多元介孔MOFs:通过调节表面亲水/疏水性实现酶固定化的多功能平台

作为高效的生物催化剂,酶在食品加工、医药制造、化学工业和能源开发等众多领域发挥着重要作用.然而,游离状态的酶由于存在稳定性差、易失活、难以回收利用及不适用于连续化生产等问题,限制了其广泛应用.为了解决上述难题,人们探索发展... 作为高效的生物催化剂,酶在食品加工、医药制造、化学工业和能源开发等众多领域发挥着重要作用.然而,游离状态的酶由于存在稳定性差、易失活、难以回收利用及不适用于连续化生产等问题,限制了其广泛应用.为了解决上述难题,人们探索发展了固定化酶技术.固定化酶不仅保留了原有的催化活性,而且在稳定性方面有了显著提升,从而极大地提高了酶的利用效率.随着固定化酶材料和酶工程领域的不断发展,固定化酶技术为酶的应用开辟了更广阔的前景.沸石-咪唑骨架材料-8(ZIF-8)作为一种金属-有机骨架(MOFs)材料,因其独特的结构特性而被广泛用作酶固定化载体.然而,ZIF-8的微孔结构和固有的疏水性,以及前体物2-甲基咪唑(2-MeIm)的质子化现象,常常影响固定化酶的活性.为克服这些限制,本文采用多元配体竞争和孔隙工程相结合的策略,设计并制备了一种具有可调节亲水/疏水表面的新型多元介孔MOFs(mMOFs),并用作酶固定化载体.在mMOFs的制备过程中,采用亲水性和配位能力更强的3-甲基-1H-1,2,4-三唑(3-MTZ)和5-甲基四唑(5-MTA)配体,以部分替代传统的2-MeIm配体.随后,这些多元配体(包括2-MeIm,3-MTZ和5-MTA)与七水硫酸锌结合,在甲醇溶液中通过自凝聚作用形成透明的胶体溶液,进而完全溶解在水中.在此过程中,七水硫酸锌发挥了软模板的作用,引导介孔的形成,从而成功制备出新型多元介孔mMOFs.接着,利用这些mMOFs作为酶固定化的载体,固定了辣根过氧化物酶(HRP)和葡萄糖氧化酶(GOx),并详细研究了其催化性能.结果表明,与传统的ZIF-8载体相比,以mMOFs为载体的固定化酶展现出了更高的酶活性和稳定性.催化性能提升主要归因于mMOFs的亲水性和介孔结构改善了酶和底物间的传质,同时2-MeIm的质子化效应减弱也为酶提供了优良的微环境,稳定了酶的构象.综上,本文采用多元配体竞争与孔隙工程相结合的策略,成功构建了一种具有可调节亲水/疏水表面的新型多元介孔mMOFs,并用于酶固定化,所得固定化酶表现出较好的催化性能.通过深入的研究,揭示了该固定化酶表现出良好催化性能的作用机制,为可控设计制备性能优越的、以MOFs为载体的固定化酶提供了新思路,并为进一步拓宽MOFs材料的应用范围提供了参考. 展开更多

析氢反应是电解水产制氢的关键反应之一.在碱性条件下,由于催化剂表面与反应过程中产生的氧物种、氢物种与催化剂的吸附未处于最佳状态,析氢反应动力学往往比较缓慢,比在酸性条件下慢2-3个数量级.目前,铂基纳米催化剂被认为是最优的析... 析氢反应是电解水产制氢的关键反应之一.在碱性条件下,由于催化剂表面与反应过程中产生的氧物种、氢物种与催化剂的吸附未处于最佳状态,析氢反应动力学往往比较缓慢,比在酸性条件下慢2-3个数量级.目前,铂基纳米催化剂被认为是最优的析氢催化剂,但因价格昂贵、稳定性较差,限制了其在电解水器件上的大规模应用.因此,设计一种价格较为低廉、活性高和稳定性好的碱性析氢催化剂尤为必要.钌作为铂族金属之一,其价格约为铂的三分之一,但其与氢的结合能却与铂类似.因此,钌基催化剂被认为是有望替代铂作为析氢催化剂.本文结合模板法、静电纺丝和碳化热解策略构筑了一种负载无定型钌纳米簇的原子级钴掺杂一维碳纳米笼催化剂,有效提升了碱性条件下析氢反应性能.该催化剂具有较高的比表面积,丰富的缺陷结构,原子级分散的金属Co掺杂以及无定型钌纳米簇结构,并在碱性条件下催化析氢反应时,表现出了低起始过电位,低Tafel斜率(62 mV dec^(-1))和高稳定性.X射线衍射(XRD)结果表明,该催化剂中钌和钴以无定型形式存在.选区电子衍射结果表明了无定型结构的存在,并且钌以纳米簇形式存在,其平均粒径为1.48 nm.X射线光电子能谱结果表明该催化剂含有较高的氮掺杂,拉曼光谱证明了其存在碳缺陷结构.进一步通过X射线近边吸收谱和扩展边精细结构谱研究了钌和钴的配位环境,结果发现,钌带轻微正电荷,并且存在Ru-Ru键;钴带正电荷,价态介于0与+2价之间,其主要与N配位.在1 mol/L氢氧化钾电解液中,该杂化催化剂的电催化活性明显高于没有钴掺杂的催化剂以及商业化铂/碳催化剂.在电流密度为10 mA cm^(-2)时,过电位仅为40 mV,在过电位为100 mV时,该催化剂的质量催化活性达到了3.77 mAμg_(Ru)^(-1),远超商业化铂/碳(2.01 mAμg_(Ru)^(-1))和钌/碳(1.66 mAμg_(Ru)^(-1)).该催化剂也表现出了优异的析氢催化稳定性,5000圈循环后,10 mA cm^(-2)下的过电位仅增加了2 mV,在恒电流测试中,10 h后电位仅下降了95 mV. 展开更多

Establishing agro-ecological compensation mechanism is the objective requirement for protecting agricultural ecological environment on the prerequisite of stabilizing and benefiting farmers,to realize protection in the process of development and promote development through protection.This study analyzed existing problems in the practice of establishing agro-ecological compensation mechanism in Jiangsu Province,including failure to form systematic compensation system due to limited technological reserve,failure to bring into play expected compensation effect due to scant attention,and failure to set up perfect input mechanism due to insufficient financing channel.In line with these problems,it came up with feasible policy recommendations,including strictly implementing top frame design for agro-ecological compensation and giving impetus to standardized and institutionalized work,building pilot demonstration area for agro-ecological compensation,setting up diversified agro-ecological compensation approaches,establishing long-term security mechanism for agro-ecological compensation,and strengthening propaganda and education of agro-ecological compensation.It is expected to provide reference for establishing agro-ecological compensation mechanism in Jiangsu Province,and also provide system guarantee for promoting ecological progress and agricultural modernization in Jiangsu Province. 展开更多

Expensive instruments and complicated data processing are often required to discriminate solvents with similar structures and properties.Colorimetric sensors with high selectivity,low cost,and good portability are highly desirable to simplify such detection tasks.Herein,we report the fabrication of a photonic crystal sensor based on the self-assembled resorcinol formaldehyde(RF)hollow spheres to realize colorimetric sensing of polar solvents,including homologs and isomers based on the saturated diffusion time.The diffusion of solvent molecules through the photonic crystal film exhibits a unique three-step diffusion profile accompanied by a dynamic color change,as determined by the physicochemical properties of the solvent molecules and their interactions with the polymer shells,making it possible to accurately identify the solvent type based on the dynamic reflection spectra or visual perception.With its superior selectivity and sensitivity,this single-component colorimetric sensor represents a straightforward tool for convenient solvent detection and identification. 展开更多

The specific surface area and total pore volume of the product re... A sol-gel procedure in a water/oil emulsion was introduced for the synthesis of porous silica spheres. Tetraethoxysilane was used as the silica source. The specific surface area and total pore volume of the product reached 772.3 m2/g and 0.663 cm3/g, respectively. The electrolyte washing process conferred a surface charge to the product, which displayed self-dispersal properties in water. The porous spheres have potential applications in the fields of drug delivery, controlled release capsules, indoor air pollutant scavengers, and hydrogen storage agents. The oil phase, which accounts for over 8O% of the chemical cost of the procedure, could largely be recycled by filtering, standing, and layering. The whole procedure is suitable for application as an industrial process. 展开更多
