a long-pulse echo signal with coherent degradation is matched with random phase noise of a certain frequency and achieves coherence restoration.Simulation and field experiment results show that this proposed method can recover the intrapulse coherence in long-pulse echo signals.In addition,for the real target echo signal at 4.2 and 19.8 km,the peak signal-to-noise ratio processed by this method is increased by 0.35 times and 4 times after pulse compression,respectively.

The Impact of Seasonal Variation on Clinical Pregnancy and Live Birth Rates in Assisted Reproductive Technology: A Retrospective Cohort Study in Hainan

Objective:To investigate the influence of season on live birth and clinical pregnancy rates,as well as assisted reproductive technology(ART)outcomes,in the Hainan region.Methods:Patients were categorized into four groups based on the dates of artificial insemination and transplantation:spring,summer,autumn,or winter.The main outcome measures were clinical pregnancy rates and live birth rates.Secondary outcomes included body mass index(BMI),oocyte number,two pronuclei(2PN)cleavage rate,total gonadotropin(Gn)dosage and days,age,2PN fertilization rate,sperm concentration,sperm PR rate,anti-Müllerian hormone(AMH),and endometrial thickness.Outpatient semen quality indicators included sperm PR rate,total sperm count,sperm concentration,and total sperm motility.Results:This retrospective cohort study analyzed 2,016 artificial insemination cycles and 1,783 ovarian retrieval cycles from January 2017 to October 2022,and assessed the semen quality of 6,651 outpatients from May 2017 to October 2022.In artificial insemination cycles,sperm PR rate and clinical pregnancy rate were highest in winter,with a statistically significant difference between groups(P<0.05).Clinical pregnancy rate was influenced by both age and sperm PR rate(P<0.05).In ovarian retrieval cycles,the winter group had significantly higher clinical pregnancy,2PN fertilization,and 2PN cleavage rates than the other groups.The autumn group had higher live birth rates,though not significantly different.Additionally,winter months showed higher total sperm concentration and total sperm number compared to other seasons.Conclusion:Seasonality affected clinical pregnancy and live birth rates in artificial insemination cycles but not in ovarian retrieval cycles in the Hainan region.These findings suggest that while there is no need to choose a specific season for ovarian retrieval cycles,artificial insemination in winter may be preferable for patients.

Three-dimensional coherent ladar based on FMCW and its flight demonstration

A scanning three-dimensional coherent laser radar(ladar) based on the frequency modulated continuous wave(FMCW) is proposed and demonstrated, which can obtain many three-dimensional high-quality images. The system utilizes an electro-optic modulator and an optical filter to output a linear FMCW with a bandwidth of 2 GHz. The flexible and variable rotating double prism is used for beam scanning. The images of flight demonstration are formed by attitude compensation correction. The experiment result validates the performance of our system for airborne three-dimensional scanning imaging.
