The Wnt signaling pathway plays a critical role in bone homeostasis,and the related protein therapy strategies have been reported to have great potential in osseointegration;however,they face formidable challenges suc...The Wnt signaling pathway plays a critical role in bone homeostasis,and the related protein therapy strategies have been reported to have great potential in osseointegration;however,they face formidable challenges such as complex external environments and unavoidable protein denaturation.In this work,we report a novel approach combining the synthesis of metal–organic frameworks(MOFs)and protein encapsulation in a one-pot process based on zeolitic imidazolate framework-8(ZIF-8)and Wnt3a protein,with improved biomechanical behavior and enhanced protein biological response.This combination was designed to enhance the Wnt3a protein function through the improved chemical stability provided by the ZIF-8 crystals.Additionally,the zinc ions contained in the ZIF-8 crystals induced bone homeostasis,further favoring the osteogenesis.The results showed that the Wnt3a protein-loaded ZIF-8 crystals served as efficient drug delivery vehicles to promote osteogenesis,preventing protein denaturation.In particular,Wnt3a-loaded ZIF-8 nanoparticles(Wnt3a@ZIF-8 NPs)had higher efficacy on bone marrow mesenchymal stem cells(BMSCs)than ZIF-8 NPs or Wnt3a proteins,contributing to the osteogenesis through ZIF-8 crystals and intracellular Wnt3a proteins released from Wnt3a@ZIF-8 NPs.Furthermore,polymerase chain reaction(PCR)analysis showed that the osteogenic pathways were upregulated.Overall,the present one-pot process can open up new avenues to develop signaling protein-delivery systems for applications in protein therapy strategies.展开更多
Oral and maxillofacial diseases are a group of high-incidence disorders that affect people’s life quality to a great extent,while the wet and highly movable environment of the related regions brings challenges to tra...Oral and maxillofacial diseases are a group of high-incidence disorders that affect people’s life quality to a great extent,while the wet and highly movable environment of the related regions brings challenges to traditional therapies.Faced with the obstacles of insufficient adhesive strength and ensuing short drug retention time,conventional oral therapeutic agents often have difficulty in achieving their desired efficacy.Oral and maxillofacial wet-adhesive materials have the advantages of excellent wet environment retention,internal stability,plasticity,and clinical potential,thus have become a significant research direction in the field of oral related disorders healing.In the past decade,the development of oral adhesive materials with good wet adhesion has accelerated based on the chemical molecular interaction,physical interlocking,and biological adhesion mechanisms,including biomimetic-inspired materials,naturally derived polymer–based materials and adhesive electrospun fiber films.These fancy wet-adhesive materials can be used for oral mucosal drug delivery,oral vaccination,wound healing,and bone defects treatments.Despite their numerous novel applications,wet-adhesive materials in stomatology still face unresolved challenges from material and biological aspects.Here,advances in designs of oral and maxillofacial wetadhesive materials are reviewed in terms of design backgrounds,attachment mechanisms,and common classifications.Recent demonstrations of wet-adhesive materials for oral and maxillofacial region medical applications from drug delivery to multifunctional tissue treatments are presented.To conclude,current challenges and prospects on potential applications of oral and maxillofacial wet-adhesive materials are also briefly discussed.展开更多

The modulation of triplet-involved emissions, particularly room temperature phosphorescence(RTP),however, is scarcely reported. Herein, polymorphism is reported to tune the triplet-involved emissions of 2 CZBZL, a newly designed pure organic luminogen consisting of twisted benzil and two planar carbazole moieties. Other than the conventional modulation through changing molecular conformation and packing, vibration can also finely tune the triplet-involved emissions. Besides prompt fluorescence(PF),polymorph B with relatively extended conformation emits thermally activated delayed fluorescence(TADF), whereas the others(A, C–E) with similarly more twisted conformations generate predominant RTP or simultaneous DF and RTP. Herein, based on our previous strategy for the construction of efficient and multifunctional...Fabrication of efficient solid luminogens with tunable emission is both fundamentally significant and technically important. Herein, based on our previous strategy for the construction of efficient and multifunctional solid luminogens through the combination of diverse aggregation-induced emission (ALE) units with other functional moieties, a group of luminophores with electron donor-acceptor (D-A) structure and typical intramolecular charge transfer (ICT) characteristics, namely CZ-DCDPP, DPA-DCDPP and DBPA-DCDPP were synthesized and investigated. The presence of twisting and AlE-active 2,3- dicyano-S,6-diphenylpyrazine (DCDPP) moiety endows them highly emissive in the solid states, whereas the introduction of arylamines with varied electron-donating capacity and different conjugation render them with tunable solid emissions from green to red. While CZ-DCDPP and DPA-DCDPP solids exhibit distinct mechanochromism, both DPA-DCDPP and DBPA-DCDPP solids can generate efficient red emission. Owing to their high efficiency, remarkable thermal and morphological stabilities and moreover red emission, they are promising for diverse optoelectronic and biological applications.展开更多
