Effects of Polysaccharides from Dicliptera chinensis(L.)Nees.on TLR4/NF-κB Signaling Pathway in Cell Model of Non-alcoholic Fatty Liver

[Objectives]To observe the effects of polysaccharides from Dicliptera chinensis(L.)Nees.on the expression of TLR/NF-κB pathway related proteins in HepG2 cells induced by oleic acid,and to explore the possible mechanism of polysaccharides from D.chinensis(L.)Nees.in the treatment of non-alcoholic fatty liver disease(NAFLD).[Methods]HepG2 cells were induced with oleic acid to establish a non-alcoholic fatty liver cell model.After intervention with 0.25 and 0.5 mg/mL of D.chinensis(L.)Nees.polysaccharides,the ALT and AST activity and TG and TC contents were detected with kits,and the changes in the expression of CDK5,TLR4,p-NF-κB and NF-κB were analyzed using Western-blotting.[Results]In the HepG2 cells induced with oleic acid,the ALT and AST activity increased significantly,the TG and TC contents increased significantly,and the expression levels of CDK5,TLR4 and p-NF-κB proteins up-regulated significantly.In the HepG2 cells intervened with D.chinensis(L.)Nees.polysaccharides,the activity of ALT and AST,the contents of TG and TC,and the expression levels of CDK5,TLR4 and p-NF-κB proteins all reduced significantly.[Conclusions]Polysaccharides from D.chinensis(L.)Nees.may interfere with NAFLD by inhibiting the TLR4/NF-κB pathway. 展开更多

The production of face towels is growing at an annual rate of about 4%in China,reaching 1.13 million tons by 2021.Phthalates(PAEs)are widely used in textiles,and face towels,as an important household textile,may expose people to PAEs via the skin,further leading to health risks.We collected new face towels and analyzed the distribution characterization of PAEs in them.The changes of PAEs were explored in a face towel use experiment and a simulated laundry experiment.Based on the use of face towels by 24 volunteers,we calculated the estimated daily intake(EDI)and comprehensively assessed the hazard quotient(HQ),hazard index(HI),and dermal cancer risk(DCR)of PAEs exposure in the population.PAEs were present in new face towels at total concentrations of<MDL–2388 ng/g,with a median of 173.2 ng/g,which was a lower contamination level compared with other textiles.PAE contents in used face towels were significantly higher than in new face towels.The concentrations of PAEs in coral velvet were significantly higher than those in cotton.Water washing removed some PAEs,while detergent washing increased the PAE content on face towels.Gender,weight,use time,and material were the main factors affecting EDI.The HQ and HIwere less than 1,which proved PAEs had no significant non-carcinogenic health risks.Among the five target PAEs studied,DEHPwas the only carcinogenic PAE andmay cause potential health risks after long-term exposure.Therefore,we should pay more attention to DEHP. 展开更多

It has been proven that inflammation ... Nowadays great progress has been made in targeted therapy and comprehensive therapy for a few cancers. But the prognosis of the vast majority of cancer patients is still weakness. It has been proven that inflammation can affect the development and treatment of cancer. Some previous studies have suggested that inflammation-related interleukins (ILs) play a significant role in the development of some cancers.1,2 ILs act as a considerable role in immunity,3 and inflammation affects the development of cancer. Therefore, the expression profile of IL gene may also be one of the vital markers of early pathological changes, occurrence and progression of cancer.4,5 Currently, studies on the expression of IL27 in cancer are still extremely limited. Our work comprehensively and systematically studied the IL27 gene expression in the generic cancer when compared with the existing IL27 gene related research experiment. We screened out cancer species linked with poor prognosis, and explored the correlation between IL27 gene expression and immune cell infiltration and immune microenvironment in these cancers further, providing an increasing number of the direct basis for subsequent studies. Our findings showed that IL27 can be used to assess prognosis and immune infiltration in patients with colon adenocarcinoma (COAD), glioblastoma multiforme (GBM), head and neck squamous cell carcinoma (HNSC), skin cutaneous melanoma (SKCM) and testicular germ cell tumors (TGCT) as a biomarker. 展开更多

监测土地利用/覆盖及生态系统服务价值变化是评估生态工程效益最直接有效的方式。以京津风沙源治理工程区为研究区,基于土地利用/覆盖变化面积和植被覆盖度等指标,从土地利用/覆盖类型转换和土地覆盖渐变两个角度揭示区域土地利用/覆盖... 监测土地利用/覆盖及生态系统服务价值变化是评估生态工程效益最直接有效的方式。以京津风沙源治理工程区为研究区,基于土地利用/覆盖变化面积和植被覆盖度等指标,从土地利用/覆盖类型转换和土地覆盖渐变两个角度揭示区域土地利用/覆盖变化过程;并运用改进的当量因子法,评估同期生态系统服务价值的变化。结果表明:区域草地面积超过总面积的一半,总体上呈现草地和耕地集中分布,林地、沙丘零散镶嵌的格局。1990—2018年,区域沙地、草地面积减少,耕地、林地面积增加;同时,沙地上表现出植被覆盖增加的渐变特征,增速为每10年约增长4.22%。同期区域生态系统服务价值呈现出先减少后增加的趋势,生态工程实施后,2010—2018年生态系统服务价值增加明显。1990—2018年,生态系统服务价值总体上增加了3655.21亿元,其中,由土地利用/覆盖类型变化导致的价值增加量为120.53亿元,而由土地覆盖渐变导致的增加量为5355.04亿元。土地覆盖渐变对生态系统服务价值的影响不容忽视,我们建议在生态建设过程中,不仅要重视退耕还林、未利用地造林种草等土地利用/覆盖方式的改变,更要关注草地和林地的修复和恢复,注重生态工程成果的维持和质量的提升。 展开更多
