NDFIP1 has been previously reported as a tumor suppressor in multiple solid tumors,but the function of NDFIP1 in NSCLC and the underlying mechanism are still unknown.Besides,the WW domain containing proteins can be recognized by NDFIP1,resulted in the loading of the target proteins into exosomes.However,whether WW domain-containing transcription regulator 1(WWTR1,also known as TAZ)can be packaged into exosomes by NDFIP1 and if so,whether the release of this oncogenic protein via exosomes has an effect on tumor development has not been investigated to any extent.Here,we first found that NDFIP1 was low expressed in NSCLC samples and cell lines,which is associated with shorter OS.Then,we confirmed the interaction between TAZ and NDFIP1,and the existence of TAZ in exosomes,which requires NDFIP1.Critically,knockout of NDFIP1 led to TAZ accumulation with no change in its mRNA level and degradation rate.And the cellular TAZ level could be altered by exosome secretion.Furthermore,NDFIP1 inhibited proliferation in vitro and in vivo,and silencing TAZ eliminated the increase of proliferation caused by NDFIP1 knockout.Moreover,TAZ was negatively correlated with NDFIP1 in subcutaneous xenograft model and clinical samples,and the serum exosomal TAZ level was lower in NSCLC patients.In summary,our data uncover a new tumor suppressor,NDFIP1 in NSCLC,and a new exosome-related regulatory mechanism of TAZ. 展开更多

To reduce the impact of fire, the authors propose a new method for predicting and rating fire in video through deep-learning models in this work such that rescue team can save lives of people. The proposed method explores a hybrid deep convolutional neural network, which involves motion detection and maximally stable extremal region for detecting and rating fire in video. Further, the authors propose to use a channel-wise attention mechanism of the deep neural network for detecting rating of fire level. Experimental results on a large dataset show the proposed method outperforms the existing methods for detecting and rating fire in video. 展开更多

Oscillation of Surface PM2.5 Concentration Resulting from an Alternation of Easterly and Southerly Winds in Beijing: Mechanisms and Implications 被引量:3

We used simultaneous measurements of surface PM_(2.5) concentration and vertical profiles of aerosol concentration,temperature, and humidity, together with regional air quality model simulations, to study an episode of aerosol pollution in Beijing from 15 to 19 November 2016. The potential effects of easterly and southerly winds on the surface concentrations and vertical profiles of the PM_(2.5) pollution were investigated. Favorable easterly winds produced strong upward motion and were able to transport the PM_(2.5) pollution at the surface to the upper levels of the atmosphere. The amount of surface PM_(2.5) pollution transported by the easterly winds was determined by the strength and height of the upward motion produced by the easterly winds and the initial height of the upward wind. A greater amount of PM_(2.5) pollution was transported to upper levels of the atmosphere by upward winds with a lower initial height. The pollutants were diluted by easterly winds from clean ocean air masses. The inversion layer was destroyed by the easterly winds and the surface pollutants and warm air masses were then lifted to the upper levels of the atmosphere, where they re-established a multi-layer inversion. This region of inversion was strengthened by the southerly winds, increasing the severity of pollution. A vortex was produced by southerly winds that led to the convergence of air along the Taihang Mountains. Pollutants were transported from southern–central Hebei Province to Beijing in the boundary layer. Warm advection associated with the southerly winds intensified the inversion produced by the easterly winds and a more stable boundary layer was formed. The layer with high PM_(2.5) concentration became dee-per with persistent southerly winds of a certain depth. The polluted air masses then rose over the northern Taihang Mountains to the northern mountainous regions of Hebei Province. 展开更多

Spinal cord injury(SCI)is an overwhelming and incurable disabling event accompanied by complicated inflammation-related pathological processes,such as excessive reactive oxygen species(ROS)produced by the infiltrated inflammatory immune cells and released to the extracellular microenvironment,leading to the widespread apoptosis of the neuron cells,glial and oligodendroctyes.In this study,a thioketal-containing and ROS-scavenging hydrogel was prepared for encapsulation of the bone marrow derived mesenchymal stem cells(BMSCs),which promoted the neurogenesis and axon regeneration by scavenging the overproduced ROS and re-building a regenerative microenvironment.The hydrogel could effectively encapsulate BMSCs,and played a remarkable neuroprotective role in vivo by reducing the production of endogenous ROS,attenuating ROS-mediated oxidative damage and downregulating the inflammatory cytokines such as interleukin-1 beta(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-alpha(TNF-α),resulting in a reduced cell apoptosis in the spinal cord tissue.The BMSCs-encapsulated ROS-scavenging hydrogel also reduced the scar formation,and improved the neurogenesis of the spinal cord tissue,and thus distinctly enhanced the motor functional recovery of SCI rats.Our work provides a combinational strategy against ROS-mediated oxidative stress,with potential applications not only in SCI,but also in other central nervous system diseases with similar pathological conditions. 展开更多

This study analyzes the impact of circulation types(CTs)on ozone(O_(3))pollution in Beijing.The easterly high-pressure(SWW)circulation occurred most frequently(30%;276 day),followed by northwesterly high-pressure(AN)circulation(24.3%;224 day).The SWW type had the highest O_(3) anomaly of+17.28μg/m^(3),which was caused by excellent photochemical reactions,poor diffusion ability and regional transport.Due to the higher humidity and precipitation in the low-pressure type(C),the O_(3) increase(+8.02μg/m^(3))was less than that in the SWW type.Good diffusion/wet deposition and weak formation ability contributed to O_(3) decrease in AN(-12.54μg/m^(3))and northerly high-pressure(ESN)CTs(-12.26μg/m^(3)).The intra-area transport of O_(3) was significant in polluted circulations(SWW-and C-CTs).In addition,higher temperature,radiation and less rainfall also contributed to higher O_(3) in northern Beijing under the SWW type.For the clean CTs(AN and ESN CTs),precursor amount and intra-area transport played a dominant role in O_(3) distribution.Under the northeasterly low-pressure CT,better formation conditions and higher precursor amount combined with the intra-area southerly transport to cause higher O_(3) values in the south than in the north.The higher O_(3) in the northwestern area under the northeasterly high-pressure type was influenced by weaker titration loss and high O_(3) concentration in previous day.Annual variation in the CTs contributed up to 86.1%of the annual variation in O_(3).About 78%-83%of the diurnal variation in O_(3) resulted from local meteorological factors. 展开更多

Sunshine duration(SD) is adopted widely to study global dimming/brightening. However, long-term simultaneous measurements of SD and closely related impact factors require further analysis to elucidate how and why SD has varied during the past decades. In this study, a long-term(1958–2021) SD data series obtained from the Shangdianzi Global Atmosphere Watch(GAW) station in China was analyzed to detect linear trends, climatic jumps, and climatic periods in SD using linear fitting, the Mann–Kendall trend test, and the continuous wavelet transform method. Annual SD exhibited steady dimming(-67.3 h decade-1) before 2010, followed by a period of brightening(189.9 h decade-1)during 2011–2020. An abrupt jump in annual SD occurred in 1995, and the annual SD anomaly exhibited significant oscillation with ~3-yr periodicity during 1960–1978. Partial least squares analysis revealed that annual SD anomaly was associated with variations in relative humidity, gale days, cloud cover, and black carbon(BC). Further analysis of the clear-sky daily sunshine percentage(DSP) and simultaneous measurements of aerosol properties, including aerosol optical depth, aerosol extinction coefficient, single scattering albedo(SSA), BC, and total suspended particulates, suggested that variation in DSP was affected primarily by aerosol scattering and absorption. Furthermore, the hourly clear-sky SD at high aerosol loading was approximately 60% and 56% of that at middle and low aerosol loadings, respectively. The pattern of diurnal variation in clear-sky hourly SD, as well as the actual values, can be affected by the fine particulate concentration, aerosol extinction coefficient, and SSA. 展开更多

Both of the surface topographical features and distribution of biochemical cues can influence the cell-substrate interactions and thereby tissue regeneration in vivo.However,they have not been combined simultaneously onto a biodegradable scaffold to demonstrate the synergistic role so far.In this study,a proof-of-concept study is performed to prepare micropatterns and peptide gradient on the inner wall of a poly(D,L-lactide-co-caprolactone)(PLCL)guidance conduit and its advantages in regeneration of peripheral nerve in vivo.After linear ridges/grooves of 20/40μm in width are created on the PLCL film,its surface is aminolyzed in a kinetically controlled manner to obtain the continuous gradient of amino groups,which are then transferred to CQAASIKVAV peptide density gradient via covalent coupling of glutaraldehyde.The Schwann cells are better aligned along with the stripes,and show a faster migration rate toward the region of higher peptide density.Implantation of the nerve guidance conduit made of the PLCL film having both the micropatterns and peptide gradient can significantly accelerate the regeneration of sciatic nerve in terms of rate,function recovery and microstructures,and reduction of fibrosis in muscle tissues.Moreover,this nerve conduit can also benefit the M2 polarization of macrophages and promote vascularization in vivo. 展开更多
