特异性蛋白1在胰腺癌中的生物学作用及对细胞因子网络的影响 被引量:1

目的探讨特异性蛋白1(Sp1)在胰腺癌中表达和生物学功能并初步分析Sp1调控的细胞因子网络。方法Western blotting检测不同胰腺癌细胞系(BxPC3、AsPC1、CFPAC1、PANC1、Capan1、HS-766T、SW1990和HPAF-II)的Sp1水平;慢病毒感染构建稳定干... 目的探讨特异性蛋白1(Sp1)在胰腺癌中表达和生物学功能并初步分析Sp1调控的细胞因子网络。方法Western blotting检测不同胰腺癌细胞系(BxPC3、AsPC1、CFPAC1、PANC1、Capan1、HS-766T、SW1990和HPAF-II)的Sp1水平;慢病毒感染构建稳定干扰和过表达Sp1的胰腺癌细胞系,IncuCyte生长曲线和平板克隆形成实验检测增殖活力,划痕实验和Boyden小室实验检测迁移和侵袭能力。悬液芯片进一步研究干扰Sp1前后的细胞因子变化并分析调控的信号通路和生物学功能。结果与正常胰腺上皮细胞HPDE相比,胰腺癌细胞系中Sp1呈不同程度的表达,其中BxPC3细胞的最高,而AsPC1细胞的最低。接下来成功构建稳定干扰和过表达Sp1的胰腺癌细胞系,体外实验证明Sp1促进胰腺癌细胞体外增殖、迁移和侵袭能力。悬液芯片结果:干扰Sp1后一组炎性因子、趋化因子和基质金属蛋白酶类发生变化,其中51个上调(>2倍)、26个下调(<0.5倍)。GO分析显示,差异表达的因子与细胞生长增殖、炎症反应、免疫反应、淋巴细胞迁移和趋化反应等密切相关。信号通路分析显示差异表达基因涉及细胞因子与其受体反应、趋化因子信号通路、TNF、TGF-β、JAK/STAT和uPAR等。成功构建Sp1调控差异细胞因子的网络模式图。结论胰腺癌高表达Sp1可通过分泌大量细胞因子来激活相应信号通路,进而调控微环境,共同促进胰腺癌侵袭和转移。下一步需要深入研究Sp1在细胞因子调控网络中的核心作用机制。 展开更多

目的·探索应用程序性死亡蛋白-1(programmed death-1,PD-1)抑制剂行免疫治疗的进展期胃癌患者的免疫相关不良反应(immune-related adverse events,irAEs)的特征及预测因素,并分析irAEs与患者预后的相关性。方法·选择2018年6月... 目的·探索应用程序性死亡蛋白-1(programmed death-1,PD-1)抑制剂行免疫治疗的进展期胃癌患者的免疫相关不良反应(immune-related adverse events,irAEs)的特征及预测因素,并分析irAEs与患者预后的相关性。方法·选择2018年6月—2021年10月于上海交通大学医学院附属仁济医院应用PD-1抑制剂治疗的进展期胃癌患者140例。根据患者有无irAEs发生,将其分为irAEs组和非irAEs组。收集并分析2组患者的临床特征、irAEs的表现及预后情况。采用多因素Logistic回归模型分析影响irAEs发生的相关因素,并建立irAEs的预测模型。运用受试者操作特征曲线(receiver operating characteristic curve,ROC curve,ROC曲线)对不同指标预测irAEs发生的能力进行评估。采用Kaplan-Meier生存曲线分析irAEs与预后的相关性。运用Cox比例风险模型分析影响患者预后的相关因素。结果·共计132例患者完成随访,其中有63例(47.7%)患者发生irAEs。比较2组患者的临床特征的结果显示,年龄≥65岁、Ki-67指数、白细胞计数、中性粒细胞计数、调节性T细胞(regulatory T cell,Treg)计数的组间差异具有统计学意义(均P<0.05)。多因素Logistic回归分析显示,Treg计数为影响irAEs发生的保护因素(P=0.030)。ROC曲线提示,Treg+Ki-67+年龄(≥65岁)联合指标可较好地预测irAEs的发生(AUC=0.753,95%CI 0.623~0.848,P=0.000)。Kaplan-Meier生存曲线的结果显示,irAEs组患者的无进展生存期(progression-free survival,PFS)较非irAEs组有所延长(P=0.001)。Cox比例风险回归分析提示,irAEs是患者PFS的独立影响因素(P=0.006)。结论·Treg计数是行PD-1抑制剂免疫治疗的进展期胃癌患者发生irAEs的独立影响因素,且irAEs的发生可延长患者的PFS;Treg+Ki-67+年龄(≥65岁)联合指标可对该不良反应的发生进行较好的预测。 展开更多

研究了蟹腿关节壳聚糖溶液的流变行为,以及该种壳聚糖湿法纺丝成形中喷丝头拉伸倍数、凝固浴组成、干燥温度和脱水介质对壳聚糖纤维结构性能的影响。结果表明,最佳成形条件:喷丝头拉伸倍数为1.05,凝固浴为NaOH质量分数为10%的溶液和乙... 研究了蟹腿关节壳聚糖溶液的流变行为,以及该种壳聚糖湿法纺丝成形中喷丝头拉伸倍数、凝固浴组成、干燥温度和脱水介质对壳聚糖纤维结构性能的影响。结果表明,最佳成形条件:喷丝头拉伸倍数为1.05,凝固浴为NaOH质量分数为10%的溶液和乙醇的复配液(体积比为6∶4),干燥温度为70℃。在此条件下制备得到的壳聚糖初生纤维力学性能较好,进一步分别以甲醇和乙醇溶液作为脱水剂对初生纤维进行浸泡脱水处理。结果表明:经醇脱水处理后,壳聚糖纤维的直径和平衡溶胀率减小、强度和结晶度增大,并且纤维表面更加致密,丝束之间易于分离;甲醇溶液脱水处理效果更好,可使壳聚糖纤维的断裂强度提高24.1%。 展开更多

目的:报道1例罕见的胃癌合并肾平滑肌肉瘤病例,探讨其临床特点及诊治方法,以提高对独立多部位原发恶性肿瘤的认识。方法:通过对1例胃癌合并肾平滑肌肉瘤的同时性双原发肿瘤患者的诊治过程分析,结合国内外相关文献报道,总结该类患者的临... 目的:报道1例罕见的胃癌合并肾平滑肌肉瘤病例,探讨其临床特点及诊治方法,以提高对独立多部位原发恶性肿瘤的认识。方法:通过对1例胃癌合并肾平滑肌肉瘤的同时性双原发肿瘤患者的诊治过程分析,结合国内外相关文献报道,总结该类患者的临床诊治经验。结果:本例胃癌合并肾平滑肌肉瘤患者在体检时发现右肾占位,外科手术切除后病理诊断为右肾平滑肌肉瘤。术后胃镜活检确诊为胃窦腺癌。化疗3个周期后行胃肿瘤手术切除。术后发现颈部淋巴结和肺转移,穿刺病理诊断为平滑肌肉瘤转移。经化疗、放疗和靶向治疗后,患者病情稳定,生存期已超过3年。结论:同时性双原发胃癌和肾平滑肌肉瘤极为罕见,诊断需排除原发肿瘤转移可能。本例患者在肾平滑肌肉瘤术后随访期间确诊原发胃窦腺癌。左侧颈部淋巴结为胃癌常见转移部位,而该例患者病理确诊为肾平滑肌肉瘤转移。虽处于肿瘤晚期,经多学科协作综合治疗,在保证患者较好的生活质量的同时,延长了生存期。 展开更多

Sensory information arising from somatic and visceral area is thought to be respectively transmitted to the brain by two distinct sensory neuronal systems, the somatic and visceral sensory pathway. A novel and unique spinal sensory system, the spinosolitary tract-dorsal column postsynaptic neuronal system (SST-DCPS), was physiologically and anatomically identified. The spinal neurons project to both visceral (the solitary tract nucleus) and somatic sensory (the dorsal column nuclei) nuclei via their branched axons and receive both visceral and somatic sensory information ascending from periphery through dichotomized primary afferents and descending from their targets via their branched axons. The brain might thus be aware of both visceral and somatic sensation via a single SST-DCPS neuronal channel. The finding of SST-DCPS system, as an example, might be considered as an intersection or fuzzy set of the SST and DCPS system and the concept of dichotomy in classification of neurons and neuronal pathways 展开更多

The direct conversion of methanol into methane is the main process in anaerobic treatment of methanol containing wastewater.However,acetic acid can also be produced from methanol theoretically,which may probably result in an abrupt pH drop and deteriorate the anaerobic process.Therefore,it is interesting to know what would really happen in an anaerobic reactor treating methanol wastewater.In this study,an up-flow anaerobic sludge bed(UASB)reactor treating methanol wastewater was operated.The chemical oxygen demand(COD),acetic acid and pH of the effluent were monitored at different loadings and influent alkalinity.The results showed that the anaerobic reactor could be operated steadily at as low as 119 mg/L of influent alkalinity and high organic loading rate with no obvious pH drops.Volatile fatty acids accumulation was not observed even at strong shock loadings.The microorganisms in the sludge at the end of the test became homogeneous in morphology,which were mainly spherical or spheroidal in shape. 展开更多

Experiments were performed on 13 Wister rats. Fastblus (FB) was injected into subcutaneous tissue in perineum area and nuclear yellow (NY) was injected into subserous lamina of the bladder wall, respectively. FB, NY and FB+NY were mostly found in the spinal ganglions of L6, S1 ans S2 segments. The numbers of FB,NY and FB+NY labeled cells were 146, 186 and 81, in a total of 463 labeled cells and their proportion was 40%, 51% and 9%, respectively. The result indicates that the spinal ganglion neurons dually innervate both the somatic and visceral tissues, and the convergence of somato_visceral sensory pathways might occur in the spinal ganglion cells. 展开更多
